2Observational study of the circulation in the southern South China Sea显示文摘CTD data from two oceanographic survey cruises, conducted by 'Shiyan No.3' R/V during two reversing monsoons, were employed to calculate and analyze the distributions of temperature, salinity and geopotential anomaly in the southern South China Sea (SSCS). Based on these distributions, the horizontal structures of the SSCS circulation are described systematically. Study results show that the SSCS circulation structure could be classified as a two-layer (the upper and the lower) pattern, and that the main circulation in the upper layer (0-400 m) is driven by the South China Sea monsoons and is opposite in direction to the reversal of the monsoons. The distributions of principal current systems and their features are presented. 6Genome-and transcriptome-wide association studies provide insights into the genetic basis of natural variation of seed oil content in Brassica napus显示文摘Seed oil content(SOC)is a highly important and complex trait in oil crops.Here,we decipher the genetic basis of natural variation in SOC of Brassica napus by genome-and transcriptome-wide association studies using 505 inbred lines.We mapped reliable quantitative trait loci(QTLs)that control SOC in eight environments,evaluated the effect of each QTL on SOC,and analyzed selection in QTL regions during breeding.Six-hundred and ninety-two genes and four gene modules significantly associated with SOC were identified by analyzing population transcriptomes from seeds.A gene prioritization framework,POCKET(prioritizing the candidate genes by incorporating information on knowledge-based gene sets,effects of variants,genome-wide association studies,and transcriptome-wide association studies),was implemented to determine the causal genes in the QTL regions based on multi-omic datasets.A pair of homologous genes,BnPMT6s,in two QTLs were identified and experimentally demonstrated to negatively regulate SOC.This study provides rich genetic resources for improving SOC and valuable insights toward understanding the complex machinery that directs oil accumulation in the seeds of B.napus and other oil crops.Shan Tang Hu Zhao Shaoping Lu Liangqian Yu Guofang Zhang Yuting Zhang Qing-Yong Yang Yongming Zhou Xuemin Wang Wei Ma Weibo Xie Liang Guo 2021Molecular Plant2021,14,3:6
7Epithelial-mesenchymal transition status of circulating tumor cells in breast cancer and its clinical relevance显示文摘Objective:Circulating tumor cells(CTCs)play a critical role in cancer metastasis,but their prevalence and significance remain unclear.This study attempted to track the epithelial-mesenchymal transition(EMT)status of CTCs in breast cancer patients and investigate their clinical relevance.Methods:In this study,the established negFACS-IF:E/M platform was applied to isolate rare CTCs and characterize their EMT status in breast cancer.A total of 89 breast cancer patients were recruited,including stage 0–III(n=60)and late stage(n=29)cases.Results:Using the negFACS-IF:E/M platform,it was found that in human epidermal growth factor receptor 2(HER2)+patients,mesenchymal CTCs usually exhibited a high percentage of HER2+cells.Stage IV breast cancer patients had considerably more CTCs than stage 0–III patients.Among stage 0–III breast cancers,the HER2 subtype included a significantly higher percentage of mesenchymal and biphenotypic(epithelial and mesenchymal)CTCs than the luminal A or B subtypes.Among stage IV patients,CTCs were predominantly epithelial in cases with local recurrence and were more mesenchymal in cases with distant metastasis.By applying a support vector machine(SVM)algorithm,the EMT status of CTCs could distinguish between breast cancer cases with metastasis/local recurrence and those without recurrence.Conclusions:The negFACS-IF:E/M platform provides a flexible and generally acceptable method for the highly sensitive and specific detection of CTCs and their EMT traits in breast cancer.This study demonstrated that the EMT status of CTCs had high clinical relevance in breast cancer,especially in predicting the distant metastasis or local recurrence of breast cancer.Jiaojiao Zhou Xuan Zhu Shijie Wu Jingxin Guo Kun Zhang Chunjing Xu Huihui Chen Yuxi Jin Yuting Sun Shu Zheng Yiding Chen 2020Cancer Biology & Medicine2020,17,1:3
