| 2 | Analysis of bacterial community in bulking sludge using culture-dependent and -independent approaches显示文摘The bacterial community of a bulking sludge from a municipal wastewater treatment plant with anoxic-anaerobic-oxic process was investigated by combination of cultivation and 16S rRNA gene clone library analysis for understanding the causes of bulking.A total of 28 species were obtained from 63 isolates collected from six culture media.The most cultivable species belonged to γ-Proteobacteria including Klebsiella sp.,Pseudomonas sp.,Aeromonas sp.and Acinetobacter sp.Further analysis of these strains by repetitive sequence based on polymerase chain reaction (rep-PCR) technology showed that rep-PCR yielded discriminatory banding patterns within the same genus using REP and BOX primer sets.While the culture-independent assessment revealed that β-Proteobacteria was the dominant group in the bulking sample.Sequence analysis revealed that the highest proportion (14.7%) of operational taxonomic units was 98% similar to Candidatus Accumulibacter phosphatis,which is used to remove phosphorous from wastewater.Our results indicated that combining different approaches can produce complementary information,thus generate a more accurate view of microbial community in bulking sludge. | Decai Jin Ping Wang Zhihui Bai Xinxin Wang Hong Peng Rong Qi Zhisheng Yu Guoqiang Zhuang | 2011 | Journal of Environmental Sciences2011,23,11: | 11 |
| 3 | A MEMS Device for in-situ TEM Test of SCS Nanobeam显示文摘In-situ tensile testing in TEM (transmission electron microscopy) is a useful tool for studying mechanical properties of nano-structures because it can provide quanti-tative information on sample deformation at atomic scale. To facilitate in-situ TEM tensile tests of SCS (single crystal silicon) nanobeam, a MEMS tensile-testing chip was designed and fabricated. The chip was fabricated by means of bulk micro-machining and wafer bonding techniques. An SCS nanobeam, a comb drive ac-tuator, a force sensor beam and an electron beam window were integrated into the chip. With the on-chip comb-drive-actuator stretching the nanobeam and in-situ TEM observation, tensile test on a 90 nm-thick nanobeam was performed and the strain-stress relationship was obtained. | 4 | ADAR-mediated RNA editing in non-coding RNA sequences显示文摘Adenosine to inosine(A-to-I)RNA editing is the most abundant editing event in animals.It converts adenosine to inosine in double-stranded RNA regions through the action of the adenosine deaminase acting on RNA(ADAR)proteins.Editing of pre-mRNA coding regions can alter the protein codon and increase functional diversity.However,most of the A-to-I editing sites occur in the non-coding regions of pre-mRNA or mRNA and non-coding RNAs.Untranslated regions(UTRs)and introns are located in pre-mRNA non-coding regions,thus A-to-I editing can influence gene expression by nuclear retention,degradation,alternative splicing,and translation regulation.Non-coding RNAs such as microRNA(miRNA),small interfering RNA(siRNA)and long non-coding RNA(lncRNA)are related to pre-mRNA splicing,translation,and gene regulation.A-to-I editing could therefore affect the stability,biogenesis,and target recognition of non-coding RNAs.Finally,it may influence the function of non-coding RNAs,resulting in regulation of gene expression.This review focuses on the function of ADAR-mediated RNA editing on mRNA non-coding regions(UTRs and introns)and non-coding RNAs(miRNA,siRNA,and lncRNA). | YANG Yun ZHOU XinXin JIN YongFeng | 2013 | Science China(Life Sciences)2013,56,10: | 4 |
| 5 | Sphingosine-1-phosphate,a novel TREM2 ligand,promotes microglial phagocytosis to protect against ischemic brain injury显示文摘The mechanism of sphingosine-1-phosphate(S1P)-mediated phagocytosis remains unknown.Here,we found that S1P or FTY720(an analog of S1P)promoted microglial phagocytosis in stroke independent of S1PRs.First,we used computer simulation of molecular docking to predict that S1P might be a ligand for triggering receptor expressed on myeloid cells 2(TREM2).Next,microscale thermophoresis(MST),surface plasmon resonance(SPR)and liquid chromatography–tandem mass spectrometry(LC–MS/MS)were performed to reveal that S1P was a novel TREM2 ligand.Then,we confirmed the pro-phagocytosis of S1P targeting in Trem2-Dap12 transfected CHO cells and TREM2 knockdown microglia.Point mutation analysis showed that D104 was the critical binding residue.Trem2^(−/−)mice