| 1 | 3D bioactive composite scaffolds for bone tissue engineering显示文摘Bone is the second most commonly transplanted tissue worldwide,with over four million operations using bone grafts or bone substitute materials annually to treat bone defects.However,significant limitations affect current treatment options and clinical demand for bone grafts continues to rise due to conditions such as trauma,cancer,infection and arthritis.Developing bioactive three-dimensional(3D)scaffolds to support bone regeneration has therefore become a key area of focus within bone tissue engineering(BTE).A variety of materials and manufacturing methods including 3D printing have been used to create novel alternatives to traditional bone grafts.However,individual groups of materials including polymers,ceramics and hydrogels have been unable to fully replicate the properties of bone when used alone.Favourable material properties can be combined and bioactivity improved when groups of materials are used together in composite 3D scaffolds.This review will therefore consider the ideal properties of bioactive composite 3D scaffolds and examine recent use of polymers,hydrogels,metals,ceramics and bio-glasses in BTE.Scaffold fabrication methodology,mechanical performance,biocompatibility,bioactivity,and potential clinical translations will be discussed. | Gareth Turnbull Jon Clarke Frederic Picard Philip Riches Luanluan Jia Fengxuan Han Bin Li Wenmiao Shu | 2018 | Bioactive Materials2018,3,3: | 40 |
| 2 | Phase Ⅱ study of apatinib in combination with oral vinorelbine in heavily pretreated HER2-negative metastatic breast cancer and clinical implications of monitoring ctDNA显示文摘Objective:Apatinib is an oral TKI targeting VEGFR-2.Single-agent apatinib treatment has been shown to produce an objective response in patients with pretreated m BC.Oral vinorelbine also holds promise as a treatment of choice in patients with m BC.This study aimed to investigate the efficacy and safety of the oral vinorelbine-apatinib combination in patients with pretreated m BC.In addition,we detected gene variants in ct DNA to explore the therapeutic implications.Methods:This study enrolled patients with HER2-negative m BC who were pretreated with anthracycline/taxanes.Patients were treated with apatinib at 500 mg/425 mg daily plus oral vinorelbine 60 mg/m2 on days 1,8,and 15 of every cycle(3 weeks).The primary endpoint was PFS.The secondary endpoints were ORR,CBR,OS,and safety.Patients eligible for ct DNA detection were evaluated before and during treatment.Results:Forty patients were enrolled.The median PFS was 5.2 months(95%CI,3.4–7.0 months),and the median OS was 17.4 months(95%CI,8.0–27.0 months).The ORR was 17.1%(6/35),and the CBR was 45.7%(16/35).The most common AEs included gastrointestinal reaction,myelosuppression,and hypertension.In 20 patients,ct DNA was detected at baseline and during treatment.A significant difference was found in PFS for undetected vs.detected baseline ct DNA(13.9 months vs.3.6 months,P=0.018).Conclusions:All-oral therapy with apatinib plus vinorelbine displayed objective efficacy in patients with heavily pretreated HER2-negative m BC,with acceptable and manageable toxicity profiles.Patients with no gene variant detected and lower variant allele frequencies in ct DNA at baseline showed longer PFS. | Anjie Zhu Peng Yuan Nanlin Hu Mingzhou Li Wenmiao Wang Xue Wang Jian Yue Jiayu Wang Yang Luo Fei Ma Pin Zhang Qing Li Binghe Xu Shanbo Cao Giuseppe Lippi Yoichi Naito Mohammed A.Osman Gustavo N.Marta Gianluca Franceschini Armando Orlandi | 2021 | Cancer Biology & Medicine2021,18,3: | 4 |
| 3 | Building Osteogenic Microenvironments with a Double-Network Composite Hydrogel for Bone Repair显示文摘The critical factor determining the in vivo effect of bone repair materials is the microenvironment,which greatly depends on their abilities to promote vascularization and bone formation.However,implant materials are far from ideal candidates for guiding bone regeneration due to their deficient angiogenic and osteogenic microenvironments.Herein,a double-network composite hydrogel combining vascular endothelial growth factor(VEGF)-mimetic peptide with hydroxyapatite(HA)precursor was developed to build an osteogenic microenvironment for bone repair.The hydrogel was prepared by mixing acrylatedβ-cyclodextrins and octacalcium phosphate(OCP),an HA precursor,with gelatin solution,followed by ultraviolet photo-crosslinking.To improve the angiogenic potential of the hydrogel,QK,a VEGF-mimicking peptide,was loaded in acrylatedβ-cyclodextrins.The QK-loaded hydrogel promoted tube formation of human umbilical vein endothelial cells and upregulated the expression of angiogenesis-related genes,such as Flt1,Kdr,and VEGF,in bone marrow mesenchymal stem cells.Moreover,QK could recruit bone marrow mesenchymal stem cells.Furthermore,OCP in the composite hydrogel could be transformed into HA and release calcium ions facilitating bone regeneration.The double-network composite hydrogel integrated QK and OCP showed obvious osteoinductive activity.The results of animal experiments showed that the composite hydrogel enhanced bone regeneration in skull defects of rats,due to perfect synergistic effects of QK and OCP on vascularized bone regeneration.In summary,improving the angiogenic and osteogenic microenvironments by our double-network composite hydrogel shows promising prospects for bone repair. | Jiaying Li Jinjin Ma Qian Feng En Xie Qingchen Meng Wenmiao Shu Junxi Wu Liming Bian Fengxuan Han Bin Li | 2023 | Research2023,,3: | 3 |

