4MicroRNA-125b regulates Th17/Treg cell differentiation and is associated with juvenile idiopathic arthritis显示文摘Background Juvenile idiopathic arthritis(JIA)is the most common rheumatic disease in childhood driven by aberrant pathways of T-cell activation.T helper 17(Th17)/regulatory T cell(Treg)imbalance plays critical roles in the pathogenesis of arthritis.MicroRNA-125b(miR-125b)was upregulated after the activation of the initial CD4^+T cells,and could regulate the differentiation of CD4^+T cells.However,the effects of miR-125b on Th17/Treg imbalance and differentiation of Th 17/Treg cells remain unknown.Methods In this study,we evaluated the expression of miR-125b in the peripheral blood mononuclear cells(PBMCs)of children with JIA,and the relationship of miR-125b with Th17/Treg imbalance.Then,we used lentivirus vector-mediated overexpression technology to investigate the regulatory function of miR-125b in CD4^+T cells or dendritic cell/CD4^+T co-culture system.Results Decreased miR-125b expression in PBMCs and CD4^+T cells of JIA patients was negatively correlated with the ratio of Th17/Treg cells.It also correlated negatively with retinoic acid receptor-related orphan receptorγt but positively with Forkhead box protein 3 at transcriptional levels.Furthermore,we found that miR-125b overexpression inhibited Th17 cell differentiation,whereas facilitated the differentiation of Treg cells.MiR-125b upregulation led to the decrease of Th 17-secreting cytokines but the increase of the Treg-secreting cytokines.Conclusions Our results demonstrate that miR-125b participated in regulating Thl7/Treg cell differentiation and imbalance in JIA patients.These findings provide novel insight into the critical role of miR-125b in the Th17/Treg imbalance of JIA,and raise the distinct possibility that miR-125b may prove to be a potential therapeutic target for JIA.Zhi-Dan Fan Qian Cao Na Huang Le Ma Hui-Hui Ma Ya-Yuan Zhang Hai-Guo Yu Guo-Ping Zhou 2020World Journal of Pediatrics2020,16,1:7

9Understanding the role of rumen epithelial host-microbe interactions in cattle feed efficiency显示文摘Feed efficiency is one of the economically important traits for the cattle industry that affects profit(feed costs)and the environment(production of manure and methane).Due to that feed efficiency is driven by multi-factors,mechanisms contributing to the animal to animal variation in this trait have not been well defined,limiting the development of precision feeding strategies to improve the herd production efficiency.Rumen microbial fermentation and volatile fatty acids(VFA)production have been recently reported to be associated with cattle feed efficiency,however the roles of rumen epithelial function in feed efficiency are less studied although the rumen epithelium has an important function in VFA absorption and metabolism which can affect host feed efficiency.Rumen epithelium is colonized with a diverse microbial population,termed epimural microbiota,which has proposed functions in tissue development,barrier and inflammation,urea transport,and oxygen scavenging,suggesting that they can affect rumen epithelial functions and subsequently cattle feed efficiency.Especially,prospective functions of epimural microbiota,enhanced rumen immunity and increased rumen epithelial thickness,might contribute to less nutritional requirement for tissue recuperation.Thus,the understanding of the functions of rumen epithelium,epimural microbiota,and rumen epithelial host-microbe interactions is essential to identify their roles in contributing to feed efficiency.In this review,we will focus on to date research findings on the structure of rumen epithelium,epimural microbiota,and epithelial host-microbe interactions together with their functions and how these are associated with feed efficiency,aiming to provide insights on future directions to study rumen epithelial host-microbe interactions and improve the rumen functions in cattle.Sang Weon Na Le Luo Guan 2022Animal Nutrition2022,,3:2
