1Signaling pathways in cancer-associated fibroblasts and targeted therapy for cancer显示文摘To flourish,cancers greatly depend on their surrounding tumor microenvironment(TME),and cancer-associated fibroblasts(CAFs)in TME are critical for cancer occurrence and progression because of their versatile roles in extracellular matrix remodeling,maintenance of sternness,blood vessel formation,modulation of tumor metabolism,immune response,and promotion of cancer cell proliferation,migration,invasion,and therapeutic resistance.CAFs are highly heterogeneous stromal cells and their crosstalk with cancer cells is mediated by a complex and intricate signaling network consisting of transforming growth factor-beta,phosphoinositide 3-kinase/AKT/mammalian target of rapamycin,mitogen-activated protein kinase,Wnt,Janus kinase/signal transducers and activators of transcription,epidermal growth factor receptor,Hippo,and nuclear factor kappa-light-chain-enhancer of activated B cells,etc.,signaling pathways.These signals in CAFs exhibit their own special characteristics during the cancer progression and have the potential to be targeted for anticancer therapy.Therefore,a comprehensive understanding of these signaling cascades in interactions between cancer cells and CAFs is necessary to fully realize the pivotal roles of CAFs in cancers.Herein,in this review,we will summarize the enormous amounts of findings on the signals mediating crosstalk of CAFs with cancer cells and its related targets or trials.Further,we hypothesize three potential targeting strategies,including,namely,epithelial-mesenchymal common targets,sequential target perturbation,and crosstalk-directed signaling targets,paving the way for CAF-directed or host cell-directed antitumor therapy.Fanglong Wu Jin Yang Junjiang Liu Ye Wang Jingtian Mu Qingxiang Zeng Shuzhi Deng Hongmei Zhou 2021Signal Transduction and Targeted Therapy2021,6,7:9
2Water-use efficiency in response to simulated increasing precipitation in a temperate desert ecosystem of Xinjiang, China显示文摘Water-use efficiency(WUE) is a key plant functional trait that plays a central role in the global cycles of water and carbon. Although increasing precipitation may cause vegetation changes, few studies have explored the linkage between alteration in vegetation and WUE. Here, we analyzed the responses of leaf WUE, ecosystem carbon and water exchanges, ecosystem WUE, and plant community composition changes under normal conditions and also under extra 15% or 30% increases in annual precipitation in a temperate desert ecosystem of Xinjiang, China. We found that leaf WUE and ecosystem WUE showed inconsistent responses to increasing precipitation. Leaf WUE consistently decreased as precipitation increased. By contrast, the responses of the ecosystem WUE to increasing precipitation are different in different precipitation regimes: increasing by 33.9% in the wet year(i.e., the normal precipitation years)and decreasing by 4.1% in the dry year when the precipitation was about 30% less than that in the wet year.We systematically assessed the herbaceous community dynamics, community composition, and vegetation coverage to explain the responses of ecosystem WUE, and found that the between-year discrepancy in ecosystem WUE was consistent with the extent to which plant biomass was stimulated by the increase in precipitation. Although there was no change in the relative significance of ephemerals in the plant community, its greater overall plant biomass drove an increased ecosystem WUE under the conditions of increasing precipitation in 2011. However, the slight increase in plant biomass exerted no significant effect on ecosystem WUE in 2012. 4Beam shaping with limited amplitude weight values for satellite active phased array antenna显示文摘To gain the tradeoff between lower sidelobe and higher power amplifiers efficiency,a transmitting beam shaping scheme with limited amplitude weight values for satellite active phased array antenna is presented. The scheme is implemented by a dual coding genetic algorithm(GA). Phase and amplitude of array weight vectors for beam shaping are encoded by real coding and finite length binary coding,respectively,which,maintaining accuracy of results,reduces the amplitude dynamic range and improves the efficiency of power amplifiers. The presented algorithm,compared with complex-coded GA,increases the convergence rate due to the search space's decrease. 7Rapid accuracy determining DNA purity and concentration in heavy oils by spectrophotometry methods显示文摘DNA analysis is the core of biotechnology applied in petroleum resources and engineering. Traditionally accurate determination of DNA purity and concentration by spectrometer is the first and critical step for downstream molecular biology research. In this study, three different spectrophotometry methods, BPM, NDTT and NPMTTZ were compared for their performance in determining DNA concentration and purity in 32 oil samples, and molecule methods like quantitative real-time PCR (qPCR) and high-throughput sequence were also performed to help assess the accuracy of the three methods in determining DNA concentration and purity. For ordinary heavy oil (OHO), extra heavy oil (EHO) and super heavy oil (SHO), the characteristics of high viscosity (η), density (ρ) and resin plus asphaltene content will affect the DNA extraction and UV determination. The DNA concentration was decreased as density increased: OHO (11.46 ± 18.34 ng/μL), EHO (6.68 ± 9.67 ng/μL) and SHO (6.20 ± 7.83 ng/μL), and the DNA purity was on the reverse: OHO (1.31 ± 0.27), EHO (1.54 ± 0.20), and SHO (1.83 ± 0.32). 