2Isolation and characterization of ferrous-and sulfur-oxidizing bacteria from Tengchong solfataric region,China显示文摘Microbial oxidation and reduction of iron and sulfur are important parts of biogeochemical cycles in acidic environments such as geothermal solfataric regions.Species of Acidithiobacillus and Leptospirillum are the common ferrous-iron and sulfur oxidizers from such environments.This study focused on the Tengchong sofataric region,located in Yunnan Province,Southwest China.Based on cultivation,9 strains that grow on ferrous-iron and sulfuric compounds were obtained.Analysis of 16S rRNA genes of the 9 strains indicated that they were affliated to Acidithiobacillus,Alicyclobacillus,Sulfobacillus,Leptospirillum and Acidiphilium.Physiological and phylogenetic studies indicated that two strains(TC-34 and TC-71) might represent two novel members of Alicyclobacillus.Strain TC-34 and TC-71 showed 94.8%-97.1%16S rRNA gene identities to other species of Alicyclobacillus.Different from the previously described Alicyclobacillus species,strains TC-34 and TC-71 were mesophilic and their cellular fatty acids do not containω-cyclic fatty acids.Strain TC-71 was obligately dependent on ferrous-iron for growth.It was concluded that the ferrous-iron oxidizers were diversified and Alicyclobacillus species were proposed to take part in biochemical geocycling of iron in the Tengchong solfataric region.JIANG Chengying LIU Ying LIU Yanyang GUO Xu LIU Shuang-Jiang 2009Journal of Environmental Sciences2009,21,9:5
3Proteomic and molecular investigations revealed that Acidithiobacillus caldus adopts multiple strategies for adaptation to NaCl stress显示文摘Acidithiobacillus caldus plays an important role in commercial bioleaching.To understand how NaCl stress adaptation occurs in A.caldus,we grew A.caldus strain SM-1 in media containing high NaCl concentrations.SM-1 grew at concentrations of up to 1.0-mol L^(-1)NaCl,but growth was severely inhibited at higher concentrations.Proteomic analysis showed that SM-1 used multiple strategies to respond to NaCl stress.In addition to several heatshock proteins,enzymes involved in proline biosynthesis increased under NaCl stress.In addition,two DNA-binding proteins and a third protein of unknown function(Atc_(1291)),which was subsequently identified as a putative single-stranded DNA-binding protein,were up-regulated in the presence of NaCl stress.These DNA-binding proteins might play a role in response to osmotic stress.Atc_(1291)was cloned and expressed in Escherichia coli.Surprisingly,we found that E.coli BL21/pET28a-atc_(1291)grew to higher cell densities than E.coli BL21/pET28a,regardless of NaCI stress.Homologs to Atc_(1291)were identified in several groups of Proleohacleria.The role of Atc_(1291)in enhancing cell growth needs further investigation.Xu Guo Chengying Jiang Yuanming Luo Mingjiang Zhang Ansgar Poetsch Shuangjiang Liu 2014Chinese Science Bulletin2014,59,3:2

8ISOLATION AND CHARACTERIZATION OF BACTERIA FOR REMOVAL OF SULFUR IN ORGANIC COMPOUNDS显示文摘Four bacterial strains have been isolated for their ability to growth on dibenzothiophene (DBT), dibenzothiophene sulfone (DBTO2). The results show that different bacteria strains oxidize the DBT to different products through different pathways. R-6 and R-16 metabolize DBT to DBTO2 and HBP; they are identified as species of Bacillus brevis and Bacillus sphaericus. For long time storage, R-9 identified as Nocardia globerula oxidizes DBT to DBT-sulfone (DBTO2) and benzenamine n-phenyl. The optimum temperature and pH for growth of R-16 are 32℃ and 7.02 respectively. 