2Pharmacogenetic Correlates of Antipsychotic-Induced Weight Gain in the Chinese Population显示文摘Antipsychotic-induced weight gain(AIWG) is a common adverse effect of this treatment, particularly with second-generation antipsychotics, and it is a major health problem around the world. We aimed to review the progress of pharmacogenetic studies on AIWG in the Chinese population to compare the results for Chinese with other ethnic populations, identify the limitations and problems of current studies, and provide future research directions in China. Both English and Chinese electronic databases were searched to identify eligible studies. We determined that [ 25 single-nucleotide polymorphisms in19 genes have been investigated in association with AIWG in Chinese patients over the past few decades. HTR2 C rs3813929 is the most frequently studied single-nucleotide polymorphism, and it seems to be the most strongly associated with AIWG in the Chinese population. However, many genes that have been reported to be associated with AIWG in other ethnic populations have not beenincluded in Chinese studies. 3Proteome analysis of hepatic non-parenchymal cells of immune liver fibrosis rats显示文摘Elucidation of the mechanisms of liver fibrogenesis is important to treat liver fibrosis.In this study,we established rat models of liver fibrosis with stages from 0–1,2,and 3–4 to 4 at 2,4,6,and 8 weeks,respectively,by injection of pig serum.Liver fibrogenesis was detected by Masson’s trichrome staining.Rat non-parenchymal cells(NPCs)were enriched 4-fold by Percoll density gradient centrifugation.Protein extracts from NPCs were prepared at 4 and 8 weeks,separated by two-dimensional electrophoresis,and then stained with Coomassie Blue G-250.At 4 weeks,we identified 18 non-redundant differentially expressed proteins of which protein disulfide-isomerase associated protein 3(PDIA3)and NDUV showed consistent expression at protein and mRNA levels from 4 to 8 weeks.PDIA3 was found to be down-regulated by Western blotting in the rat model and immunohistochemically in human liver.Our results revealed important aspects of the pathogenesis/progression of liver fibrosis and demonstrated important changes in protein expression levels of NPCs at various stages of liver fibrosis.ZHAO QianQian FENG YanLing JIA XiaoFang YIN Lin ZHENG Ye OUYANG DongSheng ZHOU HongHao ZHANG LiJun 2014Science China(Life Sciences)2014,57,3:5

7Critical involvement of lysyl oxidase in seizure-induced neuronal damage through ERK-Alox5-dependent ferroptosis and its therapeutic implications显示文摘Recent insights collectively suggest the important roles of lysyl oxidase(LysOX)in the pathological processes of several acute and chronic neurological diseases,but the molecular regulatory mechanisms remain elusive.Herein,we explore the regulatory role of LysOX in the seizure-induced ferroptotic cell death of neurons.Mechanistically,LysOX promotes ferroptosis-associated lipid peroxidation in neurons via activating extracellular regulated protein kinase(ERK)-dependent 5-lipoxygenase(Alox5)signaling.In addition,overexpression of LysOX via adeno-associated viral vector(AAV)-based gene transfer enhances ferroptosis sensitivity and aggravates seizure-induced hippocampal damage.Our studies show that pharmacological inhibition of LysOX withβ-aminopropionitrile(BAPN)significantly blocks seizure-induced ferroptosis and thereby alleviates neuronal damage,while the BAPN-associated cardiotoxicity and neurotoxicity could further be reduced through encapsulation with bioresponsive amorphous calcium carbonate-based nanocarriers.These findings unveil a previously unrecognized LysOX-ERK-Alox5 pathway for ferroptosis regulation during seizure-induced neuronal damage.Suppressing this pathway may yield therapeutic implications for restoring seizure-induced neuronal injury.Xiaoyuan Mao Xuan Wang Mingzhu Jin Qin Li Jining Jia Menghuan Li Honghao Zhou Zhaoqian Liu Weilin Jin Yanli Zhao Zhong Luo 2022Acta Pharmaceutica Sinica B2022,12,9:1

