1Sequence framework of two different kinds of margins and their response to tectonic activity during the Middle-Late Triassic, Ordos Basin显示文摘Two kinds of margin respectively occur in the Ordos Basin during the Middle-Late Triassic (Yanchang Age), one is foreland margin developed under the background of flexural subsidence by thrusting intensively in the southwest margin, and the other is intracratonic basin margin by stable subsidence in northern and central parts of the basin. The Middle-Late Triassic Yanchang Formation can be divided into four regional third-order sequences, which are separated by gentle angular unconformity or regional erosion surface, made up of lowstand system tract (LST), expanding system tract (EST) and highstand system tract (HST) from lower to upper within a sequence. But there are distinct differences of the sequence framework between the southwest margin and northern and central parts of the basin. The southwest margin develops heavy conglomerate layer and unconformity as a result of orogeny by thrusting, and the intracratonic basin margin by stable subsidence in the northern and central parts grows aggradational sandstone, conglomerate in fluvio-delta system and parallel unconformity. The depositional framework of southwest margin reflects the tectonic evolution from flexural subsidence by thrusting to rebounded uplift. The formation of sequence boundary is related to the resilient uplift and erosion. The sequence stratigraphic framework and depositional system tract configuration in the foreland basin are controlled by structural activity of the fold and thrust belt, and the sequence succession reflects episodic thrusting of the Middle-Late Triassic toward the foreland basin. The sequence evolution in northern and central parts reflects the depositional succession of fluvio-delta system under intracratonic background, composed of coarse-grained sediment in braided channel deposit at the lower, meandering channel deposit in the middle and fine-grained sediment in the flood plain at the upper, dominated by lake level fluctuation. 3NUMERICAL SOLUTION OF FLUID-STRUCTURE INTERACTION IN LIQUID-FILLED PIPES BY METHOD OF CHARACTERISTICS显示文摘Fluid-structure interaction (FSI) is essentially a dynamic phenomenon and always exists in fluid-filled pipe system. The four-equation model, which has been proved to be effective to describe and predict the phenomenon of FSI due to friction coupling and Poisson coupling being taken into account, is utilized to describe the FSI of fluid-filled pipe system. Terse compatibility equations are educed by the method of characteristics (MOC) to describe the fluid-filled pipe system. To shorten computing time needed to get the solutions under the condition of keeping accuracy requirement, two steps are adopted, firstly the time step Δt and divided number of the straight pipe are optimized, sec-ondly the mesh spacing Δz close to boundary is subdivided in several submeshes automatically ac-cording to the speed gradient of fluid. 5Preparation of ZrB_(2)-MoSi_(2) high oxygen resistant coating using nonequilibrium state powders by self-propagating high-temperature synthesis显示文摘To achieve high oxygen blocking structure of the ZrB_(2)-MoSi_(2) coating applied on carbon structural material,ZrB_(2)-MoSi_(2) coating was prepared by spark plasma sintering(SPS)method utilizing ZrB_(2)-MoSi_(2) composite powders synthesized by self-propagating high-temperature synthesis(SHS)technique as raw materials.The oxygen blocking mechanism of the ZrB_(2)-MoSi_(2) coatings at 1973 K was investigated.Compared with commercial powders,the coatings prepared by SHS powders exhibited superior density and inferior oxidation activity,which significantly heightened the structural oxygen blocking ability of the coatings in the active oxidation stage,thus characterizing higher oxidation protection efficiency.The rise of MoSi_(2) content facilitated the dispersion of transition metal oxide nanocrystals(5-20 nm)in