3PhaseⅡstudy of induction chemotherapy followed by concurrent chemoradiotherapy with raltitrexed and cisplatin in locally advanced nasopharyngeal carcinoma显示文摘Objective:For locally advanced nasopharyngeal carcinoma(LA-NPC)patients,high incidences of distant metastases and severe treatment related toxicities are the main obstacles needed to be overcome.Raltitrexed,a specific thymidylate synthase inhibitor with a convenient administration schedule,has an acceptable and manageable toxicity,and possesses radio-sensitizing properties.To investigate the efficacy and safety of raltitrexed and cisplatin induction chemotherapy and concurrent chemoradiotherapy(IC+CCRT)in patients with LA-NPC,a phaseⅡclinical study was conducted.Methods:Sixty eligible patients with LA-NPC were enrolled into this study.A raltitrexed-cisplatin combination was used as part of an IC+CCRT regimen.Raltitrexed-cisplatin IC was given once every 3 weeks(q3 w)for two cycles,followed by raltitrexed-cisplatin based CCRT q3 w for two cycles.Intensity-modulated radiotherapy(IMRT)was given for all enrolled patients.Results:All patients were included in survival analysis according to the intent-to-treat principle.The objective response rate(ORR)3 months after treatment was 98%.The 2-year overall survival(OS)rate was 92%.The median relapse-free survival(RFS)time was 30.5[95%confidence interval(95%CI),28.4-32.3]months.The 2-year RFS rate was 85%.The 2-year local failure-free survival(LFFS)rate was 97%and the 2-year distant metastasis-free survival(DMFS)rate was 88%.Acute toxicities were mostly grade 2 and 3 reactions in bone marrow suppression,gastrointestinal side effect and oropharyngeal mucositis.Only two patients occurred grade 4 acute toxicities,one was bone marrow suppression and the other was dermatitis radiation.Conclusions:The combination of raltitrexed and cisplatin has a comparable efficacy to those in standard firstline therapy.Yuan Wu Xueyan Wei Zilong Yuan Hongbin Xu Yanping Li Ying Li Liu Hu Guang Han Yu Qian Desheng Hu 2020Chinese Journal of Cancer Research2020,32,5:4
4Effects of Shuanghuanglian Propolis Oral Liquid on Growth Performance,Slaughter Performance and Meat Quality of Broilers显示文摘[Objective] The paper was to investigate the effects of adding Shuanghuanglian propolis oral liquid in drinking water on growth performance,slaughter performance and meat quality of broilers. 5Effects of Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum on chemotactic migration of alveolar macrophages and inters regions macrophages in rats显示文摘OBJECTIVE: To observe the effects of serum containing Mahuang(Herba Ephedra Sinica) or Wuweizi(Fructus Schisandrae Chinensis) on the migration of alveolar macrophages(AM) and interstitial macrophages(IM) from normal rats, and to analyze and compare the mechanisms leading to cell migration differences.METHODS: Rats were randomly divided into three groups: Mahuang(Herba Ephedra Sinica), Wuweizi(Fructus Schisandrae Chinensis), and blank serum. After treatment with the herbs, serum was extracted from the rats. AM and IM were isolated from normal rats and cultured. The effects of Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum on normal rat AM and IM chemotactic migration were determined by transwell assays. The CC chemokine receptor(CCR) 2, CCR5, voltage-gated Kvl. 3 K^+channel(Kv1. 3), and voltage-gated Kvl. 5 K^+channel(Kv1. 5) protein levels were analyzed by western blotting.RESULTS: The migration quantities of AM and IM in the Mahuang(Herba Ephedra Sinica) and Wuweizi(Fructus Schisandrae Chinensis) medicated serum groups were significantly higher than those in the blank serum group(P < 0.01). Compared with the Wuweizi(Fructus Schisandrae Chinensis) medicated serum group, the migration quantity of cultured rat AM in the Mahuang(Herba Ephedra Sinica) medicated serum group was significantly increased(P <0.01). Meanwhile, compared with the Mahuang(Herba Ephedra Sinica) medicated serum group, the migration quantity of cultured rat IM in the Wuweizi(Fructus Schisandrae Chinensis) medicated serum group was significantly increased(P < 0.01).CCR2, CCR5, Kv1. 3, and Kv1. 