Rheumatoid arthritis: pathological mechanisms and modern pharmacologic therapies显示文摘Rheumatoid arthritis(RA) is a chronic systemic autoimmune disease that primarily affects the lining of the synovial joints and is associated with progressive disability, premature death, and socioeconomic burdens. A better understanding of how the pathological mechanisms drive the deterioration of RA progress in individuals is urgently required in order to develop therapies that will effectively treat patients at each stage of the disease progress. Here we dissect the etiology and pathology at specific stages:(i) triggering,(ii) maturation,(iii) targeting, and(iv) fulminant stage, concomitant with hyperplastic synovium, cartilage damage, bone erosion, and systemic consequences. Modern pharmacologic therapies(including conventional, biological, and novel potential small molecule disease-modifying anti-rheumatic drugs) remain the mainstay of RA treatment and there has been significant progress toward achieving disease remission without joint deformity. Despite this, a significant proportion of RA patients do not effectively respond to the current therapies and thus new drugs are urgently required. This review discusses recent advances of our understanding of RA pathogenesis, disease modifying drugs, and provides perspectives on next generation therapeutics for RA.Qiang Guo Yuxiang Wang Dan Xu Johannes Nossent Nathan J.Pavlos Jiake Xu 2018Bone Research2018,6,2:71
Wnt/b-catenin signaling plays an ever-expanding role in stem cell self-renewal,tumorigenesis and cancer chemoresistance显示文摘Wnt signaling transduces evolutionarily conserved pathways which play important roles in initiating and regulating a diverse range of cellular activities, including cell proliferation, calcium homeostasis, and cell polarity. The role of Wnt signaling in controlling cell proliferation and stem cell self-renewal is primarily carried out through the canonical pathway, which is the best-characterized the multiple Wnt signaling branches. The past 10 years has seen a rapid expansion in our understanding of the complexity of this pathway,as many new components of Wnt signaling have been identified and linked to signaling regulation, stem cell functions, and adult tissue homeostasis.Additionally,a substantial body of evidence links Wnt signaling to tumorigenesis of cancer types and implicates it in the development of cancer drug resistance. Thus,a better understanding of the mechanisms by which dysregulation of Wnt signaling precedes the development and progression of human cancer may hasten the development of pathway inhibitors to augment current therapy. This review summarizes and synthesizes our current knowledge of the canonical Wnt pathway in development and disease. We begin with an overview of the components of the canonical Wnt signaling pathway and delve into the role this pathway has been shown to play in stemness, tumorigenesis, and cancer drug resistance.Ultimately,we hope to present an organized collection of evidence implicating Wnt signaling in tumorigenesis and chemoresistance to facilitate the pursuit of Wnt pathway modulators that may improve outcomes of cancers in which Wnt signaling contributes to aggressive disease and/or treatment resistance.Maryam K.Mohammed Connie Shao Jing Wang Qiang Wei Xin Wang Zachary Collier Shengli Tang Hao Liu Fugui Zhang Jiayi Huang Dan Guo Minpeng Lu Feng Liu Jianxiang Liu Chao Ma Lewis L.Shi Aravind Athiviraham Tong-Chuan He Michael J.Lee 2016Genes & Diseases2016,3,1:70

Optical vortices 30 years on:OAM manipulation from topological charge to multiple singularities显示文摘Thirty years ago,Coullet et al.proposed that a special optical field exists in laser cavities bearing some analogy with the superfluid vortex.Since then, optical vortices have been widely studied, inspired by the hydrodynamics sharing similar mathematics.Akin to a fluid vortex with a central flow singularity,an optical vortex beam has a phase singularity with a certain topological charge, giving rise to a hollow intensity distribution.Such a beam with helical phase fronts and orbital angular momentum reveals a subtle connection between macroscopic physical optics and microscopic quantum optics. These amazing properties provide a new understanding of a wide range of optical and physical phenomena, including twisting photons, spin–orbital interactions,Bose-Einstein condensates, etc.,while the associated technologies for manipulating optical vortices have become increasingly tunable and flexible.Hitherto, owing to these salient properties and optical manipulation technologies, tunable vortex beams have engendered tremendous advanced applications such as optical tweezers,high-order quantum entanglement, and nonlinear optics. This article reviews the recent progress in tunable vortex technologies along with their advanced applications.Yijie Shen Xuejiao Wang Zhenwei Xie Changjun Min Xing Fu Qiang Liu Mali Gong Xiaocong Yuan 2019Light(Science & Applications)2019,8,1:77
Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. Gene transfectionmediated by lipofectamine was used to transfectthe eukaryotic expression vector pCDNA3. 1-HBXof HBV x gene into human hepatocellularcarcinoma cell line HCC-9204 and NF-κB wasdetected.RESULTS NF-κB was widely expressed inhuman hepatocellular carcinoma tissues in atotal of 52 cases and its expression was relatedto the X protein of HBV. NF-KB was localizedboth in the cytoplasm and . The nuclei ofhepatocellular carcinoma cells in 11 cases whichwere positive for the X protein of HBV while in 41cases negative for the X protein of HBV, NF-Kbwas only localized in the cytoplasm ofhepatocellular carcinoma cells but translocatedto the nuclei of hepatocellular carcinoma cellsafter the eukaryotic expression vectorpCDNA3.1-HBX was transfected into HCC-9204cells.CONCLUSION This study strongly suggeststhat the nuclear factor NF-KB is widely expressedin hepatocellular carcinoma tissues in differentstyles according to the expression of the Xprotein of HBV. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. The X protein ofHBV can activate NF-κB to translocate into nucleiof hepatocellular carcinoma cells.Shuang Ping Guo~1 Wen Liang Wang~1 Yu Qiang Zhai~2 Yi Ling Zhao~1 ~1Department of Pathology,Xijing Hospital of the Fourth Military Medical University,Xi’an,China ~2Department of Urology, the Central Hospital of Xi’an,China 2001World Journal of Gastroenterology2001,7,3:55
OsWRKY03, a rice transcriptional activator that functions in defense signaling pathway upstream of OsNPR1显示文摘WRKY family proteins are a class of plant specific transcription factors that involve in many stress response pathways. It has been shown that one Arabidopsis WRKY protein, AtWRKY29/22, is activated by MAP kinase signaling cascade and confers resistance to both bacterial and fungal pathogens. However, little is known about the biological roles of WRKY proteins in rice. In this study, we investigated the expression patterns of rice AtWRKY29/22 homolog, OsWRKY03, under different conditions, and also its possible role involved in plant defense. Our results showed that OsWRKY03 was up-regulated by several defense signaling molecules or different treatments. Further analysis revealed that the expres- sion of OsWRKY03 was light dependent. Transcriptional activation activity of OsWRKY03 was also demonstrated by yeast functional assay. Transient expression of OsWRKY03-GFP fusion protein in onion epidermis cells showed that OsWRKY03 was a nuclear localized protein. OsNPR1 as well as several other pathogenesis-related genes, such as OsPR1b, phenylalanine ammonia-lyase (ZB8) and peroxidase (POX22.3), were induced in OsWRKY03-overexpressing transgenic plants. These results indicated that OsWRKY03 is located upstream of OsNPR1 as a transcriptional activator in salicylic acid (SA)-dependent or jasmonic acid (JA)-dependent defense signaling cascades.Xiao Qiang LIU Xian Quan BAI Qian QIAN Xiu Jie WANG Ming Sheng CHEN Cheng Cai CHU 2005Cell Research2005,15,8:55

Correlations Between Foliar Stable Carbon Isotope Composition and Environmental Factors in Desert Plant Reaumuria soongorica (Pall.) Maxim.显示文摘Leaves of 407 individuals of Reaumuria soongorica (Pall.) Maxim. collected from the major distribution areas were measured to investigate the distribution characteristics of the stable carbon isotope in this desert plant, as well as correlations between δ13C values and environmental factors. Results showed that δ13C values in R. soongorica ranged from-22.77‰. to-29.85‰. and that the mean δ13C value (-26.52‰)was higher than a previously reported δ13C value for a different desert ecosystem. This indicates that R.soongorica belongs to the C3 photosynthetic pathway and has higher water use efficiency than other species. The correlations between δ13C values and environmental factors demonstrated that the foliar δ13C values in R. soongorica increased significantly with decreasing mean annual precipitation and mean relative humidity, and decreased with decreasing duration of sunshine and evaporation. The spatial distribution trend of δ13C values in R. soongorica was not obvious and there was no significant correlation between the δ13C values and mean annual temperature. We conclude that different distribution trends in δ13C values for R. soongorica were likely caused by stomatal limitation rather than by nutrient-related changes in photosynthetic efficiency and that precipitation played an important role in the wide distribution range of R.soongorica. This pattern of δ13C values for R. soongorica reinforced that it is a super-xerophil in terms of its adaptive strategies to a desert environment.Jian-Ying MA Tuo CHEN Wei-Ya QIANG Gang WANG 2005Journal of Integrative Plant Biology2005,47,9:42

