1A USM-Θ two-phase turbulence model for simulating dense gas-particle flows显示文摘A second-order moment two-phase turbulence model for simulating dense gas-particle flows (USM-Θ model), combining the unified second-order moment twophase turbulence model for dilute gas-particle flows with the kinetic theory of particle collision, is proposed. The interaction between gas and particle turbulence is simulated using the transport equation of two-phase velocity correlation with a two-time-scale dissipation closure. The proposed model is applied to simulate dense gas-particle flows in a horizontal channel and a downer. Simulation results and their comparison with experimental results show that the model accounting for both anisotropic particle turbulence and particle-particle collision is obviously better than models accounting for only particle turbulence or only particle-particle collision.The USM-Θ model is also better than the k-ε-kp-Θ model and the k-ε-kp-εp-Θ model in that the first model can simulate the redistribution of anisotropic particle Reynolds stress components due to inter-particle collision, whereas the second and third models cannot.Yong Yu Lixing Zhou Baoguo Wang Feipeng Cai 2005Acta Mechanica Sinica2005,21,3:10
2Discriminated sgRNAs-Based SurroGate System Greatly Enhances the Screening Efficiency of Plant Base-Edited Cells显示文摘The development of CRISPR/Cas9-mediated base editing has made genomic modification more efficient. However, selection of genetically modified cells from millions of treated cells, especially plant cells, is still challenging. In this study, an efficient surrogate reporter system based on a defective hygromycin resistance gene was established in rice to enrich base-edited cells. After step-by-step optimization, the Discriminated sgRNAs-based SurroGate system (DisSUGs) was established by artificially differentiating the editing abilities of a wild-type single guide RNA (sgRNA) targeting the surrogate reporter gene and an enhanced sgRNA targeting endogenous sites. The DisSUGs enhanced the efficiency of screening base-edited cells by 3- to 5-fold for a PmCDA1-based cytosine-to-tyrosine base editor (PCBE), and 2.5- to 6.5-fold for an adenine base editor (ABE) at endogenous targets. These targets showed editing efficiencies of <25% in the conventional systems. The DisSUGs greatly enhanced the frequency of homozygous substitutions and expanded the activity window slightly for both a PCBE and an ABE. Analyses of the total number of single-nucleotide variants from whole-genome sequencing revealed that, compared with the no-enrichment PCBE strategy, the DisSUGs did not alter the frequency of genome-wide sgRNA-independent off-target mutations, but slightly increased the frequency of target-dependent off-target mutations. 6Expanding base editing scope to near-PAMless with engineered CRISPR/Cas9 variants in plants显示文摘Dear Editor,Base editors(BEs),including cytosine base editor(CBE)and adenine base editor(ABE),have been widely used to generate irreversible nucleotide substitution in plants and animals.However,their wide applications are largely hindered by the strict NG protospacer adjacent motif(PAM)sequences recognized by Streptococcus pyogenes Cas9(SpCas9)and its engineered variants,such as SpCas9-NG and xCas9(Hua et al.,2019;Ren et al.,2019;Wu et al.,2019;Zhong et al.,2019;Zhang et al.,2020).Most recently,it was reported that three new SpCas9 variants,SpCas9-NRRH,SpCas9-NRTH,and SpCas9-NRCH,could recognize non-G PAMs(NRNH,where R is A or G and H is A,C,or T)in human cells(Miller et al.,2020).Meanwhile,SPRY,another new SpCas9 variant,was developed to greatly expand the editing scope of BEs to nearly PAMless(Walton et al.,2020).In this study,we generated a series of efficient BE toolkits and almost achieved C-to-T mutation without PAM restriction except for NTG PAM,and largely expanded A-to-G mutation scope in stable transformed rice,providing a reference for application in other plants.Chengwei Zhang Yao Wang Feipeng Wang Si Zhao Jinling Song Feng Feng Jiuran Zhao Jinxiao Yang 2021Molecular Plant2021,14,2:4
7Developing high-efficiency base editors by combining optimized synergistic core components with new types of nuclear localization signal peptide显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)–CRISPR-associated protein(Cas) system has been widely used for genome editing. In this system, the cytosine base editor(CBE) and adenine base editor(ABE) allow generating precise and irreversible base mutations in a programmable manner and have been used in many different types of cells and organisms. However, their applications are limited by low editing efficiency at certain genomic target sites or at specific target cytosine(C) or adenine(A) residues. Using a strategy of combining optimized synergistic core components, we developed a new multiplex super-assembled ABE(sABE) in rice that showed higher base-editing efficiency than previously developed ABEs. We also designed a new type of nuclear localization signal(NLS) comprising a FLAG epitope tag with four copies of a codon-optimized NLS(F4NLS^(r2)) to generate another ABE named F4NLS-sABE. This new NLS increased editing efficiency or edited additional A at several target sites. A new multiplex super-assembled CBE(sCBE) and F4NLS^(r2) involved F4NLS-sCBE were also created using the same strategy. F4NLS-sCBE was proven to be much more efficient than sCBE in rice. 9Revisiting the thermal ageing on the metallised polypropylene film capacitor:from device to dielectric film显示文摘Metallised polypropylene film capacitors(MPPFCs)are widely used in power electronics and are generally degraded by elevated temperatures.This work aims to determine the relationships between the structural changes of MPPFC and the microstructural variations of the PP film during the thermal ageing of MPPFC at 100℃ for 38 days.The capacitance of MPPFC has a slight decrease during thermal ageing.However,the breakdown voltage of the MPPFC decreases by 39.4%by the ageing.The partial discharge(PD)number of MPPFC increases linearly with ageing time.The tear-down analysis of the MPPFC reveals that the molecular structure of the PP film has not been altered but has led to molecular chain scission and the generation of some polar fragments/groups.Meanwhile,the relative permittivity of the PP films rises as the ageing time increases.Moreover,thermal ageing causes the conversion of aluminum to alumina in the metallised electrode,which is hydrophilic for polar groups and leads to an adhesion effect between the metallised electrodes and the PP film.Contact angle measurements prove that the surface hydrophilicity of the PP sample increased after thermal ageing.Therefore,the PD/breakdown voltage in the MPPFC increases/decreases due to the uneven adhesion of the metallised PP film.Yushuang He Feipeng Wang Guoqiang Du Lei Pan Kaizheng Wang Reimund Gerhard Ronald Plath Pawel Rozga Pavel Trnka 2023High Voltage2023,8,2:2
10Novel nanometer scaffolds regulate the biological behaviors of neural stem cells显示文摘Ideal tissue-engineered scaffold materials regulate proliferation, apoptosis and differentiation of cells seeded on them by regulating gene expression. In this study, aligned and randomly oriented collagen nanofiber scaffolds were prepared using electronic spinning technology. Their diameters and appearance reached the standards of tissue-engineered nanometer scaffolds. The nanofiber scaffolds were characterized by a high swelling ratio, high porosity and good mechanical properties. The proliferation of spinal cord-derived neural stem cells on novel nanofiber scaffolds was obviously enhanced. The proportions of cells in the S and G 2 /M phases noticeably increased. Moreover, the proliferation rate of neural stem cells on the aligned collagen nanofiber scaffolds was high. The expression levels of cyclin D1 and cyclin-dependent kinase 2 were increased. Bcl-2 expression was significantly increased, but Bax and caspase-3 gene expressions were obviously decreased. There was no significant difference in the differentiation of neural stem cells into neurons on aligned and randomly oriented collagen nanofiber scaffolds. These results indicate that novel nanofiber scaffolds could promote the proliferation of spinal cord-derived neural stem cells and inhibit apoptosis without inducing differentiation. 11A synthetical methodology for identifying pr ior ity pollutants in reclaimed water based on meta-analysis显示文摘Wastewater reclamation and reuse is an increasing global project,while the reclamation treatment on wastewater does not completely remove all pollutants in water.The residual pollutants in reclaimed water would cause potential risk on human health and ecosystem safety during the long-term use.It is impossible to analyze and control all pollutants one by one in practice,therefore,identification and control of priority pollutants will be efficient strategy to ensure the safe use of reclaimed water.An integrated three-step methodology for identifying priority pollutants in reclaimed water was proposed in this study.First,a comprehensive literature survey on the occurrence of pollutants in reclaimed water was conducted,and a dataset D PR for pollutants occurrence in reclaimed water was established,containing 1,113 pollutants.Second,611 chemicals that had been recommended as hazardous pollutants for various water bodies in previous literatures were summarized,and a dataset D HP for hazardous pollutants in water was obtained.Third,meta-analysis on these two datasets(D PR and D HP)was performed,a new dataset D HPR for hazardous pollutants in reclaimed water was established,including 265 candidates.Finally,59 substances out of dataset D HPR were identified as priority pollutants for reclaimed water based on their recommendation frequency.It is expected that this synthetical methodology will provide powerful support for scientific evaluating and managing water pollution and ensuring safe use of reclaimed water.Feipeng Wang Dongbin Wei Miao Chen Shuang Peng Qiaorong Guo Xinyi Zhang Jun Liu Yuguo Du 2022Journal of Environmental Sciences2022,34,2:2

