1Orthogonal transformation operation theorem of a spatial universal uniform rotating magnetic field and its application in capsule endoscopy显示文摘According to the anti-phase sine current superposition theorem, the orientation, the magnetic flux density, the angular speed and the rotational direction of the spatial universal rotating magnetic field (SURMF) can be controlled within the tri-axial orthogonal square Helmholtz coils (TOSHC). Nevertheless, three coupling direction angles of the normal vector of the SURMF in the Descartes coordinate system cannot be separately controlled, thus the adjustment of the orientation of the SURMF is difficult and the flexibility of the robotic posture control is restricted. For the dimension reduction and the decoupling of control variables, the orthogonal transformation operation theorem of the SURMF is proposed based on two independent rotation angular variables, which employs azimuth and altitude angles as two variables of the three-phase sine current superposition formula derived by the orthogonal rotation inverse transformation. Then the unique control rules of the orientation and the rotational direction of the SURMF are generalized in each spatial quadrant, thus the scanning of the normal vector of the SURMF along the horizontal or vertical direction can be achieved through changing only one variable, which simplifies the control process of the orientation of the SURMF greatly. To validate its feasibility and maneuverability, experiments were conducted in the animal intestine utilizing the innovative dual hemisphere capsule robot (DHCR) with active and passive modes. It was demonstrated that the posture adjustment and the steering rolling locomotion of the DHCR can be realized through single variable control, thus the orthogonal transformation operation theorem makes the control of the orientation of the SURMF convenient and flexible significantly. 2Research on Low Cycle Fatigue Reliability-based Robust Design Optimization of Turbine Blade显示文摘针对涡轮叶片低周疲劳可靠性稳健设计优化问题,对叶片材料进行了高温疲劳试验,采用定量方程随机化方法处理试验数据,获得叶片材料的概率-应变-寿命曲线。采用贝塞尔曲线描述叶片型线方程,建立了涡轮叶片结构及流场的参数化模型,采用热-流-固耦合有限元法对涡轮流场和叶片进行了数值分析,得到叶片动能效率和应力应变分布特性。建立了叶片疲劳可靠性稳健设计优化模型,并采用响应面法获得叶片结构性能函数和极限状态函数,将叶片低周疲劳可靠性作为基本约束条件,采用序列二次规划优化法得到设计优化结果。研究结果表明,优化后的叶片低周疲劳可靠性以及稳健性显著提高,模型及方法正确可行,可用于涡轮叶片以及其他复杂结构的低周疲劳可靠性稳健设计优化。PENG Maolin YANG Zichun CAO Yueyun CHU Zhuli 2013中国电机工程学报2013,33,11:8
3Salmonella flagella confer anti-tumor immunological effect via activating Flagellin/TLR5 signalling within tumor microenvironment显示文摘Salmonella:mediated cancer therapy has achieved remarkable anti-tumor effects in experimental animal models,but the detailed mechanism remains unsolved.In this report,the active involvement of the host immune response in this process was confirmed by comparing the tumor-suppressive effects of Salmonella in immunocompetent and immunodeficient mice bearing melanoma allografts.Since flagella are key inducers of the host immune response during bacterial infection,flagella were genetically disrupted to analyse their involvement in Salmonella-mediated cancer therapy.The results showed that flagellum-deficient strains failed to induce significant anti-tumor effects,even when more bacteria were administered to offset the difference in invasion efficiency.Flagella mainly activate immune cells via Flagellin/Toll-like receptor 5(TLR5)signalling pathway.Indeed,we showed that exogenous activation of TLR5 signalling by recombinant Flagellin and exogenous expression of TLR5 both enhanced the therapeutic efficacy of flagellum-deficient Salmonella against melanoma.Our study highlighted the therapeutic value of the interaction between Salmonella and the host immune response through Flagellin/TLR5 signalling pathway during Salmonella-mediated cancer therapy,thereby suggesting the potential application of TLR5 agonists in the cancer immune therapy.Jianxiang Chen Yiting Qiao Guo Chen Cunjie Chang Heng Dong Bo Tang Xiawei Cheng Xiufeng Liu Zichun Hua 2021Acta Pharmaceutica Sinica B2021,11,10:6