9Downstream signaling of reactive oxygen species,protein kinase C epsilon translocation and delayed neuroprotection in sevoflurane preconditioned rats following cerebral ischemia/reperfusion显示文摘BACKGROUND:Brief exposure to the anesthetic sevoflurane results in delayed neuroprotection. However,few studies have addressed delayed neuroprotection after preconditioning with a single administration of sevoflurane. OBJECTIVE:To explore the relationship between a single preconditioning administration of sevoflurane and reactive oxygen species production and protein kinase C-epsilon(PKC-ε) translocation. DESIGN,TIME,AND SETTING:The randomized,controlled,animal experiment was conducted at the Central Laboratory,Xiangya Hospital,Central South University,China from November 2007 to April 2008. MATERIALS:A total of 120 healthy,male,Sprague Dawley rats were equally and randomly assigned into five groups:sham operation,ischemia/reperfusion,sevoflurane,2-mercaptopropionylglycine (2-MPG,a selective reactive oxygen species scavenger) + sevoflurane(MPG + sevoflurane),and MPG.Sevoflurane(Baxter,USA) and MPG(Sigma,USA) were used in this study. METHODS:Intervention consisted of three procedures.(1) MPG injection:a selective reactive oxygen species scavenger,MPG(20 mg/kg),was infused into the rat caudal vein in the MPG and MPG + sevoflurane groups.(2) Sevoflurane preconditioning:30 minutes following MPG injection, rats in the sevoflurane and MPG + sevoflurane groups breathed a mixed gas of 2.4%sevoflurane and 97.6%oxygen for 60 minutes.Rats in the sham operation,ischemia/reperfusion,and MPG groups breathed 100%pure oxygen for 60 minutes.(3) Ischemia/reperfusion:24 hours after sevoflurane or pure oxygen preconditioning,middle cerebral artery occlusion models were established in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane,and MPG groups. Following 2 hours ischemia/6 hours and 24 hours reperfusion,the carotid artery was separated,but the middle cerebral artery was not occluded,in the sham operation group. MAIN OUTCOME MEASURES:In the ischemic hemisphere,PKC-εtranslocation in the rat parietal cortex was measured by Western blot analysis.Infarct volume was calculated using the TTC assay.Neurological deficits were evaluated in rats using a scoring system of 8 points. RESULTS:After 6 hours reperfusion,the ratio of PKC-εin membrane/(cytosol + membrane) was significantly less in the sham operation group than in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane),and MPG groups(P<0.05).The ratio of PKC-εin membrane/(cytosol + membrane) was significantly greater in the sevoflurane group than in the sham operation,ischemia/reperfusion, MPG + sevoflurane,and MPG groups(P<0.05).No significant differences were observed in the ischemia/reperfusion,MPG + sevoflurane,and MPG groups(P>0.05).Following 24 hours reperfusion, the ratio of PKC-εin membrane/(cytosol + membrane) was significantly less in the sham operation group than in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane,and MPG groups(P<0.05). No significant differences were detected in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane, and MPG groups(P>0.05).Compared with the ischemia/reperfusion,MPG + sevoflurane,and MPG groups,infarct volume was significantly smaller,and neurological deficits were significantly improved, in the sevoflurane group(P<0.05).No significant differences in infarct volume and neurological deficits were observed among the ischemia/reperfusion,MPG + sevoflurane,and MPG groups(P>0.05).Infarcts or neurological deficits were not detected in the sham operation group. 18Drug-microbiota interactions: an emerging priority for precision medicine显示文摘Individual variability in drug response(IVDR)can be a major cause of adverse drug reactions(ADRs)and prolonged therapy,resulting in a substantial health and economic burden.Despite extensive research in pharmacogenomics regarding the impact of individual genetic background on pharmacokinetics(PK)and pharmacodynamics(PD),genetic diversity explains only a limited proportion of IVDR.The role of gut microbiota,also known as the second genome,and its metabolites in modulating therapeutic outcomes in human diseases have been highlighted by recent studies.Consequently,the burgeoning field of pharmacomicrobiomics aims to explore the correlation between microbiota variation and IVDR or ADRs.This review presents an up-to-date overview of the intricate interactions between gut microbiota and classical therapeutic agents for human systemic diseases,including cancer,cardiovascular diseases(CVDs),endocrine diseases,and others.We summarise how microbiota,directly and indirectly,modify the absorption,distribution,metabolism,and excretion(ADME)of drugs.Conversely,drugs can also modulate the composition and function of gut microbiota,leading to changes in microbial metabolism and immune response.We also discuss the practical challenges,strategies,and opportunities in this field,emphasizing the critical need to develop an innovative approach to multi-omics,integrate various data types,including human and microbiota genomic data,as well as translate lab data into clinical practice.To sum up,pharmacomicrobiomics represents a promising avenue to address IVDR and improve patient outcomes,and further research in this field is imperative to unlock its full potential for precision medicine.Qing Zhao Yao Chen Weihua Huang Honghao Zhou Wei Zhang 2023Signal Transduction and Targeted Therapy2023,8,11:0

20Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND
