| 1 | Effects of high temperature on the microstructure and mechanical behavior of hard coal显示文摘Coal is a common porous sedimentary rock whose microstructure and mechanical behavior are often affected by high temperature.In order to deeply investigate the effect of high temperature on the structure and mechanical properties of coal,six groups of coal samples with different heating temperatures were prepared.Various testing methods,including ultrasonic test,scanning electron microscopy,nuclear magnetic resonance and uniaxial compression test,were used in this study.The results showed that the physical and chemical change processes of coal samples under high temperature can be divided into two stages:the stage of drying and degassing and the stage of pyrolysis of the coal matrix.With the increase of heating temperature,the total porosity of coal samples increases,while the integrity and the deformation resistance of coal samples decrease.In addition,the elastic modulus and uniaxial compressive strength of the coal sample decrease with the increase of heating temperature.The Poisson’s ratio decreases firstly and then increases with the increase of heating temperature. | Chengdong Su Jiadong Qiu Qiuhong Wu Lei Weng | 2020 | International Journal of Mining Science and Technology2020,30,5: | 11 |
| 2 | Wheat powdery mildew resistance gene Pm64 derived from wild emmer (Triticum turgidum var.dicoccoides) is tightly linked in repulsion with stripe rust resistance gene Yr5显示文摘Stripe rust and powdery mildew are both devastating diseases for durum and common wheat.Pyramiding of genes conferring resistance to one or more diseases in a single cultivar is an important breeding approach to provide broader spectra of resistances in wheat improvement. A new powdery mildew resistance gene originating from wild emmer(Triticum turgidum var.dicoccoides) backcrossed into common wheat(T. aestivum) line WE35 was identified. It conferred an intermediate level of resistance to Blumeria graminis f. sp. tritici isolate E09 at the seedling stage and a high level of resistance at the adult plant stage. Genetic analysis showed that the powdery mildew resistance in WE35 was controlled by a dominant gene designated Pm64. Bulked segregant analysis(BSA) and molecular mapping indicated that Pm64 was located in chromosome bin 2 BL4-0.50–0.89. Polymorphic markers were developed from the corresponding genomic regions of Chinese Spring wheat and wild emmer accession Zavitan to delimit Pm64 to a 0.55 cM genetic interval between markers WGGBH1364 and WGGBH612, corresponding to a 15 Mb genomic region on Chinese Spring and Zavitan 2 BL, respectively. The genetic linkage map of Pm64 is critical for fine mapping and cloning. Pm64 was completely linked in repulsion with stripe rust resistance gene Yr5. | 3 | Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). | 4 | Mapping stripe rust resistance genes by BSR-Seq:YrMM58 and YrHY1 on chromosome 2AS in Chinese wheat lines Mengmai 58 and Huaiyang 1 are Yr17显示文摘Stripe rust(yellow rust), caused by Puccinia striiformis f. sp. tritici(PST),is one of the most devastating fungal diseases in common wheat(Triticum aestivum L.) in China and worldwide. Resistance breeding is the most effective strategy to control diseases in crop plants. Chinese wheat lines Mengmai 58 and Huaiyang 1 are highly resistant to PST race CYR34(V26) at the adult plant stage. To genetically map the underlying resistance genes we developed segregating populations by crossing Mengmai 58 and Huaiyang 1 with the susceptible cultivar Nongda 399. The stripe rust resistances in Mengmai 58 and Huaiyang 1 were both controlled by single dominant genes, provisionally designated YrMM58 and YrHY1, respectively. Bulked segregant RNA-Seq(BSR-Seq) analysis showed that YrMM58 and YrHY1 were located in the same distal ~16 Mb region on chromosome 2 AS.Comparative genomics analysis with the physical map of Aegilops tauschii proved useful for developing additional markers to saturate the genetic linkage map. YrMM58 and YrHY1 were mapped to the distal end of chromosome arm 2 AS, with the closest marker WGGB148 being 7.7 cM and 3.8 cM from the resistance