1Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
2Numerical investigation on freeze-drying of aqueous material frozen with pre-built pores显示文摘Freeze-drying of the initially porous frozen material with pre-built pores from liquid material was found experimentally to save drying time by over 30% with an initial saturation being 0.28 compared with the conventional operation with the initial saturation being 1, using mannitol as the solid material. In order to understand the mass and heat transfer phenomena of this novel process, a two-dimensional mathematical model of coupled mass and heat transfer was derived with reference to the cylindrical coordinate system. Three adsorption–desorption equilibrium relationships between the vapour pressure and saturation value namely, power-law, Redhead's style and Kelvin's style equation, were tested. Kelvin's style in exponential form of adsorption equilibrium relation gave an excellent agreement between the model prediction and experimental measurement when the equation parameter, γ, of 5000 was applied. Analyses of temperature and ice saturation profiles show that additional heat needs to be supplied to increase the sample temperature in order to promote the desorption process. Simulation also shows that there is a threshold initial porosity after which the drying time decreased with the increase in the initial porosity. 4Hd1,Ghd7,and DTH8 synergistically determine the rice heading date and yield-related agronomic traits显示文摘Heading date determines the seasonal and regional adaptation of rice(Oryza sativa L.)varieties and is mainly controlled by photoperiod sensitivity(PS).The core heading date genes Hd1,Ghd7,DTH8,and PRR37 act synergistically in regulating the PS.In this study,we systematically analyze the heading date,PS,and agronomic traits of eight homozygous lines with various combinations of Hd1,Ghd7,and DTH8 alleles in the prr37 background under long-day(LD)and short-day(SD)conditions,respectively.We find that Hd1 alone promotes heading,regardless of the day length.However,under LDs,Hd1 suppresses flowering,in coordination with functional Ghd7 or with Ghd7 and DTH8.These loci cooperate to negatively regulate the Ehd1-Hd3 a/RFT1 pathway and delay heading.Under SDs,Hd1 competes with various heading suppressors to promote heading.Therefore,the dual function of Hd1 is vital for PS.The lines carrying Hd1 alone show reduced plant height with fewer primary and secondary branches in panicles.Lines carrying Ghd7 and DTH8(with hd1)show delayed heading and improve agronomic traits.Overall,our results reveal the regulation of rice PS flowering by the core heading date genes and their effects on agronomic traits,providing valuable information for the selection of rice varieties for adaptation to different light and temperature conditions.Kangli Sun Minghui Huang Wubei Zong Dongdong Xiao Chen Lei Yanqiu Luo Yingang Song Shengting Li Yu Hao Wanni Luo Bingqun Xu Xiaotong Guo Guangliang Wei Letian Chen Yao-Guang Liu Jingxin Guo 2022Journal of Genetics and Genomics2022,49,5:4
5A recombinant receptor-binding domain in trimeric form generates protective immunity against SARSCoV-2 infection in nonhuman primates显示文摘A safe and effective vaccine is critical to combat the COVID-19 pandemic.Here,we developed a trimeric SARS-CoV-2 receptor-binding domain(RBD)subunit vaccine candidate that simulates the natural structure of the spike(S)trimer glycoprotein.Immunization with the RBD trimer-induced robust humoral and cellular immune responses,and a high level of neutralizing antibodies was maintained for at least 4.5 months.Moreover,the antibodies that were produced in response to the vaccine effectively cross-neutralized the SARS-CoV-2501Y.V2 variant(B.1.351).Of note,when the vaccine-induced antibodies dropped to a sufficiently low level,only one boost quickly activated the anamnestic immune response,conferring full protection against a SARSCoV-2 challenge in rhesus macaques without typical histopathological changes in the lung tissues.These results demonstrated that the SARS-CoV-2 RBD trimer vaccine candidate is highly immunogenic and safe,providing long-lasting,broad,and significant immunity protection in nonhuman primates,thereby offering an optimal vaccination strategy against COVID-19.Limin Yang Deyu Tian Jian-bao Han Wenhui Fan Yuan Zhang Yunlong Li Wenqiang Sun Yanqiu Wei Xiaodong Tian Dan-dan Yu Xiao-li Feng Gong Cheng Yuhai Bi Yong-tang Zheng Wenjun Liu 2021The Innovation2021,2,3:3