the SiO_(2) glass layer and conduced to the increasing viscosity,thus strengthening the inerting impact of the compound glass layer in the inert oxidation stage.Nevertheless,the ZrB_(2)-40 vol% MoSi_(2) coating sample prepared by SHS powders presented the lowest oxygen permeability of 0.3% and carbon loss rate of 0.29×10^(6)g·cm^(-2)·s^(-1).Owing to the gradient oxygen partial pressure inside the coatings,the Si-depleted layer was developed under the compound glass layer,which brought about acute oxygen erosion.Menglin ZHANG Xuanru REN Mingcheng ZHANG Songsong WANG Li WANG Qingqing YANG Hongao CHU Peizhong FENG 2021Journal of Advanced Ceramics2021,10,5:2
6Signaling pathways in rheumatoid arthritis:implications for targeted therapy显示文摘Rheumatoid arthritis(RA)is an incurable systemic autoimmune disease.Disease progression leads to joint deformity and associated loss of function,which significantly impacts the quality of life for sufferers and adds to losses in the labor force.In the past few decades,RA has attracted increased attention from researchers,the abnormal signaling pathways in RA are a very important research field in the diagnosis and treatment of RA,which provides important evidence for understanding this complex disease and developing novel RA-linked intervention targets.The current review intends to provide a comprehensive overview of RA,including a general introduction to the disease,historical events,epidemiology,risk factors,and pathological process,highlight the primary research progress of the disease and various signaling pathways and molecular mechanisms,including genetic factors,epigenetic factors,summarize the most recent developments in identifying novel signaling pathways in RA and new inhibitors for treating RA.therapeutic interventions including approved drugs,clinical drugs,pre-clinical drugs,and cutting-edge therapeutic technologies.These developments will hopefully drive progress in new strategically targeted therapies and hope to provide novel ideas for RA treatment options in the future.Qian Ding Wei Hu Ran Wang Qinyan Yang Menglin Zhu Meng Li Jianghong Cai Peter Rose Jianchun Mao Yi Zhun Zhu 2023Signal Transduction and Targeted Therapy2023,8,3:2

10Relaxin-encapsulated polymeric metformin nanoparticles remodel tumor immune microenvironment by reducing CAFs for efficient triple-negative breast cancer immunotherapy显示文摘Cancer-associated fibroblasts(CAFs)are one of the most abundant stromal cells in the tumor microenvironment which mediate desmoplastic response and are the primary driver for an immunosuppressive microenvironment,leading to the failure of triple-negative breast cancer(TNBC)immunotherapy.Therefore,depleting CAFs may enhance the effect of immunotherapy(such as PD-L1 antibody).Relaxin(RLN)has been demonstrated to significantly improve transforming growth factor-β(TGF-β)induced CAFs activation and tumor immunosuppressive microenvironment.However,the short half-life and systemic vasodilation of RLN limit its in vivo efficacy.Here,plasmid encoding relaxin(pRLN)to locally express RLN was delivered with a new positively charged polymer named polymeric metformin(PolyMet),which could increase gene transfer efficiency significantly and have low toxicity that have been certified by our lab before.In order to improve the stability of pRLN in vivo,this complex was further formed lipid poly-γ-glutamic acid(PGA)/PolyMetpRLN nanoparticle(LPPR).The particle size of LPPR was 205.5±2.9 nm,and the zeta potential was+55.4±1.6 mV.LPPR displayed excellent tumor penetrating efficacy and weaken proliferation of CAFs in 4T1luc/CAFs tumor spheres in vitro.In vivo,it could reverse aberrantly activated CAFs by decreasing the expression of profibrogenic cytokine and remove the physical barrier to reshape the tumor stromal microenvironment,which enabled a 2.2-fold increase in cytotoxic T cell infiltration within the tumor and a decrease in immunosuppressive cells infiltration.Thus,LPPR was observed retarded tumor growth by itself in the 4T1 tumor bearing-mouse,and the reshaped immune microenvironment further led to facilitate antitumor effect when it combined with PD-L1 antibody(aPD-L1).Altogether,this study presented a novel therapeutic approach against tumor stroma using LPPR to achieve a combination regimen with immune checkpoint blockade therapy against the desmoplastic TNBC model.Hongyan Zhang Liying Chen Yue Zhao Ningchao Luo Jingbin Shi Shujun Xu Lisha Ma Menglin Wang Mancang Gu Chaofeng Mu Yang Xiong 2023Asian Journal of Pharmaceutical Sciences2023,18,2:1
