| 3 | Distinct Effect of CD40 and TNF-Signaling on the Chemokine/Chemokine Receptor Expression and Function of the Human Monocyte-Derived Dendritic Cells显示文摘A key and limiting step in the process of human monocyte-derived dendritic cells (mDCs) for clinical use is their in vitro maturation and in vivo migration. We previously observed that CD40 signal facilitated human mDC growth and maturation. To further explore this process,mDCs generated with GM-CSF and IL-4 were co-cultured with apoptotic tumor cells for 24 hours,followed by incubating with anti-CD40 monoclonal antibody or TNF-α for 48 hours to generate mature DCs. The chemokine/chemokine receptor expression and functions of mature DCs upon various stimuli were determined. The expression of costimulatory molecules on apoptotic tumor cell-loaded mature DCs co-cultured with either anti-CD40 antibody (anti-CD40-DCs) or TNF-α (TNF-DCs) were up-regulated compared to immature DCs,consistent with the abilities of these cytokine to drive DC maturation in vitro. The mRNA levels of chemokines such as stromal cell-derived factor-1α (SDF-1α),EBV-induced molecule 1 ligand chemokine (ELC),and IFN inducible protein-10 (IP-10) in anti-CD40 activated DCs were increased and the dendritic cell-specific chemokine 1 (DC-CK1) was moderately up-regulated as compared with other mature DCs. The corresponding chemokine receptors CXCR4 and CCR7 of anti-CD40-DCs were significantly expressed. The CXCR3 expression on activated T cells stimulated by anti-CD40-DCs was also increased. Moreover,the anti-CD40-DCs had a stronger ability to stimulate T cell proliferation than any other DCs. The NF-κB activity was much higher in anti-CD40-DCs than that of TNF-DCs. | 4 | FoxO directly regulates the expression of TOR/S6K and vitellogenin to modulate the fecundity of the brown planthopper显示文摘As a conserved transcription factor,FoxO plays a crucial role in multiple physiological processes in vivo,including stress resistance,longevity,growth and reproduction.Previous studies on FoxO have focused on human,mouse,Drosophila melanogaster and Caenorhabditis elegans,while there are few reports on agricultural pests and little is known about how FoxO modulates insect fecundity.In Asia,the brown planthopper(BPH)Nilaparvata lugens(St?l)is one of the most serious pests in rice production and high fecundity is the basis of the outbreak of BPH.Here,using the genome-wide ChIP-seq of NlFoxO in BPH,we found that NlFoxO binds to the promoters of ribosomal protein S6 kinase(NlS6K)and serine/threonine-protein kinase mTOR(NlTOR)and increases their expression levels.We also found that NlFoxO directly binds to the exon of vitellogenin(NlVg)and has a specific inhibitory effect on its expression.In addition,the number of eggs laid and their hatching rate decreased significantly after injection of NlFoxO double-stranded RNA into BPH adults.Our findings provide direct evidence that FoxO modulates insect fecundity through binding to the promoters of NlS6K,NlTOR and the exon of NlVg and affecting their gene expression in the Vg network. | Yi Dong Weiwen Chen Kui Kang Rui Pang Yipei Dong Kai Liu Wenqing Zhang | 2021 | Science China(Life Sciences)2021,64,1: | 6 |
| 5 | Water quality criteria for 4-nonylphenol in protection of aquatic life显示文摘Here reproduction and death toxicity data were selected for 4-nonylphenol based on endocrine disrupting properties of the reproductive system. Assessment factor (AF) and species sensitivity distribution (SSD) curve were employed to derive criteria maximum concentration (CMC) and criteria continuous concentration (CCC) of three different 4-nonylphenols for protection of aquatic life. The results showed that the CMC and CCC based on SSD method and death toxicity data for three different 4-nonylphenols (CAS No: 104405, 25154523, 84852163) were 26.7, 13.6, 3.84 g L 1 and 8.86, 2.21, 0.97 g L 1, respectively. Based on SSD and reproductive toxicity data, the CCC values of different 4-nonylphenols (CAS No: 104405, 25154523) were 1.59 and 1.34 g L 1, respectively. The CCC values obtained by the AF for three different 4-nonylphenol (CAS No: 104405, 25154523, 84852163) were 0.165, 1.03, 0.74 g L 1 and 0.5, 0.5, 0.1 g L 1, respectively, based on death toxicity data and reproductive toxicity data. The CCC values obtained by AF were all lower than the corresponding criteria values obtained by SSD, and the CCC values based on reproductive toxicity data were less than those based on the death toxicity data. | 6 | High efficiency synthesis of HKUST-1 under mild conditions with high BET surface area and CO_2 uptake capacity显示文摘This study focuses on the development of a hydrothermal method for the rapid synthesis of good quality copper benzene-1,3,5-tricarboxylate(referred to as HKUST-1)with high yield under mild preparation conditions to address the issues associated with reported methods.Different synthesis conditions and activation methods were studied to understand their influence on the properties of HKUST-1.It was found that mixing the precursors at50°C for 3 h followed by activation via methanol refluxing led to the formation of a product with the highest BET specific surface area of 1615 m^2/g and a high yield of 84.1%.The XRD and SEM data illustrated that the product was highly crystalline.The sample was also tested on its capacity in CO_2 adsorption.The results showed strong correlation between surface area of the sample and its CO_2 uptake at 1 bar and 27°C.The HKUST-1 prepared in this study demonstrated a high CO_2 uptake capacity of 4.2 mmol/g.It is therefore concluded that this novel and efficient method can be used in the rapid preparation of HKUST-1 with high surface area and CO_2 uptake capacity. | Yipei Chen Xueliang Mu Edward Lester Tao Wu | 2018 | Progress in Natural Science:Materials International2018,28,5: | 3 |
