1Effect of subsoil tillage depth on nutrient accumulation, root distribution, and grain yield in spring maize显示文摘A four-year field experiment was conducted to investigate the effect of subsoiling depth on root morphology, nitrogen(N), phosphorus(P), and potassium(K) uptake, and grain yield of spring maize. The results indicated that subsoil tillage promoted root development,increased nutrient accumulation, and increased yield. Compared with conventional soil management(CK), root length, root surface area, and root dry weight at 0–80 cm soil depth under subsoil tillage to 30 cm(T1) and subsoil tillage to 50 cm(T2) were significantly increased, especially the proportions of roots in deeper soil. Root length, surface area, and dry weight differed significantly among three treatments in the order of T2 > T1 > CK at the12-leaf and early filling stages. The range of variation of root diameter in different soil layers in T2 treatment was the smallest, suggesting that roots were more likely to grow downwards with deeper subsoil tillage in soil. The accumulation of N, P, and K in subsoil tillage treatment was significantly increased, but the proportions of kernel and straw were different. In a comparison of T1 with T2, the grain accumulated more N and P, while K accumulation in kernel and straw varied in different years. Grain yield and biomass were increased by 12.8% and 14.6% on average in subsoil tillage treatments compared to conventional soil treatment. 3High-quality genome assembly of 'Cuiguan' pear (Pyrus pyrifolia) as a reference genome for identifying regulatory genes and epigenetic modifications responsible for bud dormancy显示文摘Dormancy-associated MADS-box(DAM)genes serve as crucial regulators of the endodormancy cycle in rosaceous plants.Although pear DAM genes have been identified previously,the lack of a high-quality reference genome and techniques to study gene function have prevented accurate genome-wide analysis and functional verification of such genes.Additionally,the contribution of other genes to the regulation of endodormancy release remains poorly understood.In this study,a high-quality genome assembly for'Cuiguan'pear(Pyrus pyrifolia),which is a leading cultivar with a low chilling requirement cultivated in China,was constructed using PacBio and Hi-C technologies.Using this genome sequence,we revealed that pear DAM genes were tandemly clustered on Chr8 and Chr15 and were differentially expressed in the buds between'Cuiguan'and the high-chilling-requirement cultivar'Suli'during the dormancy cycle.Using a virus-induced gene silencing system,we determined the repressive effects of DAM genes on bud break.Several novel genes potentially involved in the regulation of endodormancy release were identified by RNA sequencing and H3K4me3 chromatin immunoprecipitation sequencing analyses of‘Suli'buds during artificial chilling using the new reference genome.Our findings enrich the knowledge of the regulatory mechanism underlying endodormancy release and chilling requirements and provide a foundation for the practical regulation of dormancy release in fruit trees as an adaptation to climate change.Yuhao Gao Qinsong Yang Xinhui Yan Xinyue Wu Feng Yang Jianzhao Li Jia Wei Junbei Ni Mudassar Ahmad Songling Bai Yuanwen Teng 2021Horticulture Research2021,8,1:5
4Quantifying the Cloud Water Resource:Methods Based on Observational Diagnosis and Cloud Model Simulation显示文摘Based on the concepts of cloud water resource(CWR)and related variables proposed in the first part of this study,this paper provides details of two methods to quantify the CWR.One is diagnostic quantification(CWR-DQ)based on satellite observations,precipitation products,and atmospheric reanalysis data;and the other is numerical quantification(CWR-NQ)based on a cloud resolving model developed at the Chinese Academy of Meteorological Sciences(CAMS).The two methods are applied to quantify the CWR in April and August 2017 over North China,and the results are evaluated against all available observations.Main results are as follows.(1)For the CWR-DQ approach,reference cloud profiles are firstly derived based on the Cloud Sat/CALIPSO joint satellite observations for 2007–2010.The NCEP/NCAR reanalysis data in 2000–2017 are then employed to produce three-dimensional cloud fields.The budget/balance equations of atmospheric water substance are lastly used,together with precipitation observations,to retrieve CWR and related variables.It is found that the distribution and vertical structure of clouds obtained by the diagnostic method are consistent with observations.