| 3 | SUMO E3 Ligase AtMMS21 Regulates Drought Tolerance in Arabidopsis thaliana显示文摘Post-translational modifications of proteins by small ubiquitin-like modifiers (SUMOs) play crucial roles in plant growth and development, and in stress responses. The MMS21 is a newly-identified Arabidopsis thaliana L. SUMO E3 ligase gene aside from the SIZ1, and its function requires further elucidation. Here, we show that MMS21 deficient plants display improved drought tolerance, and constitutive expression of MMS21 reduces drought tolerance. The expression of MMS21 was reduced by abscisic acid (ABA), polyethylene glycol (PEG) or drought stress. Under drought conditions, mms21 mutants showed the highest survival rate and the slowest water loss, and accumulated a higher level of free proline compared to wild-type (WT) and MMS21 over-expression plants. Stomatal aperture, seed germination and cotyledon greening analysis indicated that mms21 was hypersensitive to ABA. Molecular genetic analysis revealed that MMS21 deficiency led to elevated expression of a series of ABA-mediated stress-responsive genes, including COR15A, RD22, and P5CS1 The ABA and drought-induced stress-responsive genes, including RAB18, RD29A and RD29B, were inhibited by constitutive expression of MMS21. Moreover, ABA-induced accumulation of SUMO-protein conjugates was blocked in the mms21 mutant. | 5 | Physical model test and numerical simulation on the failure mechanism of the roadway in layered soft rocks显示文摘To explore the failure mechanism of roadway in layered soft rocks,a physical model with the physically finite elemental slab assemblage(PFESA)method was established.Infrared thermography and a video camera were employed to capture thermal responses and deformation.The model results showed that layered soft roadway suffered from large deformation.A three-dimensional distinct element code(3 DEC)model with tetrahedral blocks was built to capture the characteristics of roadway deformation,stress,and cracks.The results showed two failure patterns,layer bending fracture and layer slipping after excavation.The layer bending fracture occurred at positions where the normal direction of layers pointed to the inside of the roadway and the layer slipping occurred in the ribs.Six schemes were proposed to investigate the effects of layered soft rocks.The results showed that the deformation of ribs was obviously larger than that of the roof and floor when the roadway passed through three types of strata.When the roadway was completely in a coal seam,the change of deformation in ribs was not obvious,while the deformation in the roof and floor increased obviously.These results can provide guidance for excavation and support design of roadways in layered soft rocks. | Xiaoming Sun Chengwei Zhao Yong Zhang Feng Chen Shangkun Zhang Kaiyuan Zhang | 2021 | International Journal of Mining Science and Technology2021,31,2: | 9 |
| 6 | Prediction of Cross-Tension Strength of Self-Piercing Riveted Joints Using Finite Element Simulation and XGBoost Algorithm显示文摘Self-piercing riveting(SPR)has been widely used in automobile industry,and the strength prediction of SPR joints always attracts the attention of researchers.In this work,a prediction method of the cross-tension strength of SPR joints was proposed on the basis of finite element(FE)simulation and extreme gradient boosting decision tree(XGBoost)algorithm.An FE model of SPR process was established to simulate the plastic deformations of rivet and substrate materials and verified in terms of cross-sectional dimensions of SPR joints.The residual mechanical field from SPR process simulation was imported into a 2D FE model for the cross-tension testing simulation of SPR joints,and cross-tension strengths from FE simulation show a good consistence with the experiment result.Based on the verified FE model,the mechanical properties and thickness of substrate materials were varied and then used for FE simulation to obtain cross-tension strengths of a number of SPR joints,which were used to train the regression model based on the XGBoost algorithm in order to achieve prediction for cross-tension strength of SPR joints.Results show that the cross-tension strengths of SPR steel/aluminum joints could be successfully predicted by the XGBoost regression model with a respective error less than 7.6%compared to experimental values. | Jianping Lin Chengwei Qi Hailang Wan Junying Min Jiajie Chen Kai Zhang Li Zhang | 2021 | Chinese Journal of Mechanical Engineering2021,34,2: | 8 |

