1Liraglutide alleviates cardiac fibrosis through inhibiting P4hα-1 expression in STZ-induced diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy is an important contributor to morbidity and mortality of diabetic patients by causing heart failure.Interstitial and perivascular fibrosis plays a crucial role in diabetic cardiomyopathy.However,there is a lack of effective specific treatments available for diabetic cardiomyopathy.In the present study,we aim to explore the effects of Liraglutide,a GLP-1 analogue,on diabetic cardiomyopathy in STZ-induced diabetic rats fed with high-fat diet.A total of 60 male Wistar rats were randomly assigned to three groups,i.e.normal group,model group,and Liraglutide group,with 20 rats in each group.Serum levels of TC,TG,LDL-C,NEFA,and hydroxyproline were measured using commercial kits.Cardiac function was evaluated by QRS waves,LVEDd,LVESd,and LVEF.Myocardial fibrosis was measured by immunohistochemistry.Our results demonstrated that chronic administration of Liraglutide decreased the level of blood glucose and significantly alleviated lipid metabolic disturbance compared with the model group.Furthermore,Liraglutide was found to improve the damaged cardiac function.In line with this,we also found that the alleviation of cardiac dysfunction was associated with the decreased fibrosis in diabetic myocardial tissues,which was reflected by the decreased expressions of P4hα-1,COL-1,COL-3,MMP-1,and MMP-9.Our results thus suggest that Liraglutide might have a myocardial protective effect by inhibiting P4hα-1-mediated myocardial fibrosis.Tong Zhao Huiqiang Chen Fei Xu Juan Wang Yusheng Liu Xiaowei Xing Linlin Guo Mingxiang Zhang Qinghua Lu 2019Acta Biochimica et Biophysica Sinica2019,51,3:9
2Probabilistic ecological risk assessment for three chlorophenols in surface waters of China显示文摘Individual and combined assessment of risks of adverse effects to aquatic ecosystems of three chlorophenols(CPs),including 2,4dichlorophenol(2,4-DCP),2,4,6-trichlorophenol(2,4,6-TCP) and pentachlorophenol(PCP),were conducted.A probabilistic approach based on the concentrations of CPs in surface waters of China was used to determine the likelihood of adverse effects.The potential risk of CPs in surface waters of China was determined to be of concern,especially PCP and mixtures of CPs.The risks of adverse effects were examined as the joint probabilities of exposure and response.The joint probability for PCP was 0.271 in the worst case and 0.111 in the median case,respectively.Based on the cumulative probability,5% of aquatic organisms included in the assessment would be affected 21.36% of the time in the worst case and 5.99% of the time in median case,respectively.For the mixtures of CPs,the joint probability were 0.171 in the worst case and 0.503 in median case,respectively and 5% of species would be affected 49.83% of the time for the worst case and 12.72% in the median case,respectively.Risks of effects of the individual CPs,2,4-DCP and 2,4,6-TCP were deemed to be acceptable with a overlapping probability of < 0.1 with 5% of species being affected less than 4% of the time.Liqun Xing Hongling Liu John E Giesy Xiaowei Zhang Hongxia Yu 2012Journal of Environmental Sciences2012,24,2:7
3Global analysis of CCT family knockout mutants identifies four genes involved in regulating heading date in rice显示文摘Many genes encoding CCT domain-containing proteins regulate flowering time. In rice(Oryza sativa), 41 such genes have been identified, but only a few have been shown to regulate heading date. Here, to test whether and how additional CCT family genes regulate heading date in rice, we classified these genes into five groups based on their diurnal expression patterns. The expression patterns of genes in the same subfamily or in close phylogenetic clades tended to be similar. We generated knockout mutants of the entire gene family via CRISPR/Cas9. The heading dates of knockout mutants of only 4 of 14 genes previously shown to regulate heading date were altered, pointing to functional redundancy of CCT family genes in regulating this trait. Analysis of mutants of four other genes showed that OsCCT22, OsCCT38, and OsCCT41 suppress heading under long-day conditions and promote heading under short-day conditions. OsCCT03 promotes heading under both conditions and upregulates the expression of Hd1 and Ehd1, a phenomenon not previously reported for other such genes. 