| 1 | Correlation of E-selectin gene polymorphisms with risk of ischemic stroke A meta-analysis显示文摘OBJECTIVE: To investigate the association of E-selectin S128R polymorphisms with ischemic stroke. DATA SOURCES: A computer-based online search was conducted in PubMed, Elsevier, Ovid Database, the China National Knowledge Infrastructure, and Wanfang Database between January 1998 and December 2010. STUDY SELECTION: Case-controlled studies addressing the association of the E-selectin polymorphism and ischemic stroke were included in this review. The genotype distribution complied with the Hardy-Weinberg genetic equilibrium. The included reports were evaluated by two authors for strict quality screening. Meta-analysis software, REVMAN 5.1, was used to investigate heterogeneity, pooled odds ratio (OR) and 95% confidence interval (CI) in individual studies. MAIN OUTCOME MEASURES: Genotype and allele distributions at the E-selectin S128R site. RESULTS: Six case-controlled studies were included after screening and application of inclusion and exclusion criteria. There was no heterogeneity in the genotype and allele frequencies, and no publication bias was found. Meta-analysis of the pooled data showed that the OR value of the (AC+CC)/AA genotype was 1.93 (95% CI: 1.55-2.41, Z = 5.80, P < 0.000 01), and the OR for the C/A allele was 1.80 (95% CI: 1.47-2.22, Z = 5.59, P < 0.000 01) in the ischemic stroke group, compared with control group. Results of pooled data in Chinese subjects showed that the OR value of (AC+CC)/AA was 2.36 (95% CI: 1.68-3.31, Z = 4.99, P < 0.000 01), and the OR value of the C/A allele was 2.25 (95% CI: 1.63-3.12, Z = 4.89, P < 0.000 01). | 6 | Characteristics and stability of slope movement response to underground mining of shallow coal seams away from gullies显示文摘Underground pressure is abnormal during mining of shallow coal seams under gullies. We studied gully slope movements, subject to underground mining, with physical simulation and theoretical analysis. The rules disclose that the slope rock slides horizontally in response to mining in the direction of gullies and rotates reversely with the appearance of a polygon block in mining away from gullies. We focused our attention on the case of mining away from a gully. We built a mechanical model in terms of a polygon block hinged structure and investigated the variation of horizontal thrust and shear force at the hinged point in relation to the rotation angle under different fragmentations. The Sliding-Rotation instability conditions of the polygon block hinged structure are presented based on the analyses of sliding instability and rotation instability. | 7 | Applicable conditions for a classification system of aquifer-protective mining in hallow coal seams显示文摘Based on the conclusions of domestic and foreign research, we have analyzed the collapse-fall characteristics of overlying strata and the mechanism of aquifer-protective mining in shallow coal seam working faces at the Shendong Mine. We have selected the height of the water-conducting fracture zone in overlying strata as a composite index and established the applicable conditions of aquifer-protective mining in shallow coal seams with a multi-factor synthetic-index classification method. From our calculations and analyses of variance, we used factors such as the overlying strata strength, mining disturbing factors and rock integrity as related factors of the composite index. We have classified the applicable conditions of aquifer-protective mining in shallow coal seams into seven types by comparing the result of the height of water-conducting fractured zones of long-wall and short-wall working faces with the thickness of the bedrock, the thickness of the weathered zone and the size of safety coal-rock pillars. As a result, we propose the preliminary classification system of aquifer-protective mining in shallow coal seams. | 8 | Optimized pre-treatment of high strength food waste digestate by high content aluminum-nanocluster based magnetic coagulation显示文摘Coagulation-based pre-treatment efficiency of high strength digestate of food waste(HSDFW) anaerobic digestion is negated by organic ligand-catalyzed decomposition of coagulants. In this study, an efficient HSDFW pre-treatment method, magnetic seeds(MS) coagulation, was employed by using highly stable Keggin Al_(30) nanocluster(PAC_(30)), MS and polyacrylamide(PAM), and its operation was optimized by evaluating the performance of removing turbidity, total suspended solids(TSS), chemical oxygen demand(COD), and total phosphorous(TP) phosphate. Results showed that at the optimum dosage of 4.82 g/L, PAC_(30) demonstrated excellent removals as high as 98.93% ± 0.1% of turbidity, 98.04% ± 0.1% of TSS, 58.28% ± 0.3% of total COD, 99.98% ± 0.01% of TP and 99.50% ± 0.01% of dissolved phosphate, respectively. Apparent molecular weight(AMW) and three-dimensional excitation-emission matrix(3 D-EEM) fluorescence spectroscopy analyses demonstrated more efficient removal of dissolved organic matter(DOM), particularly non-biodegradable and hydrophobic components by PAC_(30) than commercial coagulant. The sedimentation was much improved from 40 min by coagulation/flocculation to about 5 min settling by MS coagulation. | 10 | Scientific literature addressing brain glioma in the Web of Science A 10-year bibliometric analysis显示文摘Brain glioma is a hot topic in recent years; however, brain glioma remains poorly understood. A bibliometric analysis based on the Science Citation Index (SCI) published by the Institute of Scientific Information (ISI) was performed to