To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://gffzzdbc7b6aaae734bddhkpnqxbwno5uv6xub.ffgz.tsg.suse.edu.cn/gsa or http://gffzz4eaf941a184140e3hkpnqxbwno5uv6xub.ffgz.tsg.suse.edu.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 8 | Gray bootstrap method for estimating frequency-varying random vibration signals with small samples显示文摘During environment testing, the estimation of random vibration signals(RVS) is an important technique for the airborne platform safety and reliability. However, the available methods including extreme value envelope method(EVEM), statistical tolerances method(STM) and improved statistical tolerance method(ISTM) require large samples and typical probability distribution. Moreover, the frequency-varying characteristic of RVS is usually not taken into account. Gray bootstrap method(GBM) is proposed to solve the problem of estimating frequency-varying RVS with small samples. Firstly, the estimated indexes are obtained including the estimated interval, the estimated uncertainty, the estimated value, the estimated error and estimated reliability. In addition, GBM is applied to estimating the single flight testing of certain aircraft. At last, in order to evaluate the estimated performance, GBM is compared with bootstrap method(BM) and gray method(GM) in testing analysis. | 9 | Effect of allogeneic umbilical cord mesenchymal stem cell transplantation in a rat model of hepatic cirrhosis显示文摘OBJECTIVE: To determine the effects of human umbilical cord mesenchymal stem cell(UCMSC) transplantation, alone or in combination with tanshinone IIA(Tan IIA) on hepatic cirrhosis in rats.METHODS: A rat model of cirrhosis was established. Rats were divided into control, UCMSC, and UCSMC plus Tan IIA groups. Rats in the UCMSC group were injected via the tail vein with 0.2 m L Di I-labeled UCMSC suspension. Intraperitoneal Tan IIA injections(20 mg/kg) were started on the day ofUCMSC transplantation in the UCMSC plus Tan IIA group, and continued for 7 consecutive days thereafter. Rats were sacrificed 1 day, 3 days, 1 month,and 3 months after transplantation and the numbers of Di I-labeled UCMSCs colonizing the liver were determined. Albumin(ALB) and alanine aminotransferase(ALT) levels were measured in venous blood, and m RNA and protein expression levels of human ALB and cytokeratin(CK)-18 in liver tissues were determined by reverse transcription-polymerase chain reaction and western blotting, respectively.RESULTS: Serum ALT levels were significantly lower and serum ALB levels significantly higher in rats in the UCMSC group compared with the control group(P < 0.05). Hepatic CK-18 and ALB m RNA and protein expression levels increased after transplantation, and were significantly higher in the UCMSC plus Tan IIA group compared with the UCMSC group(P < 0.05).CONCLUSION: Human UCMSCs transplanted into rats with liver cirrhosis can grow and differentiate into hepatocyte-like cells resulting in improved liver function in vivo. | 11 | Qingnaoyizhi decoction suppresses the formation of glial fibrillary acidic protein-positive cells in cultured neural stem cells by inhibiting the Janus kinase 2/signal transducer and activator of transcription 3 signaling pathway显示文摘OBJECTIVE: Inactivation of the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3) signaling axis plays a crucial role in determining the fate of neural stem cells(NSCs).Qingnaoyizhi decoction(QNYZD) has been used for the treatment of vascular dementia and has shown to improve synaptic remodeling. The aim of this study was to evaluate the effect of cerebrospinal fluid(CSF) containing QNYZD(CSF-QNYZD) on the differentiation of cultured NSCs and the involvement of the JAK2/STAT3 pathway.METHODS: The protein expression levels of glial fibrillary acidic protein(GFAP), tubulin, drosophila mothers against decapentaplegic protein(SMAD-1), STAT3, and phosphorylated-STAT3 were detected by western immunoblot analysis in the groups: control, CSF, JAK/STAT inhibitor(AG490),CSF-QNYZD, and CSF-XDZ(CSF-Xidezhen). The differentiation of NSCs was determined by immunofluorescence