1AoATG5 plays pleiotropic roles in vegetative growth,cell nucleus development,conidiation,and virulence in the nematode-trapping fungus Arthrobotrys oligospora显示文摘Autophagy is an evolutionarily conserved process in eukaryotes,which is regulated by autophagy-related genes(ATGs).Arthrobotrys oligospora is a representative species of nematode-trapping(NT)fungi that can produce special traps for nematode predation.To elucidate the biological roles of autophagy in NT fungi,we characterized an orthologous Atg protein,AoAtg5,in A.oligospora.We found that AoATG5 deletion causes a significant reduction in vegetative growth and conidiation,and that the transcript levels of several sporulation-related genes were significantly downregulated during sporulation stage.In addition,the cell nuclei were significantly reduced in theΔAoATG5 mutant,and the transcripts of several genes involved in DNA bio-synthesis,repair,and ligation were significantly upregulated.InΔAoATG5 mutants,the autophagic process was significantly impaired,and trap formation and nematocidal activity were significantly decreased.Comparative transcriptome analysis results showed that AoAtg5 is involved in the regulation of multiple cellular processes,such as autophagy,nitrogen metabolism,DNA biosynthesis and repair,and vesicular transport.In summary,our results suggest that AoAtg5 is essential for autophagy and significantly contributes to vegetative growth,cell nucleus development,sporulation,trap formation,and pathogenicity in A.oligospora,thus providing a basis for future studies focusing on related mechanisms of autophagy in NT fungi.Duanxu Zhou Yingmei Zhu Na Bai Le Yang Meihua Xie Jiangliu Yang Meichen Zhu Ke-Qin Zhang Jinkui Yang 2022Science China(Life Sciences)2022,65,2:2
2BnbHLH92a negatively regulates anthocyanin and proanthocyanidin biosynthesis in Brassica napus显示文摘Yellow seed trait is a desirable characteristic with potential for increasing seed quality and commercial value in rapeseed,and anthocyanin and proanthocyanidins(PAs)are major seed-coat pigments.Few transcription factors involved in the regulation of anthocyanin and PAs biosynthesis have been characterized in rapeseed.In this study,we identified a transcription factor gene BnbHLH92a(BnaA06T0441000ZS)in rapeseed.Overexpressing BnbHLH92a both in Arabidopsis and in rapeseed reduced levels of anthocyanin and PAs.Correspondingly,the expression profiles of anthocyanin and PA biosynthesis genes(TT3,BAN,TT8,TT18,and TTG1)were shown by quantitative real-time PCR to be inhibited in BnbHLH92a-overexpressing Arabidopsis seeds,indicating that BnbHLH92a represses the anthocyanin and PA biosynthesis pathway in Arabidopsis.BnbHLH92a physically interacts with the BnTTG1 protein and represses the biosynthesis of anthocyanins and PAs in rapeseed.BnbHLH92a also binds directly to the BnTT18 promoter and represses its expression.These results suggest that BnbHLH92a is a novel upstream regulator of flavonoid biosynthesis in B.napus.Ran Hu Meichen Zhu Si Chen Chengxiang Li Qianwei Zhang Lei Gao Xueqin Liu Shulin Shen Fuyou Fu Xinfu Xu Ying Liang Liezhao Liu Kun Lu Hao Yu Jiana Li Cunmin Qu 2023The Crop Journal2023,11,2:0
3Magnesium Demethylcantharidate induces apoptosis in hepatocellular carcinoma cells via ER stress显示文摘Cantharidin(CTD)is a bioactive ingredient isolated from Cantharis vesicatoria(blister beetles),which has potential therapeutic value as an anticancer agent.Magnesium Demethylcantharidate(MDC)is a recently developed derivative of Cantharidin(CTD),and previous studies have illustrated its excellent anticancer activity on HCC cells.However,the effect and mechanism of MDC remains unclear and need to be further studied.In particular,whether MDC can cause ER stress in HCC is still unknown.In this study,we demonstrated that endoplasmic reticulum stress(ERS)-related proteins were changed in SMMC-7721 and Bel-7402 cells after being exposed to MDC.Moreover,we found that MDC could significantly inhibit the growth of xenograft tumor in nude mice.In summary,we confirmed that MDC could induce ERS in HCC cells and thus induce apoptosis.XINTING ZHU MENG YE KELAN FANG FANG LIU JING HUI MEICHEN LIU XIAOFEI LI RONG YAN YUN LIU 2022BIOCELL2022,46,12:0