identify the global research and to improve the understanding of research trends in the brain glioma field from 2001 to 2010. During 2001 to 2010, there were 8 413 papers addressing brain glioma added to the SCI, and this trend is increasing annually. Of these reports, 6 945 papers are written in English. Journals published in the United States had the most papers, including ten core source titles. The Journal of Neuro-Oncology published the most articles followed by Cancer Research. Furthermore, the University of California, San Francisco, is the most productive institution for publishing articles in the brain glioma field. | 11 | Diagnosis and clinical manifestations of subacute combined degeneration of the spinal cord: Analysis of 21 cases显示文摘BACKGROUND: Subacute combined degeneration of the spinal cord is caused by vitamin B12 deficiency and is a kind of degenerative disease owing the characteristics of nervous system diseases. In addition, different patients have variously clinical manifestations and various prognoses after vitamin B12 therapy. OBJECTIVE: To investigate and analyze diagnosis, clinical manifestations and prognosis of subacute combined degeneration of the spinal cord. DESIGN: Case analysis. SETTING: Department of Neurology, the Third Hospital of Peking University. PARTICIPANTS: A total of 21 subacute combined degeneration of the spinal cord patients including 14 males and 7 females aged from 33 to 82 years were selected from Department of Neurology, the Third Hospital of Peking University from January 1999 to December 2005. Duration from onset to final diagnosis lasted for 1.5–108 months. All patients had typically clinical manifestations; meanwhile, level of serum vitamin B12 was decreased and/or vitamin B12 therapy was effective. All patients provided the confirmed consent. METHODS: Clinical data of 21 subacute combined degeneration of the spinal cord patients were retrospectively analyzed, while general data and clinical characteristics were recorded at the same time. Levels of blood routine, serum vitamin B12 and homocysteine were measured at the phase of hospitalization. Normal value of serum vitamin B12 was 187–1 059 ng/L and normal value of serum homocysteine was 5– 15 μmol/L. All patients received neuroelectrophysiological examination and 15 patients received MRI examinations of spinal cord. After final diagnosis, patients were given vitamin B12 therapy. And follow-up was performed to investigate the prognosis. MAIN OUTCOME MEASURES: ① Levels of blood routine, serum vitamin B12 and homocysteine; ② results of neuroelectrophysiological examination; ③ results of MRI examination of spinal cord; ④ prognosis. RESULTS: Clinical data of 21 patients and follow-up data of 20 patients were involved in the final analysis and 1 patient was lost because of living in the other province. ① Clinical manifestations: All 21 patients had typically clinical manifestations. The original symptoms included numbness of lower and/or upper limbs (5 cases), unstable gait (3 cases), limb asthenia (4 cases), limb numbness combined with light asthenia (5 cases), limb numbness combined with unskillful activity (3 cases), and limb numbness combined with unstable gait (1 case). ② Experimental results: Eight subacute combined degeneration of the spinal cord patients accompanied with mild-severe anemia and mean corpuscular volume of 13 patients were increased. Among 13 subacute combined degeneration of the spinal cord patients not administrating vitamin B12 before hospitalization, the levels of serum vitamin B12 of 2 patients were not measured but those of other patients were decreased. After vitamin B12 therapy,the levels of serum vitamin B12 of 8 patients were normal or increased. In addition, the levels of serum homocysteine of 6 patients were not measured but those of 7 patients were increased. While, the levels of homocysteine of 5 following-up patients were normal. The levels of serum vitamin B12 of 8 patients who received with vitamin B12 therapy before hospitalization were normal or increased. Among them,the levels of homocysteine were not measured in 4 patients, those of 3 patients were increased, and that of 1 patient was normal. ③ Results of neuroelectrophysiological examination: Among all patients, 95% (20/21) patients had abnormal sensory-evoked potential, 89% (8/9) patients had abnormal motor evoked potential, 67% (10/15) patients had abnormal nerve conduction, 13% (2/15) patients had neurogenic muscle injury showed by electromyography (EMG), 70% (7/10) patients had abnormal brain-stem auditory evoked potential, and 40% (4/10) patients had abnormal visual evokedpotential. ④ Results of MRI examination of spinal cord: MRI examination demonstrated that 40% (6/15) patients had spinal cord lesion, but spinal cord lesion disappeared in 2 patients during follow up. In addition, clinical manifestations of patients were improved after standard vitamin B12 therapy. CONCLUSION: Nervous system lesion caused by vitamin B12 deficiency is not only involved in spinal cord, also in peripheral nerve, optic nerve, auditory pathway, etc. Diagnosis of the lesion depends on clinical characteristics and level of serum vitamin B12. | 12 | Tumor shrinkage by cyclopamine tartrate through inhibiting hedgehog signaling显示文摘The link of hedgehog (Hh) signaling activation to human cancer and synthesis of a variety of Hh signaling inhibitors raise great expectation that inhibiting Hh signaling may be effective in human cancer treatment. Cyclopamine (Cyc), an alkaloid from the Veratrum plant, is a specific natural product inhibitor of the Hh