| 1 | Identification of immunoreactive proteins of Brucella melitensis by immunoproteomics显示文摘Infection with Brucella causes brucellosis, a chronic disease in humans, which induces abortion and sterility in livestock. Among the different Brucella species, Brucella melitensis is considered the most virulent and is the predominant species associated with outbreaks in China. To date, no safe human vaccine is available against Brucella infection. The currently used live vaccines against Brucella in livestock induce antibodies that interfere with the diagnosis of field infection in vaccinated animals, which is harmful to eradication programs. However, there is as yet no complete profile of immunogenic proteins of B. melitensis. Towards the development of a safer, equally efficacious, and field infection-distinguishable vaccine, we used immunoproteomics to identify novel candidate immunogenic proteins from B. melitensis M5. Eighty-eight immunoreactive protein spots from B. melitensis M5 were identified by Western blotting and were assigned to sixty-one proteins by mass spectrometry, including many new immunoreactive proteins such as elongation factor G, F0F1 ATP synthase subunit beta, and OMP1. | 2 | Efficacy of tanshinone ⅡA and mesenchymal stem cell treatment of learning and memory impairment in a rat model of vascular dementia显示文摘OBJECTIVE: To investigate the efficacy of administration of tanshinone Ⅱ A(TSA) combined with mesenchymal stem cells(MSCs) for the treatment of learning and memory impairment caused by vascular dementia(Va D) and to determine the underlying mechanism.METHODS: Modified four-vessel occlusion was used to establish a Va D model in rats, and their spatial learning and memory capacity was assessed by the Morris water maze. The rats were randomized into MSCs, TSA, MSCs combined with TSA, vehicle and sham groups. Histological changes were determined by hematoxylin and eosin staining, and the hippocampal neuron apoptosis ratio was assessed by flow cytometry. Western blotting was performed to detect Bcl-2 and Bax expression. The reactive oxidative species(ROS) levels and the activity of total superoxide dismutase(T-SOD), an antioxidant enzyme in the rat hippocampus, were determined.RESULTS: TSA combined with MSCs treatment administered by intravenous injection in the tail significantly attenuated cognitive deficits in the Va D model compared with the vehicle group(P < 0.01),and its protective effect on cognitive function was greater than that obtained by treatment with MSCs or TSA alone. Furthermore, TSA combined with MSCs treatment achieved synergistic effects in suppressing neuronal apoptosis in the rat hippocampus caused by global brain ischemia via up-regulating the expression of Bcl-2, an anti-apoptosis protein, and decreasing the expression of Bax, a pro-apoptotic protein. | 3 | A novel inhibitor of N^(6)-methyladenosine demethylase FTO induces m RNA methylation and shows anti-cancer activities显示文摘N^(6)-methyladenosine(m^(6)A)modification is critical for m RNA splicing,nuclear export,stability and translation.Fat mass and obesity-associated protein(FTO),the first identified m^(6)A demethylase,is critical for cancer progression.Herein,we developed small-molecule inhibitors of FTO by virtual screening,structural optimization,and bioassay.As a result,two FTO inhibitors namely 18077 and 18097 were identified,which can selectively inhibit demethylase activity of FTO.Specifically,18097 bound to the active site of FTO and then inhibited cell cycle process and migration of cancer cells.In addition,18097 reprogrammed the epi-transcriptome of breast cancer cells,particularly for genes related to P53 pathway.18097 increased the abundance of m^(6)A modification of suppressor of cytokine signaling1(SOCS1)m RNA,which recruited IGF2 BP1 to increase m RNA stability of SOCS1 and subsequently activated the P53 signaling pathway.Further,18097 suppressed cellular lipogenesis via downregulation of peroxisome proliferator-activated receptor gamma(PPARγ),CCAAT/enhancer-binding protein alpha(C/EBPa),and C/EBPβ.Animal studies confirmed that 18097 can significantly suppress in vivo growth and lung colonization of breast cancer cells.Collectively,we identified that FTO can work as a potential drug target and the small-molecule inhibitor 18097 can serve as a potential agent against breast cancer. | Guoyou Xie Xu-Nian Wu Yuyi Ling Yalan Rui Deyan Wu Jiawang Zhou Jiexin Li Shuibin Lin Qin Peng Zigang Li Hongsheng Wang Hai-Bin Luo | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 4 |
| 4 | Responses of soil microbial communities and functions associated with organic carbon mineralization to nitrogen addition in a Tibetan grassland显示文摘Alpine grasslands with a high soil organic carbon(SOC)storage on the Tibetan Plateau are experiencing rapid climate warming and anthropogenic nitrogen(N)deposition;this is expected to substantially increase the soil N availability,which may impact carbon(C)cycling.However,little is known regarding how N enrichment influences soil microbial communities and functions relative to C cycling in this region.We conducted a 4-year field experiment on an alpine grassland to evaluate the effects of four different rates of N addition(0,25,50,and 100 kg N ha^-1 year^-1)on the abundance and community structure(phospholipid fatty acids,PLFAs)of microbes,enzyme activities,and community level physiological profiles(CLPP)in soil.We found that N addition increased the microbial biomass C(MBC)and N(MBN),along with an increased abundance of bacterial PLFAs,especially Gram-negative bacterial PLFAs,with a decreasing ratio of Gram-positive to Gram-negative bacteria.The N addition also stimulated the growth of fungi,especially arbuscular mycorrhizal fungi,reducing the ratio of fungi to bacteria.Microbial functional diversity and activity of enzymes involved in C cycling(β-1,4-glucosidase and phenol oxidase)and N cycling(β-1,4-N-acetyl-glucosaminidase and leucine aminopeptidase)increased after N addition,resulting in a loss of SOC.A meta-analysis showed that the soil C/N ratio was a key factor in the response of oxidase activity to N amendment,suggesting that the responses of soil microbial functions,which are linked to C turnover relative to N input,primarily depended upon the soil C/N ratio.Overall,our findings highlight that N addition has a positive influence on microbial communities and their associated functions,which may reduce soil C storage in alpine grasslands under global change scenarios. | Ruyi LUO Jiafa LUO Jianling FAN Deyan LIU Jin-Sheng HE Nazia PERVEEN Weixin DING | 2020 | Pedosphere2020,30,2: | 3 |
| 5 | Yield-scaled nitrous oxide emissions from nitrogen-fertilized croplands in China: A meta-analysis of contrasting mitigation scenarios显示文摘Nitrogen(N) losses in cropland resulting from the application of synthetic fertilizers decrease crop productivity and exacerbate environmental pollution.Mitigation measures, such as reduction in N fertilizer application rates, can have unintentional adverse effects on crop yield. We conducted a meta-analysis of soil N_(2)O emissions from agricultural fields across China under contrasting mitigation scenarios as a novel approach to identify the most effective strategy for the mitigation of emissions of N_(2)O derived from N fertilizer use in China. Current standard agricultural practice was used as a baseline scenario(BS), and 12 potential mitigation scenarios(S1–S12) were derived from the available literature and comprised single and combinations of management scenarios that accounted for crop yield. Mitigation scenarios S6(nitrification inhibitor 3,4-dimethylpyrazole phosphate) and S11(20% reduction in N application rate plus nitrification inhibitor dicyandiamide) in maize, rice, and wheat crops led to an average 56.0% reduction in N_(2)O emissions at the national level, whereas scenario S4(nitrification inhibitor dicyandiamide) led to yield optimization, with a 14.0% increase for maize and 8.0% increase for rice as compared to the BS.
