A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. Further efforts should focus on identifying the etiology of the SARS-associated virus and ruling out conclusively the existence of otherpossible SARS-related pathogen(s). | QIN E'de ZHU Qingyu YU Man FAN Baochang CHANG Guohui SI Bingyin YANG Bao PENG Wenming JIANG Tao LIU Bohua DENG Yongqiang LIU Hong ZHANG Yu WANG Cui LI Yuquan GAN Yonghua LI Xiaoyu L Fushuang TAN Gang CAO Wuchun, YANG Ruifu Institute of Microbiology and Epidemiology, Chinese Academy of Military Medical Sciences, Beijing 100071, China WANG Jian, LI Wei, XU Zuyuan, LI Yan, WU Qingfa, LIN Wei, CHEN Weijun, TANG Lin, DENG Yajun, HAN Yujun, LI Changfeng, LEI Meng, LI Guoqing, LI Wenjie, L Hong, SHI Jianping, TONG Zongzhong, ZHANG Feng, LI Songgang, LIU Bin, LIU Siqi, DONG Wei, WANG Jun, Gane K-S Wong, YU Jun & YANG Huanming* Beijing Genomics Institute, Chinese Academy of Sciences, Beijing 101300 National Center for Genome Information, Beijing 101300, China | 2003 | Chinese Science Bulletin2003,48,10: | 121
Clearance Influence on Dynamic Response of Intermittent Roller Chain Drive显示文摘The clearances appear in chain link hinges induced by manufacturing tolerance or wear of components are the most important factor that influences the dynamic performance of the intermittent roller chain drives.Due to the existence of the clearances in chain link hinges, the serious impact vibration phenomenon which influences the stability and the position accuracy of the chain drive system would be caused in the intermittent motion. But,the problem may be that a reasonable modeling on the chain with clearances is difficult due to the large clearance's number in chain system and the fact that the clearance' size are different.Currently, the studies on the dynamics of the intermittent roller chain considering the multi-clearance' joints are rare. Most research works have only focused on the constant moving chains.Taking the intermittent roller chain system as an object, this paper designs and builds the experimental device of this kind of mechanical system. The Longitudinal vibration response of the intermittent chain under the different motion laws was tested. The experimental study shows that the clearances presented in chain link hinges can cause severe impact vibration in the intermittent motion of chain. In subsequent work, the dynamic model of the intermittent roller chain system with the multi-clearance' joints was established.By calculating the dynamic response of this kind of mechanical system under the different motion laws, the effect of the clearances on the dynamic response of the intermittent chain drive system was analyzed. The theoretical simulation shows that the serious impact vibration phenomenon of the chain system can be caused by the clearances at the start accelerating period, and the chain drive system is often accompanied by the severe shock and vibration at the moment that the chain moves from the acceleration period to deceleration period. The research conclusions made by the experimental and theoretical studies indicate that the use of motion laws with small and continuous jerk at conversion point can effectively suppress the impact vibration at this point caused by clearances' effect.Additionally, the use of nonsymmetrical motion laws with small jerk approaching the end of the indexing period can obtain a smaller residual vibration. The presented motion law can provide an important reference for the improvement of the dynamic performance of the intermittent chain drive systems. | XU Lixin YANG Yuhu CHANG Zongyu SHEN Zhaoguang LIU Jianping | 2010 | Chinese Journal of Mechanical Engineering2010,23,6: | 11
Effect of soothing liver therapy on oocyte quality and growth differentiation factor-9 in patients undergoing in vitro fertilization and embryo transfer显示文摘OBJECTIVE:To investigate the effect of Soothing liver therapy on infertile women undergoing in vitro fertilization and embryo transfer(IVF-ET)and to explore its mechanism.METHODS:Fifty-eight women with tubal infertility were randomized into two groups:30 in an experimental group treated with Xiaoyao powder(Shugan)plus gonadotropin-releasing hormone analog(GnRHa)/follicle-stimulating hormone(FSH)/human chorionic gonadotropin(hCG)and 28 in the control group who were treated with GnRHa/FSH/hCG only. The total gonadotropin(Gn)doses required, endometrial thickness, oocyte numbers, high quality embryo production rate and pregnancy rate of the two groups were compared. The concentration of growth differentiation factor-9(GDF-9)in follicular fluid was detected by western blotting