| 7 | High-Q, low-mode-volume microsphere-integrated Fabry–Perot cavity for optofluidic lasing applications显示文摘We develop a hybrid optofluidic microcavity by placing a microsphere with a diameter ranging from 1 to 4 μm in liquid-filled plano-plano Fabry–Perot(FP) cavities, which can provide an extremely low effective mode volume down to 0.3–5.1 μm^3 while maintaining a high Q-factor up to 1 × 10~4–5 × 10~4 and a finesse of ~2000. Compared to the pure plano-plano FP cavities that are known to suffer from the lack of mode confinement, diffraction, and geometrical walk-off losses as well as being highly susceptible to mirror misalignment, our microsphere-integrated FP(MIFP) cavities show strong optical confinement in the lateral direction with a tight mode radius of only 0.4–0.9 μm and high tolerance to mirror misalignment as large as 2°. With the microsphere serving as a waveguide, the MIFP is advantageous over a fiber-sandwiched FP cavity due to the open-cavity design for analytes/liquids to interact strongly with the resonant mode, the ease of assembly, and the possibility to replace the microsphere. In this work, the main characteristics of the MIFP, including Q-factor, finesse, effective mode radius and volume, and their dependence on the surrounding medium's refractive index, mirror spacing, microsphere position inside the FP cavity, and mirror misalignment, are systematically investigated using a finite-element method.Then, by inserting dye-doped polystyrene microspheres of various sizes into the FP cavity filled with water, we experimentally realize single-mode MIFP optofluidic lasers that have a lasing threshold as low as a few microjoules per square millimeter and a lasing spot radius of only ~0.5 μm. | 10 | N-acetylcysteine protect lymphocytes and cytokines against heavy ion irradiation via counteracting the glutamate显示文摘We evaluated the protective effect of N-acetylcysteine (NAC) on immunity system irradiated by 12C6+ ion beam. Kun-Ming mice were whole-body irradiated by 12C6+ ion at doses of 0, 0.5, 1, 1.5, 2, 2.5 and 3 Gy. The results showed that in saline group, the lymphocytes DNA double-strand breaks (DSBs), maleic dialdehyde, thymocytes number in G 0 /G 1 and apoptosis percentage increased with dose increment, and the levels of interferon-γ, glutathione, superoxide radical (SOD) and natural killer cells activity decreased with dose increment. However, there were no significant changes in NAC-treated group. | 17 | The Recent Crustal Deformation on the Sichuan-Yunnan Rhombic Block Boundaries显示文摘Based on data from an across-fault survey along the Sichuan-Yunnan rhombic block boundaries, the recent deformation characteristics on each fault have been analyzed. It was found that the rate of crustal deformation is slowing down along the northern segment and increasing along the southern segment. Each fault has different features of deformation. The horizontal deformation is mainly characterized by left-lateral strike-slip. The rate of vertical deformation is less than that of the horizontal deformation. The faults have the feature of upper wall uplifting alternated with descending. | 18 | Predicting the utilization factor of blasthole in rock roadways by random forest显示文摘The utilization factor of blasthole is a crucial indicator of the effectiveness of blasting in rock roadways.A significant value means that the explosive energy is fully utilized,the single-cycle advance is high,and the excavation rate is fast.A good blasting programme is a prerequisite for improving the utilization rate and predicting the utilization rate before blasting operations can verify the feasibility of the blasting programme.Firstly,a database of rock roadway blasting covering different geological and production conditions is estab-lished.Secondly,error analysis and the Gini coefficient method are used to weight the characteristic variables,quantify the importance of the variables and identify eight key indicators affecting the blasting hole utilization rate.Then,a random forest algorithm-based model for predicting utilization factor of blasthole is proposed,and the results of the model on the test set are:root mean square error(RMSE)is 0.0137,mean absolute error(MAE)is 0.0087,and coefficient of determination(R^(2))is 0.905.The performance of this method is com-pared with that of the neural network and support vector machine models on the test sets to verify the superiority of the random forest algorithm.Finally,to verify the generalization ability and practicality of the random forest prediction model,the model is applied to the rock roadway blasting construction of Gu Bei coal mine in Anhui Province.The results show that R2 is 0.913,so the model is reliable and accurate,which can meet the actual engineering requirements and lay the foundation for the promotion and application of this technology. | Xinmin Ma Zhenyu Chen Pan Chen Haozhang Zheng Xingyu Gao Junjie Xiang Liying Chen Yipei Huang | 2023 | Underground Space2023,,4: | 0 |
| 19 | Study on the Relationship Between Recent Crustal Deformation and the Temporal-Spatial Distribution of Strong Earthquakes (M_s≥6.0) in the Sichuan-Yunnan Region显示文摘Analysis of deformation data measured across the faults, regional vertical deformation data and GPS measurements in the Sichuan-Yunnan region made since the 1980s permitted us to conclude that the crustal deformation in the region during this period of time was relatively weak and caused the occurrence of earthquakes (M S≥6.0), which were not distributed along the major boundary active faults in the region after the 1981 Dawu M S 6.9 earthquake and that the seismic activity is characterized by quasi-clockwise migration. Thus, it follows that earthquake prediction research should be focused on the central part of the Sichuan-Yunnan region in the coming years.
