| 1 | Effect of Bcl-2 and Bax on survival of side population cells from hepatocellular carcinoma cells显示文摘AIM: To understand the role and significance of side population (SP) cells from hepatocellular carcinoma (HCC) in hepatocarcinogenesis, development, relapse and metastasis, we simulated the denutrition conditions that cancer cells experience in clinical therapy, observed the different anti-apoptosis ability of SP cells and non-SP cells under such conditions, and established the possible effects of P53, Bcl-2 and Bax on survival of SP cells. METHODS: We used ? ow cytometry to analyze and sort the SP and non-SP cells in established HCC lines MHCC97 and hHCC. We evaluated cell proliferation by methyl thiazolyl tetrazolium (MTT) assay and investigated the expression of p53, bcl-2 and bax genes during denutrition, by RT-PCR and immuno? uorescence staining. RESULTS: The percentage of SP cells in the two established HCC lines was 0.25% and 0.5%, respectively. SP cells had greater anti-apoptosis and proliferation ability than non-SP cells. Expression of Bcl-2 and Bax in SP and non-SP cells differed during denutrition. The former was up-regulated in SP cells, and the latter was up-regulated in non-SP cells. CONCLUSION: It may be that different upstream molecules acted and led to different expression levels of Bcl-2 and Bax in these two cell lines. There was a direct relationship between up-regulation of Bcl-2 and down-regulation of Bax and higher anti-apoptosis ability in SP cells. It may be that the existence and activity of SP cells are partly responsible for some of the clinical phenomena which are seen in HCC, such as relapse or metastasis. | 2 | Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. | 4 | Treating chronic hepatitis B virus:Chinese physicians' awareness of the 2010 guidelines显示文摘AIM: To investigate Chinese physicians' awareness of the 2010 guidelines on the treatment of chronic hepatitis B virus(HBV) infection.METHODS: This was a quantitative survey that investigated the characteristics and practices of physicians who were treating patients with hepatitis B, the profile of their patients and physician practices regarding the diagnosis and treatment of HBV at the time of the survey. Participants were randomly selected from available databases of Chinese physicians and requested to complete either an online or paper-based survey. Data from the survey responses were analysed. For data validation and interpretation, qualitative in-depth interviews were conducted with 39 of the respondents.RESULTS: Five-hundred completed surveys, from 663 physicians were available for analysis. A mean of 175 chronic hepatitis B(CHB) patients was seen by each physician every month, of whom 85(49%) were treated in line with therapeutic indications stated in the 2010 guidelines. A total of 444(89%) physicians often(> 60% of the time) adhered to the guidelines. Most physicians used antiviral medications as recommended. For patients with compensated and decompensated cirrhosis, 342(68%) and 336(67%) of physicians, respectively, often followed the recommendation to use potent nucleos(t)ide analogues with a high genetic barrier to resistance, using the appropriate treatment more than 60% of the time. | 5 | Geothermal energy exploitation from depleted high-temperature gas reservoirs by recycling CO_(2): The superiority and existing problems显示文摘CO_(2) can be used as an alternative injectant to exploit geothermal energy from depleted high-temperature gas reservoirs due to its high mobility and unique thermal properties.However,there has been a lack of systematic analysis on the heat mining mechanism and performance of CO_(2),as well as the problems that may occur during geothermal energy exploitation at specific gas reservoir conditions.In this paper,a base numerical simulation model of a typical depleted high-temperature gas reservoir was established to simulate the geothermal energy exploitation processes via recycling CO_(2) and water,with a view to investigate whether and/or at which conditions CO_(2) is more suitable than water for geothermal energy exploitation.The problems that may occur during the CO_(2)-based geothermal energy exploitation were also analyzed along with proposed feasible solutions.The results indicate that,for a depleted low-permeability gas reservoir with dimensions of 1000 m×500 m×50 m and temperature of 150℃ using a single injection-production well group