2A journey to understand wheat Fusarium head blight resistance in the Chinese wheat landrace Wangshuibai显示文摘Fusarium head blight(FHB) or scab caused by Fusarium graminearum is a major threat to wheat production in China as well as in the world. To combat this disease, multiple efforts have been carried out internationally. In this article, we review our long-time effort in identifying the resistance genes and dissecting the resistance mechanisms by both forward and reverse genetics approaches in the last two decades. We present recent progress in resistance QTL identification, candidate functional gene discovery, marker-assisted improvement of FHB resistant varieties, and findings in investigating association of signal molecules, such as Ca^(++),SA, JA, and ET, with FHB response, with the assistance from rapidly growing genomics platforms. 3Efficient manipulations of circularly polarized terahertz waves with transmissive metasurfaces显示文摘The unrestricted control of circularly polarized(CP)terahertz(THz)waves is important in science and applications,but conventional THz devices suffer from issues of bulky size and low efficiency.Although Pancharatnam-Berry(PB)metasurfaces have shown strong capabilities to control CP waves,transmission-mode PB devices realized in the THz regime are less efficient,limiting their applications in practice.Here,based on Jones matrix analysis,we design a trilayer structure(thickness of ~λ/5)and experimentally demonstrate that the structure can serve as a highly efficient transmissive meta-atom(relative efficiency of ~90%)to build PB metadevices for manipulating CP THz waves.Two ultrathin THz metadevices are fabricated and experimentally characterized with a z-scan THz imaging system.The first device can realize a photonic spin Hall effect with an experimentally demonstrated relative efficiency of ~90%,whereas the second device can generate a high-quality background-free CP Bessel beam with measured longitudinal and transverse field patterns that exhibit the nondiffracting characteristics of a Bessel beam.All the experimental results are in excellent agreement with full-wave simulations.Our results pave the way to freely manipulate CP THz beams,laying a solid basis for future applications such as biomolecular control and THz signal transportation.Min Jia Zhuo Wang Heting Li Xinke Wang Weijie Luo Shulin Sun Yan Zhang Qiong He Lei Zhou 2019Light(Science & Applications)2019,8,1:9
4Aptamer-wrapped gold nanoparticles for the colorimetric detection of omethoate显示文摘Organophosphorous pesticide(OP) contamination has serious adverse effects on human health and the environment. Due to the toxicity of OPs and the threat presented by their accidental or intentional release in populated areas, the determination and monitoring of these OPs in food products and environment is of great importance. OPs are present in very small quantities and therefore, methods for their detection need to be highly sensitive and selective. Here, we aimed to develop a simple and selective aptamer-based colorimetric assay for the detection of omethoate, which is one of the commonly used OPs. The principle of the assay is that single-stranded DNA(ss DNA)-wrapped gold nanoparticles(Au NPs) are resistant to salt-induced aggregation. By employing an 'artificial antibody' organophosphorous pesticide-binding aptamer(OBA) as the recognition element, aptamer-wrapped Au NPs(Au-apta) show high selectivity towards omethoate, resulting in the disconnection of aptamers from Au NPs and the aggregation of Au NPs. As there is a significant color change from the interparticle plasmon coupling during the aggregation of Au NPs, the established assay showed good linearity between 0.1 and 10 μmol/L, with a low detection limit of 0.1 μmol/L. Other OPs such as profenofos, phorate, and isocarbophos would not interfere with the detection of omethoate despite having similar structures. 5Engineering an enthesis-like graft for rotator cuff repair:An approach to fabricate highly biomimetic scaffold capable of zone-specifically releasing stem cell differentiation inducers显示文摘Rotator cuff(RC)attaches to humerus across a triphasic yet continuous tissue zones(bone-fibrocartilage-tendon),termed“enthesis”.Regrettably,rapid and functional enthesis regeneration is challenging after RC tear.The existing grafts bioengineered for RC repair are insufficient,as they were engineered by a scaffold that did not mimic normal enthesis in morphology,composition,and tensile property,meanwhile cannot simultaneously stimulate the formation of bone-fibrocartilage-tendon tissues.Herein,an optimized decellularization approach based on a vacuum aspiration