4Non-deterministic fatigue life analysis using convex set models显示文摘The non-probabilistic approach to fatigue life analysis was studied using the convex models-interval, ellipsoidal and multiconvex models. The lower and upper bounds of the fatigue life were obtained by using the second-order Taylor series and Lagrange multiplier method. The solving process for derivatives of the implicit life function was presented. Moreover, a median ellipsoidal model was proposed which can take into account the sample blind zone and almost impossibility of concurrence of some small probability events. The Monte Carlo method for multi-convex model was presented, an important alternative when the analytical method does not work. A project example was given. The feasibility and rationality of the presented approach were verified. 5Galangin and TRAIL cooperate to suppress A549 lung cancer proliferation via apoptosis and p38 MAPK activation显示文摘Tumor necrosis factor-related apoptosis inducing ligand(TRAIL)is a promising antitumor therapy against lung cancer which is currently undergoing a phase II clinical trial in China.Unfortunately some cancer patients in the clinical trial displayed resistance to TR AIL treatment.In investigating ways to overcome this resistance,we evaluated the inhibitory effect of galangin on TRAIL resistant A549 human lung adenocarcinoma cells.Here we report that,in comparison with the single agents,the combination of galangin and TRAIL markedly suppressed proliferation of A549 cells and induced apoptosis as shown by DAPI and JC-1 staining.The combination of galangin and TRAIL induced PARP cleavage,activation of caspase-8 and p38 MAPK(mitogen-activated protein kinases).These findings indicate that the combination of galangin and TRAIL may constitute a promising strategy for the treatment of lung cancer.Wenjing Zhang Qilai Huang Zichun Hua 2012Acta Pharmaceutica Sinica B2012,2,6:4
6SARS-CoV-2 nsp12 attenuates type Ⅰ interferon production by inhibiting IRF3 nuclear translocation显示文摘SARS-CoV-2 is the pathogenic agent of COVID-19,which has evolved into a global pandemic.Compared with some other respiratory RNA viruses,SARS-CoV-2 is a poor inducer of type Ⅰ interferon(IFN).Here,we report that SARS-CoV-2 nsp12,the viral RNA-dependent RNA polymerase(RdRp),suppresses host antiviral responses.SARS-CoV-2 nsp12 attenuated Sendai virus(SeV)-or poly(I:C)-induced IFN-β promoter activation in a dose-dependent manner.It also inhibited IFN promoter activation triggered by RIG-I,MDA5,MAVS,and IRF3 overexpression.Nsp12 did not impair IRF3 phosphorylation but suppressed the nuclear translocation of IRF3.Mutational analyses suggested that this suppression was not dependent on the polymerase activity of nsp12.Given these findings,our study reveals that SARS-CoV-2 RdRp can antagonize host antiviral innate immunity and thus provides insights into viral pathogenesis.Wenjing Wang Zhuo Zhou Xia Xiao Zhongqin Tian Xiaojing Dong Conghui Wang Li Li Lili Ren Xiaobo Lei Zichun Xiang Jianwei Wang 2021Cellular & Molecular Immunology2021,18,4:4
7Preparation of hapten-specific monoclonal antibody for cadmium and its ELISA application to aqueous samples显示文摘High-affinity and specific monoclonal antibodies against cadmium-ethylene diamine tetraacetic acid(EDTA)complex have been produced using the hybridoma technique.A hapten was synthesized and characterized by Fourier Transform Infrared Spectroscopy(FT-IR)and UVVis.Competitive enzyme-linked immunosorbent assay(ELISA)for quantitative detection of cadmium in aqueous sample was developed.The monoclonal antibody with high level of binding affinity for Cd-IEDTA-BSA and high specificity for soluble Cd-EDTA complex showed less than 0.99%cross-reactivity with other 11 metals.The limit of detection was 0.10μg·L^(-1),and the effective linear range was 10^(-1)-10^(3)μg·L^(-1).The intra-and inter-assay coefficient variations were 1.5%-6.3%and 3.2%-7.4%,respectively.The spike recovery in different water samples were between 98.5%and 110.3%.The detection limit of this assay was well below the allowable concentration of cadmium(3μg·L^(-1)),and the working range was wider than that of other methods which showed the range of 2.19-86.38 and 0-10^(3)μg·L^(-1).The competitive ELISA established in this paper was sensitive and accurate in the screening of cadmium in aqueous samples.The results will lay a solid foundation for construction of an immunoassay kit for cadmium.Huan HE Bo TANG Cheng SUN Shaogui YANG Weijuan ZHENG Zichun HUA 2011Frontiers of Environmental Science & Engineering2011,5,3:4