(2)For the CWR-NQ approach,it assumes that the cloud resolving model is able to describe the cloud microphysical processes completely and precisely,from which four-dimensional distributions of atmospheric water vapor,hydrometeors,and wind fields can be obtained.The data are then employed to quantify the CWR and related terms/quantities.After one-month continuous integration,the mass of atmospheric water substance becomes conserved,and the tempospatial distributions of water vapor,hydrometeors/cloud water,and precipitation are consistent with observations.(3)Diagnostic values of the difference in the transition between hydrometeors and water vapor(Cvh-Chv)and the surface evaporation(Es)are well consistent with their numerical values.(4)Correlation and bias analyses show that the diagnostic CWR contributors are well correlated with observations,and match their numerical counterparts as well,indicating that the CWR-NQ and CWR-DQ methods are reasonable.(5)Underestimation of water vapor converted from hydrometeors(Chv)is a shortcoming of the CWR-DQ method,which may be rectified by numerical quantification results or by use of advanced observations on higher spatiotemporal resolutions.Miao CAI Yuquan ZHOU Jianzhao LIU Chao TAN Yahui TANG Qianrong MA Qi LI Jietai MAO Zhijin HU 2020Journal of Meteorological Research2020,34,6:4
5Carbon inputs regulate the temperature sensitivity of soil respiration in temperate forests显示文摘Litter and root activities may alter the temperature sensitivity(Q_(10))of soil respiration.However,existing studies have not provided a comprehensive understanding of the effects of litter and root carbon inputs on the Q_(10)of soil respiration in different seasons.In this study,we used the trench method under in situ conditions to measure the total soil respiration(R_(total)),litter-removed soil respiration(R_(no-litter)),root-removed soil respiration(R_(no-root)),and the decomposition of soil organic matter(i.e.,both litter and root removal;R_(SOM))in different seasons of pioneer(Populus davidiana Dode)and climax(Quercus liaotungensis Mary)forests on the Loess Plateau,China.Soil temperature,soil moisture,litter biomass,fine root biomass,litter carbon,and root carbon were analyzed to obtain the drive mechanism of the Q_(10)of soil respiration in the two forests.The results showed that the Q_(10)of soil respiration exhibited seasonality,and the Q_(10)of soil respiration was higher in summer.The litter enhanced the Q_(10)of soil respiration considerably more than the root did.Soil temperature,soil moisture,fine root biomass,and litter carbon were the main factors used to predict the Q_(10)of different soil respiration components.These findings indicated that factors affecting the Q_(10)of soil respiration highly depended on soil temperature and soil moisture as well as related litter and root traits in the two forests,which can improve our understanding of soil carbon–climate feedback in global warming.The results of this study can provide reference for exploring soil respiration under temperate forest restoration.LIU Yulin LI Jiwei HAI Xuying WU Jianzhao DONG Lingbo PAN Yingjie SHANGGUAN Zhouping WANG Kaibo DENG Lei 2022Journal of Arid Land2022,14,9:3
6Cryo-EM structures of human m^(6)A writer complexes显示文摘N6-methyladenosine(m^(6)A)is the most abundant ribonucleotide modification among eukaryotic messenger RNAs.The m^(6)A“writer”consists of the catalytic subunit m^(6)A-METTL complex(MAC)and the regulatory subunit m^(6)A-METTL-associated complex(MACOM),the latter being essential for enzymatic activity.Here,we report the cryo-electron microscopy(cryo-EM)structures of MACOM at a 3.0-Åresolution,uncovering that WTAP and VIRMA form the core structure of MACOM and that ZC3H13 stretches the conformation by binding VIRMA.Furthermore,the 4.4-Åresolution cryo-EM map of the MACOM–MAC complex,combined with crosslinking mass spectrometry and GST pull-down analysis,elucidates a plausible model of the m^(6)A writer complex,in which MACOM binds to MAC mainly through WTAP and METTL3 interactions.In combination with in vitro RNA substrate binding and m^(6)A methyltransferase activity assays,our results illustrate the molecular basis of how MACOM assembles and interacts with MAC to form an active m^(6)A writer complex.Shichen Su Shanshan Li Ting Deng Minsong Gao Yue Yin Baixing Wu Chao Peng Jianzhao Liu Jinbiao Ma Kaiming Zhang 2022Cell Research2022,32,11:3