5 proteins were expressed on the AM cell surface, and showed significantly higher expression in the Mahuang(Herba Ephedra Sinica) medicated serum group compared with the Wuweizi(Fructus Schisandrae Chinensis)medicated serum group. In contrast, CCR5, Kv1.3,and Kv1.5 proteins were expressed on the IM cell surface, and showed significantly higher expression in the Wuweizi(Fructus Schisandrae Chinensis)medicated serum group compared with the Mahuang(Herba Ephedra Sinica) medicated serum group.CONCLUSION: Mahuang(Herba Ephedra Sinica)and Wuweizi(Fructus Schisandrae Chinensis) can promote AM and IM migration ability, with Mahuang(Herba Ephedra Sinica) targeting AM more apparently and Wuweizi(Fructus Schisandrae Chinensis) targeting IM more apparently. 7Fiber-tip polymer clamped-beam probe for high-sensitivity nanoforce measurements显示文摘Micromanipulation and biological,material science,and medical applications often require to control or measure the forces asserted on small objects.Here,we demonstrate for the first time the microprinting of a novel fiber-tip-polymer clamped-beam probe micro-force sensor for the examination of biological samples.The proposed sensor consists of two bases,a clamped beam,and a force-sensing probe,which were developed using a femtosecond-laser-induced two-photon polymerization(TPP)technique.Based on the finite element method(FEM),the static performance of the structure was simulated to provide the basis for the structural design.A miniature all-fiber micro-force sensor of this type exhibited an ultrahigh force sensitivity of 1.51 nmμN−1,a detection limit of 54.9 nN,and an unambiguous sensor measurement range of~2.9 mN.The Young’s modulus of polydimethylsiloxane,a butterfly feeler,and human hair were successfully measured with the proposed sensor.To the best of our knowledge,this fiber sensor has the smallest force-detection limit in direct contact mode reported to date,comparable to that of an atomic force microscope(AFM).This approach opens new avenues towards the realization of small-footprint AFMs that could be easily adapted for use in outside specialized laboratories.As such,we believe that this device will be beneficial for high-precision biomedical and material science examination,and the proposed fabrication method provides a new route for the next generation of research on complex fiber-integrated polymer devices.Mengqiang Zou Changrui Liao Shen Liu Cong Xiong Cong Zhao Jinlai Zhao Zongsong Gan Yanping Chen Kaiming Yang Dan Liu Ying Wang Yiping Wang 2021Light(Science & Applications)2021,10,9:3
8Efficacy of Rebixiao Chinese herbal tablets and Chinese formula granules in acute gout arthritis patients:a randomized,multicenter,double-blind,controlled trial显示文摘OBJECTIVE:To evaluate the clinical efficacy and safety of Rebixiao(RBX)Chinese herbal tablets(CHT)and Chinese formula granules(CFG)in the treatment of acute gout arthritis(AGA).METHODS:This randomized,multicenter,double-blind,controlled trial included 165 AGA patients with the damp-heat symptom pattern who were randomly divided into an RBX CHT group and an RBX CFG group and treated for 7 d at three centers.The total effective rates of the joint symptom score,Traditional Chinese Medicine(TCM)symptoms score,erythrocyte sedimentation rate(ESR),and C-reactive protein(CRP)were used to evaluate the clinical efficacy.Safety assessments were also performed.RESULTS:Of the 165 enrolled patients,147 completed the clinical observation.There was no difference in baseline between the two groups.The total effective rates of the joint symptom score were 94.36%and 97.36%,and the total effective rates of the TCM symptoms score were 95.77%and 97.36%in the CFG group and CHT group,respectively.No statistical difference was found between the two groups(P>0.05).Additionally,ESR and CRP were similar in both groups(P>0.05).Furthermore,treatment efficacy regarding TCM and joint symptoms,the ESR,and CRP were consistent within each center and among the different centers(P>0.05).In addition,the incidence of adverse events was 4.22%and 2.63%in the CFG group and CHT group,respectively,and no difference was observed between the two groups(P>0.05).CONCLUSION:RBX CFG and CHT have significant and similar efficacy in the treatment of AGA,and CFG did not increase adverse side effects.Wang Shasha Zhang Ying Dai Min Yang Min Leng Wenfei Peng Xiufeng Li Guojun Yang Jinbing Gan Jianping Zhu Fenglin Li Yanping Wu Bin 2020Journal of Traditional Chinese Medicine2020,40,4:3