gene, which was considered to be Yr17. | 5 | Mapping a leaf senescence gene els1 by BSR-Seq in common wheat显示文摘Leaf senescence is normally the last stage of plant development. Early senescence of functional leaves significantly reduces the photosynthetic time and efficiency, seriously affecting grain yield and quality in wheat. Discovering genes responsible for early leaf senescence(els) are necessary for developing novel germplasms and cultivars with delayed leaf-senescence through molecular manipulation and marker assisted selection. In this study, we identified an early leaf senescence line M114 in a derivative of a wheat breeding population. Genetic analysis indicated that early leaf senescence in M114 is controlled by a single recessive gene, provisionally designated els1. By applying bulked segregant analysis and RNA-Seq(BSR-Seq), seven polymorphic markers linked to els1 were developed and the gene was located on chromosome arm 2 BS in a 1.5 c M genetic interval between markers WGGB303 and WGGB305. | 7 | Mapping of wheat stripe rust resistance gene Yr041133 by BSR-Seq analysis显示文摘Puccinia striiformis Westend. f. sp. tritici(Pst) pathotype CYR34 is widely virulent and prevalent in China.Here, we report identification of a strpie rust resistance(Yr) gene, designated Yr041133, in winter wheat line 041133. This line produced a hypersensitive reaction to CYR34 and conferred resistance to 13 other pathotypes. Resistance to CYR34 in line 041133 was controlled by a single dominant gene. Bulked segregant RNA sequencing(BSR-Seq) was performed on a pair of RNA bulks generated by pooling resistant and susceptible recombinant inbred lines. Yr041133 was mapped to a 1.7 c M genetic interval on the chromosome arm 7 BL that corresponded to a 0.8 Mb physical interval(608.9–609.7 Mb) in the Chinese Spring reference genome. | 19 | Comparative effects of enzymatic soybean,fish meal and milk powder in diets on growth performance,immunological parameters,SCFAs production and gut microbiome of weaned piglets显示文摘Background:The objective of this study was to evaluate the replacement effects of milk powder(MK)and fish meal(FM)by enzymatic soybean(ESB)in diets on growth performance,immunological parameters,SCFAs production and gut microbiome of weaned piglets.Methods:A total of 128 piglets with initial body weight at 6.95±0.46 kg,were randomly assigned into 4 dietary treatments with 8 replicates per treatment and 4 piglets per replicate for a period of 14 d.Piglets were offered isonitrogenous and iso-energetic diets as follows:CON diet with MK and FM as high quality protein sources,ESB plus FM diet with ESB replacing MK,ESB plus MK diet with ESB replacing FM,and ESB diet with ESB replacing both MK and FM.Results:No significant differences were observed in growth performance among all treatments(P>0.05).However,piglets fed ESB plus FM or ESB diet had increased diarrhea index(P<0.01),and lower digestibility of dry matter(DM),gross energy(GE)or crude protein(CP),relative to piglets fed CON diet(P<0.01).Moreover,the inclusion of ESB in diet markedly decreased the plasma concentration of HPT and fecal concentration of butyric acid(BA)(P<0.01).The High-throughput sequencing of 16S rRNA gene V3−V4 region of gut microbiome revealed that the inclusion of ESB in diet increased the alpha diversity,and the linear discriminant analysis effect size(LEfSe)showed that piglets fed with ESB plus FM or ESB diet contained more gut pathogenic bacteria,such as g_Peptococcus,g_Veillonella and g_Helicobacter.Conclusion:The inclusion of ESB in diet did not markedly affect growth performance of piglets,but the replacement of MK or both MK and FM by ESB increased diarrhea index,which could be associated with lower nutrients digestibility and more gut pathogenic bacteria.However,piglets fed diet using ESB to replace FM did not markedly affect gut health-related parameters,indicating the potential for replacing FM with ESB in weaning diet. | Yingjie Li Yang Liu Jiangnan Wu Qiuhong Chen Qiang Zhou Fali Wu Ruinan Zhang Zhengfeng Fang Yan Lin Shengyu Xu Bin Feng Yong Zhuo De Wu Lianqiang Che | 2022 | Journal of Animal Science and Biotechnology2022,13,2: | 0 |
