1Self-processing of ribozyme-flanked RNAs into guide RNAs in vitro and in vivo for CRISPR-mediated genome editing显示文摘CRISPR/Cas9 uses a guide RNA(gRNA)molecule to execute sequence-specific DNA cleavage and it has been widely used for genome editing in many organisms.Modifications at either end of the gRNAs often render Cas9/gRNA inactive.So far,production of gRNA in vivo has only been achieved by using the U6 and U3 snRNA promoters.However,the U6 and U3 promoters have major limitations such as a lack of cell specificity and unsuitability for in vitro transcription.Here,we present a versatile method for efficiently producing gRNAs both in vitro and in vivo.We design an artificial gene named RGR that,once transcribed,generates an RNA molecule with ribozyme sequences at both ends of the designed gRNA.We show that the primary transcripts of RGR undergo self-catalyzed cleavage to generate the desired gRNA,which can efficiently guide sequence-specific cleavage of DNA targets both in vitro and in yeast.RGR can be transcribed from any promoters and thus allows for cell-and tissue-specific genome editing ifappropriate promoters are chosen.Detecting mutations generated by CRISPR is often achieved by enzyme digestions,which are not very compatible with high-throughput analysis.Our system allows for the use of universal primers to produce any gRNAs in vitro,which can then be used with Cas9 protein to detect mutations caused by the gRNAs/CRISPR.In conclusion,we provide a versatile method for generating targeted mutations in specific cells and tissues,and for efficiently detecting the mutations generated.Yangbin Gao Yunde Zhao 2014Journal of Integrative Plant Biology2014,56,4:39
2阴道镜下子宫颈多点活检及颈管内膜刮取术对高级别子宫颈病变的诊断价值显示文摘目的:评价阴道镜下子宫颈多点活检及颈管内膜刮取术(ECC)对高级别子宫颈病变[指子宫颈上皮内瘤变(CIN)Ⅱ及以上病变]的诊断价值。方法2009年4月-2010年4月进行的深圳子宫颈癌筛查项目Ⅱ(SHENCCASTⅡ)是以子宫颈液基细胞学检查联合HPV检测[包括HPV DNA分型和第2代杂交捕获(HC-Ⅱ)法]方法在10000例妇女中进行子宫颈癌筛查,对于上述筛查指标任一结果阳性(即子宫颈癌筛查阳性)者同时行阴道镜下子宫颈多点活检及ECC,对其活检组织(包括多点活检和ECC)进行病理检查。实施阴道镜检查并获得有效病理检查结果及保存有清晰阴道镜图像者共2558例,其中阴道镜下子宫颈4个象限均无异常(即阴道镜检查阴性)者1790例。对照其活检组织的病理诊断,分析子宫颈细胞学检查与HPV检测结果提示高级别子宫颈病变的作用,探讨多点活检与ECC对诊断高级别子宫颈病变的价值。结果2558例行阴道镜检查妇女中,阴道镜检查阴性者1790例(69.98%,1790/2558),其中经多点活检及ECC获取的组织行病理检查后诊断为高级别子宫颈病变者共40例(2.23%,40/1790)。阴道镜检查阴性的40例高级别子宫颈病变患者中,细胞学检查为高级别鳞状上皮内病变(HSIL)和不能除外高度病变的鳞状上皮细胞(ASC-H)者与高级别子宫颈病变的患病高风险有关(OR值分别为28.37、15.07,P值分别为0.000、0.001);HPV16、52、58、31、33、18型阳性者与高级别子宫颈病变的患病高风险有关(OR=3.11,P=0.017);HC-Ⅱ法检测HPV阳性者与高级别子宫颈病变的患病高风险有关(OR=3.58,P=0.025)。2558例行阴道镜检查妇女中,≥40岁与〈40岁者ECC阳性(ECC阳性指ECC获取的组织病理诊断为高级别子宫颈病变)率分别为40.7%(44/108)和19.2%(24/125),两者比较,差异有统计学意义(χ2=13.01,P=0.000);≥40岁与〈40岁者多点活检阳性(指多点活检组织病理诊断为高级别子宫颈病变)率分别为90.7%(98/108)、88.8%(111/125),两者比较,差异无统计学意义(χ2=0.24,P>0.05)。40例阴道镜检查阴性的高级别子宫颈病变患者中,有32例在3项子宫颈癌高风险因素(即细胞学检查为HSIL或ASC-H,HPV16、18、52和58型中任一亚型阳性,HC-Ⅱ法检测HPV阳性)中至少有2项阳性。结论仅对阴道镜检查阳性者行子宫颈活检,会漏诊部分高级别子宫颈病变患者;对子宫颈癌筛查阳性者均行阴道镜下子宫颈多点活检及ECC,有利于减少漏诊。张凌 杜辉 张薇 YangBin 王纯 JeromeLBelinson 吴瑞芳 2015中华妇产科杂志2015,50,4:34
3Trustworthiness of Internet-based software显示文摘Recent years see an increasing concern over the trustworthiness of Inter- net-based software. By analyzing the trustworthiness of Internet-based software and the nature of the Internet applications, we point out that, on the one hand, due to the open- ness and dynamic nature of the Internet, the identity trustworthiness and the capability trustworthiness of the software are facing serious challenges; on the other hand, in order to ensure the trustworthiness of the whole system, emerging computing paradigms based on the collaboration of autonomous software need some impacts on the behavior of the software. Here we put forward a conceptual model for the trustworthiness of Inter- net-based software, and propose a trustworthy assurance framework for Internet-based virtual computing environment (iVCE). This framework deals with the trustworthy proper- ties of software on identity, capability and behavior in a combinated way. 4Geochemical fractionation of rare earth elements in lacustrine deposits from Qaidam Basin显示文摘Based on the concentration and distribution pattern analysis of the rare earth elements (REEs) at the Shell Bar section from Qaidam Basin, we studied the geochemical fractionations of REE in paleolake deposits and their paleo-environmental significance. Our results show that the REE concentration in AS (acid soluble) and AR (acid residual) fractions are 20.9 μg/g and 95.4 μg/g (except element Y) individually, showing a strong REE differentiation between AS and AR fractions. However, the two types of fractions (AS & AR) have similar REE distribution patterns, which are slightly rich in light earth rare elements (LREEs), with slightly right-tilting and negative Eu anomaly. The LREE of AR is richer than that of AS. There were no significant correlations between the REE in AS, AR and other proxies. It