pathway that acts by targeting smoothened (SMO) protein. However, its poor solubility, acid sensitivity, and weak potency relative to other Hh antagonists prevent the clinical development of Cyc as a therapeutic agent. Here, we report properties of cyclopamine tartrate salt (CycT) and its activities in Hh signaling-mediated cancer in vitro and in vivo. Unlike Cyc, CycT is water soluble (5-10 mg/mL). The median lethal dose (LD50) of CycT was 62.5 mg/kg body weight compared to 43.5 mg/kg for Cyc, and the plasma half-life (T1/2) of CycT was not significantly different from that of Cyc. We showed that CycT had a higher inhibitory activity for Hh signaling-dependent motor neuron differentiation than did Cyc (IC50 = 50 nmol/L for CycT vs. 300 nmol/L for Cyc). We also tested the antitumor effectiveness of these Hh inhibitors using two mouse models of basal cell carcinomas (K14cre:Ptch1neo/neo and K14cre:SmoM2YFP). After topical application of CycT or Cyc daily for 21 days, we found that all CycT-treated mice had tumor shrinkage and decreased expression of Hh target genes. | 13 | Adenosine monophosphate-activated protein kinase activation enhances embryonic neural stem cell apoptosis in a mouse model of amyotrophic lateral sclerosis显示文摘Alterations in embryonic neural stem cells play crucial roles in the pathogenesis of amyotrophic lateral sclerosis. We hypothesized that embryonic neural stem cells from SOD1G93 A individuals might be more susceptible to oxidative injury, resulting in a propensity for neurodegeneration at later stages. In this study, embryonic neural stem cells obtained from human superoxide dismutase 1 mutant(SOD1G93A) and wild-type(SOD1WT) mouse models were exposed to H2O2. We assayed cell viability with mitochondrial succinic dehydrogenase colorimetric reagent, and measured cell apoptosis by flow cytometry. Moreover, we evaluated the expression of the adenosine monophosphate-activated protein kinase(AMPK) α-subunit, paired box 3(Pax3) protein, and p53 in western blot analyses. Compared with SOD1 WT cells, SOD1G93 A embryonic neural stem cells were more likely to undergo H2O2-induced apoptosis. Phosphorylation of AMPKα in SOD1G93 A cells was higher than that in SOD1 WT cells. Pax3 expression was inversely correlated with the phosphorylation levels of AMPKα. p53 protein levels were also correlated with AMPKαphosphorylation levels. Compound C, an inhibitor of AMPKα, attenuated the effects of H2O2. | 15 | Enhanced Answer Selection in CQA Using Multi-Dimensional Features Combination显示文摘Community Question Answering(CQA) in web forums, as a classic forum for user communication,provides a large number of high-quality useful answers in comparison with traditional question answering.Development of methods to get good, honest answers according to user questions is a challenging task in natural language processing. Many answers are not associated with the actual problem or shift the subjects,and this usually occurs in relatively long answers. In this paper, we enhance answer selection in CQA using multidimensional feature combination and similarity order. We make full use of the information in answers to questions to determine the similarity between questions and answers, and use the text-based description of the answer to determine whether it is a reasonable one. Our work includes two subtasks:(a) classifying answers as good, bad, or potentially associated with a question, and(b) answering YES/NO based on a list of all answers to a question. | 18 | Brassinosteroid and gibberellin coordinate rice seed germination and embryo growth by regulating glutelin mobilization显示文摘Seed germination is the beginning of a new lifecycle,and involves many complex physiological and biochemical reactions including seed reserve mobilization in the endosperm and nutrient transport and reuse in the embryo.Although glutelin is a dominant storage protein in rice,its contribution to seed germination and its regulatory mechanisms are mostly unknown.Gibberellin (GA) and brassinosteroid (BR),two major growth-promoting phytohormones,also play positive roles in controlling seed germination.However,how GA and BR interact and coordinate seed germination and facilitate glutelin mobilization remains unclear.In the present study,biochemical and physiological analyses of seed germination indicated that both GA and BR promote seed germination and post-germination growth.Exogenous application of GA restored germination defects caused by BR deficiency or insensitivity.Proteomic and q RT-PCR results showed that the expression of several glutelin proteins and their encoding genes was induced by BR and GA in the embryo.Expression assays suggested that the increased accumulation of glutelin protein in the embryo was due to the accelerated degradation of glutelin by a cysteine proteinase (REP-1) in the endosperm.The breakdown of glutelin in the endosperm showed a strict positive correspondence with the length of the shoot.The GluA2 mutation led to reduced degradation rate of glutelin and defects in seed germination,and the promotion effect of GA on seed germination was weakened in the glua2mutant.In vitro culture assay of rice embryos showed that glutelin mobilization functioned downstream of the GA and BR pathways to promote shoot elongation.These findings suggest a mechanism that mediates crosstalk between BR and GA in co-regulating rice seed germination and embryo growth. | Min Xiong Lingyi Chu Qianfeng Li Jiawen Yu Yihao Yang Peng Zhou Yong Zhou Changquan Zhang Xiaolei Fan Dongsheng Zhao Changjie Yan Qiaoquan Liu | 2021 | The Crop Journal2021,9,5: | 1 |