8Macrophage-mediated tumor-targeted delivery of engineered Salmonella typhimurium VNP20009 in anti-PD1 therapy against melanoma显示文摘Bacterial antitumor therapy has great application potential given its unique characteristics,including genetic manipulation, tumor targeting specificity and immune system modulation. However,the nonnegligible side effects and limited efficacy of clinical treatment limit their biomedical applications. Engineered bacteria for therapeutic applications ideally need to avoid their accumulation in normal organs and possess potent antitumor activity. Here, we show that macrophage-mediated tumor-targeted delivery of Salmonella typhimurium VNP20009 can effectively reduce the toxicity caused by administrating VNP20009 alone in a melanoma mouse model. This benefits from tumor-induced chemotaxis for macrophages combined with their slow release of loaded strains. Inspired by changes in the tumor microenvironment, including a decrease in intratumoral dysfunctional CD8+T cells and an increase in PDL1 on the tumor cell surface, macrophages were loaded with the engineered strain VNP-PD1nb, which can express and secrete anti-PD1 nanoantibodies after they are released from macrophages. This novel triple-combined immunotherapy significantly inhibited melanoma tumors by reactivating the tumor microenvironment by increasing immune cell infiltration, inhibiting tumor cell proliferation, remodeling TAMs to an M1-like phenotype and prominently activating CD8+T cells. 9Adenosine monophosphate-activated protein kinase activation enhances embryonic neural stem cell apoptosis in a mouse model of amyotrophic lateral sclerosis显示文摘Alterations in embryonic neural stem cells play crucial roles in the pathogenesis of amyotrophic lateral sclerosis. We hypothesized that embryonic neural stem cells from SOD1G93 A individuals might be more susceptible to oxidative injury, resulting in a propensity for neurodegeneration at later stages. In this study, embryonic neural stem cells obtained from human superoxide dismutase 1 mutant(SOD1G93A) and wild-type(SOD1WT) mouse models were exposed to H2O2. We assayed cell viability with mitochondrial succinic dehydrogenase colorimetric reagent, and measured cell apoptosis by flow cytometry. Moreover, we evaluated the expression of the adenosine monophosphate-activated protein kinase(AMPK) α-subunit, paired box 3(Pax3) protein, and p53 in western blot analyses. Compared with SOD1 WT cells, SOD1G93 A embryonic neural stem cells were more likely to undergo H2O2-induced apoptosis. Phosphorylation of AMPKα in SOD1G93 A cells was higher than that in SOD1 WT cells. Pax3 expression was inversely correlated with the phosphorylation levels of AMPKα. p53 protein levels were also correlated with AMPKαphosphorylation levels. Compound C, an inhibitor of AMPKα, attenuated the effects of H2O2. 10High population and dispersion of pentacoordinated Al~Ⅴ species on the surface of ?ame-made amorphous silica-alumina显示文摘Pentacoordinated Al(Al~Ⅴ)species in silica-alumina are promising to promote the formation of acid sites or act as surface defects for tailoring single-atom catalysts.However,pentahedral coordination(Al~Ⅴ)is rarely observed in conventionally prepared silica-alumina.Here,we show that high population and dispersion of Al~Ⅴ species on the surface of amorphous silica-alumina(ASA)can be achieved by means of flame spray pyrolysis.High resolution TEM/EDX,high magnetic-field NMR and DFT calculations are employed to characterize the structure of as-prepared ASAs.Solid-state ^(27)Al multi-quantum MAS NMR experiments show that most of the Al~Ⅴspecies are formed independently from the alumina phase and are accessible for guest molecules on the surface.Upon water adsorption,these Al~Ⅴ species are transformed to Al~Ⅵ species,structurally similar to surface Al~Ⅳ species,as confirmed by DFT calculations.The outstanding catalytic activity of as-synthesized ASA is demonstrated using the in situ H/D exchange reaction with deuterated benzene as an example.The Al~Ⅴ-rich ASA provides a much lower activation energy(~30 kJ/mol)than that reported for zeolite H-ZSM-5(~60 kJ/mol).The superior catalytic performance is attributed to the high Al~Ⅴcontent promoting the surface active sites in ASA.The knowledge gained on the synthesis of Al~Ⅴ-rich ASAs and the nature of aluminum coordination in these materials could pave the way to more efficient silica-alumina based catalysts.Zichun Wang Yijiao Jiang Xianfeng Yi Cuifeng Zhou Aditya Rawal James Hook Zongwen Liu Feng Deng Anmin Zheng Michael Hunger Alfons Baiker Jun Huang 2019Science Bulletin2019,64,8:2
