2Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL.Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen 2023Cancer Communications2023,43,2:3
3Host HDAC4 regulates the antiviral response by inhibiting the phosphorylation of IRF3显示文摘Class II HDACs, such as HDAC4, are critical regulators of the immune response in various immune cells;however, its role in innate immunity remains largely unknown.Here, we report that the overexpression of HDAC4 suppresses the production of type I interferons triggered by pattern-recognition receptors (PRRs). HDAC4 repressed the translocation of transcription factor IRF3 to the nucleus, thereby decreasing IRF3-mediated IFN-β expression. In particular, we also determined that HDAC4 can be phosphorylated and simultaneously block the phosphorylation of IRF3 at Ser386 and Ser396 by TBK1 and IKKε, respectively, by interacting with the kinase domain of TBK1 and IKKε. Furthermore, IFN-β may stimulate the expression of HDAC4. 4Histone Deacetylase 3 Inhibitor Suppresses Hepatitis C Virus Replication by Regulating Apo-A1 and LEAP-1 Expression显示文摘Histone deacetylase (HDAC) inhibitors show clinical promise for the treatment of cancers, including hepatocellular carcinoma (HCC). In this study, we investigated the effect of HDAC inhibitor treatment on hepatitis C virus (HCV)replication in Huh7 human liver cells and in a mouse model of HCV infection. Viral replication was markedly suppressed by the HDAC3 inhibitor at concentrations below 1 mmol/L, with no cellular toxicity. This was accompanied by upregulation of liver-expressed antimicrobial peptide 1 (LEAP-1) and downregulation of apolipoprotein-A1 (Apo-A1), as determined by microarray and quantitative RT-PCR analyses. Moreover, HDAC3 was found to modulate the binding of CCAAT-enhancer-binding protein a (C/EBPa), hypoxia-inducible factor 1 a (HIF1 a), and signal transducer and activator of transcription 3 (STAT3) to the LEAP-1 promoter. HDAC3 inhibitor treatment also blocked HCV replication in a mouse model of HCV infection. 7Vector Control of the Single-Phase Inverter Based on the Extended and Virtual Circuits显示文摘A vector control based on the extended equivalent circuit and virtual circuits is proposed for the single-phase inverter.By the extended circuit,the other two phase voltages can be extended by the output voltage of the single-phase inverter so as to construct the voltage vector.The voltage outer-loop is to control the voltage vector in dq coordinate system,and the output voltage can track the target value without deviation in steady state.By designing the virtual circuit,the voltage inner-loop can achieve approximate decoupling and improve the dynamic response under the changeable load.Compared with the traditional dual closed-loop control,the proposed dual closed-loop control scheme only needs to detect and control the voltage without the current.It not only can achieve good control effect,but also reduce the complexity of the hardware.Finally,the simulation and experimental results show that the single-phase inverter has good static and dynamic characteristics regardless of stable load or changeable load.Tao Xu Ming Yang Naizhe Diao Xianrui Sun Suliang Wu Zhuangzhuang Shen Hongzhi Chen Chonghui Song 2018CES Transactions on Electrical Machines and Systems2018,2,3:0
8Smart all-time vision:The battery-free video communication for urban administration and law enforcement显示文摘The Chinese government is dedicated to enhancing the level of informatization in administrative law enforcement to ensure fairness and increase credibility.Currently,law enforcement has exposed such problems as insufficient force,difficulties in collecting evidence,and low evidential power.These problems contradict the principles of rule of law and standardization.Many local governments have implemented advanced information technologies for urban administration and law enforcement,including big data,artificial intelligence,and IoT.Among these technologies,IoT is the core communication infrastructure for innovative law enforcement platforms.However,traditional video communications rely on batteries or dedicated power sources,leading