indicated that the lacustrine deposition had different material sources and experienced varying geochemical procedures. Correlation analysis between the REE and the content of fine grain-size (<4 μm) of the sediments, Rb/Sr ratio and Mn concentration showed high correlation coefficients. Our results demonstrated that the REE in acid soluble and residual fractions bear different environmental significances, which are strongly dependent on local environment. The REE of acid soluble fraction is closely related to the paleoclimatic changes in the lake catchment and the evolutionary processes of lake itself. Our results suggest that δCe and (La/Yb)n could be used as proxies of the reduction-oxidation conditions and furthermore the temperature change and the lake level fluctuations. 6Design and fabrication of superconducting HEB mixer显示文摘This paper describes the design and fabrication of superconducting hot electron bolometer (HEB) mixer based on ultra-thin superconducting NbN films. The high quality films were epitaxially grown on high resistance Si substrates. The device was fabricated by magnetron sputtering, electron beam lithography (EBL), reactive ion etching (RIE), lithography, and so on. The device's resistance-tempera-ture (R-T) curves and current-voltage (I-V) curves were studied. The results of THz response of the device are presented. Y-factor technique was used to measure the device's noise temperature. 7Fast-Suppressor Screening for New Components in Protein Trafficking,Organelle Biogenesis and Silencing Pathway in Arabidopsis thaliana Using DEX-Inducible FREE1-RNAi Plants显示文摘Membrane trafficking is essential for plant growth and responses to external signals.The plant unique FYVE domain-containing protein FREE1 is a component of the ESCRT complex(endosomal sorting complex required for transport).FREE1 plays multiple roles in regulating protein trafficking and organelle biogenesis including the formation of intraluminal vesicles of multivesicular body(MVB),vacuolar protein transport and vacuole biogenesis,and autophagic degradation.FREE1 knockout plants show defective MVB formation,abnormal vacuolar transport,fragmented vacuoles,accumulated autophagosomes,and seedling lethality.To further uncover the underlying mechanisms of FREE1 function in plants,we performed a forward genetic screen for mutants that suppressed the seedling lethal phenotype of FREE1-RNAi transgenic plants.The obtained mutants are termed as suppressors of free1(sof).To date,229 putative sof mutants have been identified.Barely detecting of FREE1 protein with M3plants further identified 84 FREE1-related suppressors.Also145 mutants showing no reduction of FREE1 protein were termed as RNAi-related mutants.Through next-generation sequencing(NGS)of bulked DNA from F2mapping population of two RNAi-related sof mutants,FREE1-RNAi T-DNA inserted on chromosome 1 was identified and the causal mutation of putative sof mutant is being identified similarly.These FREE1-and RNAi-related sof mutants will be useful tools and resources for illustrating the underlying mechanisms of FREE1 function in intracellular trafficking and organelle biogenesis,as well as for uncovering the new components involved in the regulation of silencing pathways in plants.Qiong Zhao Caiji Gao PoShing Lee Lin Liu Shaofang Li Tangjin Hu Jinbo Shen Shuying Pan Hao Ye Yunru Chen Wenhan Cao Yong Cui Peng Zeng Sheng Yu Yangbin Gao Liang Chen Beixin Mo Xin Liu Shi Xiao Yunde Zhao Silin Zhong Xuemei Chen Liwen Jiang 2015Journal of Genetics and Genomics2015,42,6:4
8Influence of Ultra-Fine B_4C and Nano-SiO_2 on the Properties of Alumina-Graphite Refractories显示文摘The role of nano-SiO 2 and ultra-fine boron carbide on the properties of alumina-graphite materials was investigated. The study showed that the ultra-fine boron carbide added modified the microstructure of residual carbon and promoted the chemical bond between residual carbon from phenolic resin and flake graphite. The carbon white could strengthen the residual carbon from phenolic resin. These two additives improved the mechanical properties of AG refractories at both room temperature and high temperature, and thermal shock resistance was improved noticeably. When the two additives were doped together, carbon white could retard the evaporation of B 2O 3. 