9Tensile properties analysis of CFRP-titanium plate multi-bolt hybrid joints显示文摘Hybrid joints have better tensile properties than pure bonded and bolted bolts,and are increasingly used in the aerospace field.Tensile tests are carried out for the Hybrid Bonded/Bolted(HBB)joints of Carbon Fiber Reinforced Polymer(CFRP)laminate and titanium alloy plate under different bolt numbers,and the corresponding load–displacement curves are obtained.At the same time,based on Continuum Damage Mechanics(CDM)theory,which is derived from 3D Hashin failure criteria,and a Cohesive Zone Model(CZM),the tensile strength prediction model of the composite laminate-titanium alloy plate multi-bolted HBB joint was established,and the numerical simulation results were in good agreement with the experimental height,which validate the feasibility of the model.The difference in the bearing capacity of HBB joints under different numbers of bolts is compared and analyzed.On this basis,the influence of inter-bolt distance on the tensile properties of the HBB joints is explored.The results show that the double-nail HBB joints can effectively improve the end warpage and low bearing capacity of the single-nail HBB joints.The tensile failure load of the double-nail HBB joints under the standard lap width(30 mm)is 82.6%higher than that of the single nail,the tensile failure load of the three-bolt HBB joints is 34.1%higher than that of the double nail.For the three-bolt HBB joint,the joint strength is controlled by the adhesive and the external bolt,while the internal bolt is redundant,the hybrid joint can be simplified by reducing the middle bolt.The inter-bolt distance has a great influence on the failure load of the hybrid joint.Increasing the inter-bolt distance can effectively improve the bearing capacity of the structure.Yanping ZHENG Chaoyu ZHANG Ying TIE Xu WANG Mingkun LI 2022Chinese Journal of Aeronautics2022,35,3:2
10Two-grid methods for semi-linear elliptic interface problems by immersed finite element methods显示文摘In this paper,two-grid immersed finite element (IFE) algorithms are proposed and analyzed for semi-linear interface problems with discontinuous diffusion coefficients in two dimension.Because of the advantages of finite element (FE) formulation and the simple structure of Cartesian grids,the IFE discretization is used in this paper.Two-grid schemes are formulated to linearize the FE equations.It is theoretically and numerically illustrated that the coarse space can be selected as coarse as H =O(h^1/4)(or H =O(h^1/8)),and the asymptotically optimal approximation can be achieved as the nonlinear schemes.As a result,we can settle a great majority of nonlinear equations as easy as linearized problems.In order to estimate the present two-grid algorithms,we derive the optimal error estimates of the IFE solution in the L^p norm.Numerical experiments are given to verify the theorems and indicate that the present two-grid algorithms can greatly improve the computing efficiency.Yang WANG Yanping CHEN Yunqing HUANG Ying LIU 2019Applied Mathematics and Mechanics(English Edition)2019,40,11:2
11A durable 4-1BB-based CD19 CAR-T cell for treatment of relapsed or refractory non-Hodgkin lymphoma显示文摘Objective:Previous studies reported that 4-1BB-based CD19 chimeric antigen receptor(CAR)-T cells were more beneficial for the clinical outcomes than CD28-based CAR-T cells,especially the lower incidence rate of severe adverse events.However,the median progression-free survival(mPFS)of 4-1BB-based product Kymriah was shorter than that of CD28-based Yescarta(2.9 months vs.5.9 months),suggesting that Kymriah was limited in the long-term efficacy.Thus,a safe and durable 4-1BB-based CD19 CAR-T needs to be developed.Methods:We designed a CD19-targeted CAR-T(named as IM19)which consisted of an FMC63 scFv,4-1BB and CD3ζintracellular domain and was manufactured into a memory T-enriched formulation.A phase I/II clinical trial was launched to evaluate the clinical outcomes of IM19 in relapsed or refractory(r/r)B cell non-Hodgkin lymphoma(B-NHL).Dose-escalation investigation(at a dose of 5×10^(5)/kg,1×10^(6)/kg and 3×106/kg)was performed in 22 r/r B-NHL patients.All patients received a single infusion of IM19 after 