10Buyang Huanwu Tang(补阳还五汤) protects H_(2)O_(2)-induced RGC-5 cell against oxidative stress and apoptosis via reactive oxygen species-mitogen-activated protein kinase signaling pathway显示文摘OBJECTIVE: To study the molecular mechanism of Buyang Huanwu Tang( 补阳还五汤)(BHT) protecting retinal ganglion cells(RGCs) from oxygen induced oxidative stress and apoptosis after anterior ischemia. METHODS: In this study, the Chinese herbs of BHT were extracted by first boiling in water, then were filtered, concentrated, and freeze-dried. The chemical profile of BHT extract was determined by liquid chromatography mass spectrometry(LC-MS). H2O2-induced RGC-5 cells were used as a cell model to investigate the protective effect and mechanism of BHT on RGCs. RESULTS: The survival rate of damaged RGC-5 by BHT was significantly increased by the 3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazolium-romid method. Fluorescence activating cell sorter(FACS analysis) showed that BHT could significantly reduce apoptosis induced by oxidative stress via the reactive oxygen species(ROS)-mitogen-activated protein kinase(MAPK)-Caspase-3 signal pathway. 11Machine Learning for Investigation on Endocrine-Disrupting Chemicals with Gestational Age and Delivery Time in a Longitudinal Cohort显示文摘Endocrine-disrupting chemicals(EDCs)are widespread environmental chemicals that are often considered as risk factors with weak activity on the hormone-dependent process of pregnancy.However,the adverse effects of EDCs in the body of pregnant women were underestimated.The interaction between dynamic concentration of EDCs and endogenous hormones(EHs)on gestational age and delivery time remains unclear.To define a temporal interaction between the EDCs and EHs during pregnancy,comprehensive,unbiased,and quantitative analyses of 33 EDCs and 14 EHs were performed for a longitudinal cohort with 2317 pregnant women.We developed a machine learning model with the dynamic concentration information of EDCs and EHs to predict gestational age with high accuracy in the longitudinal cohort of pregnant women.The optimal combination of EHs and EDCs can identify when labor occurs(time to delivery within two and four weeks,AUROC of 0.82).Our results revealed that the bisphenols and phthalates are more potent than partial EHs for gestational age or delivery time.This study represents the use of machine learning methods for quantitative analysis of pregnancy-related EDCs and EHs for understanding the EDCs’mixture effect on pregnancy with potential clinical utilities.Hemi Luan Hongzhi Zhao Jiufeng Li Yanqiu Zhou Jing Fang Hongxiu Liu Yuanyuan Li Wei Xia Shunqing Xu Zongwei Cai 2021Research2021,,1:0
12Mediastinal Tuberculoma Mimicking Malignant Cardiac Tumor:A Case Report显示文摘Background:The clinical manifestations of cardiac masses are diverse and lack specifi city.Here we report a cardiac mass detected by transthoracic echocardiography.Multimodality imaging and pathological fi ndings after the operation confi rmed the mass as mediastinal tuberculoma.Case presentation:A 45-year-old male patient was admitted to our hospital reporting chest tightness,weight loss,and dyspnea for 3 months after exercise.Transthoracic echocardiography showed that there were a large number of pericardial effusions and a soft tissue mass measuring 7.7 cm×4.5 cm in the upper mediastinum,which oppressed the right pulmonary artery and accelerated the blood fl ow of the left pulmonary artery.Contrast-enhanced ultrasonography showed degenerative inhomogeneous high enhancement of and an unclear boundary in the mass.Contrast-enhanced chest CT revealed punctate and patchy calcifi cation in and uneven enhancement of the mass and the lymph nodes around the aortic arch.The mass was diagnosed as a malignant mediastinal tumor.Pathological analysis of the mass revealed chronic granulomatous tuberculosis.The symptoms abated signifi cantly after antituberculosis treatment.The patient remained asymptomatic during follow-up.Conclusion:This report presents a rare case of mediastinal tuberculoma mimicking a malignant cardiac tumor.Multimodality imaging should be incorporated for differentiation of cardiac masses.Yiqian Ding Wei Li Yanqiu Liu Min Ye Liangping Cheng Donghong Liu Hong Lin Fengjuan Yao 2021Cardiovascular Innovations and Applications2021,,2:0

