1Relationship of hepatitis B virus infection of placental barrier and hepatitis B virus intra-uterine transmission mechanism显示文摘AIM: To explore the mechanism of intra-uterine transmission,the HBV infection status of placental tissue and in vitro cultured placental trophoblastic cells was tested through in vivo and in vitro experiments. METHODS: A variety of methods,such as ELISA,RT- PCR,IHC staining and immunofluorescent staining were employed to test the HBV marker positive pregnant women's placenta and in vitro cultured placental trophoblastic cells. RESULTS: The HBV DNA levels in pregnant women's serum and fetal cord blood were correlated. For those cord blood samples positive for HBV DNA,their maternal blood levels of HBV DNA were at a high level. The HBsAg IHC staining positive cells could be seen in the placental tissues and the presence of HBV DNA detected. After co-incubating the trophoblastic cells and HBV DNA positive serum in vitro,the expressions of both HBsAg and HBV DNA could be detected. 2Randomized controlled trial: Moxibustion and acupuncture for the treatment of Crohn's disease显示文摘AIM: To evaluate the clinical efficacy and safety of acupuncture and moxibustion for the treatment of active Crohn's disease(CD).METHODS: Ninety-two patients were equally and randomly divided into the treatment group and received herb-partitioned moxibustion combined with acupuncture, and the control group received wheat bran-partitioned moxibustion combined with superficial acupuncture. The patients received three treatment sessions per week for 12 wk and were followed up for 24 wk. The main outcome was evaluated using the CD Activity Index(CDAI) score, and the secondary outcomes were evaluated using laboratory indicators such as hemoglobin(HGB), C-reactive protein(CRP), erythrocyte sedimentation rate, quality-of-life, endoscopic ratings, and intestinal histology scores.RESULTS: The CDAI scores of both the treatment and control groups were significantly reduced after treatment compared with those measured before treatment. However, the degree of improvement in the treatment group was significantly greater than that of the control group. The improvement in symptoms in patients of the treatment group was sustained at follow-up, whereas that of the control group was not. The overall efficacy of the treatment was significantly greater than that of the control. Both groups demonstrated significant improvements in quality-of-life ratings after treatment, but the improvement was significantly greater in the treatment group than in the control group. 3Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
4Graphitic carbon nitride (g-C3N4)-based nanosized heteroarrays: Promising materials for photoelectrochemical water splitting显示文摘Photoelectrochemical(PEC)water splitting is recognized as a sustainable strategy for hydrogen generation due to its abundant hydrogen source,utilization of inexhaustible solar energy,high-purity product,and environment-friendly process.To actualize a practical PEC water splitting,it is paramount to develop efficient,stable,safe,and low-cost photoelectrode materials.Recently,graphitic carbon nitride(g-C3N4)has aroused a great interest in the new generation photoelectrode materials because of its unique features,such as suitable band structure for water splitting,a certain range of visible light absorption,nontoxicity,and good stability.Some inherent defects of g-C3N4,however,seriously impair further improvement on PEC performance,including low electronic conductivity,high recombination rate of photogenerated charges,and limited visible light absorption at long wavelength range.Construction of g-C3N4-based nanosized heteroarrays as photoelectrodes has been regarded as a promising strategy to circumvent these inherent limitations and achieve the high-performance PEC water splitting due to the accelerated exciton separation and the reduced combination of photogenerated electrons/holes.Herein,we summarize in detail the latest progress of g-C3N4-based nanosized heteroarrays in PEC water-splitting photoelectrodes.Firstly,the unique advantages of this type of photoelectrodes,including the highly ordered nanoarray architectures and the heterojunctions,are highlighted.Then,different g-C3N4-based nanosized heteroarrays are comprehensively discussed,in terms of their fabrication methods,PEC capacities,and mechanisms,etc.To conclude,the key challenges and possible solutions for future development on g-C3N4-based nanosized heteroarray photoelectrodes are discussed.Liqun Wang Wenping Si Yueyu Tong Feng Hou Daniele Pergolesi Jungang Hou Thomas Lippert Shi Xue Dou Ji Liang 2020Carbon Energy2020,2,2:10