staining. The proliferation of NSCs was measured using the Cell Counting Kit-8 proliferation assay.RESULTS: Compared with the control group,CSF-QNYZD and AG490 significantly increased the number and expression of tubulin-positive cells, reduced the number and expression of GFAP-positive cells, and down-regulated the expression of p-STAT3. However, CSF-QNYZD also decreased the expression of SMAD-1 and STAT3.CONCLUSION: Enhanced neuronal differentiation may be associated with the down-regulation of glial differentiation instead of promoting proliferationin treated NSCs. Furthermore, QNYZD may play a direct role in suppressing the formation of GFAP-positive cells and enhancing neuronal differentiation by inhibiting JAK2/STAT3 activation. | 12 | Strengthening FCC-CoCrFeMnNi high entropy alloys by Mo addition显示文摘In order to strengthen the face-centered-cubic(FCC) type CoCrFeMnNi high entropy alloys(HEAs), different contents of Mo(0–16 at.%, similarly hereinafter) were alloyed. Phase evolution, microstructure,mechanical properties and related mechanism of these HEAs were systematically studied. The results show that sigma phase is appeared with addition of Mo, and the volume fraction of it increases gradually from 0 to 66% with increasing Mo content. It is found that Mo is enriched in sigma phase, which indicates that Mo element is beneficial to form sigma phase. Compressive testing shows that the yield strength of the alloys increases gradually from 216 to 765 MPa, while the fracture strain decreases from 50%(no fracture) to 19% with increasing of Mo. The alloy exhibits the best compressive performance when Mo content reaches 11%, the yield strength, fracture strength and fracture strain are 547 MPa, 2672 MPa and44% respectively. | 14 | Microstructures and mechanical properties of Nb-alloyed CoCrCuFeNi high-entropy alloys显示文摘Nb has a positive effect on improving the mechanical properties of metal materials, and it is expected to strengthen CoCrCuFeNi high-entropy alloys(HEAs) with outstanding ductility and relatively weak strength. In this paper, the alloying effects of Nb on the microstructural evolution and the mechanical properties of the(CoCrCuFeNi)_(100-x)Nb_xHEA were investigated systematically. The result shows that Nb promotes the phase transition from FCC(face-centered cubic) to Laves phase, and the volume fractions of Laves phase increase from 0% to 58.2% as the Nb content increases. Compressive testing shows that the addition of Nb has a positive effect on improving the strength of CoCrCuFeNi HEA. The compressive yield strength of(CoCrCuFeNi)_(100-x)Nb_xHEAs increases from 338 MPa to 1322 MPa and the fracture strain gradually reduces from 60.0%(no fracture) to 8.1% as the Nb content increases from 0 to 16 at.%. | 16 | A meta-analysis of Chinese herbal medicines for vascular dementia显示文摘OBJECTIVE: To investigate the efficacy and safety of Chinese herbal medicines in the treatment of patients with vascular dementia. DATA RETRIEVAL: We retrieved publications from Cochrane Library (2004 to July 2011), PubMed (1966 to July 2011), the Chinese Science and Technique Journals Database (1977 to July 2011), the China National Knowledge Infrastructure (1979 to July 2011), Google Scholar (July 2011), and the Chinese Biomedical Database (1977 to July 2011) using the'Chinese medicine OR Chinese herbal medicine' and 'vascular dementia OR mild cognition impair OR multi-infarct dementia OR small-vessel dementia OR strategic infarct dementia OR hypoperfusion dementia OR hemorrhagic dementia OR hereditary vascular dementia'. SELECTION CRITERIA: Randomized controlled trials comparing Chinese herbal medicines with placebo/western medicine in the treatment of patients with vascular dementia were included. Diagnostic standards included Diagnostic and Statistical Manual of Mental Disorders-IV, and National Institute of Neurological Disorders and Stroke and Association Internationale pour la Recherché et l'Enseignement en Neurosciences. Two participants independently conducted literature screening, quality evaluation and data extraction. The quality of