1Plastome-wide comparison reveals new SNV resources for the authentication of Dendrobium huoshanense and its corresponding medicinal slice(Huoshan Fengdou)显示文摘Dendrobium species and their corresponding medicinal slices have been extensively used as traditional Chinese medicine(TCM) in many Asian countries. However, it is extremely difficult to identify Dendrobium species based on their morphological and chemical features. In this study, the plastomes of D. huoshanense were used as a model system to investigate the hypothesis that plastomic mutational hotspot regions could provide a useful single nucleotide variants(SNVs) resource for authentication studies. We surveyed the plastomes of 17 Dendrobium species, including the newly sequenced plastome of D. huoshanense. A total of 19 SNVs that could be used for the authentication of D. huoshanense were detected. On the basis of this comprehensive comparison, we identified the four most informative hotspot regions in the Dendrobium plastome that encompass ccsA to ndhF, matK to 3′trnG, rpoB to psbD, and trnT to rbcL. Furthermore, to established a simple and accurate method for the authentication of D. huoshanense and its medicinal slices, a total of 127 samples from 20 Dendrobium species including their corresponding medicinal slices(Fengdous) were used in this study. 2Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing.Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang 2021Cellular & Molecular Immunology2021,18,9:5
3Differentiation of smooth muscle progenitor cells in peripheral blood and its application in tissue engineered blood vessels显示文摘Background: A major shortcoming in tissue engineered blood vessels (TEBVs) is the lack of healthy and easily attainable smooth muscle cells (SMCs). Smooth muscle progenitor cells (SPCs), especially from peripheral blood, may offer an alternative cell source for tissue engineering involving a less invasive harvesting technique. Methods: SPCs were isolated from 5-ml fresh rat peripheral blood by density-gradient centrifugation and cultured for 3 weeks in endothelial growth medium-2-MV (EGM-2-MV) medium containing platelet-derived growth factor-BB (PDGF BB). Before seeded on the synthesized scaffold, SPC-derived smooth muscle outgrowth cell (SOC) phenotypes were assessed by immuno-fluorescent staining, Western blot analysis, and reverse transcription polymerase chain reaction (RT-PCR). The cells were seeded onto the silk fibroin-modified poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (SF-PHBHHx) scaffolds by 6×104 cells/cm2 and cultured under the static con- dition for 3 weeks. The growth and proliferation of the seeded cells on the scaffold were analyzed by 3-(4,5-dimethylthiazol-2-yl)- diphenyltetrazolium bromide (MTT) assay, scanning electron microscope (SEM), and 4,6-diamidino-2-phenylindole (DAPI) staining. Results: SOCs displayed specific 'hill and valley' morphology, expressed the specific markers of the SMC lineage: smooth muscle (SM) α-actin, calponin and smooth muscle myosin heavy chain (SM MHC) at protein and messenger ribonucleic acid (mRNA) levels. RT-PCR results demonstrate that SOCs also expressed smooth muscle protein 22α (SM22α), a contractile protein, and extracellular matrix components elastin and matrix Gla protein (MGP), as well as vascular endothelial growth factor (VEGF). After seeded on the SF-PHBHHx scaffold, the cells showed excellent metabolic activity and proliferation. Conclusion: SPCs isolated from peripheral blood can be differentiated into the SMCs in vitro and have an impressive growth potential in the biodegradable synthesized scaffold. 4Fast-Suppressor Screening for New Components in Protein Trafficking,Organelle Biogenesis and Silencing Pathway in Arabidopsis thaliana Using DEX-Inducible FREE1-RNAi Plants显示文摘Membrane trafficking is essential for plant growth and responses to external signals.The plant unique FYVE domain-containing protein FREE1 is a component of the ESCRT complex(endosomal sorting complex required for transport).FREE1 plays multiple roles in regulating protein trafficking and organelle biogenesis including the formation of intraluminal vesicles of multivesicular body(MVB),vacuolar protein transport and vacuole biogenesis,and autophagic degradation.FREE1 knockout plants show defective MVB formation,abnormal vacuolar transport,fragmented vacuoles,accumulated autophagosomes,and seedling lethality.To further uncover the underlying mechanisms of FREE1 function in plants,we performed a forward genetic screen for mutants that suppressed the seedling lethal phenotype of FREE1-RNAi transgenic plants.The obtained mutants are termed as suppressors of free1(sof).To date,229 putative sof mutants have been identified.Barely detecting of FREE1 protein with M3plants further identified 84 FREE1-related suppressors.Also145 mutants showing no reduction of FREE1 protein were termed as RNAi-related mutants.Through next-generation sequencing(NGS)of bulked DNA from F2mapping population of two RNAi-related sof mutants,FREE1-RNAi T-DNA inserted on chromosome 1 was identified and the causal mutation of putative sof mutant is being identified similarly.These FREE1-and RNAi-related sof mutants will be useful tools and resources for illustrating the underlying mechanisms of FREE1 function in intracellular trafficking and organelle biogenesis,as well as for uncovering the new components involved in the regulation of silencing pathways in plants.Qiong Zhao Caiji Gao PoShing Lee Lin Liu Shaofang Li Tangjin Hu Jinbo Shen Shuying Pan Hao Ye Yunru Chen Wenhan Cao Yong Cui Peng Zeng Sheng Yu Yangbin Gao Liang Chen Beixin Mo Xin Liu Shi Xiao Yunde Zhao Silin Zhong Xuemei Chen Liwen Jiang 2015Journal of Genetics and Genomics2015,42,6:4

