| 1 | ABNORMAL FLOWER AND GRAIN 1 encodes OsMADS6 and determines palea identity and affects rice grain yield and quality显示文摘The palea and lemma are floral organ structures unique to grasses;these structures form the hull and directly affect grain size and quality. However, the molecular mechanisms controlling the development of the hull are not well understood. In this study, we characterized the rice(Oryza sativa) abnormal flower and grain1(afg1) mutant, a new allele of OsMADS6. Similar to previously characterized osmads6 alleles, in the afg1 floret, the palea lost its marginal region and acquired the lemma identity. However, in contrast to other osmads6 alleles, the afg1 mutant showed altered grain size and grain quality, with decreased total starch and amylose contents, and increased protein and soluble sugar contents. The analysis of transcriptional activity suggested that AFG1 is a transcriptional activator and may affect grain size by regulating the expression levels of several genes related to cell expansion and proliferation in the afg1 mutant. | 3 | Numerical Investigation of Pressure Distribution in a Low Specific Speed Centrifugal Pump显示文摘To study the pressure distribution of the volute casing, front casing and back casing in a prototype centrifugal pump, the pressure experiments and numerical simulations are carried out at six working conditions in this paper. The experimental results shows that the asymmetry of static pressure distribution on volute casing and front cavity is caused by the tongue of the volute and it may result in high radial and axial resultant force which can cause vibration and noise in the centrifugal pump. With the increasing of flow rate, the asymmetry of static pressure distribution and the magnitude of static pressure values reduce. The numerical results indicate that the pressure fluctuation near the tongue is strongest and it becomes slighter at point away from the tongue. With the increasing of flow rate, the local high-pressure region in impeller passage reduces and the flow becomes smoother accordingly, whereas the fluid speed becomes much higher which may cause further flow losses. The results predicted by numerical simulation are in coincident with the experimental ones. | 4 | Effects of blocking Le^Y oligosaccharide on cell surface to MMPs secreted by blastocysts and epithelial cells in mouse in vitro显示文摘In order to understand the role of Le+Y oligosaccharide antigen (Le+Y) during implantation, the relationship of Le+Y on the cell surface with matrix metalloproteinase (MMPs) secreted by blastocysts and monolayer epithelial cells during implantation in the mouse %in vitro% was studied by monoclonal antibody (mAb) AH-6, directed to Le+Y[Fuc α1-2 Gal β1-4 (Fuc α1-3) GlcNAc-], and gelatin zymography. The results showed that MMPs secretion was reduced after Le+Y on the cell surface of either epithelial cells or trophoblasts was blocked. It indicated that MMPs expression which played an important function during the process of implantation were regulated by Le+Y. | 6 | A digital energy spectroscopy based on FIR filter显示文摘We designed a universal digital energy spectroscopy based on online digital signal processing. A prototype system was built and tested. Signals from radiation detectors were processed via a digital filter whose coefficients could be modified without changing the hardware. The paper introduces the hardware design of the digital energy spectroscopy system as well as the full set of software consisting of the selection of the coefficients of the finite impulse response (FIR) filter and the coding in the field-programmable gate array (FPGA). The system was tested with the high purity germanium (HPGe) detector. | 7 | Novel cyclen-based linear polymer as a high-affinity binding material for DNA condensation显示文摘A novel cyclen-based linear polyamine (POGEC) was designed and synthesized from the reaction between 1,3-propanediol diglycidyl ether and 1,7-bis(diethoxyphosphory)-1,4,7,10-tetraazacyclod-odecane. High-affinity binding between POGEC and DNA was demonstrated by agarose gel electrophoresis and scanning electron microscopy (SEM). Moreover,the formed POGEC/DNA complex (termed polyplex) could be disassociated to release the free DNA through addition of the physiological concentration of NaCl solution. Fluorescence spectrum was used to measure the high-affinity binding and DNA condensation capability of POGEC. | 8 | Novel strategy of constructing fluorescent probe for MAO-B via cascade reaction and its application in imaging MAO-B in human astrocyte显示文摘To detect monoamine oxidase B(MAO-B), the level of which is one of the most important indicators of neurodegenerative diseases such as Parkinson's disease, a new type of cascade reaction based on the formation of coumarin was applied in this research. After the reaction with MAO-B, the protecting group of hydroxyl group in probes(DEAN-MA and DEAB-MA) was removed, and the fluorescence intensity significantly increased(l_(em) 456 nm) as the structure of coumarin was formed subsequently. The probes showed excellent sensitivity and selectivity to MAO-B. The detection limit of DEAN-MA and DEAB-MA were 0.6ng/mL and 0.2 ng/mL, respectively. | 10 | α-Lipoic Acid Exerts Its Antiviral Effect against Viral Hemorrhagic Septicemia Virus(VHSV) by Promoting Upregulation of Antiviral Genes and Suppressing VHSV-Induced Oxidative Stress显示文摘Viral hemorrhagic septicemia virus(VHSV), belonging to the genus Novirhabdovirus, Rhabdoviridae family, is a causative agent of high mortality in fish and has caused significant losses to the aquaculture industry. Currently, no effective vaccines, Food and Drug Administration-approved inhibitors, or other therapeutic intervention options are available against VHSV. α-Lipoic Acid(LA), a potent antioxidant, has been proposed to have antiviral effects against different viruses. In this study, LA(CC_(50)= 472.6 lmol/L) was repurposed to exhibit antiviral activity against VHSV. In fathead minnow cells,LA significantly increased the cell viability post-VHSV infection(EC_(50)= 42.7 lmol/L), and exerted a dose-dependent inhibitory effect on VHSV induced-plaque, cytopathic effects, and VHSV glycoprotein expression. The time-of-addition assay suggested that the antiviral activity of LA occurred at viral replication stage. Survival assay revealed that LA could significantly upregulated the survival rate of VHSV-infected largemouth bass in both co-injection(38.095% vs. 1.887%,P < 0.01) and post-injection manner(38.813% vs. 8.696%, P < 0.01) compared with the control group. Additional comparative transcriptome and q RT-PCR analysis revealed LA treatment upregulated the expression of several antiviral genes, such as IRF7, Viperin, and ISG15. Moreover, LA treatment reduced VHSV-induced reactive oxygen species production in addition to Nrf2 and SOD1 expression. Taken together, these data demonstrated that LA suppressed VHSV replication by inducing antiviral genes expression and reducing VHSV-induced oxidative stress. | 17 | Experimental and numerical study on the dynamic shear banding mechanism of HfNbZrTi high entropy alloy显示文摘High-entropy alloys(HEAs) have attracted considerable attention in recent years because of their unique mechanical properties.In this work, the mechanism of dynamic shear banding(also called adiabatic shear bands, ASBs) in a BCC HEA HfNbZrTi was investigated combining dynamic experiments and numerical simulations. The temperature evolution during dynamic shear banding, which has been believed to play a dominant role during ASB formation in the literature, was measured using high-speed infrared thermal detectors synchronized with a split Hopkinson pressure bar system. The dynamic mechanical behavior of the BCC HEA was described using the Johnson-Cook model accompanied by damage accumulation. The process of ASB formation,considering potential contributions from thermal softening and damage softening, was numerically investigated by controlling the activation of each softening mechanism separately.