3-day conditional regimen.Results:At month 3,the overall response rate(ORR)was 59.1%,the complete response rate(CRR)was 50.0%.The mPFS was 6 months and the 1-year overall survival rate was 77.8%.Cytokine release syndrome(CRS)occurred in 13 patients(59.1%),with 54.5%of grade 1−2 CRS.Only one patient(4.5%)experienced grade 3 CRS and grade 3 neurotoxicity.Conclusions:These results demonstrated the safety and durable efficacy of a 4-1BB-based CD19 CAR-T,IM19,which is promising for further development and clinical investigation.Zhitao Ying Ting He Shanzhao Jin Xiaopei Wang Wen Zheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Weiping Liu Lijuan Deng Yanping Ding Xuelian Hu Bing Bu Xin’an Lu Yuqin Song Jun Zhu 2022Chinese Journal of Cancer Research2022,34,1:2

13Free Riding Inhibition Mechanism Based on User Behavior in P2P File-Sharing System显示文摘In order to inhibit Free Riding in Peer-toPeer(P2P) file-sharing systems,the Free Riding Inhibition Mechanism Based on User Behavior(IMBUB) is proposed.IMBUB considers the regularity of user behavior and models user behavior by analyzing many definitions and formulas.In IMBUB,Bandwidth Allocated Ratio,Incentive Mechanism Based on User Online Time,Double Reward Mechanism,Incentive Mechanism of Sharing for Permission and Inhibition Mechanism of White-washing Behavior are put forward to inhibit Free Riding and encourage user sharing.A P2P file system BITShare is designed and realized under the conditions of a campus network environment.The test results show that BITShare's Query Hit Ratio has a significant increase from 22% to 99%,and the sharing process in BITShare is very optimistic.Most users opt to use online time to exchange service quality instead of white-washing behavior,and the real white-ishing ratio in BITShare is lower than 1%.We confirm that IMBUB can effectively inhibit Free Riding behavior in P2P file-sharing systems.Zhang Yu Bai Yanping Hao Ying 2012China Communications2012,9,12:2
143D printed fiber-optic nanomechanical bioprobe显示文摘Ultrasensitive nanomechanical instruments,e.g.atomic force microscopy(AFM),can be used to perform delicate biomechanical measurements and reveal the complex mechanical environment of biological processes.However,these instruments are limited because of their size and complex feedback system.In this study,we demonstrate a miniature fiber optical nanomechanical probe(FONP)that can be used to detect the mechanical properties of single cells and in vivo tissue measurements.A FONP that can operate in air and in liquids was developed by programming a microcantilever probe on the end face of a single-mode fiber using femtosecond laser two-photon polymerization nanolithography.To realize stiffness matching of the FONP and sample,a strategy of customizing the microcantilever’s spring constant according to the sample was proposed based on structure-correlated mechanics.As a proof-of concept,three FONPs with spring constants varying from 0.421 N m^(−1)to 52.6 N m^(−1)by more than two orders of magnitude were prepared.The highest microforce sensitivity was 54.5 nmμN^(−1)and the detection limit was 2.1 nN.The Young’s modulus of heterogeneous soft materials,such as polydimethylsiloxane,muscle tissue of living mice,onion cells,and MCF-7 cells,were successfully measured,which validating the broad applicability of this method.Our strategy provides a universal protocol for directly programming fiber-optic AFMs.Moreover,this method has no special requirements for the size and shape of living biological samples,which is infeasible when using commercial AFMs.FONP has made substantial progress in realizing basic biological discoveries,which may create new biomedical applications that cannot be realized by current AFMs.Mengqiang Zou Changrui Liao Yanping Chen Lei Xu Shuo Tang Gaixia Xu Ke Ma Jiangtao Zhou Zhihao Cai Bozhe Li Cong Zhao Zhourui Xu Yuanyuan Shen Shen Liu Ying Wang Zongsong Gan Hao Wang Xuming Zhang Sandor Kasas Yiping Wang 2023International Journal of Extreme Manufacturing2023,5,1:1

