| 4 | Assessment of the uncertainties in temperature change in China during the last century显示文摘We have used the China Homogenized Historic Temperature dataset and some long-term station series of the neighbor countries from CRUTEM3,a 5°×5° gridded dataset of monthly mean temperature since 1900,to provide a 107-year record of surface temperature trends and variability.We derived a comprehensive set of uncertainty estimates to accompany the data:measurement and sampling errors,uncertainties in temperature bias estimates,and uncertainties arising from limited observational coverage on large-scale averages have all been estimated.We reanalysed the temperature changes during the period of record.The best estimates of trends for 1900-2006 with uncertainties at 95% confidence range are about 0.09±0.017°C/decade for the year as a whole,and 0.14±0.021°C/decade,0.11±0.021°C/decade,0.04±0.017°C/decade,and 0.07±0.017°C/decade for winter,spring,summer and autumn respectively.For 1954-2006,the trends for annual,winter,spring,summer and autumn are:0.26±0.032°C/decade,0.35±0.046°C/decade,0.25±0.051°C/decade,0.16±0.037°C/decade and 0.22±0.055°C/decade.Winter saw the most significant warming trend in both 1900-2006 and 1954-2006,while during the most recent period(the satellite era,1979-2006),all the seasons show similar warming trends:0.45±0.13°C/decade,0.51±0.11°C/decade,0.52±0.16°C/decade,0.37±0.10°C/decade and 0.50±0.16°C/decade for annual,winter,spring,summer and autumn.Trends arising from urbanization have been evaluated as less than 5% of the total warming trend for 1951-2001,so this bias was not removed. | LI QingXiang DONG WenJie LI Wei GAO XiaoRong JONES P KENNEDY J PARKER D | 2010 | Chinese Science Bulletin2010,55,19: | 26 |

| 7 | Sorafenib inhibits growth and metastasis of hepatocellular carcinoma by blocking STAT3显示文摘AIM: To investigate the inhibitory role and the underlying mechanisms of sorafenib on signal transducer and activator of transcription 3 (STAT3) activity in hepatocellular carcinoma (HCC).METHODS: Human and rat HCC cell lines were treated with sorafenib. Proliferation and STAT3 dephosphorylation were assessed. Potential molecular mechanisms of STAT3 pathway inhibition by sorafenib were evaluated. In vivo antitumor action and STAT3 inhibition were investigated in an immunocompetent orthotopic rat HCC model.RESULTS: Sorafenib decreased STAT3 phosphorylationat the tyrosine and serine residues (Y705 and S727), but did not affect Janus kinase 2 (JAK2) and phosphatase shatterproof 2 (SHP2), which is associated with growth inhibition in HCC cells. Dephosphorylation of S727 was associated with attenuated extracellular signal-regulated kinase (ERK) phosphorylation, similar to the effects of a mitogen-activated protein kinase (MEK) inhibitor U0126, suggesting that sorafenib induced S727 dephosphorylation by inhibiting MEK/ERK signaling. Meanwhile, sorafenib could also inhibit Akt phosphorylation, and both the phosphatidylinositol-3-kinase (PI3K) inhibitor LY294002 and Akt knockdown resulted in Y705 dephosphorylation, indicating that Y705 dephosphorylation by sorafenib was mediated by inhibiting the PI3K/Akt pathway. | 13 | Utilizing gastric cancer organoids to assess tumor biology and personalize medicine显示文摘While the incidence and mortality of gastric cancer (GC) have declined due to public health programs, it remains the third deadliest cancer worldwide. For patients with early disease, innovative endoscopic and complex surgical techniques have improved survival. However, for patients with advanced disease, there are limited treatment options and survival remains poor. Therefore, there is an urgent need for more effective therapies. Since novel therapies require extensive preclinical testing prior to human trials, it is important to identify methods to expedite this process. Traditional cancer models are restricted by the inability to accurately recapitulate the primary human tumor, exorbitant costs, and the requirement for extended periods of development time. An emerging in vitro model to study human disease is the patient-derived organoid, which is a three-dimensional system created from fresh surgical or biopsy tissues of a patient’s gastric tumor. Organoids are cultured in plastic wells and suspended in a gelatinous matrix, providing a substrate for extension and growth in all dimensions. They are rapid-growing and highly representative of the molecular landscape, histology, and morphology of the various subtypes of GC. Organoids uniquely model tumor initiation and growth, including steps taken by normal stomach cells to transform into invasive, intestinal-type tumor cells. Additionally, they provide ample material for biobanking and screening novel therapies. | 17 | Detection of Drought-Related Loci in Rice at Reproductive Stage Using Selected Introgressed Lines显示文摘The present study was carried out to illustrate high-efficient detection of quantitative trait loci (QTLs) with selected introgression lines (ILs) and the existence of 'hidden genes' conferring drought tolerance (DT). 