to maintenance challenges and high power consumption.In this paper,we propose AT-Eye,a new video communication paradigm for all-time law enforcement video monitoring,which is battery-free and high-throughput.The key enabler of AT-Eye is ambient backscatter technology,which enables energy harvesting and video communication simultaneously.Unlike previous methods,our proposal introduces a novel CCK-based modulation for tag cameras and achieves 4-way concurrency.The modulation is simple because it uses phase difference and it is energyefficient because generating square waves is adequate.Moreover,to ensure full compatibility with COTS WiFi,we modulate the physical service data unit.Therefore,data recovery is possible using only commercial NICs.We conduct comprehensive experiments to examine our proposal and experiment results show that AT-Eye achieves a throughput of 10.8 Mbps with COTS radios.We also simulate a 16-way battery-free tag camera system,demonstrating AT-Eye’s feasibility of high-definition video communication.Yajie He Chonghui Xiao Shuai Wang Yun-Hao Liu Zhaoyuan Xu Haoquan Zhou Min Zhao Wei Gong 2023Digital Communications and Networks2023,9,6:0
9Overcoming Debye length limitations:Three-dimensional wrinkled graphene field-effect transistor for ultra-sensitive adenosine triphosphate detection显示文摘Adenosine triphosphate(ATP)is closely related to the pathogenesis of certain diseases,so the detection of trace ATP is of great significance to disease diagnosis and drug development.Graphene field-effect transistors(GFETs)have been proven to be a promising platform for the rapid and accurate detection of small molecules,while the Debye shielding limits the sensitive detection in real samples.Here,a three-dimensional wrinkled graphene field-effect transistor(3D WG-FET)biosensor for ultra-sensitive detection of ATP is demonstrated.The lowest detection limit of 3D WG-FET for analyzing ATP is down to 3.01 aM,which is much lower than the reported results.In addition,the 3D WG-FET biosensor shows a good linear electrical response to ATP concentrations in a broad range of detection from 10 aM to 10 pM.Meanwhile,we achieved ultra-sensitive(LOD:10 aM)and quantitative(range from 10 aM to 100 fM)measurements of ATP in human serum.The 3D WG-FET also exhibits high specificity.This work may provide a novel approach to improve the sensitivity for the detection of ATP in complex biological matrix,showing a broad application value for early clinical diagnosis and food health monitoring.Yue Ding Chonghui Li Meng Tian Jihua Wang Zhenxing Wang Xiaohui Lin Guofeng Liu Wanling Cui Xuefan Qi Siyu Li Weiwei Yue Shicai Xu 2023Frontiers of physics2023,18,5:0
10Development of a novel virus-like particle-based vaccine for preventing tick-borne encephalitis virus infection显示文摘Tick-borne encephalitis virus(TBEV)is an important tick-borne pathogen that poses as a serious public health concern.The coverage and immunogenicity of the currently available vaccines against TBEV are relatively low;therefore,it is crucial to develop novel and effective vaccines against TBEV.The present study describes a novel strategy for the assembly of virus-like particles(VLPs)by co-expressing the structural(core/prM/E)and non-structural(NS2B/NS3Pro)proteins of TBEV.The efficacy of the VLPs was subsequently evaluated in C57BL/6 mice,and the resultant IgG serum could neutralize both Far-Eastern and European subtypes of TBEV.These findings indicated that the VLP-based vaccine elicited the production of cross-subtype reactive antibodies.The VLPs provided protection to mice lacking the type I interferon receptor(IFNAR^(-/-))against lethal TBEV challenge,with undetectable viral load in brain and intestinal tissues.Furthermore,the group that received the VLP vaccine did not exhibit significant pathological changes and the inflammatory factors were significantly suppressed compared to the control group.Immunization with the VLP vaccine induced the production of multiple-cytokine-producing antiviral CD4+T cells in vivo,including TNF-α^(+),IL-2^(+),and IFN-γ^(+)T cells.Altogether,the findings suggest that noninfectious VLPs can serve as a potentially safe and effective vaccine candidate against diverse subtypes of TBEV.Jielin Tang Muqing Fu Chonghui Xu Bao Xue Anqi Zhou Sijie Chen He Zhao Yuan Zhou Jizheng Chen Qi Yang Xinwen Chen 2023Virologica Sinica2023,38,5:0

