| 1 | Cell membrane-based nanoparticles: a new biomimetic platform for tumor diagnosis and treatment显示文摘Taking inspiration from nature, the biomimetic concept has been integrated into drug delivery systems in cancer therapy. Disguised with cell membranes, the nanoparticles can acquire various functions of natural cells. The cell membrane-coating technology has pushed the limits of common nano-systems(fast elimination in circulation) to more effectively navigate within the body. Moreover, because of the various functional molecules on the surface, cell membrane-based nanoparticles(CMBNPs) are capable of interacting with the complex biological microenvironment of the tumor. Various sources of cell membranes have been explored to camouflage CMBNPs and different tumor-targeting strategies have been developed to enhance the anti-tumor drug delivery therapy. | 2 | Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. | 4 | Targets and molecular mechanisms of triptolide in cancer therapy显示文摘Triptolide(TPL/TL) is a natural drug with novel anticancer effects. Preclinical studies indicated that TPL inhibits cell proliferation, induces cell apoptosis, inhibits tumor metastasis and enhances the effect of other therapeutic methods in various cancer cell lines. Multiple molecules and signaling pathways, such as caspases, heat-shock proteins, NF-κB, and deoxyribonucleic acid(DNA) repair-associated factors, are associated with the anti-cancer effect. TPL also improves chemoradiosensitivity in cancer therapy. Phase I trials indicate the potential clinical value of TPL use. | 6 | Potential role of N-carbamoyl glutamate in biosynthesis of arginine and its significance in production of ruminant animals显示文摘Arginine(ARG) exerts many beneficial effects on animal body and enhanced angiogenesis, lactogenesis, which finally leads to the improvement in nitrogen(N) metabolism, reproduction, lactation, immunity and growth. Unfortunately, unprotected ARG will be degraded in the rumen and its price is high, thus feeding rumen-protected ARG seems to be uneconomical. Alternatively, N-carbamoyl glutamate(NCG) is structural analogue of N-acetyl glutamate, cofactor of cabamoyl phosphate synthetase 1, is lower in rumen degradation compared to ARG. Additionally, rumen epithelial and duodenal cells have potentially utilized the NCG for ureagenesis. Supplementation of NCG to high yielding dairy cows increased plasma concentration of ARG and nitric oxide, decreased the plasma ammonia N and improved lactation performance and N utilization. Supplementation of NCG enhanced pregnancy rates in rats, improved litter size and fetal survival rate, thereby improved the reproductive performance of sows. Oral NCG supplementation increases plasma ARG and somatotropin levels, and increased growth rate and muscle protein synthesis in nursing piglets. The NCG is potential a relatively cheaper source of feed additive to offer vital compensation over oral administration of ARG, resulting in improved ruminant animal health and production. | 8 | Effects of Saccharomyces cerevisiae fermentation products on performance and rumen fermentation and microbiota in dairy cows fed a diet containing low quality forage显示文摘Background: A possible option to meet the increased demand of forage for dairy industry is to use the agricultural byproducts, such as corn stover. However, nutritional value of crop residues is low and we have been seeking technologies to improve the value. A feeding trial was performed to evaluate the effects of four levels of Saccharomyces cerevisiae fermentation product(SCFP; Original XP; Diamond V) on lactation performance and rumen fermentation in mid-lactation Holstein dairy cows fed a diet containing low-quality forage. Eighty dairy cows were randomly assigned into one of four treatments: basal diet supplemented with 0, 60, 120, or 180 g/d of SCFP per head mixed with 180, 120, 60, or 0 g of corn meal, respectively. The experiment lasted for 10 wks, with the first 2 weeks for adaptation.Results: Dry matter intake was found to be similar(P > 0.05) among the treatments. There was an increasing trend in milk production(linear, P ≤ 0.10) with the increasing level of SCFP supplementation, with no effects on contents of milk components(P > 0.05). Supplementation of SCFP linearly increased(P < 0.05) the N conversion, without affecting rumen pH and ammonia-N(P > 0.05). Increasing level of SCFP linearly increased(P < 0.05) concentrations of ruminal total volatile fatty acids, acetate, propionate, and butyrate, with no difference in molar proportion of individual acids(P > 0.05). The population of fungi and certain cel ulolytic bacteria(Ruminococcus albus, R. flavefaciens and Fibrobacter succinogenes)increased linearly(P < 0.05) but those of lactate-utilizing(Selenomonas ruminantium and Megasphaera elsdeni) and lactate-producing bacteria(Streptococcus bovis) decreased linearly(P ≤ 0.01) with increasing level of SCFP. The urinary purine derivatives increased linearly(P < 0.05) in response to SCFP supplementation, indicating that SCFP supplementation may benefit for microbial protein synthesis in the rumen.Conclusions: The SCFP supplementation was effective in maintaining milk persistency of mid-lactation cows receiving diets containing low-quality forage. The beneficial effect of SCFP could be attributed to improved rumen function; 1)microbial population shift toward greater rumen fermentation efficiency indicated by higher rumen fungi and cel ulolytic bacteria and lower lactate producing bacteria, and 2) rumen microbial fermentation toward greater supply of energy and protein indicated by greater ruminal VFA concentration and increased N conversion. | 9 | The recognition of adakite-type gneisses in the North Dabie Mountain and its implication to ultrahigh pressure metamorphic geology显示文摘Adakite is an igneous rock generated by the partial melting of subducting ocean slab in an island arc setting. Meanwhile, residuals of the slab-melting are transformed to eclogite or garnet-amphibolite during a slab subduction process. This note reports the adakite-type grey gneisses discovered in the North Dabie Mountain. These adakitic rocks are characterized by high AI2O3(14.97%-17.56%), Sr (545-941 μg/g), Sr/Y (44.71-218.98), La/Yb (16.70-97.50), and low Y (3.32-12.19μg/g), Yb (0.31-1.34 μg/g), Sc (0.92-4.2μg/g), as well as positive Sr anomaly but absent or positive Eu anomaly. They are different from the low aluminum grey gneisses with low AI2O3 (13.73%-14.38%), Sr (262-409 μg/g), Sr/Y (6.46-15.97) and La/Yb (1.90-22.60), high Y(21.79-36.24 μg/g), Yb (2.00-5.59 μg/g) , Sc (8.30%-12.70%), and negative Sr, Eu anomaly in the North Dabie Mountain. | 10 | Menthol-modified casein nanoparticles loading 10-hydroxycamptothecin for glioma targeting therapy显示文摘Chemotherapy outcomes for the treatment of glioma remains unsatisfactory due to the inefficient drug transport across the blood–brain barrier(BBB) and insufficient drug accumulation in the tumor region. Although many approaches, including various nanosystems, have been developed to promote the distribution of chemotherapeutics in the brain tumor, the delivery efficiency and the possible damage to the normal brain function still greatly restrict the clinical application of the nanocarriers.Therefore, it is urgent and necessary to discover more safe and effective BBB penetration and gliomatargeting strategies. In the present study, menthol, one of the strongest BBB penetration enhancers screened from traditional Chinese medicine, was conjugated to casein, a natural food protein with brain targeting capability. Then the conjugate self-assembled into the nanoparticles to load anti-cancer drugs.The nanoparticles were characterized to have appropriate size, spheroid shape and high loading drug capacity. Tumor spheroid penetration experiments demonstrated that penetration ability of mentholmodified casein nanoparticles(M-CA-NP) into the tumor were much deeper than that of unmodified nanoparticles. In vivo imaging further verified that M-CA-NPs exhibited higher brain tumor distribution than unmodified nanoparticles. The median survival time of glioma-bearing mice treated with HCPT-MCA-NPs was significantly prolonged than those treated with free HCPT or HCPT-CA-NPs. HE staining ofthe organs indicated the safety of the nanoparticles. | 11 | MECHANICAL PROPERTIES OF FINE PITCH DEVICES SOLDERED JOINTS BASED ON CREEP MODEL显示文摘Nonlinear analyses of quad flat package (QFP) on printed circuit board (PCB) assemblies subjected to thermal cycling conditions are presented.Two different solders are considered,namely, Sn37Pb and Sn3.5Ag.The stress and strain response of fine pitch devices soldered joints was investigated by using finite element method based on Garofalo-Arrheninus model.The simulated results indicate creep distribution of soldered joints is not uniform,the heel and toe of soldered joints, the area between soldered joints and leads are the creep concentrated sites.The similar phenomena of stress curves simulated based on Garofalo-Arrhenlnus model and Anand equations is confirmed,and the creep strain value of Sn3.5Ag soldered joints is lower than that of Sn37Pb soldered joints. Thermal cycling results show that Sn3.5Ag strongly outperforms Sn37Pb for QFP devices under the studied test condition.This is well matched with the experimental outcome analyzed.In addition,the soldered devices were tested by micro-joints tester,the tensile strength of Sn3.5Ag soldered joints is found to be higher than that of Sn37Pb soldered joints.By analyzing the fracture microstructure of soldered joints,it is found that fracture mechanism of Sn3.5Ag soldered joints is toughness fracture, while fracture mechanism of Sn37Pb soldered joints includes brittle fracture and toughness fracture. | 12 | Stability analysis of the temperature cracks in Xiaowan arch dam显示文摘In the Xiaowan arch dam there are massive temperature cracks nearly parallel to the dam axis. Obviously, whether the cracks may spread or not during the water storage process is one of the crucial factors for the safety of a dam. In this paper, a new type of crack element, in which the contact component is implicitly included into the concrete component, is proposed to simulate the effects of the existing cracks. The crack element is proved by numerical example to share the merits of both conventional contact elements and joint elements. With a finite element model of the cracked arch dam together with its rock foundation established, the transient displacement and stress fields of the dam are obtained. The complicated rock foundation, the construction process of the arch dam, the massive cracks, the transient temperature field, as well as the water storage process have been taken into consideration in the simulation. In addition to the global model, several sub-models for typical crack tips are also generated with finer elements placed around the tips. Thus, more accurate displacement and stress distribution are obtained by simultaneous sub-model simulation. When the technological parameters were set as the ranges of mould temperature 1100-1300℃,cooling distance (the distance from the exit of the cast mould to the start point of cooling) 10-20 mm, casting speed 0.2-2.5 mm/s,cooling water (20-25℃) volume 1000-1320 L/h, and when these parameters matched reasonably, the CUS process was performed stably, and pure copper rods containing continuous columnar crystals with bright and smooth surface were produced. The dendritic arm spacing of the crystals in copper rods decreased with increasing the casting speed. | 17 | Transcriptomic and genetic approaches reveal an essential role of the NAC transcription factor SlNAP1 in the growth and defense response of tomato显示文摘With global climate change,plants are frequently being exposed to various stresses,such as pathogen attack,drought,and extreme temperatures.Transcription factors(TFs)play crucial roles in numerous plant biological processes;however,the functions of many tomato(Solanum lycopersicum L.)TFs that regulate plant responses to multiple stresses are largely unknown.Here,using an RNA-seq approach,we identified SlNAP1,a NAC TF-encoding gene,which was strongly induced by various stresses.By generating SlNAP1 transgenic lines and evaluating their responses to biotic and abiotic stresses in tomato,we found that SlNAP1-overexpressing plants showed significantly enhanced defense against two widespread bacterial diseases,leaf speck disease,caused by Pseudomonas syringae pv.tomato(Pst)DC3000,and root-borne bacterial wilt disease,caused by Ralstonia solanacearum.In addition,SlNAP1 overexpression dramatically improved drought tolerance in tomato.Although the SlNAP1-overexpressing plants were shorter than the wild-type plants during the early vegetative stage,eventually,their fruit yield increased by 10.7%.Analysis of different hormone contents revealed a reduced level of physiologically active gibberellins(GAs)and an increased level of salicylic acid(SA)and abscisic acid(ABA)in the SlNAP1-overexpressing plants.Moreover,EMSAs and ChIP-qPCR assays showed that SlNAP1 directly activated the transcription of multiple genes involved in GA deactivation and both SA and ABA biosynthesis.Our findings reveal that SlNAP1 is a positive regulator of the tomato defense response against multiple stresses and thus may be a potential breeding target for improving crop yield and stress resistance. | Jiao Wang Chenfei Zheng Xiangqi Shao Zhangjian Hu Jianxin Li Ping Wang Anran Wang Jingquan Yu Kai Shi | 2020 | Horticulture Research2020,7,1: | 9 |
| 18 | Contrasting responses of net primary productivity to inter-annual variability and changes of climate among three forest types in northern China显示文摘Aims A lack of explicit information on differential controls on net primary productivity(NPP)across regions and ecosystem types is largely responsible for uncertainties in global trajectories of terrestrial carbon balance with changing environment.The objectives of this study were to determine how NPP of different forest types would respond to inter-annual variability of climate and to examine the responses of NPP to future climate change scenarios across contrasting forest types in northern China.Methods We investigated inter-annual variations of NPP in relation to climate variability across three forest types in northern China,including a boreal forest dominated by Larix gmelinii Rupr.,and two temperate forests dominated by Pinus tabulaeformis Carr.and Quercus wutaishanica Mayr.,respectively,and studied the responses of NPP in these forests to predicted changes in climate for the periods 2011–40,2041–70 and 2070–100 under carbon emission scenarios A2 and B2 of Intergovernmental Panel on Climate Change.We simulated the responses of NPP to predicted changes in future climate as well as inter-annual variability of the present climate with the Biome-BGC version 4.2 based on site-and species-specific parameters.The modeled forest NPP data were validated against values in literature for similar types of forests and compared with inter-annual growth variations reflected by tree-ring width index(RWI)at the study sites.Important Findings Inter-annual variations in modeled NPP during the period 1960–06 were mostly consistent with the temporal patterns in RWI.There were contrasting responses of modeled NPP among the three forest types to inter-annual variability of the present climate as well as to predicted changes in future climate.The modeled NPP was positively related to annual mean air temperature in the L.gmelinii forest(P<0.001),but negatively in the P.tabulaeformis forest(P=0.05)and the Q.wutaishanica forest(P=0.03),while the relationships of modeled NPP with annual precipitation for the three forest types were all positive.Multiple stepwise regression analyses showed that temperature was a more important constraint of NPP than precipitation in the L.gmelinii forest,whereas precipitation appeared to be a prominent factor limiting the growth in P.tabulaeformis and Q.wutaishanica.Model simulations suggest marked,but differential increases in NPP across the three forest types with predicted changes in future climate. | Shuai Ouyang Xiangping Wang Yulian Wu Osbert Jianxin Sun | 2014 | Journal of Plant Ecology2014,7,3: | 9 |
| 19 | Drug targeting through platelet membrane-coated nanoparticles for the treatment of rheumatoid arthritis显示文摘The effective drug treatment of rheumatoid arthritis (RA) is hindered by poordelivery efficiency to the diseased site and the serious side effects caused bywide-spread drug distribution. Traditional drug-targeting strategies, such asligand modification, are complex, laborious, and inefficient. Inspired by theintrinsic relationship between platelets and RA, platelet-mimetic nanoparticles(PNPs) were developed for targeted drug delivery in RA. Through plateletreceptor-mediated adhesion, an intact platelet membrane was coated onto poly(lactic-co-glycolic add) nanopartides, endowing the resulting PNPs with variousfunctional receptors. By coating with platelet membranes, the nanoparticleswere stabilized and had a better circulation profile, providing a benefit forpassive targeting. In vitro binding of PNPs to inflamed endothelium, and in vivoaccumulation in joints of a collagen-induced arthritis (CIA) mouse model of RAwere significantly improved via P-selectin and GVPI recognition, indicating thatthe PNPs could effectively target to RA tissues through multiple mechanisms,similar to natural platelets. Moreover, FK506, a model drug, was loaded into thePNPs and used to treat RA. Pharmacodynamic studies demonstrated that theFK506-PNPs had a notable anti-arthritic effect in CIA mice. | 20 | A Survey of MRI-Based Brain Tumor Segmentation Methods显示文摘Brain tumor segmentation aims to separate the different tumor tissues such as active cells, necrotic core,and edema from normal brain tissues of White Matter(WM), Gray Matter(GM), and Cerebrospinal Fluid(CSF). MRIbased brain tumor segmentation studies are attracting more and more attention in recent years due to non-invasive imaging and good soft tissue contrast of Magnetic Resonance Imaging(MRI) images. With the development of almost two decades, the innovative approaches applying computer-aided techniques for segmenting brain tumor are becoming more and more mature and coming closer to routine clinical applications. The purpose of this paper is to provide a comprehensive overview for MRI-based brain tumor segmentation methods. Firstly, a brief introduction to brain tumors and imaging modalities of brain tumors is given. Then, the preprocessing operations and the state of the art methods of MRI-based brain tumor segmentation are introduced. Moreover, the evaluation and validation of the results of MRI-based brain tumor segmentation are discussed.