10Gut microbiota-derived metabolites contribute negatively to hindgut barrier function development at the early weaning goat model显示文摘Early weaning induces intestinal injury,leading to a series of long-term symptoms such as inflammation,malabsorption and diarrhea.In this study,we hypothesized that microbes and theirmetabolitesmodulate the host's inflammatory response to early weaning stress in a goatmodel.A total of 18 female Tibetan goat kids(n?9)wereweaned fromtheirmothers at 28 d(D28)and 60 d(D60)postpartum.D60 and D28 groupswere fed the same solid diet ad libitum fromweaning to 75 d of age.The colonic epithelium was subject to RNAsequencing,the caecal digesta metabolomics were assessed by liquid chromatographyetandem mass spectrometry(LC-MS/MS),and the caecal microbiota composition was analysed by 16S ribosomal RNA gene sequencing.We foundthatearlyweaningsubstantially increased the colonic pro-apoptotic gene expressionof B-cell lymphoma associated X(Bax),caspase-9,and caspase-3,and decreased the expression of zonula occludens-1(ZO-1)and claudin-1(P<0.01).In addition,a significant Bacteroides acidifaciens enrichmentwas observed in the hindgut of early-weaned goats(P<0.01),which negatively correlated with lysophosphatidylcholine products.Similarly,the chemokine signaling,IL-17 signaling,and peroxisome proliferatorsactivated receptor(PPAR)signaling pathways were upregulated in the colonic mucosa of the early-weaned goats.By applying caecal microbiota transplantation from goats to defaunated C57/6J mice,we confirmed that caecalmicrobiota of D28 goat kids increased the relative abundance of B.acidifaciens and significantly upregulated the genes of Bax,G proteinecoupled receptor(GPR)109A,GPR 43,fatty acid binding protein 6,nuclear receptor subfamily 1 group H member 3,angiotensin converting enzyme 2,and IL-6 expression(P<0.05),and decreased ZO-1,and claudin-1 protein expression in the mice jejunum and colon(P<0.001).These results proposed that the hindgut microbiota andmetabolites mediate the barrier functionweakening duringearlyweaning,and the relative abundance of B.acidifacienswas negatively correlatedwiththe hindgut barrier gene expression.This studydemonstrateshowweaningstress canaffectkeyhostemicrobe interaction regulators in the hindgut,in a lysophosphatidylcholine dependent and independent manner.Furthermore,based on our mice data,these results are transferable to other mammal species.Ke Zhang Yangbin Xu Yuxin Yang Mengmeng Guo Ting Zhang Bo Zong Shuhong Huang Langda Suo Baohua Ma Xiaolong Wang Yujiang Wu Daniel Brugger Yulin Chen 2022Animal Nutrition2022,,3:2

15Combustion study with synchrotron radiation single- photon ionization technique显示文摘Here we report a combustion endstation at National Synchrotron Radiation Laboratory (NSRL) and some primary experimental results. Synchrotron radiation can provide the tunable vacuum ultraviolet (VUV) photon with the high intensity and the good collimation. VUV photoioni-zation is a single-photon ionization process. Combined with molecular-beam mass spectrometry (MBMS), the VUV single-photon ionization can be applied to detect the combustion products, especially the intermediates and free radicals produced from combustion process. 16An Accurate Common-Mode Current Prediction Method for Transformerless Photovoltaic Inverters Based on Graph Theory显示文摘The common-mode current is an important indicator with transformerless photovoltaic inverters.However,up to now,there is not an accurate method to predict common-mode current in the inverter design process,resulting from inappropriate device selection or exceeded the expected common-mode current.In order to solve this problem,this paper proposes an accurate common-mode current prediction method based on graph theory for transformerless photovoltaic inverters.In this paper,the mathematic model of the common-mode current is derived using graph theory analysis method in the full-bridge topology,and it is used to predict common-mode current.The validity and correctness of the proposed prediction method are validated by simulation and experiment.The oscillation frequency and amplitude can be predicted by the proposed common-mode prediction method,whereas the traditional common-mode analysis method cannot.This paper provides a novel way to predict and analyze common-mode current in the transformerless photovoltaic inverters.Yangbin Zeng Hong Li Bo Zhang Zhichang Yang Trillion Q.Zheng 2017Chinese Journal of Electrical Engineering2017,3,3:1

20Generalization-based discovery of spatial association rules with linguistic cloud models显示文摘Extraction of interesting and general spatial association rules from large spatial databases is an important task in the development of spatial database systems. In this paper, we investigate the generalization-based knowledge discovery mechanism that integrates attribute-oriented induction on nonspatial data and spatial merging and generalization on spatial data. Furthermore, we present linguistic cloud models for knowledge representation and uncertainty handling to enhance current generalization-based method. With these models, spatial and nonspatial attribute values are well generalized at higher-concept levels, allowing discovery of strong spatial association rules.