7Effects of trace Zr on the microstructure and properties of 2E12 alloy显示文摘Al-4.0Cu-1.4Mg-0.6Mn (2E12) and Al-4.0Cu-1.4Mg-0.6Mn-0.3Zr aluminum billets were manufactured by soft-contact electromagnetic continuous casting (EMC). Subsequent forging and heat treatment were conducted and the effects of Zr on the microstructure and properties of the Al-4.0Cu-1.4Mg-0.6Mn alloy were studied. The results show that the addition of 0.3% Zr can reduce the dendrite and refine grains. During forging and solution treatment, fine and dispersive Al3Zr particles precipitated from the supersaturated α (Al) solid solution in the heating process of the billet can effectively pin dislocations and subgrain boundaries. Because of the addition of Zr, the mechanical properties are improved with the tensile strength, yield strength, elongation, and contraction of the area increasing by 5.4%, 11.3%, 9.7%, and 12.6%, respectively. 8Investigation of carbonyl amidation and O-methylation during biosynthesis of the pharmacophore pyridyl of antitumor piericidins显示文摘Piericidins are a large family of bacterialα-pyridone antibiotics with antitumor activities such as their anti-renal carcinoma activity exhibited recently in nude mice.The backbones of piericidins are derived fromβ,δ-diketo carboxylic acids,which are offloaded from a modular polyketide synthase(PKS)and putatively undergo a carbonyl amidation beforeα-pyridone ring formation.The tailoring modifications to theα-pyridone structure mainly include the verified hydroxylation and O-methylation of the C-4′position and an unidentified C-5′O-methylation.Here,we describe a piericidin producer,terrestrial Streptomyces conglobatus,which contains a piericidin biosynthetic gene cluster in two different loci.Deletion of the amidotransferase gene pieD resulted in the accumulation of two fatty acids that should be degraded from the nascent carboxylic acid released by the PKS,supporting the carbonyl amidation function of PieD duringα-pyridone ring formation.Deletion of the O-methyltransferase gene pieB1 led to the production of three piericidin analogues lacking C-5′O-methylation,therefore confirming that PieB1 specifically catalyses the tailoring modification.Moreover,bioactivity analysis of the mutant-derived products provided clues regarding the structure-function relationship for antitumor activity.The work addresses two previously unidentified steps involved in pyridyl pharmacophore formation during piericidin biosynthesis,facilitating the rational bioengineering of the biosynthetic pathway towards valuable antitumor agents.Wanlu Li Wenyu Zhang Yijia Cheng Yaoyao Shen Jianzhao Qi Hou-Wen Lin Yongjun Zhou 2022Synthetic and Systems Biotechnology2022,7,3:2
9Comparison of algorithms for monitoring wheat powdery mildew using multi-angular remote sensing data显示文摘Powdery mildew is a disease that threatens wheat production and causes severe economic losses worldwide. Its timely diagnosis is imperative for preventing and controlling its spread. In this study, the multiangle canopy spectra and disease severity of wheat were investigated at several developmental stages and degrees of disease severity. Four wavelength variable-selected algorithms: successive projection(SPA), competitive adaptive reweighted sampling(CARS), feature selection learning(Relief-F), and genetic algorithm(GA), were used to identify bands sensitive to powdery mildew. The wavelength variables selected were used as input variables for partial least squares(PLS), extreme learning machine(ELM), random forest(RF), and support vector machine(SVM) algorithms, to construct a suitable prediction model for powdery mildew. Spectral reflectance and conventional vegetation indices(VIs) displayed angle effects under several disease severity indices(DIs). The CARS method selected relatively few wavelength variables and showed a relatively homogeneous distribution across the 13 viewing zenith angles.Overall accuracies of the four modeling algorithms were ranked as follows: ELM(0.70–0.82) > PLS(0.63–0.79) > SVM(0.49–0.69) > RF(0.43–0.69). Combinations of features and algorithms generated varied accuracies, with coefficients of determination(R^(2)) single-peaked at different observation angles. The constructed CARS-ELM model extracted a predictable bivariate relationship between the multi-angle canopy spectrum and disease severity, yielding an R^(2)> 0.8 at each measured angle. Especially for larger angles,monitoring accuracies were increased relative to the optimal VI model(40% at-60°, 33% at +60°), indicating that the CARS-ELM model is suitable for extreme angles of-60° and +60°. 