and the expression of GDF-9 mRNA in granulosa cells was measured using reverse tran-scription-polymerase chain reaction amplification.RESULTS:In the experimental group, the Gn dose was significantly lower than that in the control group;the endometrial thickness, high quality embryo production and pregnancy rates were significantly higher and the expression of GDF-9 mRNA was also significantly higher than in the control group(all P<0.05).CONCLUSION:Shugan treatment can improve the pregnancy rate of women with tubal infertility;its mechanism is possibly related to the increased expression of GDF-9 in granulosa cells. | Xing Gao Xiufeng Chang Huilan Du Min Zhang Jianping Zhang Aiping Zhu | 2013 | Journal of Traditional Chinese Medicine2013,33,5: | 11
Dynamic Modeling of a Roller Chain Drive System Considering the Flexibility of Input Shaft显示文摘Roller chain drives are widely used in various high-speed, high-load and power transmission applications, but their complex dynamic behavior is not well researched. Most studies were only focused on the analysis of the vibration of chain tight span, and in these models, many factors are neglected. In this paper, a mathematical model is developed to calculate the dynamic response of a roller chain drive working at constant or variable speed condition. In the model, the complete chain transmission with two sprockets and the necessary tight and slack spans is used. The effect of the flexibility of input shaft on dynamic response of the chain system is taken into account, as well as the elastic deformation in the chain, the inertial forces, the gravity and the torque on driven shaft. The nonlinear equations of movement are derived from using Lagrange equations and solved numerically. Given the center distance and the two initial position angles of teeth on driving and driven sprockets corresponding to the first seating roller on each side of the tight span, dynamics of any roller chain drive with two sprockets and two spans can be analyzed by the procedure. Finally, a numerical example is given and the validity of the procedure developed is demonstrated by analyzing the dynamic behavior of a typical roller chain drive. The model can well simulate the transverse and longitudinal vibration of the chain spans and the torsional vibration of the sprockets. This study can provide an effective method for the analysis of the dynamic characteristics of all the chain drive systems. | XU Lixin YANG Yuhu1 CHANG Zongyu LIU Jianping | 2010 | Chinese Journal of Mechanical Engineering2010,23,3: | 7
Effects of Shuanghuanglian oral liquids on patients with COVID-19:a randomized,open-label,parallel-controlled,multicenter clinical trial显示文摘We conducted a randomized,open-label,parallel-controlled,multicenter trial on the use of Shuanghuanglian(SHL),a traditional Chinese patent medicine,in treating cases of COVID-19.A total of 176 patients received SHL by three doses(56 in low dose,61 in middle dose, and 59 in high dose)in addition to standard care. The control group was composed of 59 patients who received standard therapy alone.Treatment with SHL was not associated with a difference from standard care in the time to disease recovery.Patients with 14-day SHL treatment had significantly higher rate in negative conversion of SARS-CoV-2 in nucleic acid swab tests than the patients from the control group(93.4%vs.73.9%,P=0.006).Analysis of chest computed tomography images showed that treatment with high-dose SHL significantly promoted absorption of inflammatory focus of pneumonia, which was evaluated by density reduction of inflammatory focus from baseline,at day 7(mean difference(95%CI),−46.39(−86.83 to−5.94)HU;P=0.025)and day 14(mean difference(95%CI),−74.21(−133.35 to−15.08)HU;P=0.014).No serious adverse events occurred in the SHL groups. This study illustrated that SHL in combination with standard care was safe and partially effective for the treatment of COVID-19. | Li Ni Zheng Wen Xiaowen Hu Wei Tang Haisheng Wang Ling Zhou Lujin Wu Hong Wang Chang Xu Xizhen Xu Zhichao Xiao Zongzhe Li Chene Li Yujian Liu Jialin Duan Chen Chen Dan Li Runhua Zhang Jinliang Li Yongxiang Yi Wei Huang Yanyan Chen Jianping Zhao Jianping Zuo Jianping Weng Hualiang Jiang Dao Wen Wang | 2021 | Frontiers of Medicine2021,15,5: | 7