10The monkey microbial biobank brings previously uncultivated bioresources for nonhuman primate and human gut microbiomes显示文摘Impact statement Nonhuman primates(NHPs)such as monkeys are the closest living relatives to humans and are the best available models for causative studies of human health and diseases.Gut microbiomes are intensively involved in host health.In this study,by large-scale cultivation of microbes from fecal samples of monkeys,we obtained previously uncultured bacterial species and constructed a Macaca fascicularis Gut Microbial Biobank(MfGMB).The MfGMB consisted of 250 strains that represent 97 species of 63 genera,25 families,and 4 phyla.The information of the 250 strains and the genomes of 97 cultured species are publicly accessible.The MfGMB represented nearly 50% of core gut microbial compositions at the genus level and covered over 80% of the KO-based known gut microbiome functions of M.fascicularis.Data mining showed that the bacterial species in the MfGMB were prevalent not only in NHPs gut microbiomes but also in human gut microbiomes.This study will help the understanding and future investigations on how gut microbiomes interact with their mammalian hosts.Danhua Li Chang Liu Rexiding Abuduaini Mengxuan Du Yujing Wang Haizhen Zhu Honghe Chen Nan Zhou Yuhua Xin Linhuan Wu Juncai Ma Yuguang Zhou Yong Lu Chengying Jiang Qiang Sun Shuang-Jiang Liu 2022mLife2022,1,2:0
11Intracavity spatiotemporal metasurfaces显示文摘Optical metasurfaces are endowed with unparallel flexibility to manipulate the light field with a subwavelength spatial resolution.Coupling metasurfaces to materials with strong optical nonlinearity may allow ultrafast spatiotemporal light field modulation.However,most metasurfaces demonstrated thus far are linear devices.Here,we experimentally demonstrate simultaneous spatiotemporal laser mode control using a single-layer plasmonic metasurface strongly coupled to an epsilon-near-zero(ENZ)material within a fiber laser cavity.While the geometric phase of the metasurface is utilized to convert the laser’s transverse mode from a Gaussian beam to a vortex beam carrying orbital angular momentum,the giant nonlinear saturable absorption of the ENZ material enables pulsed laser generation via the Q-switching process.The direct integration of a spatiotemporal metasurface in a laser cavity may pave the way for the development of miniaturized laser sources with tailored spatial and temporal profiles,which can be useful for numerous applications,such as superresolution imaging,high-density optical storage,and three-dimensional laser lithography.Wenhe Jia Chenxin Gao Yongmin Zhao Liu Li Shun Wen Shuai Wang Chengying Bao Chunping Jiang Changxi Yang Yuanmu Yang 2023Advanced Photonics2023,5,2:0
12Site-directed mutagenesis reveals new and essential elements for iron-coordination of the sulfur oxygenase reductase from the acidothermophilic Acidianus tengchongensis显示文摘Previous study on refolding of sulfur oxygenase reductase (SOR) inclusion bodies from recombinant Escherichia coli showed that iron was critical to the activity of the SOR from Acidianus ambivalens. In this study, enzymatic assays showed that 2,2′-Dipyridyl, Tiron and 8-hydroxyquinoline, which are specific for chelating ferrous or ferric ions, strongly inhibited the activity of SOR from A. tengchongensis, suggesting that iron atom is essential for SOR activity. Alignment of several functionally identified SORs and SOR-like sequences from genome database revealed a conserved, putative iron binding motif, H86-X3-H90-Xn-E114-Xn-E129 (numbering according to the Acidianus tengchongensis SOR sequence). Three mutants of SOR were generated by site-directed mutagenesis of H86, H90 and E129 into phenyla-lanine or alanine residue in this study. Circular dichroism spectrum determination indicated that there was no change of the secondary structures of mutant SORs, H86F, H90F and E129A, but all mutants were completely inactive. Through determination of iron contents we found that SOR mutants of H86F, H90F and E129A completely or partially lost iron, while mutants of C31S, C101S, and C104S (generated in a previous study) did not. This result indicated that H86, H90 and E129 but not C31, C101, and C104 were involved in binding to iron atom. Based on this and previous studies, it is proposed that the conserved motifs, C31-Xn-C101-X2-C104 and H86-X3-H90-X23-E114-X14-(E/D)129, are respectively for sulfur and molecular oxygen binding and activation.