were used to demonstrate the role of S1P-induced phagocytosis targeting on TREM2 in protecting against ischemic brain injury.Finally,further studies revealed that apolipoprotein E(APOE)loaded with S1P was released by microglia and bound to apoptotic neurons via LDL receptor related protein 1B(LRP1B)and thereby induced microglia to phagocytose apoptotic neurons.Overall,the present work reveals for the first time that S1P acts as a novel endogenous ligand of TREM2 to effectively promote microglial phagocytosis.Our findings provide a new lead compound for developing immunomodulator targeting on TREM2. | Tengfei Xue Juan Ji Yuqin Sun Xinxin Huang Zhenyu Cai Jin Yang Wei Guo Ruobing Guo Hong Cheng Xiulan Sun | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 3 |
| 6 | The mechanosensitive lncRNA Neat1 promotes osteoblast function through paraspeckle-dependent Smurf1 mRNA retention显示文摘Mechanical stimulation plays an important role in bone remodeling. Exercise-induced mechanical loading enhances bone strength,whereas mechanical unloading leads to bone loss. Increasing evidence has demonstrated that long noncoding RNAs(lnc RNAs) play key roles in diverse biological, physiological and pathological contexts. However, the roles of lnc RNAs in mechanotransduction and their relationships with bone formation remain unknown. In this study, we screened mechanosensing lnc RNAs in osteoblasts and identified Neat1, the most clearly decreased lnc RNA under simulated microgravity. Of note, not only Neat1 expression but also the specific paraspeckle structure formed by Neat1 was sensitive to different mechanical stimulations, which were closely associated with osteoblast function. Paraspeckles exhibited small punctate aggregates under simulated microgravity and elongated prolate or larger irregular structures under mechanical loading. Neat1 knockout mice displayed disrupted bone formation, impaired bone structure and strength, and reduced bone mass. Neat1 deficiency in osteoblasts reduced the response of osteoblasts to mechanical stimulation. In vivo, Neat1 knockout in mice weakened the bone phenotypes in response to mechanical loading and hindlimb unloading stimulation. Mechanistically, paraspeckles promoted nuclear retention of E3 ubiquitin ligase Smurf1 m RNA and downregulation of their translation, thus inhibiting ubiquitination-mediated degradation of the osteoblast master transcription factor Runx2, a Smurf1 target. | 7 | Numerical investigation of airborne transmission of respiratory infections on the subway platform显示文摘Underground subway platforms are among the world’s busiest public transportation systems,but the airborne transmission mechanism of respiratory infections on these platforms has been rarely studied.Here,computational fluid dynamics(CFD)modeling is used to investigate the airflow patterns and infection risks in an island platform under two common ventilation modes:Mode 1-both sides have air inlets and outlets;Mode 2-air inlets are present at the two sides and outlets are present in the middle.Under the investigated scenario,airflow structure is characterized by the ventilation jet and human thermal plumes.Their interaction with the infector’s breathing jet imposes the front passenger under the highest exposure risk by short-range airborne route,with intake fractions up to 2.57%(oral breathing)or 0.63%(nasal breathing)under Mode 1;oral breathing of the infector may impose higher risks for the front passenger compared with nasal breathing.Pathogen are efficiently diluted as they travel further,in particular to adjacent crowds.The maximum and median value of intake fractions of passengers in adjacent crowds are respectively 0.093%and 0.016%(oral breathing),and 0.073%and 0.014%(nasal breathing)under Mode 1.Compared with Mode 1,the 2nd mode minimizes the interaction of ventilation jet and breathing jet,where the maximum intake fraction is only 0.34%,and the median value in the same crowd and other crowds are reduced by 23–63%.Combining published quanta generation rate data of COVID-19 and influenza infectors,the predicted maximum and median infection risks for passengers in the same crowds are respectively 1.46%–40.23%and 0.038%–1.67%during the 3–10 min waiting period,which are more sensitive to ventilation rate and exposure time compared with return air.This study can provide practical guidance for the prevention of respiratory infections in subway platforms. | Jianjian Wei Sirui Zhu Feiwu He Qianfang Guo Xinxin Huang Jianxiang Yu Lirong Zou Tao Jin Jie Wu | 2022 | Geoscience Frontiers2022,13,6: | 2 |