10定量分析乳腺癌术后化疗前外周血多药耐药基因的检测及其临床意义的研究(英文)显示文摘Objective: To explore expressions of multidrug resistance gene (MDR) product P170 and peripheral blood MDR1 mRNA in breast cancer tissue and their clinical significances. Methods: Fluorescent quantitative RT-PCR and immunohis-tochemistry were used to detect peripheral blood MDR1 mRNA before chemotherapy in breast cancer tissue samples from 32 cases with P170 positive (trial group) and 11 cases with P170 negative (control group). Results: There were 14 cases (43.75%) peripheral blood MDR1 mRNA positive and 18 cases (56.25%) negative in 32 cases P170 positive breast cancer pa-tients, while there were 6 cases (54.55%) peripheral blood MDR1 mRNA positive and 5 cases (45.45%) negative in 11 cases P170 negative breast cancer patients; there wasn't a significance difference with peripheral blood MDR1 mRNA express rate in P170 positive (43.75%) group and P170 negative (54.55%) group (χ2 = 0.383, P > 0.05). There were 12 cases (37.50%) Her-2 positive and 20 cases (62.50%) negative in P170 positive group; there were 2 cases (18.18%) Her-2 positive and 9 cases (81.82%) negative in P170 negative group; there wasn't a significance difference with Her-2 positive rate in P170 posi-tive group (37.50%) and P170 negative (18.18%) group (χ2 = 1.391, P > 0.05). There were 13 cases (40.63%) ER positive and 19 cases (59.37%) negative in P170 positive group; there were 7 cases (63.64%) ER positive and 4 cases (36.36%) negative in P170 negative group; there wasn't a significance difference with ER positive rate in P170 positive group (40.63%) and P170 negative (63.64) group (χ2 = 1.742, P > 0.05). There were 20 cases (62.50%) PR positive and 12 cases (37.50%) negative in P170 positive group; there were 4 cases (36.36%) PR positive and 7 cases (63.64%) negative in P170 negative group; there wasn't a significance difference with PR positive rate in P170 positive group (62.50%) and P170 negative (36.36%) group (χ2 = 2.267, P > 0.05). There were 9 cases (28.12%) with lymphaden metastasis and 23 cases (71.88%) without lymphaden metastasis in P170 positive group; there were 5 cases (45.45%) with lymphaden metastasis and 6 cases (54.55%) without lymphaden metastasis in P170 negative group; there wasn't a significance difference with lymphaden metastasis rate in P170 positive group (28.12%) and P170 negative (45.45%) group (χ2 = 1.120, P > 0.05). Conclusion: 1) There were possibly MDR expressing infrequently in carcinoma tissue and peripheral blood before chemotherapy. 2) FQ-RT-PCR for detecting of periph-eral blood MDR1 mRNA is special, sensitive and reliable. 11Lycium barbarum polysaccharides ameliorate canine acute liver injury by reducing oxidative stress,protecting mitochondrial function,and regulating metabolic pathways显示文摘The development of acute liver injury can result in liver cirrhosis,liver failure,and even liver cancer,yet there is currently no effective therapy for it.The purpose of this study was to investigate the protective effect and therapeutic mechanism of Lycium barbarum polysaccharides(LBPs)on acute liver injury induced by carbon tetrachloride(CCl_(4)).To create a model of acute liver injury,experimental canines received an intraperitoneal injection of 1 mL/kg of CCl_(4)solution.The experimental canines in the therapy group were then fed LBPs(20 mg/kg).CCl_(4)-induced liver structural damage,excessive fibrosis,and reduced mitochondrial density were all improved by LBPs,according to microstructure data.By suppressing Kelch-like epichlorohydrin(ECH)-associated protein 1(Keap1),promoting the production of sequestosome 1(SQSTM1)/p62,nuclear factor erythroid 2-related factor 2(Nrf2),and phaseⅡdetoxification genes and proteins downstream of Nrf2,and restoring the activity of anti-oxidant enzymes like catalase(CAT),LBPs can restore and increase