each trial was assessed according to the Cochrane Reviewers' Handbook 5.0. MAIN OUTCOME MEASURES: Effective rate, Mini-Mental State Examination scores, Hasegawa Dementia Scale scores, and incidence of adverse reactions. RESULTS: We identified 1 143 articles discussing the effects of Chinese medicine on vascular dementia. Thirty-one of these were included in the analysis. These studies involved a total of 2 868 participants (1 605 patients took Chinese medicine decoctions (treatment group); 1 263 patients took western medicine or placebo). The results of our meta-analysis revealed that Chinese herbal remedies in the treatment group were more efficacious than the control intervention (relative risk (RR)=1.27; 95% confidence interval (CI): 1.18-1.38, P<0.01). Mini-Mental State Examination scores were higher in patients taking Chinese herbal medicines than in those in the control group (weighted mean difference (WMD)=2.83; 95%CI: 2.55-3.12, P<0.01). Patients in the treatment group showed better disease amelioration than those in the control group (Hasegawa Dementia Scale scores; WMD=2.41, 95%CI: 1.48-3.34, P<0.01). There were also considerably fewer adverse reactions among those in the treatment group compared with those in the control group (RR=0.20, 95%CI: 0.08-0.47, P<0.01). CONCLUSION: Chinese herbal medicine appears to be safer and more effective than control measures in the treatment of vascular dementia. However, the included trials were generally low in quality. | 17 | Fuzzy norm method for evaluating random vibration of airborne platform from limited PSD data显示文摘For random vibration of airborne platform, the accurate evaluation is a key indicator to ensure normal operation of airborne equipment in flight. However, only limited power spectral density(PSD) data can be obtained at the stage of flight test. Thus, those conventional evaluation methods cannot be employed when the distribution characteristics and priori information are unknown. In this paper, the fuzzy norm method(FNM) is proposed which combines the advantages of fuzzy theory and norm theory. The proposed method can deeply dig system information from limited data, which probability distribution is not taken into account. Firstly, the FNM is employed to evaluate variable interval and expanded uncertainty from limited PSD data, and the performance of FNM is demonstrated by confidence level, reliability and computing accuracy of expanded uncertainty. In addition, the optimal fuzzy parameters are discussed to meet the requirements of aviation standards and metrological practice. Finally, computer simulation is used to prove the adaptability of FNM. Compared with statistical methods, FNM has superiority for evaluating expanded uncertainty from limited data. | 18 | Myt1L Promotes Differentiation of Oligodendrocyte Precursor Cells and is Necessary for Remyelination After Lysolecithin-Induced Demyelination显示文摘The differentiation and maturation of oligodendrocyte precursor cells(OPCs) is essential for myelination and remyelination in the CNS. The failure of OPCs to achieve terminal differentiation in demyelinating lesions often results in unsuccessful remyelination in a variety of human demyelinating diseases. However, the molecular mechanisms controlling OPC differentiation under pathological conditions remain largely unknown. Myt1 L(myelin transcription factor 1-like), mainly expressed in neurons,has been associated with intellectual disability, schizophrenia, and depression. In the present study, we found that Myt1 L was expressed in oligodendrocyte lineage cells during myelination and remyelination. The expression level of Myt1 L in neuron/glia antigen 2-positive(NG^(2+))OPCs was significantly higher than that in mature CC1^+ oligodendrocytes. In primary cultured OPCs,overexpression of Myt1 L promoted, while knockdown inhibited OPC differentiation. Moreover, Myt1 L was potently involved in promoting remyelination after lysolecithin-induced demyelination in vivo. Ch IP assays showed that Myt1 L bound to the promoter of Olig1 and transcriptionally regulated Olig1 expression.
