| 5 | Dispase rapidly and effectively purifies Schwann cells from newborn mice and adult rats显示文摘In the present study, Schwann cells were isolated from the sciatic nerve of neonatal mice and purified using dispase and collagenase. Results showed that after the first round of purification with dispase, most of the Schwann cells appeared round in shape and floated in culture solution after 15 minutes. In addition, cell yield and cell purity were higher when compared to the collagenase group. After the second round of purification, the final cell yield for the dispase group was higher than that for the collagenase group, but no significant difference was found in cell purity. Moreover, similar results in cell quantity and purity were observed in adult Sprague-Dawley rats. | 10 | Transumbilical laparoendoscopic single-site surgery of simple nephrectomy of nonfunctioning kidney: a two-year experience显示文摘Background: Laparoendoscopic single-site surgery (LESS) may serve as a potential alternative to conventional laparoscopy and is developing quickly, but still in its infancy. The study is to present our two-year experience in transumbilical LESS simple nephrectomy (LESS-SN) for non-functioning kidney, in an effort to evaluate its feasibility, clinical outcomes and potential advantages. Methods: From December 2008 to December 2010, a total of 11 patients with body mass index (BMI) ≤30 underwent transumbilical TriPort TM LESS-SN by a single experienced urologist at our institution. The indications for nephrectomy included nonfunctioning kidney associated with ureteropelvic junctionstricture (n=1), ureteral calculi (n=6), tuberculosis (n=3), and ureteral stricture (n=1). Patient demographics, perioperative and follow-up data were prospectively collected and analyzed. Results: Ten procedures were successfully completed with one patient converted to open surgery due to uncontrollable bleeding. The mean operative time was 189.2 (ranging 100~320 min) with an estimated blood loss of 204.5 (ranging 50-1 000 ml). There were two complications of bleeding (1- intra-, 1- post-). The mean hospitalization after surgery was 7.9 d (ranging 4-17 d). With a regular follow-up of 1, 6, 12, and 24 months after surgery, all patients remained symptom-free with an intra-umbilical scar. Conclusion: Transumbilical LESS simple nephrectomy for nonfunctioning kidney can be accomplished with favorable surgical outcomes and a superiority of cosmesis. The structure consists of discrete neutral macrobicyclic units; the two cycles share the two tertiary amine nitrogen atoms, which exhibit an endo-cndo conformation. Three identical branches formed by 1, 2-diaminoethane link the two tertiary amine groups. The protonation reactions of cryptand (COBH) and its complex formation with copper (II) were investigated by potentiometry in water and in a DMSO/water (80:20 in mass ratio) mixture as solvents. The cryptand acts as a his-base through its two N-bridgehead and exhibits a strong cooperativity that favors the first protonation and makes the second one difficult (△pK= 5.0). An inward rotation of the amide groups to form hydrogen bonds accounts for this cooperativity. | 13 | Basic Characteristics of Active Structures in Western Sichuan and Its Vicinity and Strong Earthquake Recurrence Model显示文摘Western Sichuan and its vicinity are located in the juncture of three big active blocks, namely, the Chuandian (Sichuan and Yunnan), the Bayan Har and the South China blocks, on the eastern margin of the Qinghai-Xizang(Tibet) Plateau. Many groups of active faults that are capable of generating earthquakes are developed there. Because there exist lateral secondary active faults, the Chuandian block can be further divided into the central Yunnan and northwestern Sichuan sub_blocks; while the Longmenshan sub_block can be divided on the east end of the Bayan Har block. Joint exploration of deep crustal structure shows that there exist low-velocity and high-conductivity layers in the crust of the Chuandian and Bayan Har blocks, which are one of the important factors that make the upper crust prone to earthquake. The results of geological study and modern GPS observation show that blocks of different orders all have SE-or SSE-trending sliding, clockwise rotation and upwelling movement; but there are some differences in amplitude. | 14 | Effects of long non-coding RNA GAS5 on proliferation and apoptosis of hepatocellular carcinoma cells through miR-26a-5p action显示文摘Objective Long non-coding