device(VAD)was developed to fabricate a book-shaped decellularized enthesis matrix(O-BDEM).Then,three recombinant growth factors(CBP-GFs)capable of binding collagen were synthesized by fusing a collagen-binding peptide(CBP)into the N-terminal of BMP-2,TGF-β3,or GDF-7,and zone-specifically tethered to the collagen of O-BDEM to fabricate a novel scaffold(CBP-GFs/O-BDEM)satisfying the above-mentioned requirements.After ensuring the low immunogenicity of CBP-GFs/O-BDEM by a novel single-cell mass cytometry in a mouse model,we interleaved urine-derived stem cell-sheets into this CBP-GFs/O-BDEM to bioengineer an enthesis-like graft.Its high-performance on regenerating enthesis was determined in a canine model.These findings indicate this CBP-GFs/O-BDEM may be an excellent scaffold for constructing enthesis-like graft to patch large/massive RC tears,and provide breakthroughs in fabricating graded interfacial tissue.Can Chen Qiang Shi Muzhi Li Yang Chen Tao Zhang Yan Xu Yunjie Liao Shulin Ding Zhanwen Wang Xing Li Chunfeng Zhao Lunquan Sun Jianzhong Hu Hongbin Lu 2022Bioactive Materials2022,7,10:4
6Effects of cerium substitution on phase components, microstructures and magnetic properties of Nd-Fe-Ti-B alloy显示文摘(Nd(1-x)Cex)(12)Fe(77)Ti5B6(x=0,0.2,0.3,0.4,0.6,0.8)alloys were prepared by melt-spinning and annealing techniques.The phase constitutions,microstructures and magnetic properties were investigated by powder X-ray diffraction(XRD),a differential scanning calorimeter(DSC),a vibrating sample magnetometer(VSM)and a transmission electron microscope(TEM).It is found that with the increase of Ce content,the coercivity of the(Nd(1-x)Cex)(12)Fe(77)Ti5B6 alloys reaches maximum first and then decreases.The maximum coercivity reaches 18.5 kOe obtained in the sample of 20%Ce substituted which is 34%higher than the Ce-free sample.TEM results reveal that the micro structure refinement effect is responsible for the coercivity improvement.This phenomenon implies that in some cases,Ce and Ti co-doping is more beneficial to improving of the coercivity than Ti single doping in Nd-Fe-B alloys.With further Ce addition,magnetic properties deteriorate due to the formation of CeFe2 and TbCu7-type phases.Haijun Peng Yang Luo Yakun Dou Xinyuan Bai Wenlong Yan Dunbo Yu Yuanfei Yang Shulin Diao 2019Journal of Rare Earths2019,37,8:4
7A new type of ERGIC–ERES membrane contact mediated by TMED9 and SEC12 is required for autophagosome biogenesis显示文摘Under stress,the endomembrane system undergoes reorganization to support autophagosome biogenesis,which is a central step in autophagy.How the endomembrane system remodels has been poorly understood.Here we identify a new type of membrane contact formed between the ER-Golgi intermediate compartment(ERGIC)and the ER-exit site(ERES)in the ER-Golgi system,which is essential for promoting autophagosome biogenesis induced by different stress stimuli.The ERGIC-ERES contact is established by the interaction between TMED9 and SEC12 which generates a short distance opposition(as close as 2-5 nm)between the two compartments.The tight membrane contact allows the ERES-located SEC12 to transactivate COPII assembly on the ERGIC.In addition,a portion of SEC12 also relocates to the ERGIC.Through both mechanisms,the ERGIC-ERES contact promotes formation of the ERGIC-derived COPII vesicle,a membrane precursor of the autophagosome.The ERGIC-ERES contact is physically and functionally different from the TFG-mediated ERGIC-ERES adjunction involved in secretory protein transport,and therefore defines a unique endomembrane structure generated upon stress conditions for autophagic membrane formation.Shulin Li Rui Yan Jialu Xu Shiqun Zhao Xinyu Ma Qiming Sun Min Zhang Ying Li Jun-Jie Gogo Liu Liangyi Chen Sai Li Ke Xu Liang Ge 2022Cell Research2022,32,2:3
8Palm oil protectsα-linolenic acid from rumen biohydrogenation and muscle oxidation in cashmere goat kids显示文摘Background:In ruminants,dietary C18:3n-3 can be lost through biohydrogenation in the rumen;and C18:3n-3 that by-passes the rumen still can be lost through oxidation in muscle,theoretically reducing the deposition of C18:3n-3,the substrate for synthesis of poly-unsaturated fatty acids(n-3 LCPUFA)in muscle.In vitro studies have shown that rumen hydrogenation of C18:3n-3 is reduced by supplementation with palm oil(rich in cis-9 C18:1).In addition,in hepatocytes,studies with neonatal rats have shown that cis-9 C18:1 inhibits the oxidation of C18:3n-3.It therefore seems likely that palm oil could reduce both rumen biohydrogenation of C18:3n-3 and muscle oxidation of C18:3n-3.The present experiment tested whether the addition of palm oil to a linseed