| 1 | Single-cell transcriptome analysis reveals widespread monoallelic gene expression in individual rice mesophyll cells显示文摘Monoallelic gene expression refers to the phenomenon that all transcripts of a gene in a cell are expressed from only one of the two alleles in a diploid organism. Although monoallelic gene expression has been occasionally reported with bulk transcriptome analysis in plants, how prevalent it is in individual plant cells remains unknown. Here, we developed a single-cell RNA-seq protocol in rice and investigated allelic expression patterns in mesophyll cells of indica(93-11) and japonica(Nipponbare) inbred lines, as well as their F1 reciprocal hybrids. We observed pervasive monoallelic gene expression in individual mesophyll cells, which could be largely explained by stochastic and independent transcription of two alleles. By contrast, two mechanisms that were proposed previously based on bulk transcriptome analyses, parent-oforigin effects and allelic repression, were not well supported by our data. Furthermore, monoallelically expressed genes exhibited a number of characteristics, such as lower expression levels, narrower H3K4me3/H3K9ac/H3K27me3 peaks, and larger expression divergences between 93-11 and Nipponbare. | 2 | HYD-PEP06 suppresses hepatocellular carcinoma metastasis,epithelial-mesenchymal transition and cancer stem cell-like properties by inhibiting PI3K/AKT and WNT/β-catenin signaling activation显示文摘HYD-PEP06,an endostatin-modified polypeptide,has been shown to produce effective anticolorectal carcinoma effects through inhibiting epithelial-mesenchymal transition(EMT).However,whether HYD-PEP06 has similar suppressive effect on hepatocellular carcinoma(HCC) remained unknown.In this study,HYD-PEP06 inhibited metastasis and EMT but not proliferation in vitro.Cignal finder pathway reporter array and Western blot analysis revealed that HYD-PEP06 suppressed HCCLM3 cell metastasis and EMT by inhibiting the PI3 K/AKT pathway.Moreover,HYD-PEP06 exerted antimetastasis effects in HepG2 cancer stem-like cells(CSCs) via suppressing the WNT/β-catenin signaling pathway.Finally,in HCCLM3 tumor-bearing BALB/c nu/nu nude mice,HYD-PEP06 substantially suppressed tumor growth,lung metastasis and HCC progress.Our results suggest that HYD-PEP06 inhibits the metastasis and EMT of HCC and CSCs as well,and thus has the potential as an agent for HCC treatment. | Wei Tian Jiatong Li Zhuo Wang Tong Zhang Ying Han Yanyan Liu Wenfeng Chu Yu Liu Baofeng Yang | 2021 | Acta Pharmaceutica Sinica B2021,11,6: | 5 |

| 7 | Antenna Decoupling Based on Characteristic Modes Cancellation显示文摘The demand for a high transmission data rate in 5G leads to the wide application of the multi-input,multi-output(MIMO)technique.However,the narrow design space in mobile phone causes severe unexpected coupling between antenna units and deteriorate the system performance dramatically.In this paper,we focus on the antenna coupling within the MIMO system.Some decoupling techniques are summarized,such as neutralization line,decoupling network,common/differential mode cancellation,etc.Finally,new decoupling research developments of modal currents cancellation based on the theory of characteristic mode are elaborated.Two design examples are shown to validate the benefit of the proposed decoupling method.The-6 dB impedance bandwidth of the head-to-head antenna pair can cover 3.4–3.6 GHz.The isolation level is improved from more than 3.7 dB to more than 13 dB.The tail-to-tail antenna pair can cover 3.3–5 GHz and the isolation level is improved from more than 5 dB to more than 10 dB in the whole operating bandwidth.Both antenna pairs can achieve an envelop correlation coefficient of less than 0.2 and the antenna efficiencies are more than 40%. | CHU Qingxin ZENG Wenfeng | 2022 | Chinese Journal of Electronics2022,31,6: | 0 |

| 9 | Identification of the core promoter of STK11 gene and its transcriptional regulation by p53显示文摘Peutz-Jeghers syndrome (PJS) is an autosomal dominant disease characterized by mucocutaneous pigmentation and hamartomatous polyps. Most cases of PJS involve the inactivation of germline mutations in the serine/threonine kinase gene STK11 which is also known as LKB1. The function of STK11 was previously linked to the tumor suppressor p53 and was shown to activate the p53 target p21/ WAF1. Recently, STK11 was reported to be interacting with p53 physically in the nucleus and it can directly or indirectly phosphorylate p53. Here we characterized the 50-flanking region of human STK11 gene and identified a 161-bp fragment with promoter activity. Sequence analysis, mutagenesis and gel shift studies revealed a binding site of Sp1 and p53, which affects the promoter activity. Mutation analyses showed that this fragment was required for p53-mediated transcriptional activation. This transcriptional activation was further confirmed by real-time quantitative RT-PCR and Western blot analysis. Transient transfection of p53 expression plasmid into fetal liver cell lines increased STK11 mRNA and protein levels.