11Progranulin regulation of autophagy contributes to its chondroprotective effect in osteoarthritis显示文摘Progranulin(PGRN)is a multifunctional growth factor involved in many physiolog-ical processes and disease states.The apparent protective role of PGRN and the importance of chondrocyte autophagic function in the progression of osteoarthritis(OA)led us to investi-gate the role of PGRN in the regulation of chondrocyte autophagy.PGRN knockout chondro-cytes exhibited a deficient autophagic response with limited induction following rapamycin,serum starvation,and IL-1b-induced autophagy.PGRN-mediated anabolism and suppression of IL-1b-induced catabolism were largely abrogated in the presence of the BafA1 autophagy inhibitor.Mechanistically,during the process of OA,PGRN and the ATG5eATG12 conjugate form a protein complex;PGRN regulates autophagy in chondrocytes and OA through,at least partially,the interactions between PGRN and the ATG5eATG12 conjugate.Furthermore,the ATG5eATG12 conjugate is critical for cell proliferation and apoptosis.Knockdown or knockout of ATG5 reduces the expression of ATG5eATG12 conjugate and inhibits the chondroprotective effect of PGRN on anabolism and catabolism.Overexpression of PGRN partially reversed this effect.In brief,the PGRN-mediated regulation of chondrocyte autophagy plays a key role in the chondroprotective role of PGRN in OA.Such studies provide new insights into the pathogen-esis of OA and PGRN-associated autophagy in chondrocyte homeostasis.Yiming Pan Yuyou Yang Mengtian Fan Cheng Chen Rong Jiang Li Liang Menglin Xian Biao Kuang Nana Geng Naibo Feng Lin Deng Wei Zheng Fengmei Zhang Xiaoli Li Fengjin Guo 2023Genes & Diseases2023,10,4:0
12Blockage of EGFR/AKT and mevalonate pathways synergize the antitumor effect of temozolomide by reprogramming energy metabolism in glioblastoma显示文摘Background Metabolism reprogramming plays a vital role in glioblastoma(GBM)progression and recurrence by producing enough energy for highly proliferating tumor cells.In addition,metabolic reprogramming is crucial for tumor growth and immune-escape mechanisms.Epidermal growth factor receptor(EGFR)amplification and EGFR-vIII mutation are often detected in GBM cells,contributing to the malignant behavior.This study aimed to investigate the functional role of the EGFR pathway on fatty acid metabolism remodeling and energy generation.Methods Clinical GBM specimens were selected for single-cell RNA sequencing and untargeted metabolomics analysis.A metabolism-associated RTK-fatty acid-gene signature was constructed and verified.MK-2206 and MK-803 were utilized to block the RTK pathway and mevalonate pathway induced abnormal metabolism.Energy metabolism in GBM with activated EGFR pathway was monitored.The antitumor effect of Osimertinib and Atorvastatin assisted by temozolomide(TMZ)was analyzed by an intracranial tumor model in vivo.Results GBM with high EGFR expression had characteristics of lipid remodeling and maintaining high cholesterol levels,supported by the single-cell RNA sequencing and metabolomics of clinical GBM samples.Inhibition of the EGFR/AKT and mevalonate pathways could remodel energy metabolism by repressing the tricarboxylic acid cycle and modulating ATP production.Mechanistically,the EGFR/AKT pathway upregulated the expressions of acyl-CoA synthetase short-chain family member 3(ACSS3),acyl-CoA synthetase long-chain family member 3(ACSL3),and long-chain fatty acid