| 1 | Accurate Determination of Zr Isotopic Ratio in Zircons by Femtosecond Laser Ablation MC-ICP-MS with “Wet” Plasma Technique显示文摘This work evaluates the use of femtosecond laser ablation multiple collector inductively coupled plasma mass spectrometry(fs-LA-MC-ICP-MS) for Zr isotopic analysis in zircons. The mass fractionation caused by instrumental mass discrimination was corrected by a combination of internal correction using Sr as an internal standard(coming from a NIST SRM 987 standard solution) and external correction using a matrix-matched standard. Several important instrument parameters were investigated, such as the effect of the addition of N;and 'wet' plasma condition, the mass fractionation behaviors between Zr isotopes and Sr isotopes, the position effect in laser ablation cell and the effect of laser ablation parameters(laser spot size and energy density). The Zr isotope compositions of seven zircons(GJ-1, 91500, Ple?ovice, Rak-17, Paki, Aus and Mala) were determined by the developed fs-LAMC-ICP-MS and thermal ionization mass spectrometry(TIMS). Our fs-LA-MC-ICP-MS results for Zr isotope compositions agreed with TIMS analyses within analytical uncertainties, indicating the presented method is a suitable tool to resolve isotopic zoning in natural zircons. | 2 | A new analytical mode and application of the laser ablation inductively coupled plasma mass spectrometer in the earth sciences显示文摘We proposed a new laser analytical mode,which used the high-frequency laser ablation to deliberately obtain the peak-shape signal profile,combined with the linear regression calibration(LRC)method to calculate elemental or isotopic ratios.In order to assess the performance of the new laser analytical mode for the application in the field of earth science,we systematically investigated the elemental quantitative analysis with high spatial resolution(10μm),rapid U-Pb dating in zircons and accurate Sr-Hf isotope analysis in geological samples.The new high-frequency laser analysis technology(20 Hz,100 pulses)reduced the limit of detection(LOD)of 25 elements to 0.005-0.16μg g^(-1) with crater diameters of 10μm,which are significantly lower(decrease to 2-20%)than previous researches.The quantitative analysis of five silicate glass reference materials showed that the precision and accuracy of the 25 trace elements(with the concentration ranging from 0.17 to 683μg g^(-1))were better than 16% and 15%,respectively.The new methods raised the analytical throughput of zircon U-Pb dating,Sr isotope ratios and Hf isotope ratios to 250 analyses per hour,144 analyses per hour and 120 analyses per hour in theory,while the analytical accuracy and precision were not affected.The detailed investigations showed that the proposed new laser analytical mode has good application effects in the field of earth sciences. | Yantong FENG Wen ZHANG Zhaochu HU Yongsheng LIU Tao LUO Tao HE Hong LIU Kun YU | 2022 | Science China Earth Sciences2022,65,1: | 2 |

| 5 | Improvement of High-Temperature Mechanical Properties of Low-Carbon RAFM Steel by MX Precipitates显示文摘To investigate the influence of tantalum content on high-temperature mechanical properties of low-carbon reduced activation ferritic/martensitic(RAFM) steels, RAFM steels containing different tantalum contents(0 and 0.073%) were fabricated, and the tensile tests at room temperature and high temperature were performed, as well as the creep tests were conducted at 550 °C with the applied stress of 180 and 220 MPa. It was found that 0.073% tantalum addition results in the increase in amount of stable carbonitrides(MX), and the creep rupture time of the steel under 180 MPa is obviously increased. In addition, the increase in MX caused by tantalum addition also leads to the improvement of high-temperature tensile strength. | 7 | BEAR1, a bHLH transcription factor, controls seedling growth by regulating gibberellins biosynthesis in rice显示文摘The genetic pathways of rice seedling growth have a major impact on seedling emergence from soil and development.In this study,we identified a new bHLH transcription factor,BEAR1,from rice RNAi mutant library.Both the BEAR1-RNAi and bear1 CRISPR mutants had accelerated seedling growth.Histological section of leaves showed accelerated development of lacuna and vascular bundles in bear1 mutant.GUS staining revealed that BEAR1 was highly expressed in coleoptiles and leaves at seedling stage.Expression analysis of gibberellin(GA)biosynthesis and metabolic genes and content determination of active GAs indicated that the expression of GA biosynthesis genes,especially OsKS4 and OsCPS2,were upregulated and the GAs content were significantly increased in bear1,which correlated with the seedling phenotype of bear1 mutant.Molecular and biochemical assays revealed that BEAR1 directly binds to the promoter of OsKS4,thereby repressing its expression.Haplotypes analysis showed clear differentiation in indica and japonica rice varieties,and a strong correlation with seedling height.These findings provide novel insights into the regulation of seedling growth in rice. | Yantong Teng Maohong Cai Qinyu Xie Qinglong Liu Haiwen Zhang Tao Chen | 2023 | The Crop Journal2023,11,3: | 0 |
| 8 | Wenjing Zhitong recipe exhibits potential anti-primary dysmenorrhea properties by inhibiting COX2 and PKC signaling pathway in rats induced by estradiol benzoate and oxytocin显示文摘Objective:To explore the effect and mechanism of action of the Wenjing Zhitong recipe(WZR)in primary dysmenorrhea(PD)treatment.Methods:Uterine contractions were induced by estradiol benzoate and oxytocin in a PD model and WZR was administrated.The rate of change in uterine contractility and the writhing test were used to evaluate the effects of WZR.The serum levels of prostaglandin F_(2a)(PGF_(2a))and prostaglandin E_2(PGE_2),and the activity of cyclooxygenase-2(COX2)were detected by enzyme-linked immunosorbent assay(ELISA).The changes in phosphor-phospholipase C(pPLC/PLC),phosphor-protein kinase C(pPKC/PKC),and connexin 43(CX43)expression were detected using immunohistochemistry and western blot.Results:WZR significantly reduced the rate of change in uterine contractility and writhing times in the PD model.WZR treatment inhibited the enzymatic activity of COX2 and reduced the levels of PGF_(2a),PGF_(2a)/PGE2and COX2 in the PD model.WZR also significantly reduced the expression of pPLC/PLC,pPKC/PKC and CX43.Targeting the inhibition of COX2 activity,caffeic acid and 1-acetyl-β-carboline were validated as the active ingredients in WZR responsible for reducing uterine contractions.Conclusion:WZR attenuated PD by inhibiting COX2 activity,downregulating PGF_(2a)/PGE_2 expression,and inhibiting the PKC signaling pathway. | Jialin Li Xiaoyao Liu Mei Jiang Yantong Xu Chunguo Wang Yujie Hu Song Yang Jian Gao Weiling Wang Ting Wang | 2023 | Journal of Traditional Chinese Medical Sciences2023,10,3: | 0 |