High efficiency generation of tunable ellipse perfect vector beams显示文摘We present a highly efficient method of generating and shaping ellipse perfect vector beams (EPVBs) with a prescribed ellipse intensity profile and continuously variant linear polarization state. The scheme is based on the coaxial superposition of two orthogonally polarized ellipse laser beams of controllable phase vortex serving as the base vector components. The phase-only computer-generated hologram is specifically designed by means of a modified iteration algorithm involving a complex amplitude constraint, which is able to generate an EPVB with high diffraction efficiency in the vector optical field generator. We experimentally demonstrate that the efficiency of generating the EPVB has a notable improvement from 1.83% in the conventional complex amplitude modulation based technique to 11.1% in our method. We also discuss and demonstrate the simultaneous shaping of multiple EPVBs with independent tunable ellipticity and polarization vortex in both transversal (2D) and axial (3D) focusing structures, proving potentials in a variety of polarization-mediated applications such as trapping and transportation of particles in more complex geometric circumstances. | LIN LI CHENLIANG CHANG CAOJIN YUAN SHAOTONG FENG SHOUPING NIE ZHI-CHENG REN HUI-TIAN WANG JIANPING DING | 2018 | Photonics Research2018,6,12: | 5
Age Structure of Females in a Breeding Population of Echinotriton chinhaiensis(Caudata:Salamandridae) and Its Conservation Implication显示文摘Knowledge of life history is important for understanding possible connections to population declines. Here,we investigated the female age structure and fecundity of Echinotriton chinhaiensis, one of the most endangered salamanders in the world, using skeletochronology based on specimens collected in 2008 and 2009 from a population in Ruiyansi, northeast of Ningbo,Zhejiang,China. The results showed that most female salamanders were between 5and 6 years of age, with the minimal reproductive age, predicted to be 3 years, and the clutch size correlated to the body size.We argue that both delayed attainment of sexual maturity and low fecundity make this species more vulnerable to extinction. | Weizhao YANG Chang LIU Jianping JIANG Cheng LI Feng XIE | 2011 | Asian Herpetological Research2011,2,2: | 4
A review of the interaction between the cryosphere and atmosphere显示文摘The interaction between the cryosphere and atmosphere is an essential and extremely sensitive mutual action process on the earth.Due to global warming and the cryospheric melting, more and more attention has been paid to the interaction process between the cryosphere and atmosphere, especially the feedback of the cryosphere change to the atmosphere.A comprehensive review of the studies on the interaction between the cryosphere and atmosphere is conducted from two aspects:(1)effects of climate change on the cryosphere or responses of the cryosphere to climate change;and(2)feedback of the cryosphere change to the climate. The response of the cryosphere to climate change is lagging.Such a lagging and cumulative effect of temperature rise within the cryosphere have resulted in a rapid change in the cryosphere in the 21st century, and its impacts have become more significant. The feedback from cryosphere change on the climate are omnifarious.Among them, the effects of sea ice loss and snow cover change, especially the Arctic sea ice loss and the Northern Hemisphere snow cover change, are the most prominent. The Arctic amplification(AA)associated with sea ice feedback is disturbing, and the feedback generated by the effect of temperature rise on snow properties in the Northern Hemisphere is also of great concern. There are growing evidence of the impact of the Arctic cryosphere melting on mid-latitude weather and climate.Weakened storm troughs, steered jet stream and amplified planetary waves associated with energy propagation become the key to explaining the links between Arctic cryosphere change and atmospheric circulation. There is still a great deal of uncertainty about how cryosphere change affects the weather and climate through different atmospheric circulation processes at different spatial and temporal scales due to observation and simulation problems. | YongJian Ding JianPing Yang ShengXia Wang YaPing Chang | 2020 | Research in Cold and Arid Regions2020,12,6: | 4