oil supplement for goat kids would prevent the losses of C18:3n-3 and thus improve the FA composition in two muscles,Longissimus dorsi and Biceps femoris.To investigate the processes involved,we studied the rumen bacterial communities and measured the mRNA expression of genes related to lipid metabolism in Longissimus dorsi.Sixty 4-month-old castrated male Albas white cashmere kids were randomly allocated among three dietary treatments.All three diets contained the same ingredients in the same proportions,but differed in their fat additives:palm oil(PMO),linseed oil(LSO)or mixed oil(MIX;2 parts linseed oil plus 1 part palm oil on a weight basis).Results:Compared with the LSO diet,the MIX diet decreased the relative abuandance of Pseudobutyrivibrio,a bacterial species that is positively related to the proportional loss rate of dietary C18:3n-3 and that has been reported to generate the ATP required for biohydrogenation(reflecting a decrease in the abundance of rumen bacteria that hydrogenate C18:3n-3 in MIX kids).In muscle,the MIX diet increased concentrations of C18:3n-3,C20:5n-3,C22:6n-3,and n-3 LCPUFA,and thus decreased the n-6/n-3 ratio;decreased the mRNA expression of CPT1β(a gene associated with fatty acid oxidation)and increased the mRNA expression of FADS1 and FADS2(genes associated with n-3 LCPUFA synthesis),compared with the LSO diet.Interestingly,compared to Longissimus dorsi,Biceps femoris had greater concentrations of PUFA,greater ratios of unsaturated fatty acids/saturated fatty acids(U/S),and poly-unsaturated fatty acids/saturated fatty acids(P/S),but a lesser concentration of saturated fatty acids(SFA).Conclusions:In cashmere goat kids,a combination of linseed and palm oils in the diet increases the muscle concentration of n-3 LCPUFA,apparently by decreasing the relative abundance of rumen bacteria that are positively related to the proportional loss rate of dietary C18:3n-3,by inhibiting mRNA expression of genes related to C18:3n-3 oxidation in muscle,and by up-regulating mRNA expression of genes related to n-3 LCPUFA synthesis in muscle,especially in Longissimus dorsi.Xue Wang Graeme B.Martin Qi Wen Shulin Liu Yinhao Li Binlin Shi Xiaoyu Guo Yanli Zhao Yangdong Guo Sumei Yan 2021Journal of Animal Science and Biotechnology2021,12,2:3

10An improved U-Net for cell confluence estimation显示文摘Cell confluence is an important metric to determine the growth and the best harvest time of adherent cells. At present, the evaluation of cell confluence mainly relies on experienced labor, and thus it is not conducive to the automated cell culture. In this paper, we proposed an improved U-Net algorithm(called DU-Net) for the segmentation of adherent cells. First, the general convolution was replaced by the dilated convolution to expand the receptive fields for feature extraction. Then, the convolutional layers were combined with the batch normalization layers to reduce the dependence of the network on initialization. As a result, the segmentation accuracy and F1-score of the proposed DU-Net for adherent cells with low confluence(<50%) reached 96.94% and 93.87%, respectively, and for those with high confluence(≥50%), they reached 98.63% and 98.98%, respectively. 17Polystyrene nanoplastics aggravated ecotoxicological effects of polychlorinated biphenyls in on zebrafish(Danio rerio)embryos显示文摘In view of the accumulation of nanoplastics(NPs)in the food chain of environment and animals,and the good adsorption properties of nano-plastics to toxic substances,it is necessary to explore the influence of NPs in living organisms.In this study,single and joint toxicological effects of polystyrene nanoplastics(PS-NPs,size 80 nm)and polychlorinated biphenyls(PCBs),were explored in freshwater aquatic animal model zebrafish(Danio rerio).Our study found that exposure to single PS-NPs induced mild acute toxicity,albeit the combined exposure of PS-NPs and polychlorinated biphenyls aggravated the toxicity of PCBs in a dose-dependent manner.Results from gene expression profiling showed that NPs exposure could activate detoxification process,resulting in a slight up-regulation of antioxidant genes(sod1,gstp1),bone development genes(bmp2,bmp4)and cardiac gene(tbx20);while PCBs suppressed the detoxification through down-regulation of these genes,and the addition of NPs will exacerbate the impact of PCBs on gene suppression.Importantly,the results of in vivo purification experiments found that NPs showed prolonged retention in liver,intestine and gills of zebrafish and they might have crossed biological barrier and accumulate in lipid-rich tissues and excretion does not appear as the significant pathway for their elimination.In conclusion,the toxic effects of polychlorinated biphenyls on chorionic protected embryos were not significant as zebrafish chorion plays an important role in resisting the invasion of pollutants;PCBs can seriously damage the bone and heart development of zebrafish,while the presence of NPs significantly enhanced the toxicity of PCBs in zebrafish,which is an alarming concern for growing NPs levels and ecological safety in aquatic environment.Yizheng Li Shulin Liu Qiuping Wang Yanling Zhang Xikun Chen Lei Yan Muhammad Junaid Jun Wang 2022Geoscience Frontiers2022,13,3:1