elongation-related gene ELOVL fatty acid elongase 2(ELOVL2)in an NF-κB-dependent manner.Moreover,inhibition of the mevalonate pathway reduced the EGFR level on the cell membranes,thereby affecting the signal transduction of the EGFR/AKT pathway.Therefore,targeting the EGFR/AKT and mevalonate pathways enhanced the antitumor effect of TMZ in GBM cells and animal models.Conclusions Our findings not only uncovered the mechanism of metabolic reprogramming in EGFR-activated GBM but also provided a combinatorial therapeutic strategy for clinical GBM management.Xiaoteng Cui Jixing Zhao Guanzhang Li Chao Yang Shixue Yang Qi Zhan Junhu Zhou Yunfei Wang Menglin Xiao Biao Hong Kaikai Yi Fei Tong Yanli Tan Hu Wang Qixue Wang Tao Jiang Chuan Fang Chunsheng Kang 2023Cancer Communications2023,43,12:0
13Lactobacillus fermentum as a new inhibitor to control advanced glycation end-product formation during vinegar fermentation显示文摘The inhibitory activity of lactic acid bacteria(LAB)toward advanced glycation end-products(AGEs)during vinegar fermentation was studied,and its relationships with the substrate consumption,antioxidant capacity,total phenolic content,total flavonoid compounds,α-glucosidase,andα-amylase activity inhibition were evaluated.The vinegar was made from rice powder flour by liquid-state fermentation(LSF).The selected LAB strains were separately co-cultivated with Saccharomyces cerevisiae and Acetobacter pasteurianus 1.41 in alcoholic and acetic acid fermentation,respectively.Among 3 strains,Lactobacillus fermentum showed the strongest inhibitory effect on the formation of total fluorescent AGEs and carboxymethyl lysine(CML)/carboxyethyl lysine(CEL)in the fermentation process.The corresponding mechanisms included the acceleration of substrate consumtion,improvement of antioxidant activities,and inhibition ofα-glucosidase andα-amylase.In addition,the fluorescent AGEs and the CML/CEL were negatively correlated with the antioxidant activities,while theα-glucosidase andα-amylase activities were positively correlated with the total phenols and total flavonoids.Moreover,the variety of main flavor compounds increased,including esters,alcohols,phenols and acids.The results of the study support the potential use of screened LAB strains to inhibit the formation of fluorescent AGEs,CML and CEL on fermented products and in the food processing industry,without associated risks to consumers.Qin Li Leibing Li Hanjian Zhu Fan Yang Ke Xiao Lin Zhang Menglin Zhang Yongsheng Peng Chao Wang Dongsheng Li Qian Wu Mengzhou Zhou 2022Food Science and Human Wellness2022,11,5:0

15Integrative Identification by Hi‑C Revealed Distinct Advanced Structural Variations in Lung Adenocarcinoma Tissue显示文摘Advanced three-dimensional structure variations of chromatin in large genome fragments,such as conversion of A/B compartment,topologically associated domains(TADs)and chromatin loops are related closely to occurrence of malignant tumors.However,the structural characteristics of lung cancer still remain uncovered.In this study,we used high-throughput chromosome(Hi-C)conformation capture technology to detect the advanced structural variations in chromatin of two nonsmoking lung adenocarcinoma(LUAD)tumor and paired normal tissues.The results indicate that significant chromatin variations are detected in tumor tissues compared with normal tissues.At compartment scale,the main conversion type of compartment is A→B in tumor tissues,which are concentrated mainly on chromosome 3(Chr3)(33.6%).A total of 216 tumor-specific TADs are identified in tumor tissues,which are distributed mainly in Chr1(19),Chr2(15)and Chr3(17).Forty-one distinct enhancer-promoter loops are observed in tumor tissue,which are associated closely to tumor-related pathways including mitogen-activated protein kinase(MAPK),Phosphatidylinositol-3-kinase-Protein kinase B(PI3K-AKT),Ras,Wnt and Ras1.The most important observation in this study is that we identify five important genes(SYT16,NCEH1,NXPE3,MB21D2,and DZIP1L),which are detected in both A→B compartment,TADs and chromatin loops in tumor samples,and four of these genes(NCEH1,NXPE3,MB21D2,and DZIP1L)locate on q arm of Chr3.Further gene expression and invasion experiment analysis show that NCEH1,MB21D2 and SYT16 are involved in the tumor development.Thus,we provide a comprehensive overview of advanced structures in LUAD for the first time and provide a basis for further research on the genetic variation of this tumor.Tingting Song Menglin Yao Ying Yang Zhiqiang Liu Li Zhang Weimin Li 2023Phenomics2023,3,4:0
