| 1 | Inhibition of CDK9 by voruciclib synergistically enhances cell death induced by the Bcl-2 selective inhibitor venetoclax in preclinical models of acute myeloid leukemia显示文摘Venetoclax,an FDA-approved Bcl-2 selective inhibitor for the treatment of chronic lymphocytic leukemia and acute myeloid leukemia(AML),is tolerated well in elderly patients with AML and has good overall response rates;however,resistance remains a concern.In this study,we show that targeting CDK9 with voruciclib in combination with venetoclax results in synergistic antileukemic activity against AML cell lines and primary patient samples.CDK9 inhibition enhances venetoclax activity through downregulation of Mcl-1 and c-Myc.However,downregulation of Mcl-1 is transient,which necessitates an intermittent treatment schedule to allow for repeated downregulation of Mcl-1.Accordingly,an every other day schedule of the CDK9 inhibitor is effective in vitro and in vivo in enhancing the efficacy of venetoclax.Our preclinical data provide a rationale for an intermittent drug administration schedule for the clinical evaluation of the combination treatment for AML. | Daniel A.Luedtke Yongwei Su Jun Ma Xinyu Li Steven ABuck Holly Edwards Lisa Polin Juiwanna Kushner Sijana HDzinic Kathryn White Hai Lin Jeffrey WTaub Yubin Ge | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3 |

| 5 | Surface storm flow prediction on hillslopes based on topography and hydrologic connectivity显示文摘Background:Hillslopes provide critical watershed ecosystem services such as soil erosion control and storm flow regulation through collecting,storing,and releasing rain water.During intense rainstorms,rainfall intensity and infiltration capacity on the hillslope control Hortonian runoff while the topographic attributes of the hillslope(e.g.,slope,aspect,curvature)and the channel network define the structural hydraulic connectivity that determines how rapidly excess water is transferred.This paper discusses literature on the link between topographic attributes and hydrologic connectivity and demonstrates how this link can be used to define a parsimonious model for predicting surface runoff during high intensity rainfall.Main text:First,we provide a topographic characterization of the hillslope necessary to determine the structural hydrologic connectivity of surface flow based on existing literature.Subsequently,we demonstrate a hydrologic surface response model that routes the geomorphologic unit hydrograph(GIUH)through a spatial domain of representative elementary hillslopes reflecting the structural hydrologic connectivity.Topographic attributes impact flow and travel time distributions by affecting gravitational acceleration of overland flow and channel,solar irradiance,flow deceleration by vegetation,and flow divergence/convergence.Conclusions:We show with an example where we apply the GIUH-based model to hypothetical hillslopes that the spatial organization of the channel network is critical in the flow and travel time distribution,and that topographic attributes are key in obtaining simple yet accurate representations of hydrologic connectivity.Parsimonious GIUH models of surface runoff that use this hydrologic connectivity have the advantage of low data requirements,being scalable and applicable regardless of the spatial complexity of the hillslope,and have the potential to fundamentally improve flood forecasting tools used in the assessment of ecosystem services. | Dennis W.Hallema Roger Moussa Ge Sun Steven G.McNulty | 2016 | Ecological Processes2016,5,1: | 1 |

| 12 | Elimination of the yellow pigment gene PSY-E2 tightly linked to the Fusarium head blight resistance gene Fhb7 from Thinopyrum ponticum显示文摘Fhb7 is a major gene that was transferred from Thinopyrum ponticum to chromosome 7D of wheat(Triticum aestivum)and confers resistance to both Fusarium head blight(FHB)and Fusarium crown rot(FCR).However,Fhb7 is tightly linked to the PSY-E2 gene,which causes yellow flour,limiting its application in breeding.To break this linkage,marker K-PSY was developed for tagging PSY-E2 and used with Fhb7 markers to identify recombination between the two genes.Screening 21,000 BC1F2 backcross progeny(Chinese Spring ph1bph1b*2/SDAU 2028)revealed two Fhb7^(+)wheat-Tp7el_(2)L lines,Shannong 2–16and Shannong 16–1,that carry a desired truncated Fhb7^(+)translocation segment without PSY-E2.The two lines show levels of resistance to FHB and FCR similar to those of the original translocation line SDAU 2028,but have white flour.To facilitate Fhb7 use in wheat breeding,STS markers were developed and used to isolate Fhb7 on a truncated Tp7el_(2) translocation segment.Near-isogenic lines carrying the Fhb7^(+)segment were generated in the backgrounds of three commercial cultivars,and Fhb7^(+)lines showed increased FHB and FCR resistance without yield penalty.The breakage of the tight linkage between Fhb7 and PSY-E2 via homoeologous recombination provides genetic resources for improvement of wheat resistance to FHB and FCR and permit the large-scale deployment of Fhb7 in breeding using marker-assisted selection. | Xuefeng Li Dong Li Yu Xuan Ziming He Lanfei Zhao Yongchao Hao Wenyang Ge Shoushen Xu Bingqian Hou Biao Wang Jun Guo Wenwen Liu Mingzhu Li Yi Han Cunyao Bo Yinguang Bao Zengjun Qi Steven S.Xu Guihua Bai Hongwei Wang Lingrang Kong | 2023 | The Crop Journal2023,11,3: | 0 |

These include flaring stars, supernova remnants, accreting white dwarfs, low and high mass X-ray binaries, radio quiet and radio loud active galactic nuclei, tidal disruption events, and gamma-ray bursts. eXTP will be excellently suited to study one common aspect of these objects: their often transient nature.