5Dendrobium officinale Kimura et Migo and American ginseng mixture: A Chinese herbal formulation for gut microbiota modulation显示文摘Dendrobium officinale Kimura et Migo(D. officinale) is a famous traditional Chinese medicine(TCM). A mixture of D. officinale and American ginseng has been shown to enhance cell-mediated immunity, humoral immunity, and monocyte/macrophage functions in mice. Here, the effects of a D. officinale and American ginseng mixture on the structure of gut microbial community in dogs were examined using high-throughput 16S rRNA gene amplicon sequencing. The data revealed that while the mixture did not change the diversity of gut microbial community significantly, differences among individuals were significantly reduced. Furthermore, the mixture-responsive operational taxonomic units(OTUs) exhibited a phase-dependent expression pattern. Fifty-five OTUs were found to exhibit a mixture-induced expression pattern, among which one third were short-chain fatty acid(SCFA)-producing genera and the others were probiotic genera included Lactobacillus spp., Sutterella, Alistipes, Anaerovorax, Bilophila, Coprococcus, Gordonibacter, Oscillibacter, among others. By contrast, 36% of the OTUs exhibiting a mixture-repressed expression pattern were disease-associated microorganisms, and six genera, namely Actinomyces, Escherichia/Shigella, Fusobacterium, Slackia, Streptococcus and Solobacterium, were associated with cancer. In addition, five genera were closely associated with diabetes, namely Collinsella,Rothia, Howardella, Slackia and Intestinibacter. 6High molecular karyotype variation revealed in indigenous Elymus nutans in the Qinghai Plateau显示文摘The karyotypes of 27 individuals of Elymus nutans from eight wild populations in the Qinghai Plateau were analyzed using sequential FISH and GISH. High FISH pattern polymorphism and karyotype variation were detected within and among populations. The chromosome variations were mainly characterized as repeat deletions and amplifications along with inter-genomic translocations. The chromosomes of the St and Y genomes demonstrated higher polymorphism than those of the H genome. Six different intergenomic translocations were identified in 33.3% of individuals; type Ⅰ and Ⅱ translocations were detected with higher frequency. Further analysis revealed that type Ⅰ and Ⅱ translocations were distributed in different geographic regions. 7Cordycepin protects renal ischemia/reperfusion injury through regulating inflammation,apoptosis,and oxidative stress显示文摘Cordycepin(3-deoxyadenosine)is a naturally occurring adenosine analog and one of the bioactive constituents isolated from Cordyceps sinensis,species of the fungal genus Cordyceps.It has traditionally been a prized Chinese folk medicine for the human well-being.However,the actions of cordycepin against renal ischemia/reperfusion injury(/R)are still unknown.In the present study,rats were subject to VR and cordycepin was intragastrically administered for seven consecutive days before surgery to investigate the effects and mechanisms of cordycepin against renal/R injury.The test results of kidney and peripheral blood samples of experimental animals showed that cordycepin significantly decreased serum blood urea nitrogen and creatinine levels and markedly attenuated cell injury.Mechanistic studies showed that cordycepin significantly regulated inflammation,apoptosis,and oxidative stress.These data provide new insights for investigating the natural product with the nephroprotective effect against I/R,which should be developed as a new therapeutic agent for the treatment of I/R in the future.Feng Han Meng Dou Yuxiang Wang Cuixiang Xu Yang Li Xiaoming Ding Wujun Xue Jin Zheng Puxun Tian Chenguang Ding 2020Acta Biochimica et Biophysica Sinica2020,52,2:9