16Haploidentical transplantation has a superior graft-versus-leukemia effect than HLA-matched sibling transplantation for Ph- high-risk B-cell acute lymphoblastic leukemia显示文摘Background: Compared with human leukocyte antigen (HLA)-matched sibling donor (MSD) transplantation, it remains unclear whether haploidentical donor (HID) transplantation has a superior graft-versus-leukemia (GVL) effect for Philadelphia-negative (Ph-) high-risk B-cell acute lymphoblastic leukemia (B-ALL). This study aimed to compare the GVL effect between HID and MSD transplantation for Ph- high-risk B-ALL.Methods: This study population came from two prospective multicenter trials (NCT01883180, NCT02673008). Immunosuppressant withdrawal and prophylactic or pre-emptive donor lymphocyte infusion (DLI) were administered in patients without active graft-versus-host disease (GVHD) to prevent relapse. All patients with measurable residual disease (MRD) positivity posttransplantation (post-MRD+) or non-remission (NR) pre-transplantation received prophylactic/pre-emptive interventions. The primary endpoint was the incidence of post-MRD+.Results: A total of 335 patients with Ph- high-risk B-ALL were enrolled, including 145 and 190, respectively, in the HID and MSD groups. The 3-year cumulative incidence of post-MRD+ was 27.2% (95% confidence interval [CI]: 20.2%-34.7%) and 42.6% (35.5%-49.6%) in the HID and MSD groups(P = 0.003), respectively. A total of 156 patients received DLI, including 60 (41.4%) and 96 (50.5%), respectively, in the HID and MSD groups (P= 0.096). The 3-year cumulative incidence of relapse was 18.6% (95% CI: 12.7%-25.4%) and 25.9% (19.9%-32.3%;P = 0.116) in the two groups, respectively. The 3-year overall survival (OS) was 67.4% (95% CI: 59.1%-74.4%) and 61.6% (54.2%-68.1%;P = 0.382), leukemia-free survival (LFS) was 63.4% (95% CI: 55.0%-70.7%) and 58.2% (50.8%-64.9%;P= 0.429), and GVHD-free/relapse-free survival (GRFS) was 51.7% (95% CI: 43.3%-59.5%) and 37.8% (30.9%-44.6%;P= 0.041), respectively, in the HID and MSD groups.Conclusion: HID transplantation has a lower incidence of post-MRD+ than MSD transplantation, suggesting that HID transplantation might have a superior GVL effect than MSD transplantation for Ph- high-risk B-ALL patients.Trial registration: ClinicalTrials.gov: NCT01883180, NCT02673008.Fan Menglin Wang Yu Lin Ren Lin Tong Huang Fen Fan Zhiping Xu Yajing Yang Ting Xu Na Shi Pengcheng Nie Danian Lin Dongjun Jiang Zujun Wang Shunqing Sun Jing Huang Xiaojun Liu Qifa Xuan Li 2022Chinese Medical Journal2022,,8:0
17Heat Pumps in Denmark:Current Situation in Providing Frequency Control Ancillary Services显示文摘-The Danish government set an ambitious goal to achieve a fully renewable-based energy system by 2050.In this context,the integrated electricity and heating system is undergoing rapid development in Denmark as a promising way to accommodate the ever-growing renewable energy sources(RESs).The electric heat pumps(HPs),coupled with the power and heat sectors,can propagate the flexibility on the heat consumer side to the power system operations,playing an important role of improving system flexibility and balancing the variability of the RES.In this paper,the current development situation of HPs in Denmark is analyzed,including both the large-scale HPs in the district heating system and individual HPs on the residential side.The possibility of using HPs to provide frequency control ancillary service(FCAS)is analyzed according to the market and technical requirements of the FCAS in the Danish transmission system and experimental results of representative demonstration projects of HPs.A comprehensive analysis of the advantages,barriers,future prospects,and challenges for using HPs to provide the FCAS are carried out from the perspectives of different entities.Menglin Zhang Qiuwei Wu Theis Bo Harild Rasmussen Xiaodong Yang Jinyu Wen 2022CSEE Journal of Power and Energy Systems2022,8,3:0