4A flexible carbon nanotube@V_(2)O_(5) film as a high-capacity and durable cathode for zinc ion batteries显示文摘Aqueous zinc-ion batteries(ZIBs)are receiving a continuously increasing attention for mobile devices,especially for the flexible and wearable electronics,due to their non-toxicity,non-flammability,and low-cost features.Despite the significant progress in achieving higher capacities for electrode materials of ZIBs,to endow them with high flexibility and economic feasibility is,however,still a significant challenge remaining unsolved.Herein,we present a highly flexible composite film composed of carbon nanotube film and V_(2)O_(5)(CNTF@V_(2)O_(5))with high strength and high conductivity,which is prepared by simply impregnating a porous CNT film with an aqueous V_(2)O_(5)sol under vacuum.For this material,intimate incorporation between V_(2)O_(5)and CNTs has been achieved,successfully integrating the high zinc ion storage capability with high mechanical flexibility.As a result,this CNTF@V_(2)O_(5)film delivers a high capacity of 356.6 m Ah g^(-1)at 0.4 A g^(-1)and excellent cycling stability with 80.1%capacity retention after 500 cycles at 2.0 A g^(-1).The novel strategy and the outstanding battery performance presented in this work should shed light on the development of high-performance and flexible ZIBs.Xiaowei Wang Liqun Wang Bao Zhang Jianmin Feng Jiafeng Zhang Xing Ou Feng Hou Ji Liang 2021Journal of Energy Chemistry2021,30,8:5

6Far-field coseismic gravity changes related to the 2015 MW7.8 Nepal(Gorkha)earthquake observed by superconducting gravimeters in Chinese mainland显示文摘Using data from five SGs at four stations in Chinese mainland,obvious permanent gravity changes caused by the 2015 MW7.8 Nepal(Gorkha)earthquake were detected.We analyzed the gravity effects from ground vertical deformation(VD)using co-site continuous GPS(cGPS)data collocated at the Lijiang and the Wuhan station,and hydrological effects using GLDAS models and groundwater level records.After removing these effects,SG observations before and after the earthquake revealed obvious permanent gravity changes:−3.0μGal,7.3μGal and 8.0μGal at Lhasa,Lijiang and Wuhan station,respectively.We found that the gravity changes cannot be explained by the results of dislocation theory.LeLin Xing ZiWei Liu JianGang Jia ShuQing Wu ZhengSong Chen XiaoWei Niu 2021Earth and Planetary Physics2021,5,2:4

8Fabrication of iridium oxide neural electrodes at the wafer level显示文摘Electro-deposition, electrical activation, thermal oxidation, and reactive ion sputtering are the four primary methods to fabricate iridium oxide film. Among these methods, reactive ion sputtering is a commonly used method in standard micro-fabrication processes. In different sputtering conditions, the component, texture, and electrochemistry character of iridium oxide varies considerably. To fabricate the iridium oxide film compatible with the wafer-level processing of neural electrodes, the quality of iridium oxide film must be able to withstand the mechanical and chemical impact of post-processing, and simultaneously achieve good performance as a neural electrode. In this study, parameters of sputtering were researched and developed to achieve a balance between mechanical stability and good electrochemical characteristics of iridium oxide film on electrode. Iridium oxide fabricating process combined with fabrication flow of silicon electrodes, at wafer-level, is introduced to produce silicon based planar iridium oxide neural electrodes. 10Different structural origins for different sized surface pits observed on a-plane GaN film显示文摘A correlation study between the observed surface morphology using high-resolution scanning electron microscopy(HRSEM) and the observed structural imperfections using transmission electron microscopy(TEM) has been conducted for a-plane Ga N. There are three different sized asymmetric surface pits: large pit of 500 nm–2 ?m in side length, medium pit of 50 nm in side length, and small pit with side lengths of less than 5 nm, which originate from incomplete island coalescence, screw dislocation, and partial dislocation(PD), respectively. Both screw dislocation and PD can produce pits on the free surface because they have a perpendicular line tension to the surface, which must remain in balance with the surface tension. The two types of dislocation lead to distinctive pit sizes because the PD has a smaller Burgers vector component along the dislocation line than the pure screw dislocation. A pit that is produced in the island-coalescing process is much larger than those caused by dislocations because island coalescence is a kinetic process that involves large-scale mass transportation, whereas the dislocation mediates the surface in the local area. These three types of surface pits sometimes interact with one another in space. 