10AoATG5 plays pleiotropic roles in vegetative growth,cell nucleus development,conidiation,and virulence in the nematode-trapping fungus Arthrobotrys oligospora显示文摘Autophagy is an evolutionarily conserved process in eukaryotes,which is regulated by autophagy-related genes(ATGs).Arthrobotrys oligospora is a representative species of nematode-trapping(NT)fungi that can produce special traps for nematode predation.To elucidate the biological roles of autophagy in NT fungi,we characterized an orthologous Atg protein,AoAtg5,in A.oligospora.We found that AoATG5 deletion causes a significant reduction in vegetative growth and conidiation,and that the transcript levels of several sporulation-related genes were significantly downregulated during sporulation stage.In addition,the cell nuclei were significantly reduced in theΔAoATG5 mutant,and the transcripts of several genes involved in DNA bio-synthesis,repair,and ligation were significantly upregulated.InΔAoATG5 mutants,the autophagic process was significantly impaired,and trap formation and nematocidal activity were significantly decreased.Comparative transcriptome analysis results showed that AoAtg5 is involved in the regulation of multiple cellular processes,such as autophagy,nitrogen metabolism,DNA biosynthesis and repair,and vesicular transport.In summary,our results suggest that AoAtg5 is essential for autophagy and significantly contributes to vegetative growth,cell nucleus development,sporulation,trap formation,and pathogenicity in A.oligospora,thus providing a basis for future studies focusing on related mechanisms of autophagy in NT fungi.Duanxu Zhou Yingmei Zhu Na Bai Le Yang Meihua Xie Jiangliu Yang Meichen Zhu Ke-Qin Zhang Jinkui Yang 2022Science China(Life Sciences)2022,65,2:2
11Combination of a biopharmaceutic classification system and physiologically based pharmacokinetic models to predict absorption properties of baicalein in vitro and in vivo显示文摘Objective: To determine the in vitro and in vivo absorption properties of active ingredients of the Chinese medicine, baicalein, to enrich mechanistic understanding of oral drug absorption.Methods: The Biopharmaceutic Classification System(BCS) category was determined using equilibrium solubility, intrinsic dissolution rate, and intestinal permeability to evaluate intestinal absorption mechanisms of baicalein in rats in vitro. Physiologically based pharmacokinetic(PBPK) model commercial software GastroPlus~(TM) was used to predict oral absorption of baicalein in vivo.Results: Based on equilibrium solubility, intrinsic dissolution rate, and permeability values of main absorptive segments in the duodenum, jejunum, and ileum, baicalein was classified as a drug with low solubility and high permeability. Intestinal perfusion with venous sampling(IPVS) revealed that baicalein was extensively metabolized in the body, which corresponded to the low bioavailability predicted by the PBPK model. Further, the PBPK model predicted the key indicators of BCS, leading to reclassification as BCS-II. Predicted values of peak plasma concentration of the drug(C_(max)) and area under the curve(AUC)fell within two times of the error of the measured results, highlighting the superior prediction of absorption of baicalein in rats, beagles, and humans. The PBPK model supported in vitro and in vivo evidence and provided excellent prediction for this BCS class II drug.Conclusion: BCS and PBPK are complementary methods that enable comprehensive research of BCS parameters, intestinal absorption rate, metabolism, prediction of human absorption fraction and bioavailability, simulation of PK, and drug absorption in various intestinal segments across species. 18PKC-SWI6 signaling regulates asexual development,cell wall integrity,stress response,and lifestyle transition in the nematode-trapping fungus Arthrobotrys oligospora显示文摘Predatory fungi possess intricate signal transduction systems that regulate their development and support successful infection of the host.Herein,we characterized three components of the cell wall integrity-controlling pathway,namely protein kinase C(Ao PKC),SLT2-MAPK(Ao SLT2),and SWI6(Ao SWI6),in a representative nematode-trapping fungus Arthrobotrys oligospora,using gene disruption and multi-omics approaches.The phenotypic traits(such as mycelia development,conidiation,stress response,and trap morphogenesis) and metabolic profiles of ΔAopkc and ΔAoswi6 mutants were similar but differed from those of the ΔAoslt2 mutants.Transcriptomic analysis indicated that the genes differentially expressed in the absence of Aoswi6 were involved in DNA replication,repair,and recombination during trap formation.Moreover,the yeast two-hybrid assay showed that Ao PKC interacted with Ao SWI6,suggesting that in A.oligospora,PKC can directly regulate SWI6,bypassing the SLT2signaling cascade.Conclusively,our findings deepen our understanding of the regulatory mechanism of asexual development and lifestyle switching in nematode-trapping fungi.Meihua Xie Ni Ma Na Bai Le Yang Xuewei Yang Ke-Qin Zhang Jinkui Yang 2022Science China(Life Sciences)2022,65,12:1