the antioxidant capacity of liver.To lessen mitochondrial damage,LBPs can also enhance mitochondrial respiration,raise tissue adenosine triphosphate(ATP)levels,and reactivate the respiratory chain complexes I–V.According to serum metabolomics,the therapeutic impact of LBPs on acute liver damage is accomplished mostly by controlling the pathways to lipid metabolism.9-Hydroxyoctadecadienoic acid(9-HODE),lysophosphatidylcholine(Lyso PC/LPC),and phosphatidylethanolamine(PE)may be potential indicators of acute liver injury.This study confirmed that LBPs,an effective hepatoprotective drug,may cure acute liver injury by lowering oxidative stress,repairing mitochondrial damage,and regulating metabolic pathways.JIANJIA HUANG YUMAN BAI WENTING XIE RONGMEI WANG WENYUE QIU SHUILIAN ZHOU ZHAOXIN TANG JIANZHAO LIAO RONGSHENG SU 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,2:2
12Starch supplementation improves the reproductive performance of sows in different glucose tolerance status显示文摘This study was to evaluate the effects of glucose tolerance status,maternal starch supplementation and soybean substitution in diets on the performance of dams and their offspring.Eighty-eight pregnant sows(Landrace x Large White)were selected from an initial total of 120 sows,based on blood glucose test values,and assigned to 4 experimental treatments in a 2×2 factorial design.The factors were glucose tolerance status(glucose intolerant[GIT]vs.normal glucose tolerant[NGT])or dietary treatments(corn starch diet[CS]vs.soybean substitution diet[SSI]).A higher area under the curve(AUC)for post-meal glucose was observed(P<0.05)in the GIT group than in the NGT group on d 109 of gestation.The CS group had a lower value of homeostasis model assessment-insulin resistance than the SS group(P<0.05)on d 109 of gestation.Corn starch supplementation for sows decreased the stillbirth rate(P<0.05),regardless of the sows’glucose tolerance status.The villus height of the jejunum and the villus height to crypt depth ratio of the ileum were greater in normal birth weight piglets from the CS group than from the SS group(P<0.01),and so was the activity of sucrase in the jejunum and ileum(P<0.01).Compared with the SS group,the CS group showed a reduction in pre-weaning mortality rate,an increase in the number of high-birth-weight piglets,and a decrease in the number of low-birth-weight piglets(P<0.05)under GIT status.In conclusion,sows fed CS decreased stillbirth rate and improved insulin resistance,as well as improving the intestinal morphology and digestive enzyme activities of their progeny,regardless of glucose tolerance status.Additionally,the CS group improved birth weight distribution and decreased pre-weaning mortality rate of piglets under GIT status.Yunyu Yang Ming Deng Jianzhao Chen Xichen Zhao Kaili Xiao Wenliang He Xinggang Qiu Yanzhen Xu Yulong Yin Chengquan Tan 2021Animal Nutrition2021,,4:2

16Dietary fibers with low hydration properties exacerbate diarrhea and impair intestinal health and nutrient digestibility in weaned piglets显示文摘Background:This study aimed to investigate the hydration properties of different-source fibrous materials by com-paring their water-binding capacity(WBC),water swelling capacity(WSC),viscosity,and in vivo effects of selected samples on growth performance,nutrient digestibility,diarrhea,and intestinal health in weaned piglets.Methods:A total of 13 commercially available fibrous materials were first compared in chemical composition and in vitro hydration property.Subsequently,40 weaned piglets were randomized to five experimental dietary groups(8 piglets per group):control diet(a basal diet without dietary fiber,CON),basal diet supplemented with 5%microcrys-talline cellulose(MCC),5%wheat bran(WB),5%Moringa oleifera leaf powder(MOLP),or 5%sugar beet pulp(SBP),fol-lowed by analyzing their growth performance and diarrhea rate in a 28-d experiment.After the feeding experiment,anaesthetized piglets were killed,and their intestinal and colon content or plasma samples were analyzed in nutrient digestibility,intestinal morphology,intestinal barrier,short-chain fatty acids(SCFAs),and bacterial population.Results:In vitro studies showed low hydration properties for WB and MCC,while medium hydration properties for MOLP and SBP.In vivo studies indicated that compared with medium hydration property groups,low