18The FAST all sky HI survey(FASHI):The first release of catalog显示文摘The FAST All Sky HI survey(FASHI)was designed to cover the entire sky observable by the Five-hundred-meter Aperture Spherical radio Telescope(FAST),spanning approximately 22000 square degrees of declination between-14°and+66°,and in the frequency range of 1050-1450 MHz,with the expectation of eventually detecting more than 100000 HI sources.Between August 2020 and June 2023,FASHI had covered more than 7600 square degrees,which is approximately 35%of the total sky observable by FAST.It has a median detection sensitivity of around 0.76 m Jy beam-1and a spectral line velocity resolution of~6.4 km s-1at a frequency of~1.4 GHz.As of now,a total of 41741 extragalactic HI sources have been detected in the frequency range 1305.5-1419.5 MHz,corresponding to a redshift limit of z■0.09.By cross-matching FASHI sources with the Siena Galaxy Atlas(SGA)and the Sloan Digital Sky Survey(SDSS)catalogs,we found that 16972(40.7%)sources have spectroscopic redshifts and 10975(26.3%)sources have only photometric redshifts.Most of the remaining 13794(33.0%)HI sources are located in the direction of the Galactic plane,making their optical counterparts difficult to identify due to high extinction or high contamination of Galactic stellar sources.Based on current survey results,the FASHI survey is an unprecedented blind extragalactic H I survey.It has higher spectral and spatial resolution and broader coverage than the Arecibo Legacy Fast ALFA Survey(ALFALFA).When completed,FASHI will provide the largest extragalactic HI catalog and an objective view of HI content and large-scale structure in the local universe.Chuan-Peng Zhang Ming Zhu Peng Jiang Cheng Cheng Jing Wang Jie Wang Jin-Long Xu Xiao-Lan Liu Nai-Ping Yu Lei Qian Haiyang Yu Mei Ai Yingjie Jing Chen Xu Ziming Liu Xin Guan Chun Sun Qingliang Yang Menglin Huang Qiaoli Hao FAST Collaboration 2024Science China(Physics,Mechanics & Astronomy)2024,67,1:0
19Self-assembled nanochaperones enable the disaggregation of amyloid insulin fibrils显示文摘The deposition of highly ordered amyloid fibrils is recognized as a hallmark of amyloidosis diseases such as Alzheimer’s disease and Parkinson’s disease.Disaggregating the amyloid fibrils is considered as one of the effective strategies for the control and treatment of amyloidosis diseases.In this article,by simulating the function of natural molecular chaperones,co-assembled block copolymer micelles with coordination groups of nitrilotriacetic acid(NTA)and hydrophobic microdomains of poly(Nisopropylacrylamide)(PNIPAM)on the surface were used as nanochaperones(n Chaps)to disaggregate amyloid insulin fibrils.Zinc ions chelated by NTA can bind the histidine imidazole residues while the PNIPAM microdomains can interact with the exposed hydrophobic sites on the amyloid insulin fibrils,which synergistically perturb the stability of amyloid insulin fibrils,loosen their structure,and finally promote their disaggregation.A combination of characterizations with fluorescence spectroscopy,transmission electron microscopy(TEM),dynamic hight scattering(DLS),and quartz crystal microbalance(QCM)demonstrated that mature amyloid insulin fibrils were completely disaggregated after incubating with n Chaps for 90 h.This study may provide a promising strategy for the development of n Chaps for the treatment of amyloidosis diseases.Hui Wang Ang Li Menglin Yang Yu Zhao Linqi Shi Rujiang Ma 2022Science China Chemistry2022,65,2:0