Disordered Translocation is Hastening Local Extinction of the Chinese Giant Salamander显示文摘Biodiversity is declining globally by an unprecedented extinction rate. This is especially true for amphibians, accounting for 24.3%of all threatened vertebrates. As the largest extant amphibian species in the world, wild populations of the Chinese giant salamander(Genus Andrias)(CGS)have decreased dramatically because of overexploitation and habitat degradation.Translocation has become an important strategy for restoring threatened wild populations worldwide. However, disordered tra nsloca tion usually has negative effects on the native populations. We provide an overview of CGS translocation and show that disordered translocation can increase local population extinction.Nearly four times the estimated number of wild individuals have been released across China. There a re three types of translocation used for CGS,namely, reinforcement, reintroduction and ecological replacement, the last of which accounts for over one-third of translocations. Our genetic screening revealed that most released individuals were not from local populations, with one to four lineages detected in every release site(n=6).This disordered translocation can potentially reduce the genetic integrity of original populations.Hence,we suggest suspending current CGS translocation activities immediately, until more robust measures can be developed and implemented to improve the current translocation program, especially with respect to lineage identifica tion a nd the identifica tion of appropriate release sites. | Guocheng SHU Ping LIU Tian ZHAO Cheng LI Yinmeng HOU Chunlin ZHAO Jie WANG Xiaoxiao SHU Jiang CHANG Jianping JIANG Feng XIE | 2021 | Asian Herpetological Research2021,12,3: | 4
First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. We for the first time extended the limit to the m_(χ)<6 GeV/c^(2) region. | Yi Wang Zhi Zeng Qian Yue LiTao Yang KeJun Kang YuanJing Li Mehment Agartioglu HaiPeng An JianPing Chang JingHan Chen YunHua Chen JianPing Cheng Cheng Yi Chiang WenHan Dai Zhi Deng ChangHao Fang XinPing Geng Hui Gong QiuJu Guo XuYuan Guo HongJian He Li He ShengMing He JinWei Hu TuChen Huang HanXiong Huang HaiTao Jia LiPing Jia Xi Jiang HauBin Li JianMin Li Jin Li MingXuan Li RenMingJie Li Xia Li YuLan Li Bin Liao FongKay Lin ShinTed Lin ShuKui Liu YanDong Liu Yu Liu YuanYuan Liu ZhongZhi Liu Hao Ma YuCai Mao QiYuan Nie JinHua Ning Hui Pan NingChun Qi Jie Ren XiChao Ruan ChangSong Shang Vivek Sharma Ze She Lakhwinder Singh Monoj Kumar Singh TianXi Sun ChangJian Tang WeiYou Tang Yang Tian GuangFu Wang Li Wang Qing Wang Yu Chen Wang YunXiang Wang Zhen Wang Henry Tsz-King Wong ShiYong Wu YuCheng Wu HaoYang Xing Yin Xu Tao Xue YuLu Yan Nan Yi ChunXu Yu HaiJun Yu JianFeng Yue Ming Zeng BingTao Zhang Lei Zhang FengShou Zhang ZhenYu Zhang KangKang Zhao MingGang Zhao JiFang Zhou ZuYing Zhou JingJun Zhu | 2021 | Science China(Physics,Mechanics & Astronomy)2021,64,8: | 3
Seasonal Interaction of River Water-Groundwater-Salt Lake Brine and Its Influence on Water-Salt Balance in the Nalenggele River Catchment in Qaidam Basin,NW China显示文摘Identifying interactions among river water, groundwater and salt lake brine is important for sustainable exploitation of brine mineral resources. In this study,we investigated the water exchange rate in dry and wet seasons, and assessed the influence of seasonal water exchange on brine concentration and crystallization process in the Nalenggele(NLGL)catchment of Qaidam Basin,China. The results show that the surface water infiltration and groundwater recharge rates in the wet season were 2.81×10^(-3)and 1.15×10^(-3)m^(3)/(s·m)in upstream,1.63×10^(-2)and 1.53×10^(-2)m^(3)/(s·m)in midstream, and 2.83×10^(-4)and 6.82×10^(-5)m^(3)/(s·m)in downstream, respectively;while their counterparts in the dry season were 9.81×10^(-4)and 5.05×10^(-4)m^(3)/(s·m)in upstream, and 8.34×10^(-3)and 7.78×10^(-3)m^(3)/(s·m)in midstream, respectively. The water exchange strongly influenced brine concentration and crystallinzation process, with brine chemistry belonging to 3K_(2)SO_(4)·Na_(2)SO_(4) and KCl types in wet and dry seasons, respectively. The strong water exchange in wet season destroyed the water-salt balance, while the low water exchange rate in dry season facilitated preparation of KCl products. | Jibin Han Jianxin Xu Lei Yi Zheng Chang Jianping Wang Haizhou Ma Baoyun Zhang Hongchen Jiang | 2022 | Journal of Earth Science2022,33,5: | 3

Effect of BaNH,CaNH,Mg3N2 on the activity of Co in NH3 decomposition catalysis显示文摘Development of active and non-noble metal-based catalyst for H2 production via NH3 decomposition is crucial for the implementation of NH3 as a H2 carrier.Co-based catalysts have received increasing attention because of its high intrinsic activity and moderate cost.In this work,we examined the effect of BaNH,CaNH and Mg3 N2 on the catalytic activity of Co in the NH3 decomposition reaction. The H2 formation rate ranks the order as Co-BaNH>Co-CaNH>Co-Mg3 N2≈Co/CNTs within a reaction temperature range of 300-550℃.It is worth pointing out that the H2 formation rate of Co-BaNH at 500℃reaches20 mmolH2 gcat-1 min-1,which