| 18 | Spatial-temporal variations and driving factors of soil organic carbon in forest ecosystems of Northeast China显示文摘Forest soil carbon is a major carbon pool of terrestrial ecosystems,and accurate estimation of soil organic carbon(SOC)stocks in forest ecosystems is rather challenging.This study compared the prediction performance of three empirical model approaches namely,regression kriging(RK),multiple stepwise regression(MSR),random forest(RF),and boosted regression trees(BRT)to predict SOC stocks in Northeast China for 1990 and 2015.Furthermore,the spatial variation of SOC stocks and the main controlling environmental factors during the past 25 years were identified.A total of 82(in 1990)and 157(in 2015)topsoil(0–20 cm)samples with 12 environmental factors(soil property,climate,topography and biology)were selected for model construction.Randomly selected80%of the soil sample data were used to train the models and the other 20%data for model verification using mean absolute error,root mean square error,coefficient of determination and Lin's consistency correlation coefficient indices.We found BRT model as the best prediction model and it could explain 67%and 60%spatial variation of SOC stocks,in 1990,and 2015,respectively.Predicted maps of all models in both periods showed similar spatial distribution characteristics,with the lower SOC in northeast and higher SOC in southwest.Mean annual temperature and elevation were the key environmental factors influencing the spatial variation of SOC stock in both periods.SOC stocks were mainly stored under Cambosols,Gleyosols and Isohumosols,accounting for 95.6%(1990)and 95.9%(2015).Overall,SOC stocks increased by 471 Tg C during the past 25 years.Our study found that the BRT model employing common environmental factors was the most robust method for forest topsoil SOC stocks inventories.The spatial resolution of BRT model enabled us to pinpoint in which areas of Northeast China that new forest tree planting would be most effective for enhancing forest C stocks.Overall,our approach is likely to be useful in forestry management and ecological restoration at and beyond the regional scale. | Shuai Wang Bol Roland Kabindra Adhikari Qianlai Zhuang Xinxin Jin Chunlan Han Fengkui Qian | 2023 | Forest Ecosystems2023,10,2: | 0 |

| 20 | Experimental Characterization and Model Verification of Thermal Conductivity from Mesoporous to Macroporous SiOC Ceramics显示文摘Based on the chemical cross-linking method,this paper uses polydimethylsiloxane with various viscosities of 10 cSt,20 cSt,50 cSt,and 100 cSt to synthesize mesoporous and macroporous SiOC ceramics.Their thermal conductivities are measured by using 3ωmethod with high accuracy.Three typical models for their thermal conductivities,i.e.,series model(SM),maxwell-Eucken 1 model(ME1),and effective medium theory(EMT)model,are utilized to derive the empirical formula through the multi-parameter linear optimization algorithm,which agrees well with the experimental results.The effects of pore size and specific surface area on the overall thermal conductivity of the porous structure are explored.Interestingly,it is found that the thermal conductivities of both gas phase and solid phase inside the porous structure increase with the increasing pore size at the nanometer scale,but the overall thermal conductivity of the porous structure decreases with the increasing pore size.Scanning electron microscopy graphs corroborate that the extension of the heat transfer route and the barrier of more pores between the solid phases together cause the reduction of the gas-solid coupling thermal conductivity of SiOC ceramics with larger pore size.On the contrary,the miniaturization of individual particles through modulating the synthesis parameters can increase the number of small pores in the sample itself to meet the pseudo-lattice vibration conditions,which results in the increment of the gas-solid coupling thermal conductivity and the overall thermal conductivity of the porous structure.These findings would provide meaningful guidance for designing SiOC porous ceramic super-insulation materials with extremely low thermal conductivity. | QIU Lin DU Yanbo BAI Yangyang FENG Yanhui ZHANG Xinxin WU Jin WANG Xiaotian XU Caihong | 2021 | Journal of Thermal Science2021,30,2: | 0 |