19Physical Activity,Sedentary Behavior,and the Risk of Cardiovascular Disease in Type 2 Diabetes Mellitus Patients:The MIDiab Study显示文摘The aim of this study was to explore the associations of moderate-to-vigorous-intensity physical activity(MVPA)time and sedentary(SED)time with a history of cardiovascular disease(CVD)and multifactorial(i.e.,blood pressure(BP),body mass index(BMI),low-density lipoprotein cholesterol(LDL-C),and glycated hemoglobin A1c(HbA1c))control status among type 2 diabetes mellitus(T2DM)patients in China.A cross-sectional analysis of 9152 people with type 2 diabetes from the Multifactorial Intervention on Type 2 Diabetes(MIDiab)study was performed.Patients were grouped according to their self-reported MVPA time(low,<150 min·week−1;moderate,150 to<450 min·week−1;high,≥450 min·week−1)and SED time(low,<4 h·d–1;moderate,4 to<8 h·d–1;high,≥8 h·d–1).Participants who self-reported a history of CVD were identified as having a CVD risk.Odds ratios(ORs)and 95%confidence intervals(CIs)of CVD risk and multifactorial control status associated with MVPA time and SED time were estimated using mixed-effect logistic regression models,adjusting for China’s geographical region characteristics.The participants had a mean±standard deviation(SD)age of(60.87±8.44)years,44.5%were women,and 25.1%had CVD.After adjustment for potential confounding factors,an inverse association between high MVPA time and CVD risk that was independent of SED time was found,whereas this association was not observed in the moderate-MVPA group.A higher MVPA time was more likely to have a positive effect on the control of BMI.Compared with the reference group(i.e.,those with MVPA time≥450 min·week−1 and SED time<4 h·d–1),CVD risk was higher in the low-MVPA group:The OR associated with an SED time<4 h·d–1 was 1.270(95%CI,1.040–1.553)and that associated with an SED time≥8 h·d–1 was 1.499(95%CI,1.149–1.955).We found that a high MVPA time(i.e.,≥450 min·week−1)was associated with lower odds of CVD risk regardless of SED time among patients with T2DM.Yafei Wu Guijun Qin Guixia Wang Libin Liu Bing Chen Qingbo Guan Zhongshang Yuan Xu Hou Ling Gao Chao Xu Haiqing Zhang Xu Zhang Qiu Li Yongfeng Song Fei Jing Shizhan Ma Shanshan Shao Meng Zhao Qingling Guo Nanwei Tong Hongyan Zhao Xiaomin Xie Chao Liu Zhongyan Shan Zhifeng Cheng Xuefeng Yu Shulin Chen Tao Yang Yangang Wang Dongmei Li Zhaoli Yan Lixin Guo Qiuhe Ji Wenjuan Wang Jiajun Zhao 2023Engineering2023,,1:1
20Restructuring Tilth Layers Can Change the Microbial Community Structure and Affect the Occurrence of Verticillium Wilt in Cotton Field显示文摘Restructuring tilth layers(RTL)is a tillage method that exchanges the 0–20 and 20–40 cm soil layers that can be applied during cotton cultivation to increase cotton yield,eliminate weeds and alleviate severe disease,including Verticillium wilt.However,the mechanism by which RTL inhibits Verticillium wilt is unclear.Therefore,we investigated the distribution of microbial communities after rotary tillage(CK)and RTL treatments to identify the reasons for the reduction of Verticillium wilt in cotton fields subjected to RTL.Illumina high-throughput sequencing was used to sequence the bacterial and fungal genes.The disease incidence and severity of Verticillium wilt decreased by 28.57%and 42.64%,respectively,after RTL.Moreover,RTL significantly enhanced bacterial richness and evenness at 20–40 cm and-reduced the differences in fungal evenness and richness between soil depths of 0–20 and 20–40 cm.The number of Verticillium dahliae decreased,while the relative abundance of biocontrol bacteria such as Bacillus and Pseudoxanthomonas increased significantly following RTL.Overall RTL improved bacterial diversity,decreased the number of Verticillium dahliae and increased the relative abundance of biocontrol bacteria,which may have suppressed the occurrence of Verticillium wilt in cotton fields.Ming Dong Yan Wang Shulin Wang Guoyi Feng Qian Zhang Yongzeng Lin Qinglong Liang Yongqiang Wang Hong Qi 2023Phyton-International Journal of Experimental Botany2023,92,10:0
