| 1 | PEITC triggers multiple forms of cell death by GSH-iron-ROS regulation in K7M2 murine osteosarcoma cells显示文摘Phenethyl isothiocyanate(PEITC)is an isothiocyanate that largely exists in cruciferous vegetables and exhibits chemopreventive and chemotherapeutic potential against various cancers.However,it is little known about the molecular mechanisms of its antitumor action against osteosarcoma,which is the second highest cause of cancer-related death in children and adolescents.In this study,we investigated the effects of PEITC on K7M2murine osteosarcoma both in vitro and in vivo.We found that treatment with PEITC dose-dependently inhibited the viability of K7M2 murine osteosarcoma cells with an lCso value of 33.49μM at 24h.PEITC(1,15,30μM)dose-dependently inhibited the cell proliferation,caused G/M cell cycle arrest,depleted glutathione(GSH)generated reactive oxygen species(ROS),altered iron metabolism,and triggered multiple forms of cell death,namely ferroptosis apoptosis,and autophagy in K7M2 cells.We further revealed that PEITC treatment activated MAPK signaling pathway,and ROS generation was a major cause ofPEITC-induced cell death.In a syngeneic orthotopic osteosarcoma mouse model,administration of PEITC(30,60 mg/kg every day,ig,for 24 days)significantly inhibited the tumor growth,but higher dose of PEITC(90 mg/kg every day)compromised its ant-osteosarcoma effect.Histological examination showed that multiple cell death processes were initiated,iron metabolism was altered and MAPK signaling pathway was activated in the tumor tissues.In conclusion,we demonstrate that PEITC induces ferroptosis,autophagy,and apoptosis in K7M2 osteosarcoma cell by activating the ROS-elated MAPK signaling pathway.PEITC has promising anti-osteosarcoma activity.This study sheds light on,the redox signaling-based chemotherapeutics for cancers. | Huan-huan Lv Chen-xiao Zhen Jun-yu Liu Peng Shang | 2020 | Acta Pharmacologica Sinica2020,41,8: | 12 |

| 7 | Antitumor effects of the molecule-downsized immunocon-jugate composed of lidamycin and Fab' fragment of monoclonal antibody directed against type IV collagenase显示文摘Type IV collagenase plays an important role in tumor invasion and metastasis through cleaving type IV collagen in the basement membrane and extracellular matrix. In this study a molecule-downsized immunoconjugate (Fab′-LDM) was constructed by linking lidamycin (LDM), a highly potent antitumor antibiotic, to the Fab′ fragment of a monoclonal antibody directed against type IV collagenase and its antitumor effect was investigated. As assayed in 10% SDS-PAGE gel, the molecular weight of Fab′-LDM conjugate was 65 kD with a 1:1 molecular ratio of Fab′ and LDM. The Fab′-LDM conjugate maintained most part of the immunoreactivity of Fab′ fragment to both type IV collagense and mouse hepatoma 22 cells by ELISA. By MTT assay, Fab′-LDM conjugate showed more potent cytotoxicity to hepatoma 22 cells than that of LDM. Administered intravenously, Fab′-LDM conjugate proved to be more effective against the growth of subcutaneously transplanted hepatoma 22 in mice than free LDM in two experiment settings. In Experiment I, the drugs were given intravenously on day 1 and day 8. Fab′-LDM at the doses of 0.025 mg/kg, 0.05 mg/kg and 0.1 mg/kg inhibited tumor growth by 76.7%, 93.3% and 94.8%, while free LDM at 0.05 mg/kg inhibited tumor growth by 76.1%, respectively. In experiment II, the drugs were given intravenously on day 4 and day 11, Fab′-LDM at the doses of 0.025 mg/kg and 0.05 mg/kg inhibited tumor growth by 74.2%, 80.9%, while free LDM at 0.05 mg/kg inhibited tumor growth by 60.5%, respectively. In terms of survival time, Fab′-LDM was more effective than free LDM. | 9 | Prevalence of late-onset hypogonadism among middle-aged and elderly males in China:results from a national survey显示文摘This study aimed to propose an operational definition of late-onset hypogonadism(LOH)that incorporates both clinical symptoms and serum testosterone measurements to evaluate the prevalence of LOH in aging males in China.A population-based sample of 6296 men aged 40 years-79 years old was enrolled from six representative provinces in China.Serum total testosterone(TT),sex hormone-binding globulin(SHBG),and luteinizing hormone(LH)were measured and free testosterone(cFT)was calculated.The Aging Males’Symptoms(AMS)scale was used to evaluate the LOH symptoms.Finally,5078 men were included in this analysis.The TT levels did not decrease with age(P=0.59),and had no relationship with AMS symptoms(P=0.87 for AMS total score,P=0.74 for≥3 sexual symptoms).The cFT levels decreased significantly with age(P<0.01)and showed a negative association with the presence of≥3 sexual symptoms(P=0.03).The overall estimated prevalence of LOH was 7.8%(395/5078)if a cFT level<210 pmol l−1 combined with the presence of≥3 sexual symptoms was used as the criterion of LOH.Among them,26.1%(103/395)and 73.9%(292/395)had primary and secondary hypogonadism,respectively.After adjustment for confounding factors,primary and secondary hypogonadism was positively related to age and comorbidities.Body mass index was an independent risk factor for secondary hypogonadism.The results suggest that the AMS total score is not an appropriate indicator for decreased testosterone,and that the cFT level is more reliable than TT for LOH diagnosis.Secondary hypogonadism is the most common form of LOH. | Yi-Jun Liu Xu-Bo Shen Na Yu Xue-Jun Shang Yi-Qun Gu Lian-Dong Zuo Cheng-Liang Xiong Zhen Ye Yuan-Zhong Zhou | 2021 | Asian Journal of Andrology2021,23,2: | 6 |