8A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively. 9Dissipative particle dynamics thermostat: a novel thermostat for molecular dynamics simulation of liquid crystals with Gay–Berne potential显示文摘The Gay–Berne(GB) model has been proved to be highly successful in the simulation of liquid crystal phases via both molecular dynamics(MD) and nonequilibrium molecular dynamics(NEMD). However, the conventional thermostats used in the simulations of GB systems, such as Nosé–Hoover and Langevin thermostats, have serious shortcomings especially in NEMD simulations. Recently, dissipative particle dynamics(DPD) has established itself as a useful thermostat for soft matter simulations, whereas the application of DPD thermostat in(NE)MD simulations is limited to the spherically isotropic potential models, such as the Lennard–Jones model. Considering the virtues of the DPD thermostat, that is, local, momentum conserved, and Galilean invariant, we extend the DPD thermostat to the non-spherical GB model. It is interesting to find that the translational DPD and rotational DPD thermostats can be used in the GB system independently and both can achieve the thermostatting effects. Also, we compared the performance of the DPD thermostat with other commonly used thermostats in NEMD simulations by investigating the streaming velocity profiles and the dynamics of phase separation in a typical but simple binary GB mixture under shear field. It is revealed that the known virtues of DPD thermostats, such as Galilean invariant, shear velocity profile-unbiased, and unscreened hydrodynamic interactions, are still intact when applying to GB systems. 10Modulating the optical and electrical properties of MAPbBr_(3) single crystals via voltage regulation engineering and application in memristors显示文摘Defect density is one of the most significant characteristics of perovskite single crystals(PSCs)that determines their optical and electrical properties,but few strategies are available to tune this property.Here,we demonstrate that voltage regulation is an efficient method to tune defect density,as well as the optical and electrical properties of PSCs.A three-step carrier transport model of MAPbBr_(3) PSCs is proposed to explore the defect regulation mechanism and carrier transport dynamics via an applied bias.Dynamic and steady-state photoluminescence measurements subsequently show that the surface defect density,average carrier lifetime,and photoluminescence intensity can be efficiently tuned by the applied bias.In particular,when the regulation voltage is 20 V(electrical poling intensity is 0.167 Vμm^(−1)),the surface defect density of MAPbBr_(3) PSCs is reduced by 24.27%,the carrier lifetime is prolonged by 32.04%,and the PL intensity is increased by 112.96%.Furthermore,a voltage-regulated MAPbBr_(3) PSC memristor device shows an adjustable multiresistance,weak ion migration effect and greatly enhanced device stability.Voltage regulation is a promising engineering technique for developing advanced perovskite optoelectronic devices.Jun Xing Chen Zhao Yuting Zou Wenchi Kong Zhi Yu Yuwei Shan Qingfeng Dong Ding Zhou Weili Yu Chunlei Guo 2020Light(Science & Applications)2020,9,1:3

12Inflammation-induced inhibition of chaperone-mediated autophagy maintains the immunosuppressive function of murine mesenchymal stromal cells显示文摘Macroautophagy has been implicated in modulating the therapeutic function of mesenchymal stromal cells(MSCs).However,the biological function of chaperone-mediated autophagy(CMA)in MSCs remains elusive.Here,we found that CMA was inhibited in MSCs in response to the proinflammatory cytokines interferon-γ(IFN-γ)and tumor necrosis factor-α(TNF-α).In addition,suppression of CMA by knocking down the CMA-related lysosomal receptor lysosomal-associated membrane protein 2(LAMP-2A)in MSCs significantly enhanced the immunosuppressive effect of MSCs on T cell proliferation,and as expected,LAMP-2A overexpression in MSCs exerted the opposite effect on T cell