| 1 | A Study on Tribological Properties of Polypropylene Nanocomposites Reinforced with Pretreated HNTs显示文摘The polypropylene(PP) nanocomposites filled with pretreated halloysite nanotubes(HNTs) were prepared by the melt-blending method. Before filling, the as-received HNTs powder was at first purified and then modified. The characterization tests showed that the purified HNTs had less impurity and more uniform pore size distribution and the surface hydrophobicity of the modified HNTs was obviously improved. The mechanical and tribological properties of the PP/HNTs nanocomposites were extensively investigated. | 2 | The role of a drug-loaded poly(lactic co-glycolic acid)(PLGA) copolymer stent in the treatment of ovarian cancer显示文摘Objectives:Cisplatin(CDDP)is a widely used and effective basic chemotherapeutic drug for the treatment of a variety of tumors,including ovarian cancer.However,adverse side effects and acquired drug resistance are observed in the clinical application of CDDP.Identifying a mode of administration that can alleviate side effects and reduce drug resistance has become a promising strategy to solve this problem.Methods:In this study,3 D printing technology was used to prepare a CDDP-poly(lactic-co-glycolic acid)(CDDP-PLGA)polymer compound stent,and its physicochemical properties and cytotoxicity were evaluated both in vitro and in vivo.Results:The CDDP-PLGA stent had a significant effect on cell proliferation and apoptosis and clearly decreased the size of subcutaneous tumors in nude mice,whereas the systemic side effects were mild compared with those of intraperitoneal CDDP injection.Compared with the control group,CDDP-PLGA significantly increased the mRNA and protein levels of p-glycoprotein(P<0.01;P<0.01)and decreased vascular endothelial growth factor mRNA(P<0.05)and protein levels(P<0.01),however,CDDP-PLGA significantly decreased the mR NA and protein levels of p-glycoprotein(P<0.01;P<0.01)and vascular endothelial growth factor(P<0.01;P<0.01),which are associated with chemoresistance,in subcutaneous tumor tissue.Immunohistochemistry assay results revealed that,in the CDDP-PLGA group,the staining of the proliferation-related genes Ki67 and PCNA were lightly,and the apoptosis-related gene caspase-3 stained deeply.Conclusions:PLGA biomaterials loaded with CDDP,as compared with the same amount of free CDDP,showed good efficacy in terms of cytotoxicity,as evidenced by changes in apoptosis.Continuous local CDDP release can decrease the systemic side effects of this drug and the occurrence of drug resistance and angiogenesis,and improve the therapeutic effect.This new approach may be an effective strategy for the local treatment of epithelial ovarian cancer. | Yanqing Wang Xiaoyin Qiao Xiao Yang Mengqin Yuan Shu Xian Li Zhang Dongyong Yang Shiyi Liu Fangfang Dai Zhikai Tan Yanxiang Cheng | 2020 | Cancer Biology & Medicine2020,17,1: | 2 |
| 3 | RNA干扰技术裸鼠体内沉默缺氧诱导因子1α的表达对宫颈癌的抑瘤效应(英文)显示文摘Objective: The aim of our study was to observe the anti-tumor effect of silencing the expression of HIF-1α on cervical cancer in nude mice and to explore its mechanism of action. Methods: Human cervical cancer cell line Siha cells were divided into 3 groups: mock control group, control group transfected with scrambled sequence plasmid, and experimental group transfected with pU-HIF-1a-shRNA eukaryotic expression plasmid. Cultured cells of the three groups were inoculated in nude mice to establish cervical cancer-bearing nude mice. HIF-1α RNAi assay was performed to evaluate the tumor-suppressive effect of HIF-1α silencing on cervical cancer-bearing nude mice. Immunohistochemistry and Western blot were used to observe the distribution and protein expression of HIF-1α and GLUT1, while RT-PCR was adopted to detect the gene expression of HIF-1α, GLUTl and HK II. The product of glycolysis (lactic acid) and apoptosis in tumor cells were detected by colorimetry and semi-quantitative TUNEL staining, respectively. Results: The tumor growth in experimental group was significantly slower than that in the two