10Extracts from Huangqi(Radix Astragali Mongoliciplus) and Ezhu(Rhizoma Curcumae Phaeocaulis) inhibit Lewis lung carcinoma cell growth in a xenograft mouse model by impairing mitogen-activated protein kinase signaling, vascular endothelial growth factor prod显示文摘OBJECTIVE: To study the anti-tumor effects of the extracts from Huangqi(Radix Astragali Mongolici)and Ezhu(Rhizoma Curcumae Phaeocaulis) on the growth of Lewis lung carcinoma(LLC) in a xenograft mouse model and to investigate the possible underlying mechanism.METHODS: LLC tumor-bearing C57 BL/6 mice were treated with normal saline, cisplatin(2 mg/kg intraperitoneally every other day), or Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis)(1∶1, 2∶1, or 3∶1 ratio;5, 8, or 11 g/kg crude drug intragastrically every day) for 15 d.Body weights and tumor volumes were measured every other day. Tumors were excised on day 15 and analyzed. Tumor microvessel density(MVD)was assessed by immunohistochemical staining of CD34;and expression of vascular endothelial cell growth factor(VEGF), the mitogen-activated protein kinases p38 mitogen-activated protein kinase(MAPK), extracellular signal-regulated kinases 1 and 2(ERK1/2), and Jun N-terminal kinase(JNK)and their phosphorylated forms were assessed by Western blotting.RESULTS: Treatment with cisplatin caused a significant loss of body weight compared with controls,whereas Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) extract combinations had no effect. Extracts from Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) significantly decreased tumor weight and tumor MVD compared with controls,and at the 3∶1 treatment group had similar efficacy to cisplatin in reducing MVD. Tumors from Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) treatments also showed decreased p38 MAPK, p-p38 MAPK, ERK1/2, p-ERK1/2,JNK, and p-JNK expression compared with the control group(all P < 0.01). 11Characteristics of volatile compounds removal in biogas slurry of pig manure by ozone oxidation and organic solvents extraction显示文摘Biogas slurry is not suitable for liquid fertilizer due to its high amounts of volatile materials being of complicated composition and peculiar smell. In order to remove volatiles from biogas slurry efficiently, the dynamic headspace and gas chromatography-mass spectrometry were used to clear the composition of volatiles. Nitrogen stripping and superfluous ozone were also used to remove volatiles from biogas slurry. The results showed that there were 21 kinds of volatile compounds in the biogas slurry, including sulfur compounds, organic amines, benzene, halogen generation of hydrocarbons and alkanes, some of which had strong peculiar smell. The volatile compounds in biogas slurry can be removed with the rate of 53.0% by nitrogen stripping and with rate of 81.7% by the oxidization and stripping of the superfluous ozone. On this basis, the removal rate of the volatile compounds reached 99.2% by chloroform and n-hexane extraction, and almost all of odor was eliminated. 12Effect of Modified Xijiao Dihuang Decoction (加味犀角地黄汤) on Intestinal Flora and Th17/Treg in Rats with Radiation Enteritis显示文摘Objective To observe the effect of Modified Xijiao Dihuang Decoction(加味犀角地黄汤,MXDD)on rats with radiation enteritis,and explore its action mechanism.Methods Thirty female Sprague Dawley rats were divided into the control,model,dexamethasone(DXM),golden bifid(GB)and MXDD groups using random number table,6 rats in each group.Except the control group,the other rats were developed into radiation enteritis model by exposing to a single 60Co-γray at a dose of 11 Gy.The rats in the DXM,GB and MXDD groups were treated with DXM(1.425 mg/kg),GB(0.8 g/kg)and MXDD(36.0 g/kg)for 3 days,respectively.Body weight and diarrhea condition of rats were evaluated daily.On day 3,the feces of rats were collected for intestinal flora detection and the small intestinal tissues were also collected.Bacterial species annotation,alpha and beta diversities as well as composition of intestinal flora were detected and compared.The protein and mRNA expressions of interleukin 17(IL-17),retinoid-related orphan nuclear receptor gamma t(ROR-γt)and forkhead/winged helix transcription factor p3(FoxP3)were determined by Western blot and polymerase chain reaction,respectively.The abundance and diversity of intestinal flora as well as the proportion at the phylum and genus levels were assayed by 16S rRNA metagenome sequencing.Correlation