52 selected DT ILs, derived from BC2F2 population developed by crossing and backcrossing the susceptible recurrent parent (RP) IR64 with the susceptible donor Khazar were planted under irrigation and drought condition. Four important agronomic traits, e.g., grain yield (GY), heading date (HD), panicle numbers per plant (PN), and plant height (PH) were evaluated and 83 SSR polymorphic molecular markers were used for genotypic analysis. Chi-square test based on genetic hitch-hiking and one-way analysis of variances (ANOVA) were used to detect drought-related loci. Nine and 36 loci were detected by chi-square test and one-way ANOVA, respectively. Five common loci were observed by comparing the results of the two methods, among which two QTLs linked with RM7, and RM241 were detected under irrigation condition, both of the favorable alleles were from RP and explained 13% phenotypic variation (PV) for GY and 28% PV for PH, respectively. The other three QTLs linked with RM163, RM18, and RM270 were detected under drought condition, the favorable alleles were all from the donor and explained 10, 24, and 19% PV for HD, PH, and PH, respectively. Five common loci were observed by comparing the results of chi-square test and one-way ANOVA including two QTLs (one for GY and one for PH) under irrigation condition and three QTLs (one for HD and two for PH) under drought condition. By combining phenotypic and genotypic analysis, drought escape could be inferred as the main mechanism for drought tolerance in the present study. | 19 | Pollinator attraction in Cornus capitata (Cornaceae): the relative role of visual and olfactory cues显示文摘Aims it is generally accepted that visual displays and floral scent play important roles in communication between flowering plants and their pollinators.However,the relative role of visual and olfactory cues in pollinator attraction is largely unknown.in this study,we determined the roles of both types of cue in attracting pollinators to Cornus capitata,a medium sized tree with each capitulum sur-rounded by four large,white,petaloid bracts.Methods Pollinator observations and pollination experiments were con-ducted in a natural population;the inflorescences’visual and olfactory signals were characterized by spectral and chemical analyses;the responses of pollinators to visual and olfactory cues were tested using dual-choice behavioural bioassays;the relative roles of visual and olfactory cues in pollinator attrac-tion were tested by comparing the responses of pollinators to inflorescences subjected to three experimental treatments(intact,all bracts removed,and capitulum removed)within the natural population.Important Findings For fruit set,C.capitata is entirely dependent on pollinators,with a bee,Anthophora sp.,being the main pollinator.bracts present high colour distance and green contrast against the leaves.Twelve volatile compounds in the floral scent were detected,most of which have previously been reported to be attractive to a broad spectrum of bee species.behavioural bioassays showed that both,visual cues alone and olfactory cues alone,are attractive to pollinating bees.However,vis-ual cues alone attracted significantly more approaches than olfactory cues alone,while olfactory cues alone elicited a significantly higher landing percentage than visual cues alone.The finding suggests that,in the C.capitata-Anthophora sp.interaction,visual cues are mainly used for location from long distances,while olfactory cues mainly aid landing from short distances.our results indicate that different modalities of floral cues should be considered together to understand fully the communication between flowering plant and pollinators. | Bo Song Yang Niu Jürg Stöcklin Gao Chen De-Li Peng Yong-Qian Gao Hang Sun | 2015 | Journal of Plant Ecology2015,8,2: | 3 |

In the current study, we tested the hypothesis that common non-synonymous polymorphisms in BARD1 are associated with breast cancer susceptibility in a case-control study of 507 patients with incident breast cancer and 539 frequency-matched cancer-free controls in Chinese women. We genotyped all three common(minor allele frequency (MAF) > 0.10) non-synonymous polymorphisms(Pro24Ser, Arg378Ser, and Val507Met) in BARD1. We found that the BARD1 Pro24Ser variant genotypes(24Pro/Ser and 24Ser/Ser) and Arg378Ser variant homozygote 378Ser/Ser were associated with a significantly decreased breast cancer risk, compared with their wild-type homozygotes, respectively. Furthermore, a significant locus-locus interaction was evident between Pro24Ser and Arg378Ser (P-int = 0.032). Among the 378Ser variant allele carriers, the 24Pro/Pro wild-type homozygote was associated with a significantly increased breast cancer risk (adjusted OR = 1.81, 95% CI = 1.11-2.95), but the subjects having 24Pro/Ser or Ser/Ser variant genotypes had a significantly decreased risk(adjusted OR = 0.74, 95% CI = 0.56-0.99). In stratified analysis, this locus-locus interaction was more evident among subjects without family cancer history, those with positive estrogen receptor (ER) and individuals with negative progesterone receptor(PR). These findings indicate that the potentially functional polymorphisms Pro24Ser and Arg378Ser in BARD1 may jointly contribute to the susceptibility of breast cancer. | Huo, X Hu, Z. B Zhai, X.