control groups (P < 0.05). In the 50th day after transplantation, the tumor weight in the experimental group was (1.90 ± 0.28) g, significantly lower than (2.95 ± 0.77) g in the control group and (2.54 ± 0.56) g in the mock group (P < 0.01). In the experimental group, the gene and protein levels of HIF-1α were 0.45 ± 0.04 and 1.25 ± 0.92, and the levels of GLUT1 were 0.32 ± 0.02 and 1.25 ± 0.48, respectively. Both indicators in HIF-1α and GLUT1 were lower than that in the two control groups (P < 0.05). The expression levels of HK II gene and lactic acid in the experimental group were lower than that in the two control groups (P < 0.05), but the apoptotic cells were much more numerous in the experimental group than that in matched control groups (P < 0.01). | 9 | Clinical significance of survivin in the diagnosis and prognosis of endometrial carcinoma显示文摘Objective:To investigate the clinical significance of survivin in endometrial carcinoma and to investigate the relationship between the expression of survivin and Ki-67.Methods:Immunohistochemical S-P(streptavidin-biotin-peroxidase complex)method was performed to detect the expression of survivin and Ki-67 antigen in 15 cases of normal endometrium,21 cases of endometrial simple and complex hyperplasia,22 cases of endometrial atypical hyperplasia,and 61 cases of endometrial carcinoma.Results:Survivin was hardly detected in some normal endometrium in the proliferative phase and in the secretory phase.However,the level of survivin expression in atypical hyperplasia endometrium(72.73%)was higher than that in normal endometrium(7.14%)(P < 0.05),including simple and complex hyperplasia(42.38%)(P < 0.01),and was lower than that in endometrial carcinoma(90.17%)(P < 0.05).Moreover,significant correlation was present between the expression of survivin and the characteristics of endometrial carcinoma,including clinical stage,histological grade and the presence of invasion to myometrium(P < 0.05).In addition,Ki-67 antigen expression was positively correlated with survivin expression in all specimen.Ki-67 labeled indexes(LIs)in hyperplasia endometrium were significantly lower than those in atypical hyperplasia endometrium and endometrial carcinoma(P < 0.01),while there was no significant difference in Ki-67 LIs between atypical hyperplasia endometrium and endometrial carcinoma(P > 0.05).There was no significant relationship between Ki-67 LIs and the characteristics of endometrial carcinoma,including histological grade,clinical stage or the invasion to myometrium(P > 0.05).Conclusion:Survivin may participate in the onset and progression of endometrial carcinoma through inhibiting apoptosis and promoting proliferation.Survivin expression is correlated with the malignant degree and prognosis of tumor.Ki-67 is also associated with carcinogenesis and progression of endometrial carcinoma.The results suggest that survivin could be a diagnostic and prognostic marker for endometrial carcinoma and might provide pathways to treat the patients with recurrent or refractory or rudimental endometrial carcinoma. | Yanxiang Cheng Gantao Chen Yanjun Cheng Demin Pu | 2006 | Journal of Nanjing Medical University2006,20,4: | 0 |
| 10 | Carbazole ring: A delicate rack for constructing thermally activated delayed fluorescent compounds with through-space charge transfer显示文摘Three carbazole derivatives, Ac PTC, Px PTC and Pt PTC, consisting of two 9,9-dimethyl-9,10-dihydroacridine,phenoxazine or phenothiazine donor groups and one diphenyltriazine acceptor group fixed at 1,8,9-positions of a single carbazole ring via phenylene, are designed and synthesized. X-ray diffraction analysis of Ac PTC reveals that there exist multiple π-π interactions between the donor and acceptor groups to form a sandwich-like structural unit with edge-to-face interaction model. The compounds thus show obvious thermally activated delayed fluorescence with through-space charge transfer character and possess considerable photoluminescence quantum yields of up to 73% in doped films with sky-blue to yellow emissions.