8Preliminary Screening and Characterization of Microsatellite Markers in RAD-seq Data of Datnioides pulcher显示文摘[Objective]The paper was to develop the microsatellite markers of Datnioides pulcher,and to screen polymorphic SSR primers.[Method]D.pulcher was performed reduced-representation genome sequencing using RAD-seq(Restriction-site associated DNA sequencing).[Result]A total of 13308806 high-quality clean reads(HQ clean reads)lengths were obtained and 26359 simple sequence repeats(SSR)loci were detected in the obtained sequence by SSR search software,which consisted of 496 repeat motifs.The most common types of repeat were A/T in single base repeat sequence,and AC/GT and A G/GT repeat units in di-base repeat sequence.AGG/CCT,AGC/CTG and AGG/CCT were the dominant types in tri-base repeat sequence.In quantity,the di-base repeat type SSR loci were the largest(19492),accounting for 73.95%of the total SSR loci.Except for mono-nucleotide type,the repetition frequency of SSR loci was mainly ranged from 4 to 9,and the number of polymorphic SSR loci was 2783.About 20066 pairs of SSR primers were successfully designed with Primer 5.0.The success rate of primer design was 76.13%.According to the repetition type and repetition number in the sequencing data,100 di-to penta-base repetition type SSRs were randomly selected and validated by PCR and polyacrylamide gel in six D.pulcher samples.A total of 73 pairs of primers were amplified by electrophoresis,and 20 of them were polymorphic.[Conclusion]The developed SSR sequences can be used for genetic analysis,genetic map construction and molecular marker assisted breeding of D.pulcher and its related species.Qu Zhengwei Song Hongmei Wang Xuejie Liu Yi Mu Xidong Liu Chao Hu Yinchang 2020Animal Husbandry and Feed Science2020,12,2:0
9Identification of Fusarium wilt resistance gene SiRLK1 in Sesamum indicum L.显示文摘Sesame Fusarium wilt(SFW),caused by Fusarium oxysporum f.sp.sesami(Fos),is one of the most devastating diseases affecting sesame cultivation.Deciphering the genetic control of SFW resistance is pivotal for effective disease management in sesame.An inheritance study on a cross between the highly resistant variety Yuzhi 11 and the highly susceptible accession Sp1 using a Fos pathogenicity group 1 isolate indicated that resistance was conferred by a single dominant allele.The target locus was located in a 1.24 Mb interval on chromosome 3 using a combination of cross-population association mapping and bulked segregant analysis.Fine genetic mapping further narrowed the interval between 21,350 and 21,401 kb.The locus Sindi_0812400 was identified as the SFW resistance gene and officially designated SiRLK1.This gene encodes a specific malectin/receptor-like protein kinase with three putative tandem kinase domains and is considered a kinase fusion protein.Sequence analysis revealed that a high proportion(49.44%)of variants within the locus was located within the kinase domainⅢ,and several of which were evidently associated with the diversity in SFW response,indicating the critical role of kinase domainⅢin expression of disease resistance.These findings provide valuable information for further functional analysis of SFW resistance genes and marker-assisted resistance breeding in sesame.Yinghui Duan Wenwen Qu Shuxian Chang Ming Ju Cuiying Wang Cong Mu Hengchun Cao Guiting Li Qiuzhen Tian Qin Ma Zhanyou Zhang Haiyang Zhang Hongmei Miao 2024The Crop Journal2024,12,1:0
10Visualizing Wallerian degeneration in the corticospinal tract after sensorimotor cortex ischemia in mice显示文摘Stroke can cause Wallerian degeneration in regions outside of the brain,particularly in the corticospinal tract.To investigate the fate of major glial cells and axons within affected areas of the corticospinal tract following stroke,we induced photochemical infarction of the sensorimotor cortex leading to Wallerian degeneration along the full extent of the corticospinal tract.We first used a routine,sensitive marker of axonal injury,amyloid precursor protein,to examine Wallerian degeneration of the corticospinal tract.An antibody to amyloid precursor protein mapped exclusively to proximal axonal segments within the ischemic cortex,with no positive signal in distal parts of the corticospinal tract,at all time points.To improve visualization of Wallerian degeneration,we next utilized an orthograde virus that expresses green fluorescent protein to label the corticospinal tract and then quantitatively evaluated green fluorescent protein-expressing axons.Using this approach,we found that axonal degeneration began on day 3 post-stroke and was almost complete by 7 days after stroke.In addition,microglia mobilized and activated early,from day 7 after stroke,but did not maintain a phagocytic state over time.Meanwhile,astrocytes showed relatively delayed mobilization and a moderate response to Wallerian degeneration.Moreover,no anterograde degeneration of spinal anterior horn cells was observed in response to Wallerian degeneration of the corticospinal tract.In conclusion,our data provide evidence for dynamic,pathogenic spatiotemporal changes in major cellular components of the corticospinal tract during Wallerian degeneration.Jiao Mu Liufang Hao Zijue Wang Xuyang Fu Yusen Li Fei Hao Hongmei Duan Zhaoyang Yang Xiaoguang Li 2024Neural Regeneration Research2024,19,3:0