12Comparison of viral propagation and drug response among SARS-CoV-2 VOCs using replicons capable of recapitulating virion assembly and release显示文摘Several variants of concern(VOCs)have emerged since the WIV04 strain of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)was first isolated in January 2020.Due to mutations in the spike(S)protein,these VOCs have evolved to enhance viral infectivity and immune evasion.However,whether mutations of the other viral proteins lead to altered viral propagation and drug resistance remains obscure.The replicon is a noninfectious viral surrogate capable of recapitulating certain steps of the viral life cycle.Although several SARS-CoV-2 replicons have been developed,none of them were derived from emerging VOCs and could only recapitulate viral genome replication and subgenomic RNA(sgRNA)transcription.In this study,SARS-CoV-2 replicons derived from the WIV04 strain and two VOCs(the Beta and Delta variants)were prepared by removing the S gene from their genomes,while other structural genes remained untouched.These replicons not only recapitulate viral genome replication and sgRNA transcription but also support the assembly and release of viral-like particles,as manifested by electron microscopic assays.Thus,the S-deletion replicon could recapitulate virtually all the post-entry steps of the viral life cycle and provides a versatile tool for measuring viral intracellular propagation and screening novel antiviral drugs,including inhibitors of virion assembly and release.Through the quantification of replicon RNA released into the supernatant,we demonstrate that viral intracellular propagation and drug response to remdesivir have not yet substantially changed during the evolution of SARS-CoV-2 from the WIV04 strain to the Beta and Delta VOCs.Lingqian Tian Qiuhong Liu Rongjuan Pei Yingshan Chen Chonghui Xu Jielin Tang Hao Sun Kunpeng Liu Qi Yang Lei Yang Leshan Li Yongli Zhang Yuan Zhou Chao Shan Xue Hu Xinwen Chen Yun Wang 2022Virologica Sinica2022,37,5:0
13Histone deacetylase 4 inhibits NF-κB activation by facilitating IκBα sumoylation显示文摘Protein modification by small ubiquitin-like modifier(SUMO)is an important regulatory mechanism for multiple cellular pro-cesses.Although the canonical pathway involving the ubiquitylation or phosphorylation of IκBα has been well characterized,little is known about the sumoylation of IkBa in the control of NF-κB activity.Here,we find that histone deacetylase 4(HDAC4)negatively regulates tumor necrosis factor-alpha-or lipopolysaccharide-triggered NF-κB activation.HDAC4 belongs to the SUMO E3 ligase family and can directly sumoylate IκBα.The cytoplasm location of HDAC4 is essential for IκBαsumoylation.The Cys292 of HDAC4 is a key site for its SUMO E3 ligase activity.The sumoylation of IkBc prevents its polyubiquitination and degradation be-cause these two modifications occur both at the Lys21.Our findings reveal a previously undiscovered role for HDAC4 in the inflammatory response as a SUMO E3 ligase for IκBαsumoylation.Our work provides insight into mechanisms ensuring optimal mediation of the NF-κB pathway.Qi Yang Jielin Tang Chonghui Xu He Zhao Yuan Zhou Yanyi Wang Min Yang Xinwen Chen Jizheng Chen 2020Journal of Molecular Cell Biology2020,12,12:0
14Aero-Elastic Flutter Characteristics of Laminated Composite Panel Embedded with Shape Memory Alloy Wires显示文摘The effects of shape memory alloy( SMA) wires on the natural frequency,flutter boundary and amplitude of limit cycle oscillation( LCO) of a laminated composite panel are investigated. The classical plate theory as well as von-Karman strain-displacement relation are used to formulate the nonlinear dynamic model of the smart laminated panel. The aerodynamic loadings are simulated by the third order piston theory. The thermomechanical behavior of SMA wires is estimated according to one-dimensional Brinson SMA model.