proliferation.This effect of CMA on the immunosuppressive function of MSCs was attributed to its negative regulation of the expression of chemokine C-X-C motif ligand 10(CXCL10),which recruits inflammatory cells,especially T cells,to MSCs,and inducible nitric oxide synthase(iNOS),which leads to the subsequent inhibition of T cell proliferation via nitric oxide(NO).Mechanistically,CMA inhibition dramatically promoted IFN-γplus TNF-α-induced activation of NF-κB and STAT1,leading to the enhanced expression of CXCL10 and iNOS in MSCs.Furthermore,we found that IFN-γplus TNF-α-induced AKT activation contributed to CMA inhibition in MSCs.More interestingly,CMA-deficient MSCs exhibited improved therapeutic efficacy in inflammatory liver injury.Taken together,our findings established CMA inhibition as a critical contributor to the immunosuppressive function of MSCs induced by inflammatory cytokines nd highlighted a previously unknown function of CMA.Jie Zhang Jiefang Huang Yuting Gu Mingxing Xue Fengtao Qian Bei Wang Wanlin Yang Hongshuang Yu Qiwei Wang Xin Guo Xinyuan Ding Jina Wang Min Jin Yanyun Zhang 2021Cellular & Molecular Immunology2021,18,6:3
13Study of establishing disease-syndrome combined with animal model for immune thrombocytopenic purpura without additional conditions显示文摘Objective:To explore the feasibility of establishing the disease-syndrome combined animal model for immune thrombocytopenic purpura(ITP)without additional conditions.Methods:Three batches of data related to the ITP model mice obtained by replication at different time were analyzed,and whether the APS-injected model mice replicated through the passive immune modeling method could simulate the pathogenesis and clinical characteristics of human ITP was evaluated according to the differentiation criteria for diseasesyndrome combined model.Results:The APS-injected replicated ITP model mice possessed the following traits:(1)Compared with the normal group,the platelet count was significantly decreased,and coagulation time was significantly increased in the model group(P<.01).(2)Compared with the normal group,the medullary thrombocytogenous megakaryocytes were significantly decreased(P<.05,.01,.001).(3)The APS-injected sites and other parts of the model mice had spontaneous hemorrhage.(4)Behavioral changing signs were observed 1 week after the modeling(i.e.low activity,delayed activity,poor appetite,skin petechia/hemorrhage and spontaneous hemorrhage at the injected sites or other parts),and were getting more and more severe.Conclusion:According to the syndrome differentiation criteria for disease-syndrome combined model of ITP,the APS-injected animal model of ITP replicated through the passive immune modeling method without additional conditions possesses the characteristics of disease-syndrome combined model.It provides an ideal tool for the development of traditional Chinese medicine pharmacology experiment.Haiyan Lang Ming Guo Yuting Chu Wei Ma Yayue Zhang Ling Zhang Xinyi Chen 2016Journal of Traditional Chinese Medical Sciences2016,3,3:2
14A general and simple method for evaluating the electrical transport performance of graphene by the van der Pauw–Hall measurement显示文摘Expected for many promising applications in the field of electronics and optoelectronics, a reliable method for the characterization of graphene electrical transport properties is desired to predict its device performance or provide feedback for its synthesis.However, the commonly used methods of extracting carrier mobility from graphene field effect transistor or Hall-bar is time consuming, expensive, and significantly affected by the device fabrication process other than graphene itself.Here we reported a general and simple method to evaluate the electrical transport performance of graphene by the van der Pauw–Hall measurement.By annealing graphene in vacuum to remove the adsorbed dopants and then exposing it in ambient surroundings, carrier mobility as a function of density can be measured with the increase of carrier density due to the dopant re-adsorption from the surroundings.Further, the relationship between the carrier mobility and density can be simply fitted with a power equation to the first level approximation, with which any pair of measured carrier mobility and density can be normalized to an arbitrary carrier density for comparison.We experimentally demonstrated the reliability of the method, which is much simpler than making devices and may promote the standard making for graphene characterization.Fangzhu Qing Yang Shu Linsen Qing Yuting Niu He Guo Shuyi Zhang Chunlin Liu Changqing Shen Wanli Zhang Samuel S.Mao Wenjuan Zhu Xuesong Li 2018Science Bulletin2018,63,22:2