15Synergistic sulfonamides plus clindamycin as an alternative therapeutic regimen for HIV-associated Toxoplasma encephalitis: a randomized controlled trial显示文摘Background: The preferred therapeutic regimen for Toxoplasma encephalitis (TE) is a combination of pyrimethamine and sulfadiazine, and trimethoprim-sulfamethoxazole (TMP-SMX) plus azithromycin is the widespread alternative therapeutic regimen. The synergistic sulfonamides tablet contains TMP, sulfadiazine, and SMX and hypothetically could be used for TE treatment. This study aimed to compare the efficacy and safety of synergistic sulfonamides plus clindamycin (regimen B) with TMP-SMX plus azithromycin (regimen A) for the treatment of human immunodeficiency virus (HIV) associated TE.Methods: This was an open-labeled, multi-center randomized controlled trial recruited from 11 centers. Each recruited patient was randomly assigned to receive regimen A or regimen B for at least 6 weeks. The overall response was evaluated by assessment of the clinical response of TE-associated clinical features and the radiological response of TE-associated radiological findings. The overall response rate, clinical response rate, radiological response rate, and adverse events were assessed at 2, 6, and 12 weeks. Death events were compared between the two regimens at 6, 12, and 24 weeks.Results: A total of 91 acquired immunodeficiency syndrome (AIDS)/TE patients were included in the final analysis (44 in regimen Avs. 47 in regimen B). The overall response rate, which refers to the combined clinical and radiological response, was 18.2% (8/44) for regimen A and 21.3 % (10/47) for regimen B at week 6. The results of clinical response showed that, in comparison with regimen A, regimen B may perform better with regards to its effect on the relief of clinical manifestations (50.0% [22/44]vs. 70.2% [33/47],P = 0.049). However, no significant differences in radiological response, mortality events, and adverse events were found between the two regimens at week 6.Conclusions: Synergistic sulfonamides plus clindamycin, as a novel treatment regimen, showed no significantly different efficacy and comparable safety in comparison with the TMP-SMX plus azithromycin regimen. 17Complexation between uranyl(Ⅵ)and CMPO in a hydroxyl-functionalized ionic liquid:An extraction,spectrophotography,and calorimetry study显示文摘The extraction complexes of uranyl(Ⅵ)in HNO_(3)to a hydroxyl-functionalized ionic liquid(IL)phase,HOEtmimNTf2bearing CMPO,were investigated.Three possibly successive extraction complexes,UO_(2)L^(2+)(L=CMPO),UO2L_(2)^(2+) and UO_(2)L_(3)^(2+),were detected based on variable U/L ratios.Uranyl(Ⅵ)prefers to be extracted as complex UO_(2)L_(3)^(2+),combining with the ions from HOEtmimNTf_(2) to construct a solid material through self-assembly.The thermodynamics of complexes,UO_(2)L_(j)^(2+)(j=1-3),were studied by spectrophotometry and microcalorimetry.All the formation reactions are principally driven by entropy,although a small part of the driving force of complexes UO_(2)L_(2)^(2+)and UO_(2)L_(3)^(2+) comes from enthalpy.Based on the thermodynamic properties for complex UO_(2)L_(3)^(2+),we provide a possible coordination mode in HOEtmim NTf_(2):the first CMPO molecule coordinates with UO_(2)^(2+) in a bidentate fashion while the others do in a monodentate fashion.The results offer a thermodynamic insight into the formation behaviors of the uranyl(Ⅵ)/CMPO complexes involving the special IL HOEtmimNTf_(2),which is of significance to advance the novel IL extraction strategy.Baihua Chen Jun Liu Hongyuan Wei Yuchuan Yang Xingliang Li Shuming Peng Yanqiu Yang 2022Chinese Chemical Letters2022,33,7:0
18Expression of E-Cadherin in Oral Squamous Cell Carcinoma is Associated with Clinical Prognosis显示文摘OBJECTIVE To investigate the correlation of E-cadherin expression with clinicopathological parameters and prognosis of patients with oral squa- mous cell carcinoma. METHODS We examined the expression of the protein E-cadherin in 43 oral squamous cell carcinoma (SCC) surgical speciments by SABC im- munohistochemistry. RESULTS There was a significant correlation between the level of E- cadherin expression and tumor stage (P=0.024), invasive pattern (P= 0.009) and lymph node metastasis (P=0.023). No relation was found with age (P=0.084), sex (P= 1.356) and differentiation (P=0.877). Using the Kaplan-Meier method we demonstrated that those cases which showed E-cadherin expression (-) or (+) had a significantly poorer prognosis compared those cases which showed expression (++) or (+++) (P= 0.0146). 20Dissociation of NO_2 in femtosecond intense fields显示文摘Experimental investigations on the dis-sociation and ionization processes of NO2 in intensefields at wavelengths of 810, 405 and 270 nm, arepresented. The ratios of NO +2 /NO + are found to beindependent of the laser intensity at these threewavelengths, but show strong dependence on thewavelengths. Longer wavelength produces a largerparent-ion yield in comparison with 405 nm. Peakprofiles of all the fragment ions clearly show little ki-netic energy releasing during the dissociation. Frag-ment ions are suggested to be produced from disso-ciation of the field ionized parent ions.