15AstragalosideⅣameliorates insulin induced insulin resistance in HepG2 cells through reactive oxygen species mediated c-Jun N-terminal kinase pathway显示文摘OBJECTIVE:To investigate the effects and elucidate the mechanism of Astragaloside IV(AS-IV)for insulin resistance(IR)and type 2 diabet es mellitus(T2DM).METHODS:CCK8 kit was used to detect cell viability,glucose detection kit was used to detect the concentration of glucose in cell supernatant,reactive oxygen species(ROS)detection kit and Western blot were used to explore the mechanism of Astragaloside IV(AS-IV)in improving IR.A diabetic rat model was also established by feeding high sugar and fat diet and streptozotocin(STZ)injection.After treatment with AS-IV,rosiglitazone(ROZ),or normal saline,the fasting blood glucose(FBG),C peptide(C-P),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and the glucose tolerance were assessed.RESULTS:AS-IV could effectively reduce the content of ROS and increase the glucose uptake in high insulintreated IR-type HepG 2 cells.The results of molecular mechanisms indicated that AS-IV could improve insulin resistance by reducing JNK phosphorylation and regulating c-Jun N-terminal kinase(JNK)downstream protein expression.Additionally,AS-IV could significantly reduce the levels of FBG,TNF-α,IL-6 and the glucose tolerance in diabetic rats(P<0.05 or<0.01).The high and medium dose groups of AS-IV could significantly increase the C-P levels in diabetic rats(P<0.05 or<0.01).CONCLUSIONS:Our results indicated that AS-IV improve liver IR through the JNK pathway and ROS,which meant a new molecular target for the treatment of diabetes.The AS-IV also helped to prevent and improved the insulin resistance of rats.YE Xiaomei XING Xiaowei YUAN Kangrui WANG Dongming WU Dudu CHEN Zhi YU Zhiqiang 2023Journal of Traditional Chinese Medicine2023,43,1:1

17Identification of interaction between HIV-1 glycoprotein 41 and integrase显示文摘Human immunodeficiency virus-1(HIV-1) encodes 15 viral proteins. Protein-protein interactions play a large role in the function of these proteins. In this study, we attempted to identify novel interactions between the HIV-1 proteins to better understand the role played by viral protein-protein interactions in the life cycle of HIV-1. Genes encoding the 15 viral proteins from the HIV-1 strain AD8 were inserted into the plasmids of a yeast two-hybrid system. By screening 120 pairs of proteins, interactions between seven pairs were found. This led to the discovery of an interaction between the HIV-1 proteins integrase(IN) and glycoprotein 41(gp41), which was confirmed by both co-immunoprecipitation(Co-IP) assays and fluorescence resonance energy transfer(FRET)imaging in live cells. In addition, it was found that the amino acids at positions 76–100 of gp41 are required for it to bind to IN. Deletion of this region from gp41 prevented its interaction with IN and reduced the production of HIV-1 in 293 T cells. 18Distribution characteristics of plastic film residue in long-term mulched farmland soil显示文摘Soil contamination from film debris following the prolonged application of mulching film has emerged as a worldwide concern.However,the extent that mulching films contribute to soil microplastics,and the spatial distribution of soil contamination from film debris remain unclear.In this study,the cotton field in Xinjiang(China),which underwent film mulching for a prolonged period of 5−30 years,was selected as the research location.A total of 360 soil samples were collected,aiming to study the spatial distribution characteristics of mulching film debris pollution.The samples were extracted using the density flotation method combined with stereomicroscopic;the source,composition,abundance,and distribution characteristics of soil MPs were identified by the scanning electron microscopic,and Fourier transform infrared spectroscopic analyses.In soil mulched for a 30 year period,the abundance of microplastics across the studied soil depth(0−60 cm)was 78.51±2.57 n/(100 g).Theμ-FTIR analyses revealed that the composition of the microplastics matched that of polyethylene materials.Therefore,plastic mulching could be inferred as a major contributor to microplastic pollution in agricultural lands.Overall,it is necessary to study the distribution characteristics of plastic film remaining for further study of plastic pollution in farmland soils.Zhengxin Xu Can Hu Xufeng Wang Long Wang Jianfei Xing Xiaowei He Zaibin Wang Pengfei Zhao 2023Soil Ecology Letters2023,5,3:0

20Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND
