| 1 | Autophagy inhibition enhances apigenin-induced apoptosis in human breast cancer cells显示文摘Apigenin (4',5,7-trihydroxyflavone) is a member of the flavone subclass of flavonoids present in fruits and vegetables. The involvement of autophagy in the apigenin-induced apoptotic death of human breast cancer cells was investigated. Cell proliferation and viability were assessed by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) and clonogenic assays. Flow cytometry, fluorescent staining and Western blot analysis were employed to detect apoptosis and autophagy, and the role of autophagy was assessed using autophagy inhibitors. Apigenin dose- and time-dependently repressed the proliferation and clonogenic survival of the human breast cancer T47D and MDA-MB-231 cell lines. The death of T47D and MDA-MB-231 cells was due to apoptosis associated with increased levels of Caspase3, PARP cleavage and Bax/Bcl-2 ratios. The results from flow cytometry and fluorescent staining also verified the occurrence of apoptosis. In addition, the apigenin-treated cells exhibited autophagy, as characterized by the appearance of autophagosomes under fluorescence microscopy and the accumulation of acidic vesicular organelles (AVOs) by flow cytometry. Furthermore, the results of the Western blot analysis revealed that the level of LC3-II, the processed form of LC3-I, was increased. Treatment with the autophagy inhibitor, 3-methyladenine (3-MA), significantly enhanced the apoptosis induced by apigenin, which was accompanied by an increase in the level of PARP cleavage. Similar results were also confirmed by flow cytometry and fluorescence microscopy. These results indicate that apigenin has apoptosis-and autophagy-inducing effects in breast cancer cells. | 2 | The Ghd7 transcription factor represses ARE1 expression to enhance nitrogen utilization and grain yield in rice显示文摘The genetic improvement of nitrogen use efficiency(NUE)of crops is vital for grain productivity and sustainable agriculture.However,the regulatory mechanism of NUE remains largely elusive.Here,we report that the rice Grain number,plant height,and heading date7(Ghd7)gene genetically acts upstream of ABC1 REPRESSOR1(ARE1),a negative regulator of NUE,to positively regulate nitrogen utilization.As a transcriptional repressor,Ghd7 directly binds to two Evening Element-like motifs in the promoter and intron 1 of ARE1,likely in a cooperative manner,to repress its expression.Ghd7 and ARE1 display diurnal expression patterns in an inverse oscillation manner,mirroring a regulatory scheme based on these two loci.Analysis of a panel of 2656 rice varieties suggests that the elite alleles of Ghd7 and ARE1 have undergone diversifying selection during breeding.Moreover,the allelic distribution of Ghd7 and ARE1 is associated with the soil nitrogen deposition rate in East Asia and South Asia.Remarkably,the combination of the Ghd7 and ARE1 elite alleles substantially improves NUE and yield performance under nitrogen-limiting conditions.Collectively,these results define a Ghd7–ARE1-based regulatory mechanism of nitrogen utilization,providing useful targets for genetic improvement of rice NUE. | Qing Wang Qingmei Su Jinqiang Nian Jian Zhang Meng Guo Guojun Dong Jiang Hu Rongsheng Wang Changshuo Wei Guanwen Li Wan Wang Hui-Shan Guo Shaoyang Lin Wenfeng Qian XianzhiXie Qian Qian Fan Chen Jianru Zuo | 2021 | Molecular Plant2021,14,6: | 10 |
| 3 | A Methodology for Reliability of WSN Based on Software Defined Network in Adaptive Industrial Environment显示文摘As communication technology and smart manufacturing have developed, the industrial internet of things(IIo T)has gained considerable attention from academia and industry.Wireless sensor networks(WSNs) have many advantages with broad applications in many areas including environmental monitoring, which makes it a very important part of IIo T. However,energy depletion and hardware malfunctions can lead to node failures in WSNs. The industrial environment can also impact the wireless channel transmission, leading to network reliability problems, even with tightly coupled control and data planes in traditional networks, which obviously also enhances network management cost and complexity. In this paper, we introduce a new software defined network(SDN), and modify this network to propose a framework called the improved software defined wireless sensor network(improved SD-WSN). This proposed framework can address the following issues. 1) For a large scale heterogeneous network, it solves the problem of network management and smooth merging of a WSN into IIo T. 2) The network coverage problem is solved which improves the network reliability. 3) The framework addresses node failure due to various problems, particularly related to energy consumption.Therefore, it is necessary to improve the reliability of wireless sensor networks, by developing certain schemes to reduce energy consumption and the delay time of network nodes under IIo T conditions. | 4 | Variation of spatio-temporal distribution of on-road vehicle emissions based on real-time RFID data显示文摘High-resolution vehicular emissions inventories are important for managing vehicular pollution and improving urban air quality. This study developed a vehicular emission inventory with high spatio-temporal resolution in the main urban area of Chongqing, based on realtime traffic data from 820 RFID detectors covering 454 roads, and the differences in spatiotemporal emission characteristics between inner and outer districts were analysed. The result showed that the daily vehicular emission intensities of CO, hydrocarbons, PM2.5, PM10,and NO_(x) were 30.24, 3.83, 0.18, 0.20, and 8.65 kg/km per day, respectively, in the study area during 2018. The pollutants emission intensities in inner district were higher than those in outer district. Light passenger cars(LPCs) were the main contributors of all-day CO emissions in the inner and outer districts, from which the contributors of NO_(x) emissions were different. Diesel and natural gas buses were major contributors of daytime NO_(x) emissions in inner districts, accounting for 40.40%, but buses and heavy duty trucks(HDTs) were major contributors in outer districts. At nighttime, due to the lifting of truck restrictions and suspension of buses, HDTs become the main NO_(x) contributor in both inner and outer districts,and its three NO_(x) emission peak hours were found, which are different to the peak hours of total NO_(x) emission by all vehicles. Unlike most other cities, bridges and connecting channels are always emission hotspots due to long-time traffic congestion. | 7 | Electrochemical hydrogen storage properties of Mg100-xNix produced by hydriding combustion synthesis and mechanical milling显示文摘In this work, Mg-based hydrogen storage composites with an initial 100-x: x(x=25, 32.3, 50, 66.7) of Mg:Ni molar ratio were prepared by HCS+MM and their phase compositions and electrochemical performances were investigated in detail. The results show that the composites with desirable constituents can be achieved by adjusting the molar ratio of the starting materials in the HCS process. Particularly, the HCS product of Mg_(67.7)Ni_(32.3) consists of the principal phase Mg_2NiH_4 and minor phase Mg_2NiH_(0.3). The dominate phase varies from Mg_2NiH_(0.3) and MgH_2 for the Mg enriched sample(x < 32.3) to MgNi_2 and Ni for the Ni enriched sample(x > 32.3). The MM modification not only brings about grain refinement of the alloys, but also leads to phase transformation of part Mg_2NiH_4 to Mg_2NiH_(0.3) in the Mg_(67.7)Ni_(32.3)sample. Electrochemical tests indicate that each sample can reach its maximum discharge capacity at the first cycle. Mg_(67.7)Ni_(32.3) displays the highest discharge capacity as well as a superior electrochemical kinetics owing to its excellent H atom diffusion ability and lower charge-transfer resistance. | 15 | lncR-GAS5 upregulates the splicing factor SRSF10 to impair endothelial autophagy,leading to atherogenesis显示文摘Long noncoding RNAs(lncRNAs)play a critical role in the regulation of atherosclerosis.Here,we investigated the role of the lncRNA growth arrest-specific 5(lncR-GAS5)in atherogenesis.We found that the enforced expression of lncR-GAS5 contributed to the development of atherosclerosis,which presented as increased plaque size and reduced collagen content.Moreover,impaired autophagy was observed,as shown by a decreased LC3II/LC3I protein ratio and an elevated P62 level in lncR-GAS5-overexpressing human aortic endothelial cells.By contrast,lncR-GAS5 knockdown promoted autophagy.Moreover,serine/arginine-rich splicing factor 10(SRSF10)knockdown increased the LC3II/LC3I ratio and decreased the P62 level,thus enhancing the formation of autophagic vacuoles,autolysosomes,and autophagosomes.Mechanistically,lncR-GAS5 regulated the downstream splicing factor SRSF10 to impair autophagy in the endothelium,which was reversed by the knockdown of SRSF10.Further results revealed that overexpression of the lncR-GAS5-targeted gene miR-193-5p promoted autophagy and autophagic vacuole accumulation by repressing its direct target gene,SRSF10.Notably,miR-193-5p overexpression decreased plaque size and increased collagen content.Altogether,these findings demonstrate that lncR-GAS5 partially contributes to atherogenesis and plaque instability by impairing endothelial autophagy.In conclusion,lncR-GAS5 overexpression arrested endothelial autophagy through the miR-193-5p/SRSF10 signaling pathway.Thus,miR-193-5p/SRSF10 may serve as a novel treatment target for atherosclerosis. | Yuhua Fan Yue Zhang Hongrui Zhao Wenfeng Liu Wanqing Xu Lintong Jiang Ranchen Xu Yue Zheng Xueqing Tang Xiaohan Li Limin Zhao Xin Liu Yang Hong Yuan Lin Hui Chen Yong Zhang | 2023 | Frontiers of Medicine2023,17,2: | 1 |