between intestinal flora and Th17/Treg was analyzed by heatmap method.Results On day 1 to 3 after radiation,compared with the control group,the body weight in model group was decreased(P<0.05 or P<0.01).Compared with the model group,MXDD could alleviate weight loss and diarrhea caused by irradiation.At the phylum level,MXDD cause a significant increase in Firmicutes,and a decrease in Proteobacteria(P<0.05 or P<0.01).At the genus level,MXDD reduced the proportion of Escherichia Shigella(P<0.01).In addition,IL-17 and FoxP3 mRNA and protein expression levels were down-regulated and ROR-γt was up-regulated by MXDD treatment(P<0.05).Besides,Firmicutes and Lactobacillus were positively correlated with FoxP3(r=0.73,0.79,respectively;P<0.01),negatively correlated with IL-17(r=0.66,0.64,respectively;P<0.01 or P<0.05)and ROR-γt(r0.73,0.81,respectively;P<0.01).Proteobacteria and Escherichia Shigella both had positive correlation with IL-17(r 0.77,0.57,respectively;P<0.01 or P<0.05)and ROR-γt(r=0.94,0.79,respectively;P<0.01)and negative correlation with FoxP3(r0.74,0.65;P<0.01).Conclusion MXDD could improve the survival status of irradiated rats by regulating the richness,diversity and composition of intestinal flora,and restoring the balance of Th17/Treg.WANG Yu-guo GAO Yue FENG Jian DOU Yong-qi 2021Chinese Journal of Integrative Medicine2021,27,3:6

14Long non-coding RNA HOTAIRM1 promotes proliferation and inhibits apoptosis of glioma cells by regulating the miR-873-5p/ZEB2 axis显示文摘Background:Glioblastoma is one of the most common malignant brain tumors.Conventional clinical treatment of glioblastoma is not sufficient,and the molecular mechanism underlying the initiation and development of this disease remains unclear.Our study aimed to explore the expression and function of miR-873a-5p in glioblastoma and related molecular mechanism.Methods:We analyzed the most dysregulated microRNAs from the Gene Expression Omnibus(GEO)database and examined the expression of miR-873-5p in 20 glioblastoma tissues compared with ten normal brain tissues collected in the Zhejiang Tongde Hospital.We then overexpressed or inhibited miR-873-5p expression in U87 glioblastoma cell lines and analyzed the phenotype using the cell counting kit-8 assay,wound healing assay,and apoptosis.In addition,we predicted upstream and downstream genes of miR-873-5p in glioblastoma using bioinformatics analysis and tested our hypothesis in U87 cells using the luciferase reporter gene assay and Western blotting assay.The differences between two groups were analyzed by Student’s t test.The Kruskal-Wallis test was used for the comparison of multiple groups.A P<0.05 was considered to be significant.Results:The miR-873-5p was downregulated in glioblastoma tissues compared with that in normal brain tissues(normal vs.tumor,0.762±0.231 vs.0.378±0.114,t=4.540,P<0.01).Overexpression of miR-873-5p inhibited cell growth(t=6.095,P<0.01)and migration(t=3.142,P<0.01)and promoted cell apoptosis(t=4.861,P<0.01),while inhibition of miR-873-5p had the opposite effect.Mechanistically,the long non-coding RNA HOTAIRM1 was found to act as a sponge of miR-873-5p to activate ZEB2 expression in U87 cells.Conclusions:We uncovered a novel HOTAIRM1/miR-873-5p/ZEB2 axis in glioblastoma cells,providing new insight into glioblastoma progression and a theoretical basis for the treatment of glioblastoma.Yi-Hai Lin Liang Guo Feng Yan Zhang-Qi Dou Qian Yu Gao Chen 2020Chinese Medical Journal2020,,2:5

16Monitoring elevation change of glaciers on Geladandong Mountain using Tan DEM-X SAR interferometry显示文摘Glaciers play an important role in the climate system. The elevation change of a glacier is an important parameter in studies of glacier dynamics. Only a few ground-based measurements of high mountain glaciers are available due to their remoteness, high elevation, and complex topography. The acquisition from the German Tan DEM-X(Terra SAR-X add-on for Digital Elevation Measurement) SAR imaging configuration provides a reliable data sources for studying the elevation change of glaciers. In this study, the bistatic Tan DEM-X data that cover the Geladandong Mountain on the Tibetan Plateau were processed with SAR interferometry technique and the elevation changes of the mountain's glaciers during 2000–2014 were obtained. The results indicated that although distinct positive and negative elevation changes were found for different glacier tongues, the mean elevation change was about-0.14±0.26 m a-1. Geoscience Laser Altimeter System(GLAS) data were obtained for comparison and verification. The investigation using GLAS data demonstrated the efficacy of the proposed method in determining glacier elevation change. 