Based on the drilling results, the traps controlled two or more faults are oil-rich. However, when only one fault cut through the sand body, there is no sign for hydrocarbon accumulation in the sandstone. In terms of this phenomenon, the principle of reservoir-forming controlled by fault terrace is proposed, i.e., when the single fault activates, because of the incompressibility of pore water, the resistance of pore and the direction of buoyancy, it is impossible for hydrocarbon to accumulate in sandstone. But when there are two or more faults, one of the faults acts as the spillway so the hydrocarbon could fill in the pore of sandstone through other faults. In total five gas bearing structures and four failure traps are considered, as examples to demonstrate our findings. | 14 | High-efficiency extraction synthesis for high-purity copper nanowires and their applications in flexible transparent electrodes显示文摘Copper nanowires(Cu NWs)are considered an excellent alternative to indium tin oxide(ITO)in flexible transparency electrodes(FTEs).However,the mixed particles and surface oxidation of Cu NWs degrade the transmittance and conductivity of the electrodes.Therefore,highly purified Cu NWs without oxidation are vital for high-performance FTEs.Herein,a facile and effective purification process is introduced to purify Cu NWs in a water and n-hexane system,which takes advantage of the differences in hydrophilicity between Cu NWs and Cu NPs caused by their different adsorption affinities to octadecylamine(ODA).At the same sheet resistance,the transmittance of the purified Cu NW-based FTEs improved approximately 2%compared to that of non-purified Cu NW-based FTEs.Immersion of the electrode in glacial acetic acid removed the surface organics and oxides.After only 40 s of treatment,the sheet resistance dramatically decreased from 10^5 Ohm/sq to 31 Ohm/sq with a transmittance of 85%.In addition,the Cu NW-based FTE conductors showed excellent flexibility(remaining stable after 1000 bending cycles).The Cu NW-based FTEs were further applied to fabricate a flexible transparent heater.At a voltage of 10 V,the temperature of the heater reached 73℃,demonstrating the potential applications of this material in various fields. | He Zhang Shang Wang Yanhong Tian Jiayue Wen Chunjin Hang Zhen Zheng Yilong Huang Su Ding Chenxi Wang | 2020 | Nano Materials Science2020,2,2: | 4 |

| 17 | Generation and antitumor effects of an engineered and energized fusion protein VL-LDP-AE composed of single-domain antibody and lidamycin显示文摘Type IV collagenase plays a pivotal role in invasion, metastasis and angiogenesis of tumor. Single domain antibodies are attractive as tumor-targeting vehicle because of their much smaller size com-pared with antibody molecules produced by conventional methods. Lidamycin (LDM) is a potent enediyne-containing antitumor antibiotic. In this study an engineered and energized fusion protein VL-LDP-AE composed of lidamycin and VL domain of mAb 3G11 directed against type IV collagenase was prepared using a novel two-step method. First a VL-LDP fusion protein was constructed by DNA recombination. Secondly VL-LDP-AE was obtained by molecular reconstitution. In MTT assay, VL-LDP-AE showed potent cytotoxicity to HT-1080 cells and KB cells with IC50 values of 8.55×10-12 and 1.70×10-11 mol/L, respectively. VL-LDP-AE showed antiangiogenic activity in chick chrorioallantoic membrane (CAM) assay and tube formation assay. In in vivo experiments, VL-LDP-AE was proved to be more effective than free LDM against the growth of subcutaneously transplanted hepatoma 22 in mice. Drugs were given intravenously on day 3 and 10 after tumor transplantation. Compared in terms of maximal tolerated doses, VL-LDP-AE at 0.25 mg/kg suppressed the tumor growth by 89.5%, LDM at 0.05 mg/kg by 69.9%, and mitomycin at 1 mg/kg by 35%. Having a molecular weight of 25.2 kDa, VL-LDP-AE was much smaller than other reported antibody-based drugs. The results suggested that VL-LDP-AE would be a promising candidate for tumor targeting therapy. | 19 | Low Phase Angle Predicts Poor Survival in Patients with Hepatocellular Carcinoma:A Retrospective Study显示文摘Background The phase angle(PA)is an indicator of nutritional status derived from bioelectrical impedance analysis(BIA).Studies have shown that the PA is one of several factors related to the survival of cancer patients.However,the correlation between the PA and prognosis in patients with hepatocellular carcinoma(HCC)has not been reported.This study aimed to explore the relationship between the PA,nutrition-related indicators,and patient prognosis.Methods The data from 248 HCC patients who underwent BIA between January 2015 and December 2019 at the First Hospital of Jilin University were retrospectively analyzed.Patients were divided into two groups according to the median PA value,then the nutrition-related indicators and overall survival(OS)were compared between the two groups.The correlations between the PA and laboratory test nutrition-related indicators and the results of a body composition analysis were determined using Pearson correlation analysis.Independent predictors of the patient prognosis were identified using univariate and multivariate analyses.Results The median OS in the high and low PA groups were 70 and 54.6 months,respectively(P=0.004).The Pearson correlation analysis revealed that the PA was associated with both laboratory test nutrition-related indicators and the results of a body composition analysis.The multivariable Cox regression analysis identified the PA as an independent risk factor affecting the prognosis of patients with Barcelona Clinic Liver Cancer stage A or B.Conclusion The PA is a good biomarker for the nutritional status.We found that the PA was associated with the prognosis in patients with HCC,with a low PA indicating a poor nutritional status and worse prognosis. | Jin Yu Shi Guang Yang Bo Liu Xiao Shang Guo Zhen Cui Jia Xin Huang Wan Ting Wang Ke Yu Chen Nan Ya Wang | 2022 | Journal of Nutritional Oncology2022,7,2: | 2 |
