Developing gene-tagged molecular markers for evaluation of genetic association of apple SWEET genes with fruit sugar accumulation显示文摘Sugar content is an important component of fruit quality.Although sugar transporters are known to be crucial for sugar accumulation, the role of genes encoding SWEET sugar transporters in fruit sugar accumulation remains elusive. Here we report the effect of the SWEET genes on fruit sugar accumulation in apple.A total of 25 MdSWEET genes were identified in the apple genome, and 9 were highly expressed throughout fruit development.Molecular markers of these 9 MdSWEET genes were developed and used for genotyping of 188 apple cultivars. The association of polymorphic MdSWEET genes with soluble sugar content in mature fruit was analyzed.Three genes,MdSWEET2e,MdSWEET9b,and MdSWEET15a,were significantly associated with fruit sugar content, with MdSWEET15a and MdSWEET9b accounting for a relatively large proportion of phenotypic variation in sugar content. Moreover, both MdSWEET9b and MdSWEET15a are located on chromosomal regions harboring QTLs for sugar content.Hence,MdSWEET9b and MdSWEET15a are likely candidates regulating fruit sugar accumulation in apple. Our study not only presents an efficient way of implementing gene functional study but also provides molecular tools for genetic improvement of fruit quality in apple-breeding programs. | Qiaoling Zhen Ting Fang Qian Peng Liao Liao Li Zhao Albert Owiti Yuepeng Han | 2018 | Horticulture Research2018,5,1: | 12
In situ plasmonic optical fiber detection of the state of charge of supercapacitors for renewable energy storage显示文摘In situ and continuous monitoring of electrochemical activity is key to understanding and evaluating the operation mechanism and efficiency of energy storage devices. However, this task remains challenging. For example, the present methods are not capable of providing the real-time information about the state of charge(SOC)of the energy storage devices while in operation. To address this,a novel approach based on an electrochemical surface plasmon resonance(SPR)optical fiber sensor is proposed here. This approach offers the capability of in situ comprehensive monitoring of the electrochemical activity(the electrode potential and the SOC)of supercapacitors(used as an example). The sensor adopted is a tilted fiber Bragg grating imprinted in a commercial single-mode fiber and coated with a nanoscale gold film for high-efficiency SPR excitation.Unlike conventional“bulk”detection methods for electrode activity, our approach targets the“localized”(sub-μm-scale)charge state of the ions adjacent to the electrode interface of supercapacitors by monitoring the properties of the SPR wave on the fiber sensor surface located adjacent to the electrode.A stable and reproducible correlation between the real-time charge–discharge cycles of the supercapacitors and the optical transmission of the optical fiber has been found. Moreover, the method proposed is inherently immune to temperature cross-talk because of the presence of environmentally insensitive reference features in the optical transmission spectrum of the devices. Finally, this particular application is ideally suited to the fundamental qualities of optical fiber sensors, such as their compact size, flexible shape, and remote operation capability, thereby opening the way for other opportunities for electrochemical monitoring in various hard-to-reach spaces and remote environments. | Jiajie Lao Peng Sun Fu Liu Xuejun Zhang Chuanxi Zhao Wenjie Mai Tuan Guo Gaozhi Xiao Jacques Albert | 2018 | Light(Science & Applications)2018,7,1: | 8
Relationship between Myopia and Optical Components - A Study among Chinese Hong Kong Student Population显示文摘Purpose: To establish the prevalence and severity of myopia among the Chinese Hong Kong students and to study the relationship between myopia and optical components. Methods; One thousand and seventy-five freshmen of the 1993-1994 academic year in the Chinese University of Hong Kong underwent the eye examination including evaluation of refractive error, keratometry, and A-scan ultrasonic biometry. The data were analyzed with the SPSS/PC+4. 01 statistical package. Results: The prevalence of myopia was 91. 7% with the mean refraction being -4. 00 ± 2. 64D in this young adult population. The statistical analyses demonstrated a significant correlation between refractive value and axial length of the globe (r=-0. 78), vitreous length (r=-0. 76), anterior chamber depth (r=-0. 33), lens thickness (r = 0. 13) and corneal curvature (r = 0. 19). Conclusion: The refractive status is mainly dependent on the axial length. In general, the higher the myopia was, the longer the eyeball, the deeper the anterior chamber, | Peng Iob Lo Patrick CP Ho Joseph TF Lau Albert YK Cheung Ernst Goldschmidt Mark OM Tso | 1996 | Eye Science1996,16,3: | 2
Comparative analysis of DNA extraction protocols for ancient soft tissue museum samples显示文摘DNA studies of endangered or extinct species often rely on ancient or degraded remains. The majority of ancient DNA(aDNA)extraction protocols focus on skeletal elements, with skin and hair samples rarely explored.Similar to that found in bones and teeth,DNA extracted from historical or ancient skin and fur samples is also extremely fragmented with low endogenous content due to natural degradation processes. Thus, the development of effective DNA extraction methods is required for these materials. Here,we compared the performance of two DNA extraction protocols(commercial and custom laboratory aDNA methods)on hair and skin samples from decades-old museum specimens to Iron Age archaeological material. We found that apart from the impact sample-specific taphonomic and handling history has on the quantity and quality of DNA preservation, skin yielded more endogenous DNA than hair of the samples and protocols tested. While both methods recovered DNA from ancient soft tissue, the laboratory method performed better overall in terms of DNA yield and quality, which was primarily due to the poorer performance of the commercial binding buffer in recovering aDNA. | Ming Zhang Peng Cao Qing-Yan Dai Yong-Qiang Wang Xiao-Tian Feng Hong-Ru Wang Hong Wu Albert Min-Shan Ko Xiao-Wei Mao Yi-Chen Liu Li Yu Christian Roos Tilo Nadler Wen Xiao E.Andrew Bennett Qiao-Mei Fu | 2021 | Zoological Research2021,42,3: | 1

| 11 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment.