17Multi-scale validation strategy for satellite albedo products and its uncertainty analysis显示文摘Coarse-resolution satellite albedo products are important for climate change and energy balance research because of their capability to characterize the spatiotemporal patterns of land surface parameters at both the regional and global scales. The accuracy of coarse-resolution products is usually assessed via comparison with in situ measurements. The key issue in the comparison of remote sensing observations with in situ measurements is scaling and uncertainty. This paper presents a strategy for validating 1-km-resolution remote sensing albedo products using field measurements and high-resolution remote sensing observations. Field measurements were collected to calibrate the high-resolution(30 m) albedo products derived from HJ-1a/b images. Then, the calibrated high-resolution albedo maps were resampled(i.e., upscaled) to assess the accuracy of the coarse-resolution albedo products. The samples of field measurements and high-resolution pixels are based on an uncertainty analysis. Two types of coarse-resolution albedo datasets, from global land surface satellite(GLASS) and moderate-resolution imaging spectroradiometer(MODIS), are validated over the middle reaches of the Heihe River in China. The results indicate that the upscaled HJ(Huan Jing means environment in Chinese and this refers to a satellite constellation designed for environment and disaster monitoring by China) albedo, which was calibrated using field measurements, can provide accurate reference values for validating coarse-resolution satellite albedo products. 18Endothelial cells-targeted soluble human Delta-like 4 suppresses both physiological and pathological ocular angiogenesis显示文摘Due to its essential roles in angiogenesis, Notch pathway has emerged as an attractive target for the treatment of pathologic angiogenesis. Although both activation and blockage of Notch signal can impede angiogenesis, activation of Notch signal may be more promising because it was shown that long-term Notch signal blockage resulted in vessel neoplasm. However, an in vivo deliverable Notch ligand with highly efficient Notch-activating capacity has not been developed. Among all the Notch ligands, Delta-like4(Dll4) is specifically involved in angiogenesis. In this study, we generated a novel soluble Notch ligand h D4 R, which consists of the Delta-Serrate-Lag-2 fragment of human Dll4 and an arginine-glycine-aspartate(RGD) motif targeting endothelial cells(ECs). We demonstrated that h D4 R could bind to ECs through its RGD motif and effectively triggered Notch signaling in ECs. Further, we confirmed that h D4 R could suppress angiogenesis in vitro as manifested by network formation assay and sprouting assay. More importantly, h D4 R efficiently repressed neonatal retinal angiogenesis and laser-induced choroidal neovascularization(CNV) as well in vivo. 20Ru-doped phosphorene for electrochemical ammonia synthesis显示文摘The electrochemical ammonia synthesis has attracted increasing attention due to its energy saving characteristics.However,developing novel electrocatalysts and their mechanism remain great challenges.Here,several transition metal(TM)atoms doped on phosphorene were studied as N2 fixation electrocatalysts by using density functional theory(DFT)calculations.The results demonstrate that single Ru atom doped phosphorene shows an excellent catalytic activity for ammonia synthesis via the enzymatic pattern.A small overpotential of 0.696 V is achieved for this process.The effect of oxidation in the catalyst was also discussed in our work.Oxidation deactivates the catalyst,which should be avoided in the experiment.Our outcomes offer a novel perspective for single-atom catalytic ammonia synthesis with phosphorene as a substrate.Jian-Dong Liu Zeng-Xi Wei Yu-Hai Dou Yue-Zhan Feng Jian-Min Ma 2020Rare Metals2020,39,8:4