Satellite remote sensing of changes in NO_x emissions over China during 1996-2010显示文摘Satellite derived NO2 column data have been used to study Chinese national fossil fuel consumption and pollutant emissions.Based on NO 2 retrievals from two satellites (GOME and SCIAMACHY) for 1996-2010,we analyzed the characteristics and evolution of regional pollution related to NO x emissions in China.Satellite observations indicated that the highly polluted regions were expanding.Anthropogenic emission dominated areas have expanded from the east to central and western China, and new highly polluted regions have formed throughout the nation.Bottom-up emission estimates suggested a 133% increase in anthropogenic NOx emissions in East Central China during 1996 to 2010,which was lower than the 184% increase of the NO2 columns measured by the satellites. We found that growth rates of NOx emissions have slowed in Chinese megacities over recent years,in contrast to which, the NOx emissions were soaring in medium-sized cities, indicating that strict controls of NOx emissions from coal-fired facilities are required in China.ZHANG Qiang GENG GuanNan WANG SiWen RICHTER Andreas HE KeBin 2012Chinese Science Bulletin2012,57,22:43
The Association of Overweight and Obesity with Blood Pressure among Chinese Children and Adolescents显示文摘Objective To examine the association between obesity and high blood pressure (BP) in Chinese children and adolescents. Methods Body mass index (BMI) and blood pressure measurements of 197 191 children aged 7-17 years were obtained from a Chinese national survey in 2010. Obesity and high BP were defined according to the reference values for Chinese children. Adjusted odds ratios (ORs) and 95% confidence intervals (CIs) of different BMI categories for high BP, as well as the population attributable risk percent (PAR%), were also calculated. Results The prevalence of high BP was 16.1% for boys and 12.9% for girls in 2010. Overweight and obese children had a significantly higher prevalence of high BP than non-overweight children in both boys and girls in each age group. ORs (95% CI) for high BP were 4.1 (3.9, 4.4) in obese boys and 4.0 (3.7, 4.3) in obese girls. The overall PAR% for high BP due to overweight and obesity was 14.4%. Conclusion Overweight and obese children have a significantly higher risk of high BP than non-overweight children. Eliminating overweight and obesity could reduce 14.4% of high BP cases.DONG Bin MA Jun WANG Hai Jun WANG Zhi Qiang 2013Biomedical and Environmental Sciences2013,26,6:42
Prevalence of Atrial Fibrillation in China and Its Risk Factors显示文摘Objective To study the prevalence of atrial fibrillation(AF)and the relation with its risk factors in China. Methods A total of 19 363 participants(8635 males and 10 728 females) aged ≥35 years in geographically dispersed urban and rural regions of China were included in this cross-sectional survey. All participants received questionnaire, physical and blood examination.Echocardiography were performed for AF patients found in the survey. Results Of the 19 363 participants,199 were diagnosed with AF.The estimated age-standardized prevalence of AF was 0.78% in men and 0.76% in women. The prevalence of AF in participants aged <60 years was 0.41% in men and 0.43% in women, and was 1.83% in both men and women aged ≥60 years.About 19.0% of males and 30.9% of females with AF were diagnosed with valve disease.Age-and sex-adjusted multivariable logistic regression analysis revealed that myocardial infarction, left ventricular hypertrophy(LVH),obesity, and alcohol consumption were associated with a increased risk of AF(P<0.05).Conclusion The age standardized prevalence of AF is 0.77% in the participants enrolled in the present study. The number of AF cases aged ≥35 years is 5.26 million according to 2010 Chinese Census. Most risk factors for AF,identified mainly in Western countries, are also detected in China.LI Ying WU Yang Feng CHEN Ke Ping LI Xian ZHANG Xing XIE Gao Qiang WANG Fang Zheng ZHANG Shu 2013Biomedical and Environmental Sciences2013,26,9:48

Enhanced secondary pollution offset reduction of primary emissions during COVID-19 lockdown in China显示文摘To control the spread of the 2019 novel coronavirus(COVID-19),China imposed nationwide restrictions on the movement of its population(lockdown)after the Chinese New Year of 2020,leading to large reductions in economic activities and associated emissions.Despite such large decreases in primary pollution, there were nonetheless several periods of heavy haze pollution in eastern China, raising questions about the well-established relationship between human activities and air quality. Here, using comprehensive measurements and modeling,we show that the haze during the COVID lockdown was driven by enhancements of secondary pollution. In particular, large decreases in NOx emissions from transportation increased ozone and nighttime NO_(3)radical formation, and these increases in atmospheric oxidizing capacity in turn facilitated the formation of secondary particulate matter. Our results, afforded by the tragic natural experiment of the COVID-19 pandemic, indicate that haze mitigation depends upon a coordinated and balanced strategy for controlling multiple pollutants.Xin Huang Aijun Ding Jian Gao Bo Zheng Derong Zhou Ximeng Qi Rong Tang Jiaping Wang Chuanhua Ren Wei Nie Xuguang Chi Zheng Xu Liangduo Chen Yuanyuan Li Fei Che Nini Pang Haikun Wang Dan Tong Wei Qin Wei Cheng Weijing Liu Qingyan Fu Baoxian Liu Fahe Chai Steven JDavis Qiang Zhang Kebin He 2021National Science Review2021,8,2:47