| 8 | Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. | 10 | 3-year Treatment of Tenofovir Alafenamide vs.Tenofovir Disoproxil Fumarate for Chronic HBV Infection in China显示文摘Background and Aims:Tenofovir alafenamide(TAF)has similar efficacy to tenofovir disoproxil fumarate(TDF)but with improved renal and bone safety in chronic hepatitis B patients studied outside of China.We report 3-year results from two phase 3 studies with TAF in China(Clinicaltrials.gov:NCT02836249 and NCT02836236).Methods:Chinese hepatitis B e antigen(HBeAg)-positive and-negative chronic hepatitis B patients with viremia and elevated alanine aminotransferase were randomized 2:1 to TAF or TDF treatment groups and treated in a double-blind fashion for 144 weeks(3 years).Efficacy responses were assessed by individual study while safety was assessed by a pooled analysis.Results:Of the 334 patients(180 HBeAg-positive and 154 HBeAg-negative)randomized and treated,baseline characteristics were similar between groups.The overall mean age was 38 years and 73%were male.The mean HBV DNA was 6.4 log10 IU/mL.The median alanine aminotransferase was 88 U/L,and 37%had a history of antiviral use.At week 144,the proportion with HBV DNA<29 IU/mL was similar among the two groups,with TAF at 83%vs.TDF at 79%,and TAF at 93%vs.TDF at 92%for the HBeAg-positive and-negative patients,respectively.In each study,higher proportions of TAF than TDF patients showed normalized alanine aminotransferase(via the American Association for the Study of Liver Diseases and the China criteria)and showed loss of HBsAg;meanwhile,the HBeAg seroconversion rates were similar.Treatment was well-tolerated among the TAF patients,who showed a smaller median decline in creatinine clearance(−0.4 vs.−3.2 mL/min;p=0.014)and less percentage change in bone mineral density vs.TDF at hip(−0.95%vs.−1.93%)and spine(+0.35%vs.−1.40%).Conclusions:In chronic hepatitis B patients from China,TAF treatment provided efficacy similar to TDF but with better renal and bone safety at 3 years. | Jinlin Hou Qin Ning Zhongping Duan You Chen Qing Xie Fu-Sheng Wang Lunli Zhang Shanming Wu Hong Tang Jun Li Feng Lin Yongfeng Yang Guozhong Gong John FFlaherty Anuj Gaggar Shuyuan Mo Cong Cheng Gregory Camus Chengwei Chen Yan Huang Jidong Jia Mingxiang Zhang GS-US-320-0110 and GS-US-320-0108 China Investigators | 2021 | Journal of Clinical and Translational Hepatology2021,9,3: | 7 |

| 15 | Advanced multiple response surface method of sensitivity analysis for turbine blisk reliability with multi-physics coupling显示文摘To reasonably implement the reliability analysis and describe the significance of influencing parameters for the multi-failure modes of turbine blisk, advanced multiple response surface method(AMRSM) was proposed for multi-failure mode sensitivity analysis for reliability. The mathematical model of AMRSM was established and the basic principle of multi-failure mode sensitivity analysis for reliability with AMRSM was given. The important parameters of turbine blisk failures are obtained by the multi-failure mode sensitivity analysis of turbine blisk. Through the reliability sensitivity analyses of multiple failure modes(deformation, stress and strain) with the proposed method considering fluid–thermal–solid interaction, it is shown that the comprehensive reliability of turbine blisk is 0.9931 when the allowable deformation, stress and strain are3.7*10^(-3)m, 1.0023*10~9 Pa and 1.05*10^(-2)m/m, respectively; the main impact factors of turbine blisk failure are gas velocity, gas temperature and rotational speed. As demonstrated in the comparison of methods(Monte Carlo(MC) method, traditional response surface method(RSM), multiple response surface method(MRSM) and AMRSM), the proposed AMRSM improves computational efficiency with acceptable computational accuracy. | 18 | Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. | 19 | Voltage-gated sodium channel Nav1.5 promotes proliferation, migration and invasion of oral squamous cell carcinoma显示文摘The protein voltage-gated sodium channel Nav1.5 is highly upregulated in various types of cancer and, in general, promotes cancer cell invasiveness and metastatic progression. A previous study found that Nav1.5 was highly expressed in poorly differentiated oral squamous cell carci noma (OSCC). However, whether Nav1.5 enhances invasiveness and metastasis of OSCC are still unknown. In this study, we found that Nav1.5 was highly expressed in OSCC cell lines compared with normal oral keratinocyte HOK cell line by using western blot analysis. CCK-8 assay results revealed that downregulation of Nav1.5 expression by its specific siRNA reduced proliferation of OSCC HSC-3 cells. Moreover, transwell assay results showed Nav1.5 knockdown significantly inhibited migration and invasion of HSC-3 cells. Meanwhile, qRT-PCR and western blot analysis results showed that epidermal growth factor (EGF) induced Nav1.5 expression in a time-and dosedependent manner. In addition, EGF promoted proliferation, migration and invasion of HSC-3 cells. Importantly, the Nav1.5 inhibitor tetrodotoxin significantly inhibited the proliferation of HSC-3 cells and impeded the migration and invasion of HSC-3 cells. Furthermore, it was found that siRNAmediated knockdown of Nav1.5 also lessened the proliferation of HSC-3 cells and blocked the migration and invasion of HSC-3 cells.