is comparable to those of the active Ru/Al2 O3(ca.17 mmolH2 gcat-1 min1)and Ru/AC(21 mmolH2 gcat-1 min-1)catalysts under the similar reaction conditions.In-depth research shows that Co-BaNH exhibits an obviously higher intrinsic activity and much lower Ea(46.2 kJ mol-1)than other Co-based catalysts, suggesting that BaNH may play a different role from CaNH,Mg3 N2 and CNTs during the catalytic process.Combined results of XRD,Ar-TPD and XAS show that a[Co-N-Ba]-like intermediate species is likely formed at the interface of Co metal and BaNH,which may lead to a more energy-efficient reaction pathway than that of neat Co metal for NH3 decomposition. | Pei Yu Han Wu Jianping Guo Peikun Wang Fei Chang Wenbo Gao Weijin Zhang Lin Liu Ping Chen | 2020 | Journal of Energy Chemistry2020,29,7: | 3

Current management of chemotherapy-induced neutropenia in adults:key points and new challenges显示文摘Chemotherapy-induced neutropenia(CIN)is a potentially fatal and common complication in myelosuppressive chemotherapy. The timing and grade of CIN may play prognostic and predictive roles in cancer therapy.CIN is associated with older age, poor functional and nutritional status, the presence of significant comorbidities, the type of cancer, previous chemotherapy cycles, the stage of the disease, specific chemotherapy regimens, and combined therapies. There are many key points and new challenges in the management of CIN in adults including:(1)Genetic risk factors to evaluate the patient’s risk for CIN remain unclear. However, these risk factors urgently need to be identified.(2)Febrile neutropenia(FN)remains one of the most common reasons for oncological emergency. No consensus nomogram for FN risk assessment has been established.(3)Different assessment tools[e.g.,Multinational Association for Supportive Care in Cancer(MASCC),the Clinical Index of Stable Febrile Neutropenia(CISNE)score model, and other tools]have been suggested to help stratify the risk of complications in patients with FN.However, current tools have limitations. The CISNE score model is useful to support decision-making,especially for patients with stable FN.(4)There are still some challenges, including the benefits of granulocyte colony stimulating factor treatment and the optimal antibiotic regimen in emergency management of FN.In view of the current reports, our group discusses the key points, new challenges, and management of CIN. | Committee of Neoplastic Supportive-Care(CONS),China Anti-Cancer Association Committee of Clinical Chemotherapy,China Anti-Cancer Association Yi Ba Yuankai Shi Wenqi Jiang Jifeng Feng Ying Cheng Li Xiao Qingyuan Zhang Wensheng Qiu Binghe Xu Ruihua Xu Bo Shen Zhiguo Luo Xiaodong Xie Jianhua Chang Mengzhao Wang Yufu Li Yuerong Shuang Zuoxing Niu Bo Liu Jun Zhang Li Zhang Herui Yao Conghua Xie Huiqiang Huang Wangjun Liao Gongyan Chen Xiaotian Zhang Hanxiang An Yanhong Deng Ping Gong Jianping Xiong Qinghua Yao Xin An Cheng Chen Yanxia Shi Jialei Wang Xiaohua Wang Zhiqiang Wang Puyuan Xing Sheng Yang Chenfei Zhou | 2020 | Cancer Biology & Medicine2020,17,4: | 2
Mineral Prospectivity Prediction via Convolutional Neural Networks Based on Geological Big Data显示文摘Today’s era of big data is witnessing a gradual increase in the amount of data, more correlations between data,as well as growth in their spatial dimension.Conventional linear statistical models applied to mineral prospectivity mapping(MPM)perform poorly because of the random and nonlinear nature of metallogenic processes. To overcome this performance degradation, deep learning models have been introduced in 3 D MPM.In this study, taking the Huayuan sedimentary Mn deposit in Hunan Province as an example,we construct a 3 D digital model of this deposit based on the prospectivity model of the study area.In this approach,3 D predictor layers are converted from the conceptual model and employed in a 3 D convolutional neural network(3 D CNN).The characteristics of the spatial distribution are extracted by the 3 D CNN.Subsequently,we divide the 22 extracted ore-controlling variables into six groups for contrast experiments based on various combinations and further apply the 3 D CNN model and weight of evidence(WofE)method on each group. The predictive model is trained on the basis of the coupling correlation between the spatial distributions of the variables and the underground occurrence space of the Mn orebodies, and the correlation between different ore-controlling factors. The analysis of 12 factors indicates that the 3 D CNN model performs well in the 3 D MPM,achieving a promising accuracy of up to 100%and a loss value below 