| 11 | Research on the 'three shells' cooperative support technology of large-section chambers in deep mines显示文摘The'three shells'cooperative support technology was proposed herein according to both the large deformation of the rock surrounding large-section chambers in deep mines and the precarious stability of the support structures therein.The development range of the plastic zone in the surrounding rock was controlled by a stress shell to reduce the difficulty of controlling the surrounding rock.Additionally,the residual strength of the rock mass in the plastic zone and the self-bearing capacity of the surrounding rock were improved by a reinforced load-bearing shell.Furthermore,a passive load-bearing shell could restore the triaxial stress state of the surrounding rock on the free surface,reduce the influence of the external environment on the surrounding rock,and reinforce the surrounding rock with the strength of the shell.Reasonable layouts of large-section chambers were determined by analyzing the control effect of the stress shell on the surrounding rock under three kinds of in situ stress fields.The orthogonal test method was applied to reveal the influences of different support parameters in the reinforced loadbearing shell and passive load-bearing shell on the surrounding rock stability.The surrounding rock control effect of the'three shells'collaborative support technology was analyzed through numerical simulation and field monitoring.The results show that the maximum displacement between the roof and floor of the coal preparation chamber in the Xinjulong coal mine was approximately 48 mm,and the maximum displacement between its two sides was approximately 65 mm,indicating that the technology proposed herein could meet the long-term control requirements of the surrounding rock stability for large-section chambers in deep mines. | Cheng Zhu Yong Yuan Wenmiao Wang Zhongshun Chen Shengzhi Wang Huiwei Zhong | 2021 | International Journal of Mining Science and Technology2021,31,4: | 1 |

During first complete remission after routine chemotherapy, the patient received autologous blood stem cell transplantation, but he had relapse later. He gained a second CR after chemotherapy and underwent hi-HSCT from his daughter, who had HLA mismatched at three loci. Recovery of hemopoiesis was found at day 14 and complete donor chimerism was confirmed by PCR-STR on day 34, 95 and 238. | 20 | Three-dimensional biofabrication of nanosecond laser micromachined nanofibre meshes for tissue engineered scaffolds显示文摘There is a high demand for bespoke grafts to replace damaged or malformed bone and cartilage tissue.Three-dimensional(3D)printing offers a method of fabricating complex anatomical features of clinically relevant sizes.However,the construction of a scaffold to replicate the complex hierarchical structure of natural tissues remains challenging.This paper reports a novel biofabrication method that is capable of creating intricately designed structures of anatomically relevant dimensions.The beneficial properties of the electrospun fibre meshes can finally be realised in 3D rather than the current promising breakthroughs in two-dimensional(2D).The 3D model was created from commercially available computer-aided design software packages in order to slice the model down into many layers of slices,which were arrayed.These 2D slices with each layer of a defined pattern were laser cut,and then successfully assembled with varying thicknesses of 100μm or 200μm.It is demonstrated in this study that this new biofabrication technique can be used to reproduce very complex computer-aided design models into hierarchical constructs with micro and nano resolutions,where the clinically relevant sizes ranging from a simple cube of 20 mm dimension,to a more complex,50 mm-tall human ears were created.In-vitro cell-contact studies were also carried out to investigate the biocompatibility of this hierarchal structure.The cell viability on a micromachined electrospun polylactic-co-glycolic acid fibre mesh slice,where a range of hole diameters from 200μm to 500μm were laser cut in an array where cell confluence values of at least 85%were found at three weeks.Cells were also seeded onto a simpler stacked construct,albeit made with micromachined poly fibre mesh,where cells can be found to migrate through the stack better with collagen as bioadhesives.This new method for biofabricating hierarchical constructs can be further developed for tissue repair applications such as maxillofacial bone injury or nose/ear cartilage replacement in the future. | Ross HMcWilliam Wenlong Chang Zhao Liu Jiayuan Wang Fengxuan Han Richard ABlack Junxi Wu Xichun Luo Bin Li Wenmiao Shu | 2023 | Biomaterials Translational2023,4,2: | 0 |
