| 1 | Polycyclic aromatic hydrocarbons in soils of Beijing and Tianjin region: Vertical distribution, correlation with TOC and transport mechanism显示文摘The contents and distribution of 20 polycyclic aromatic hydrocarbons (PAHs) and heterocyclic aromatic hydrocarbons (HAHs) were investigated in 16 soil profiles of Beijing and Tianjin region. Transport of high molecular weight PAHs (HMWPAHs) and the correlation between total organic carbon (TOC) and their concentrations were also discussed. The results indicated that highly contaminated sites were located at urban or wastewater irrigation areas and pollutants mainly accumulated in topsoil (< 40 cm), with a sharp content decrease at the vertical boundary of 30-40 cm. Total PAHs/HAHs concentrations in soils from Tianjin were markedly greater than those from Beijing. Even the contents at bottoms of soil profiles in Tianjin were higher than those in topsoils of Beijing soil profile. HMWPAHs dominated the PAH profiles, exhibiting a uniform distribution of pyrogenic origin between topsoils and deep layers. Furthermore, the percentages of HMWPAHs remained relative constant with the depth of soil profiles, which were consistent with the distribution of particulate matter-associated PAHs in the local atmospheric environments. | 3 | Determination of HCH and DDT in soils following Wenchuan 5.12 Violent Earthquake in China显示文摘A large number of disinfectants and insecticides were used on a large scale after the Wenchuan 5.12 Violent Earthquake. The disinfectants and insecticides, including HCH (hexachlorocyclohexane) and DDT (dichlorodiphenyltrichoroethane) as representative OCPs, could lead to serious environmental problems. In order to disclose the residues of HCH and DDT in soils following the Wenchuan 5.12 Violent Earthquake, surface soils in Dujiangyan City and Beichuan County were sampled in 2009. The samples were analyzed by GC/MS after pre- treatment by ultrasonic solvent extraction. It has been found that the concentra-tions of γ-HCH, β-HCH and 4,4'-DDT in soils of Beichuan County were 94–102.4, 14.1–36.1 and 2.9–4.3 times high their concentrations in those of the Wolong Nature Reserve, and the concentrations in soils of Dujiangyan City were 52.4–61.6, 8.8 –45.3 and 5.4–6.2 times high their concentrations in soils of the Wolong Nature Reserve, respectively. | 7 | Rapid accuracy determining DNA purity and concentration in heavy oils by spectrophotometry methods显示文摘DNA analysis is the core of biotechnology applied in petroleum resources and engineering. Traditionally accurate determination of DNA purity and concentration by spectrometer is the first and critical step for downstream molecular biology research. In this study, three different spectrophotometry methods, BPM, NDTT and NPMTTZ were compared for their performance in determining DNA concentration and purity in 32 oil samples, and molecule methods like quantitative real-time PCR (qPCR) and high-throughput sequence were also performed to help assess the accuracy of the three methods in determining DNA concentration and purity. For ordinary heavy oil (OHO), extra heavy oil (EHO) and super heavy oil (SHO), the characteristics of high viscosity (η), density (ρ) and resin plus asphaltene content will affect the DNA extraction and UV determination. The DNA concentration was decreased as density increased: OHO (11.46 ± 18.34 ng/μL), EHO (6.68 ± 9.67 ng/μL) and SHO (6.20 ± 7.83 ng/μL), and the DNA purity was on the reverse: OHO (1.31 ± 0.27), EHO (1.54 ± 0.20), and SHO (1.83 ± 0.32).