for 40 years of operation,the heat mining rate of CO_(2) can be up to 3.8 MW at a circulation flow rate of 18 kg s^(-1)due to its high mobility along with the flow path in the gas reservoir,while the heat mining rate of water is only about 2 MW due to limitations on the injectivity and mobility.The reservoir physical property and injection-production scheme have some effects on the heat mining rate,but CO_(2)always has better performance than water at most reservoir and operation conditions,even under a high water saturation.The main problems for CO_(2) circulation are wellbore corrosion and salt precipitation that can occur when the reservoir has high water saturation and high salinity,in which serious salt precipitation can reduce formation permeability and result in a decline of CO_(2) heat mining rate (e.g.up to 24%reduction).It is proposed to apply a low-salinity water slug before CO_(2)injection to reduce the damage caused by salt precipitation.For high-permeability gas reservoirs with high water saturation and high salinity,the superiority of CO_(2) as a heat transmission fluid becomes obscure and water injection is recommended. | Guodong Cui Shaoran Ren Bin Dou Fulong Ning | 2021 | Geoscience Frontiers2021,12,6: | 3 |
| 6 | Feasibility and safety of robotic PCI in China: first in man experience in Asia显示文摘Objectives To evaluate the feasibility and safety of a second generation robotic percutaneous coronary intervention(R-PCI)system in China.Background Robotic PCI has been shown to be an effective method for conducting coronary interventions.It has further benefits of more accurate lesion measurement,improved stent deployment,reduced incidence of geographic miss and reduction of operator radiation exposure.Methods This single center evaluation enrolled 10 consecutive patients who had been selected for PCI.Clinical success was defined as residual stenosis<30%and no in-hospital major adverse cardiovascular events.Learning curve effect was assessed by comparing efficiency metrics of early vs.later cases.Results Eleven lesions were treated all successfully without manual interruption or MACE events.Most lesions(63%)were ACC/AHA class B2 and C.Mean procedure time was 57.7±26.4 min,however two procedures were part of live demonstrations.Excluding the two live cases,the mean procedure time was 51.8±23.7 min.Procedural efficiency tended to improve from early cases to later cases based on PCI time(48.3±32.9 vs.25.5±13.0 min,P=0.27),fluoroscopy time(20.3±8.2 vs.12.5±4.6 min,P=0.16),contrast volume(145.0±28.9 vs.102.5±17.1 mL,P=0.05)and Air Kerma dose(1932±978 vs.1007±70 mGy,P=0.31).Conclusions Second generation robotic PCI was safe,effective and there were trends toward improvements in procedural efficiency during this early experience in China. | Ke-Fei DOU Chen-Xi SONG Chao-Wei MU Wei-Xian YANG Cheng-Gang ZHU Lei FENG Jue CHEN Lei SONG Yu NING Bo XU | 2019 | Journal of Geriatric Cardiology2019,16,5: | 2 |
| 7 | Excellent strength-ductility synergy properties of gradient nano-grained structural CrCoNi medium-entropy alloy显示文摘Tailoring heterogeneities could bring out excellent strength-ductility synergy properties.A gradient nanograined(GNG)structure,i.e.grain size range from nanometer(~50 nm)at topest surface layer to micrometer(~1.3μm)at center layer,was successfully introduced into CrCoNi medium-entropy alloy(MEA)by means of high energy shot peening in this work.Experimental results demonstrated that this GNG CrCoNi MEA shows excellent strength and ductility combination properties,exhibiting high yield strength and ultimate tensile strength of~1215 MPa and~1524 MPa,respectively,while remaining a good ductility of~23.0%.The extraordinary hetero-deformation induced(HDI)hardening origins from heterogeneous structure,i.e.GNG structure,which contributes to the majority strength enhancement.Dynamical reinforced heterogeneous structure during tension process results in the enhanced HDI hardening effect,which facilitates excellent ductility and strain hardening capacity at high-level strength.Our work provide not only a feasible and effective way to strengthen the CrCoNi MEA,and other low stacking faults energy(SFE)materials,but also an useful insight to understanding HDI hardening in heterogeneous structure. | Wenjie Lu Xian Luo Dou Ning Miao Wang Chao Yang Miaoquan Li Yanqing Yang Pengtao Li Bin Huang | 2022 | Journal of Materials Science & Technology2022,,17: | 2 |