13Highly efficient CRISPR-SaKKH tools for plant multiplex cytosine base editing显示文摘Base editing, as an expanded clustered regularly interspaced short palindromic repeats(CRISPR)-Cas genome editing strategy, permits precise and irreversible nucleotide conversion. SaKKH, an efficient variant of a Cas9 ortholog from Staphylococcus aureus(SaCas9), is important in genome editing because it can edit sites with HHHAAT protospacer adjacent motif(PAM) that the canonical Streptococcus pyogenes Cas9(SpCas9) or its variants(e.g. xCas9, Cas9-NG) cannot. However, several technical parameters of SaKKH involved base editors have not been well defined and this uncertainty limits their application. We developed an effective multiplex cytosine base editor(SaKKHn-pBE) and showed that it recognized NNARRT, NNCRRT, NNGRGT, and NNTRGT PAMs. Based on 27 targets tested, we defined technical parameters of SaKKHn-pBE including the editing window, the preferred sequence context, and the mutation type. The editing efficiency was further improved by modification of the SaKKH sgRNA. 15Toxicity of Five Insecticides to Culicoides oxystoma Kieffer (Diptera:Ceratopogonidae)显示文摘Field-collected Culicoides oxystoma adults were exposed to different concentrations of commercial insecticides including chlorpyrifos,DDVP,deltamethyrin,beta-cypermethrin and beta-cypermethrin·emamectin benzoate via filter paper method,to determine toxicity of different insecticides on C. oxystoma.Results showed that toxicities varied significantly among insecticides. Biting midges were the most susceptible to deltamethyrin( 0. 060 mg/L),followed by chlorpyrifos( 0. 588 mg/L),beta-cypermethrin( 1. 741 mg/L),beta-cypermethrin·emamectin benzoate( 99. 670 mg/L),and DDVP( 600. 496 mg/L). After exposed to insecticides for 1 h,tested midges were transferred to recovery chamber and observed for another 24 h; increased mortality within this period was different under different concentrations of each insecticide. 17Optimization of polyurethane-bonded thin overlay mixture designation for airport pavement显示文摘This research explored the application potential of PUM thin-overlay technology on airport rapid maintenance.The rapid curing process of polyurethane binder determines the limited time window for mixing and construction of polyurethane-bonded mixture(PUM),which presents significant difference with hot-mix asphalt(HMA)technology.Therefore,this research investigated and optimized the mix design of PUM for airport thin-overlay technology based on its thermosetting characteristics.First,limestone and basalt were comprehensively compared as an aggregate for PUM.Then,the effects of molding and curing conditions were studied in terms of mixing time,molding method,molding parameters and curing temperature.Statistical analysis was also conducted to evaluate the effects of gradation and particle size on PUM performances based on gray relational analysis(GRA),thus determining the key particle size to control PUM performances.Finally,the internal structural details of PUM were captured by X-ray CT scan test.The results demonstrated that it only took 12 hours to reach 75%of maximum strength at a curing temperature of 50°C,indicating an efficient curing process and in turn allowing short traffic delay.The internal structural details of PUM presented distribution of tiny pores with few connective voids,guaranteeing waterproof property and high strength.Xianrui LI Ling XU Qidi ZONG Fu JIANG Xinyao YU Jun WANG Feipeng XIAO 2022Frontiers of Structural and Civil Engineering2022,16,8:1

20The storage and its stability of space charge in poly(ethylene naphthalene-2,6-dicarboxylate)显示文摘Poly(ethylene naphthalene-2,6-dicarboxylate) (PEN) is an aromatic polyester with molecular structure similar to that of poly(ethylene terephthalate)(PET). In this paper, the space charge storage ability and its stability in corona charged PEN were studied by measurement of surface potential decay under different storage conditions including dif- ferent temperatures and relative humidities, and analysis of open circuit thermally stimu- lated discharge (TSD) current spectra and charge TSD curves. The comparison between the charge stability of PEN and PET was carried out. The results point out that the charge stability of PEN is markedly better than that of PET under different environmental condi- tions. By means of isothermal depolarization program, the space charge lifetimes (effec- tive time constant) τ at room temperature of positively and negatively charged PEN are estimated to be 90 y.