15The Scorpion Venom Peptide Smp76 Inhibits Viral Infection by Regulating Type-Ⅰ Interferon Response显示文摘Dengue virus(DENV) and Zika virus(ZIKV) have spread throughout many countries in the developing world and infect millions of people every year, causing severe harm to human health and the economy. Unfortunately, there are few effective vaccines and therapies available against these viruses. Therefore, the discovery of new antiviral agents is critical.Herein, a scorpion venom peptide(Smp76) characterized from Scorpio maurus palmatus was successfully expressed and purified in Escherichia coli BL21(DE3). The recombinant Smp76(rSmp76) was found to effectively inhibit DENV and ZIKV infections in a dose-dependent manner in both cultured cell lines and primary mouse macrophages. Interestingly,rSmp76 did not inactivate the viral particles directly but suppressed the established viral infection, similar to the effect of interferon(IFN)-b. Mechanistically, rSmp76 was revealed to upregulate the expression of IFN-b by activating interferon regulatory transcription factor 3(IRF3) phosphorylation, enhancing the type-Ⅰ IFN response and inhibiting viral infection.This mechanism is significantly different from traditional virucidal antimicrobial peptides(AMPs). 17SERS study on the synergistic effects of electric field enhancement and charge transfer in an Ag2S quantum dots/plasmonic bowtie nanoantenna composite system显示文摘Localized surface plasmon resonance(LSPR)of nanostructures and the interfacial charge transfer(CT)of semiconductor materials play essential roles in the study of optical and photoelectronic properties.In this paper,a composite substrate of Ag2S quantum dots(QDs)coated plasmonic Au bowtie nanoantenna(BNA)arrays with a metalinsulator-metal(MIM)configuration was built to study the synergistic effect of LSPR and interfacial CT using surface-enhanced Raman scattering(SERS)in the near-infrared(NIR)region.The Au BNA array structure with a large enhancement of the localized electric field(E-field)strongly enhanced the Raman signal of adsorbed p-aminothiophenol(PATP)probe molecules.Meanwhile,the broad enhanced spectral region was achieved owing to the coupling of LSPR The as-prepared Au BNA array structure facilitated enhancements of the excitation as well as the emission of Raman signal simultaneously,which was established by finite-difference time-domain simulation.Moreover,Ag2S semiconductor QDs were introduced into the BNA/PATP system to further enhance Raman signals,which benefited from the interfacial CT resonance in the BNA/Ag2S-QDs/PATP system.As a result,the Raman signals of PATP in the BNA/Ag2S-QDs/PATP system were strongly enhanced under 785 nm laser excitation due to the synergistic effect of E-field enhancement and interfacial CT.Furthermore,the SERS polarization dependence effeas of the BNA/Ag2S-QDs/PATP system were also investigated.The SERS spectra indicated that the polarization dependence of the substrate increased with decreasing polarization angles(θpola)of excitation from p-polarized(θpola=90°)excitation to s-polarized(θpola=0°)excitation.This study provides a strategy using the synergistic effect of interfacial CT and E-field enhancement for SERS applications and provides a guidance for the development of SERS study on semiconductor QD-based plasmonic substrates,and can be farther extended to other material-nanostructure systems for various optoelectronic and sensing applications.BIN WANG CHEN ZHAO HUANYU LU TINGTING ZOU SUBHASH C.SINGH ZHI YU CHAONAN YAO XIN ZHENG JUN XING YUTING ZOU CUNZHU TONG WEILI YU BO ZHAO CHUNLEI GUO 2020Photonics Research2020,8,4:1