hydration prop-erty groups showed(1)exacerbated diarrhea,impaired intestinal health,and reduced apparent fecal digestibility of dry matter,gross energy,acid detergent fiber,and neutral detergent fiber;(2)decreased SCFAs concentration and rela-tive levels of Lactobacillus and Bifidobacterium,but increased levels of Escherichia coli and Brachyspira hyodysenteriae in colon contents.Additionally,SBP showed optimal performance in reducing diarrhea and increasing SCFAs produc-tion.Correlation analysis revealed a positive correlation of fiber hydration properties with in vitro SCFAs production,and diarrhea index and nutrient digestibility were negatively and positively correlated with SCFAs levels in the colon contents of weaned piglets,respectively.Conclusions:Different-source dietary fibers varied in their hydration properties and impacts on diarrhea,microbial composition and SCFAs production in weaned piglets.WB and MCC could exacerbate diarrhea and impair nutrient digestibility,probably because their low hydration properties were detrimental to gut microbial homeostasis and fermentation.Our findings provide new ideas for rational use of fiber resources in weaned piglets.Shuangbo Huang Zhijuan Cui Xiangyu Hao Chuanhui Cheng Jianzhao Chen Deyuan Wu Hefeng Luo Jinping Deng Chengquan Tan 2023Journal of Animal Science and Biotechnology2023,14,2:1
17Horizontal convection in a rectangular enclosure driven by a linear temperature profile显示文摘The horizontal convection in a square enclosure driven by a linear temperature profile along the bottom boundary is investigated numerically by using a finite difference method.The Prandtl number is fixed at 4.38,and the Rayleigh number Ra ranges from107 to 1011.The convective flow is steady at a relatively low Rayleigh number,and no thermal plume is observed,whereas it transits to be unsteady when the Rayleigh number increases beyond the critical value.The scaling law for the Nusselt number Nu changes from Rossby’s scaling Nu~Ra^(1/5)in a steady regime to Nu~Ra^(1/4)in an unsteady regime,which agrees well with the theoretically predicted results.Accordingly,the Reynolds number Re scaling varies from Re~Ra^(3/11)to Re~Ra^(2/5).The investigation on the mean flows shows that the thermal and kinetic boundary layer thickness and the mean temperature in the bulk zone decrease with the increasing Ra.The intensity of fluctuating velocity increases with the increasing Ra.Tianyong YANG Bofu WANG Jianzhao WU Zhiming LU Quan ZHOU 2021Applied Mathematics and Mechanics(English Edition)2021,42,8:1

19Femtosecond laser additive and subtractive micro-processing:enabling a high-channeldensity silica interposer for multicore fibre to silicon-photonic packaging显示文摘Great strides have been made over the past decade to establish femtosecond lasers in advanced manufacturing systems for enabling new forms of non-contact processing of transparent materials.Research advances have shown that a myriad of additive and subtractive techniques is now possible for flexible 2D and 3D structuring of such materials with micro-and nano-scale precision.In this paper,these techniques have been refined and scaled up to demonstrate the potential for 3D writing of high-density optical packaging components,specifically addressing the major bottleneck for efficiently connecting optical fibres to silicon photonic(SiP)processors for use in telecom and data centres.An 84-channel fused silica interposer was introduced for high-density edge coupling of multicore fibres(MCFs)to a SiP chip.Femtosecond laser irradiation followed by chemical etching was further harnessed to open alignment sockets,permitting rapid assembly with precise locking of MCF positions for efficient coupling to laser written optical waveguides in the interposer.A 3D waveguide fanout design provided an attractive balancing of low losses,modematching,high channel density,compact footprint,and low crosstalk.The 3D additive and subtractive processes thus demonstrated the potential for higher scale integration and rapid photonic assembly and packaging of micro-optic components for telecom interconnects,with possible broader applications in integrated biophotonic chips or micro-displays.Gligor Djogo Jianzhao Li Stephen Ho Moez Haque Erden Ertorer Jun Liu Xiaolu Song Jing Suoand Peter R Herman 2019International Journal of Extreme Manufacturing2019,1,4:1