RNAs(lncRNAs)regulate tumor development and progression by promoting tumor proliferation,invasion,and metastasis.The aim of the study was to investigate the effects of lncRNA growth arrest-special 5(GAS5)on proliferation and apoptosis of hepatocellular carcinoma(HCC)cells through miR-26a-5p action.Methods Expression levels of GAS5 were detected in cancerous and paracancerous tissue of 80 HCC patients by RT-qPCR.The starBase tool predicted that GAS5 had binding sites for the miRNA miR-26a-5p,which was also highly expressed in HCC tissue.The relationship between GAS5 and miR-26a-5p was confirmed using a luciferase reporter assay.The role of these lncRNAs was further explored by transfecting plasmids into SMMC-7721 cells and classifying the cells as follows:NC group,GAS5 group,anti-miR-26a-5p group,and GAS5+miR-26a-5p group.Cell proliferation,cell cycle,and apoptosis were detected in each group.The relationship between miR-26a-5p and phosphatase and tensin homolog deleted on chromosome 10(PTEN)was analyzed by TargetScan database prediction and luciferase reporter assay.Western blotting was used to quantify PTEN,phosphatidylinositol 3-kinase(PI3K),phosphorylated protein kinase B(p-Akt),cyclin D1,and human P27 protein(P27).Results GAS5 was downregulated,while miR-26a-5p was upregulated in HCC tissue compared to in paracancerous tissue.High GAS5 levels and low miR-26a-5p levels inhibited cell proliferation,increased the number of G0/G1 phase cells,promoted cell apoptosis,promoted PTEN and P27 expression,and inhibited PI3K,P-Akt,and cyclin D1 expression at the protein level.Upregulation of miR-26a-5p attenuated the effects of GAS5 upregulation on the proliferation,cell cycle,and apoptosis of HCC cells and on the expression of PTNE/PI3K/Akt signaling pathway-related proteins.Conclusion Low GAS5 levels regulate the proliferation and apoptosis of HCC cells via the PTNE/PI3K/Akt signaling pathway and are linked to upregulation of miR-26a-5p. | Zunli Yi Xiaoguang Guo Xianxue Jiang Fengmei Luo | 2022 | Oncology and Translational Medicine2022,8,3: | 0 |

| 16 | Synchronous bilateral laparoendoscopic single-site urological surgeries via the umbilicus:a report of 2 cases显示文摘Objective:To describe two cases of bilateral transumbilical laparoendoscopic single-site(LESS) surgeries in a single operative session and assess the safety,feasibility and efficacy.Methods:One patient underwent right ureterolithotomy and left varicocelectomy,and the patient was performed with right simple nephrectomy for nonfunctioning kidney due to ureteral calculus and left ureterolithotomy using a novel multichannel TriPort? via a single 2-3 cm transumbilical incision. Results:The right-side ureterolithotomy and left-side varicocelectomy were finished in 229 minutes,with a total estimated blood loss of 50 ml.The right-side simple nephrectomy and left-side ureterolithotomy in the other patient were finished in 340 minutes,with a total estimated blood loss of 100 ml.There were no major complications.At the latest follow-up,both patients remained symptom-free and there were no evidences of recurrence.Both of them were delighted for the single scar. Molecular dynamics (MD) simulation of the nanocomposite system was performed from the view-points of molecular structure and energy′s fluctuation in NVT system. The motivation of this work was to understand the stability and mechanism of the nanocomposite in different proportion. The molecular dynamics simulation results reveal that when the quantity of La in per HO-PAMAM-2.5G amounted to 0~60, the radius of gyration of HO-PAMAM-2.5/La nanocomposite is obviously decreased. The potential energy (PE) that determines the stability of nanocomposite is different according to the quantity of La. The energy of the nanocomposite fluctuated clearly within the range of 0~50 La atoms and this fluctuation gradually tends to flatness in 50~80. While the quantity of La reaches 60, the energy of the nanocomposite drops to the lowest point, the system is steady relatively. Whereafter, according to the simulation results, HO-PAMAM-2.5 G-protected lanthanum clusters were prepared and characterized by transmission electron microscopy (TEM). The structural and morphological characteristics of the target product and the precursor were analyzed by XRD, SEM, and TG-DTA. By detecting battery performance, the charged and discharged platform was over 3.6 V; the anode exhibited a discharge capacity decay of 2% from its initial ca-pacity (165 mA?h/g) after 20 cycles.