11Optimal root system strategies for desert phreatophytic seedlings in the search for groundwater显示文摘Desert phreatophytes are greatly dependent on groundwater, but how their root systems adapt to different groundwater depths is poorly understood. In the present study, shoot and root growths of Alhagi sparsifolia Shap. seedlings were studied across a gradient of groundwater depths. Leaves, stems and roots of different orders were measured after 120 days of different groundwater treatments. Results indicated that the depth of soil wetting front and the vertical distribution of soil water contents were highly controlled by groundwater depths. The shoot growth and biomass of A. sparsifolia decreased, but the root growth and rooting depth increased under deeper groundwater conditions. The higher ratios of root biomass, root/shoot and root length/leaf area under deeper groundwater conditions implied that seedlings of A. sparsifolia economized carbon cost on their shoot growths. The roots of A. sparsifolia distributed evenly around the soil wetting fronts under deeper groundwater conditions. Root diameters and root lengths of all orders were correlated with soil water availabilities both within and among treatments. Seedlings of A. sparsifolia produced finer first- and second-order roots but larger third- and fourth-order roots in dry soils. 13Elastic anisotropy and its influencing factors in organic-rich marine shale of southern China显示文摘Shale is observed to have strong anisotropy due to its unique mineralogy and microstructure, and this anisotropy property has significant impact on seismic and well-log data. The organic-rich marine shale in the southern and eastern Sichuan Basin is one of the most important shale-gas reservoir formations in China. To investigate the elastic anisotropy of this shale and its influencing factors, we performed ultrasonic velocity measurements, X-ray diffraction analysis, rock-eval pyrolysis and vitrinite reflectance measurement on the samples from the Upper Ordovician Wufeng Formation and the Lower Silurian Longmaxi Formation. The experimental results show the that:(1) the velocity anisotropy of the Wufeng-Longmaxi(WL) shale varies from 10% to 50%, and most samples have strong anisotropy;(2) the P-and S-wave anisotropy parameters(Thomsen's εand γ) increase with clay contents, but this relationship can be greatly affected by the clay orientation index;(3) organic matter content(OMC) is found to have little influence in seismic anisotropy for the over mature WL shale, whereas the OMC determines the magnitude of anisotropy of immature/mature shales(e.g. the Bakken shale or the Bazhenov shale) according to the published literatures, because organic matters in shales of different maturity have different morphologies and distributions;(4) the OMC of WL shale has positive correlation with quartz content, and this weakens the correlation between OMC and the magnitude of anisotropy to a certain extent. 14Longitudinal and lateral control methods from single vehicle to autonomous platoon显示文摘To successfully implement the platoon control of connected and automated vehicles,it is necessary to address motion control issues to achieve longitudinal and lateral collaborative control.However,due to traffic capacity limitations and the complex traffic environment in which autonomous and human-driven vehicles coexist,autonomous platoon faces significant risks and challenges.This paper investigates longitudinal and lateral control issues from the perspective of a single vehicle up to a platoon,simulating the performance and suitability of various controllers.First,a longitudinal controller based on fuzzy logic and PID control is employed for speed tracking control of a single vehicle,followed by the adoption of an MPC controller based on the vehicle kinematics model to realize the lateral motion of a single vehicle.Second,the communication methods of the autonomous platoon are discussed,and the longitudinal controller that considers the platoon's various communication topologies is developed.Thirdly,a framework for robust integrated motion control is established,which combines the robust H-infinity longitudinal controller and the APF-based MPC lateral controller.Simulation results validate the effectiveness of the aforementioned controllers and reveal their limitations.Lei Song Jun Li Zichun Wei Kai Yang Ehsan Hashemi Hong Wang 2023Green Energy and Intelligent Transportation2023,2,2:1
