1Management of cytokine release syndrome related to CAR-T cell therapy显示文摘Chimeric antigen receptor T (CAR-T) cell therapy is a novel cellular immunotherapy that is widely used to treat hematological malignancies, including acute leukemia, lymphoma, and multiple myeloma. Despite its remarkable clinical effects, this therapy has side effects that cannot be underestimated. Cytokine release syndrome (CRS) is one of the most clinically important and potentially life-threatening toxicities. This syndrome is a systemic immune storm that involves the mass cytokines releasing by activated immune cells. This phenomenon causes multisystem damages and sometimes even death. In this study, we reported the management of a patient with recurrent and refractory multiple myeloma and three patients with acute lymphocytic leukemia who suffered CRS during CAR-T treatment. The early application of tocilizumab, an anti-IL-6 receptor antibody, according to toxicity grading and clinical manifestation is recommended especially for patients who suffer continuous hyperpyrexia, hypotensive shock, acute respiratory failure, and whose CRS toxicities deteriorated rapidly. Moreover, low doses of dexamethasone (5-10 mg/day) were used for refractory CRS not responding to tocilizumab. 7Waste-to-wealth:Functional biomass carbon dots based on bee pollen waste and application显示文摘Waste utilization is not only the protection of the environment and the practice of green chemistry,but also one of the ways to develop new materials.Herein,we report two biomass carbon dots which prepared from bee pollen waste by one-step hydrothermal method.The new two carbon dots were used in sensing,cell imaging and plant growth regulation.The differences in the structure and properties of the two carbon dots were evaluated by TEM,XPS,TG and various spectroscopic methods.Both two carbon dots contain abundant functional groups,polyatomic doping,excellent water solubility and stable photoluminescence.Due to these good properties,we have demonstrated its versatile applications in Fe3+sensing,cell imaging and plant growth regulation.It shows sensitive and specific Fe^(3+) responsiveness and good biocompatibility.This research provides a green and simple method for the recycling and reuse of bee pollen waste,and also provides a reference for the application of biomass carbon dots.Feishi Shan Lijuan Fu Xiangyu Chen Xiangyan Xie Chengshuang Liao Yinxia Zhu Haoyu Xia Jing Zhang Li Yan Zhouyu Wang Xiaoqi Yu 2022Chinese Chemical Letters2022,33,6:0
8Molecular Dynamics and Preparation of HO-PAMAM-2.5G Protected Lanthanum Cluster显示文摘HO-PAMAM-2.5 G-protected lanthanum cluster was studied by Virtual Molecule(VM) software. Molecular dynamics (MD) simulation of the nanocomposite system was performed from the view-points of molecular structure and energy′s fluctuation in NVT system. The motivation of this work was to understand the stability and mechanism of the nanocomposite in different proportion. The molecular dynamics simulation results reveal that when the quantity of La in per HO-PAMAM-2.5G amounted to 0~60, the radius of gyration of HO-PAMAM-2.5/La nanocomposite is obviously decreased. The potential energy (PE) that determines the stability of nanocomposite is different according to the quantity of La. The energy of the nanocomposite fluctuated clearly within the range of 0~50 La atoms and this fluctuation gradually tends to flatness in 50~80. While the quantity of La reaches 60, the energy of the nanocomposite drops to the lowest point, the system is steady relatively. Whereafter, according to the simulation results, HO-PAMAM-2.5 G-protected lanthanum clusters were prepared and characterized by transmission electron microscopy (TEM).