Micro RNAs(mi RNAs), which function as post-transcriptional regulators, are utilized by human cytomegalovirus(HCMV) in its infection and pathogenesis. In the present study, the DNA replication inhibitor Geminin(GMNN) was identified to be a direct target of hcmv-mi R-US5-1. Overexpression of hcmv-mi R-US5-1 could block the accumulation of GMNN during HCMV infection, and the decrease of GMNN expression caused by hcmv-mi R-US5-1 or GMNN specific si RNA reduced HCMV DNA copies in U373 cells. Meanwhile, ectopic expression of hcmv-mi R-US5-1 and consequent lower expression of GMNN influenced host cell cycle and proliferation. 17A p53/CPEB2 negative feedback loop regulates renal cancer cell proliferation and migration显示文摘The tumor suppressor p53 transactivates the expression of multiple genes to exert its multifaceted functions and ultimately maintains genome stability.Thus,cancer cells develop various mechanisms to diminish p53 expression and bypass the cell cycle checkpoint.In this study,we identified the gene encoding RNAbinding protein cytoplasmic polyadenylation element-binding protein 2(CPEB2)as a p53 target.In turn,CPEB2 decreases p53 messenger RNA stability and translation to fine-tune p53 level.Specifically,we showed that CPEB2 binds the cytoplasmic polyadenylation elements in the p5330-untranslated region,and the RNA recognition motif and zinc finger(ZF)domains of CPEB2 are required for this binding.Furthermore,we found that CPEB2 was upregulated in renal cancer tissues and promotes the renal cancer cell proliferation and migration.The oncogenic effect of CPEB2 is partially dependent on negative feedback regulation of p53.Overall,we identify a novel regulatory feedback loop between p53 and CPEB2 and demonstrate that CPEB2 promotes tumor progression by inactivating p53,suggesting that CPEB2 is a potential therapeutic target in human renal cancer.Jiehui Di Guang Zhao Hui Wang Yaoyao Wu Zhongjun Zhao Bao Zhu Yanping Zhang Junnian Zheng Yong Liu Ying Hu 2021Journal of Genetics and Genomics2021,48,7:1

19Vasorelaxant effect of osthole on isolated thoracic aortic rings in rats显示文摘OBJECTIVE: To investigate the effect of osthole on isolated thoracic aortic rings, and to determine the potential mechanism of action.METHODS: Thoracic aortas were isolated from Wistar rats, and were suspended in tissue organ chambers for vascular tension measurement. The effect of cumulative osthole(10-9, 10-8, 10-7, 10-6, and 10-5 mol/L) on endothelium-intact and endothelium-denuded thoracic aortic rings pre-contracted with phenylephrine(PE, 10-6 mol/L) or KCl(6 × 10-2 mol/L) was recorded. Histomorphological changes of thoracic aorta were analyzed by hematoxylin-eosin. The effects of different osthole concentrations on endothelium-intact aortic rings, which were pre-inhibited with the non-selective nitric oxide synthase inhibitor L-Arg(NO2)-OMe·HCl(3 × 10-4 mol/L), endothelium-derived nitric oxide synthase inhibitor Nω-nitro-L-arginine(3 × 10-4 mol/L), guanylate cyclase inhibitor 1 H-[1,2,4] oxadiazolo [4,3-α]quinoxaline-1-one(10-5 mol/L), cyclooxygenase inhibitor indometacin(10-5 mol/L), and the Ca2+-activated potassium channel inhibitor tetraethylammonium nitrate(10-5 mol/L), and then contracted with PE, were examined. Aortic rings incubated with osthole(10-5 mol/L), phentolamine(10-5 mol/L), or verapamil(10-5 mol/L) in Ca2+-free Krebs-Henseleit solution(KHS) were stimulated with PE or KCl.RESULTS: There was a dose-dependent increase in vasorelaxation of isolated thoracic aortic rings(both with and without endothelium) with increasing osthole concentration. Hematoxylin-eosin staining showed that osthole significantly improved thoracic aorta ring morphology. Compared with the control group, there were also significant differences after incubation with L-Arg(NO2)-OMe ω-nitro-L-arginine, and 1 H-[1,2,4] oxadiazo·lo HCl,N [4,3-α] quinoxaline-1-one(P < 0.05 for all). The relaxation rate of the rings in the osthole group incubated with indometacin and tetraethylammonium nitrate were similar to controls. In Ca2+-free KHS, the PE-induced contraction was similar between the osthole(4.37% ± 0.41%) and control(4.21% ± 1.33%)groups. However, after cumulative CaCl2(0.5, 1, 1.5,2, 2.5, and 3 mmol/L), the Ca2+-induced contraction was significantly inhibited in the osthole and phentolamine groups compared with controls(P < 0.05).After cumulative CaCl2 was added to Ca2+-free KHS(high K+ concentration), the contraction rate was significantly higher than both of the control and the osthole groups(P < 0.05).