0.001.A comparison shows that the 3 D CNN model outperforms the WofE model in terms of predictive evaluation indexes, namely the success rate and ore-controlling rate.In particular, the 1–12 ore-controlling factors selected in experiment 5 provide a significantly better prediction effect than the other factors.Consequently,we conclude that the Mn deposit in the study area is not only related to the stratum and interlaminar anomalous bodies but also to the spatial distribution of the faults. The experimental results confirm that the proposed 3 D CNN is promising for 3 D MPM as it eliminates the interference factors. | Shi Li Jianping Chen Chang Liu Yang Wang | 2021 | Journal of Earth Science2021,32,2: | 2

Reversal of Adriamycin Resistance of Hepatocellular Carcinoma by Targeting with Recombined Adenovirus Caning Antisense mdr1 RNA显示文摘OBJECTIVE Chemotherapy is an important therapy for hepatocellular carcinoma (HCC). However, it is not effective in many cases due to recurrence and metastasis even if the initial treatment produces a response. Multidrug resistance (MDR) is considered to be one of the considerable causes. The aim of this study was to reverse MDR of HepG2/ADM cells by blocking mdr1 with an adenovirus vector carrying antisense mdr1 in a tumor transplantated in athymic mice. METHODS pCMV IE was removed from the pshuttle vector. A 0.3 kb AFP promoter was inserted into the pshuttle vector and pCMV changed into pAFP. The pAFP and asmdr1 PCR products were doubly digested with Kpnl and Apal, the digested products were ligated by T4 ligase, the asmdrl gene was inserted into pAFP and a newly plasmid pAFP-asmdr1 was constructed. Following digestion with PI-Scel/l-Ceu l, pAFP-asmdr1 was ligated with Adeno-X genome DNA and amplified in E.coli XL 1-Blue. The HEK293 cells were transfected and virus collected. The HepG2 MDR cells (HepG2/ADM) were induced by graded resistance to ADM and were inoculated into athymic mice. After adeno-asmdr1 was injected, the expression of mdr1 -mRNA and the volume of the transplantated tumor and its cells were observed. RESULTS Following injection with Adeno-asmdr1, the tumor volume in the ADM+Adeno-asmdr1 group did not increase. However the tumor volume in the PBS plus ADM group did significantly increase (P<0.05). In the tumor xenograft cells, mdr1 mRNA in the xenografts was assessed by RT-PCR and was found to be reduced at 1 week and 4 weeks in the ADM+asmdr1 group, but it was stable in the ADM group. It was only 20% in the ADM+asmdr1 group compared to the ADM group at the 4th week (P<0.05). Evidence of apoptosis was observed in the tumor xenograft cells treated with Adeno-asmdr1, but there was rare or no apoptosis in the group treated with ADM and PBS. CONCLUSION Adenovirus carrying antisense mdr1 RNA can partially reverse the MDR of HepG2/ADM cells and inhibit tumor growth by down-regulating mdr1 mRNA resulting in tumor cell apoptosis. | Xiong Ding Ying Mei Yujun Shi Jianping Gong Xuhong Li Yong Peng Yong Liu Chang'an Liu | 2006 | Chinese Journal of Clinical Oncology2006,3,1: | 1
Experimental verification of parameter m in Hoeke-Brown failure criterion considering the effects of natural fractures显示文摘HoekeBrown failure criterion is one of the widely used rock strength criteria in rock mechanics and mining engineering.Based on the theoretical expression of HoekeBrown parameter m of an intact rock, the parameter m has been modified by crack parameters for fractured rocks. In this paper, the theoretical value range and theoretical expression form of the parameter m in HoekeBrown failure criterion were discussed.A critical crack parameter B was defined to describe the influence of the critical crack when the stress was at the peak, while a parameter b was introduced to represent the distribution of the average initial fractures. The parameter m of a fractured rock contained the influences of critical crack(B),confining pressure(s3)and initial fractures(b).Then the triaxial test on naturally fractured limestones was conducted to verify the modification of the parameter m.From the ultrasonic test and loading test results of limestones, the parameter m can be obtained, which indicated that the confining pressure at a high level reduced the differences of m among all the specimens. The confining pressure s3 had an exponential impact on m,while the critical crack parameter B had a negative correlation with m.Then the expression of m for a naturally fractured limestone was also proposed. | Xu Wei Jianping Zuo Yue Shi Haiyan Liu Yunqian Jiang Chang Liu | 2020 | Journal of Rock Mechanics and Geotechnical Engineering2020,12,5: | 1