6On the critical particle size of soil with clogging potential in shield tunneling显示文摘Shield tunneling is easily obstructed by clogging in clayey strata with small soil particles.However,soil clogging rarely occurs in strata with coarse-grained soils.Theoretically,a critical particle size of soils should exist,below which there is a high risk of soil clogging in shield tunneling.To determine the critical particle size,a series of laboratory tests was carried out with a large-scale rotary shear apparatus to measure the tangential adhesion strength of soils with different particle sizes and water contents.It was found that the tangential adhesion strength at the soilesteel interface gradually increased linearly with applied normal pressure.When the particle size of the soil specimen was less than 0.15 mm,the interfacial adhesion force first increased and then decreased as the water content gradually increased;otherwise,the soil specimens did not manifest any interfacial adhesion force.The amount of soil mass adhering to the steel disc was positively correlated with the interfacial adhesion force,thus the interfacial adhesion force was adopted to characterize the soil clogging risk in shield tunneling.The critical particle size of soils causing clogging was determined to be 0.15 mm.Finally,the generation mechanism of interfacial adhesion force was explored for soils with different particle sizes to explain the critical particle size of soil with clogging risk in shield tunneling.Shuying Wang Zihao Zhou Pengfei Liu Zhao Yang Qiujing Pan Weizhong Chen 2023Journal of Rock Mechanics and Geotechnical Engineering2023,15,2:3

8Rnf20 deficiency in adipocyte impairs adipose tissue development and thermogenesis显示文摘RNF20,an E3 ligase critical for monoubiquitination of histone H2B at lysine 120(H2Bub),has been implicated in the regulation of various cellar processes;however,its physiological roles in adipocytes remain poorly characterized.Here,we report that the adipocyte-specific knockout of Rnf20(ASKO)in mice led to progressive fat toss,organomegaly and hyperinsulinemia.Despite signs of hyperinsulinemia,normal insulin sensitivity and improved glucose tolerance were observed in the young and aged CD-fed ASKO mice.In addition,high-fat diet-fed ASKO mice developed severe liver steatosis.Moreover,we observed that the ASKO mice were extremely sensitive to a cold environment due to decreased expression levels of brown adipose tissue(BAT)selective genes,including uncoupling protein 1(l/cpf),and impaired mitochondrial functions.Significantly decreased levels of peroxisome proliterator-activated receptor gamma(Ppary)were observed in the gonadal white adipose tissues(gWAT)from the ASKO mice,suggesting that Rnf20 regulates adipogenesis,at least in part,through Ppary.Rosiglitazone-treated ASKO mice exhibited increased fat mass compared to that of the non-treated ASKO mice.Collectively,our results illustrate the critical role of RNF20 in control of white and brown adipose tissue development and physiological function.Xiaojuan Liang Cong Tao Jianfei Pan Lilan Zhang Lulu Liu Ying Zhao Yiping Fan Chunwei Cao Jiali Liu Jin Zhang Sin Man Lam Guanghou Shui Wanzhu Jin Wei Li Jianguo Zhao Kui Li Yanfang Wang 2021Protein & Cell2021,12,6:1

13Plant metabolomics for studying the effect of two insecticides on comprehensive constituents of Lonicerae Japonicae Flos显示文摘Pesticides'overuse and misuse have been reported to induce ingredient variations in herbal medicine,which is now gaining attention in the medicinal field as a form of alternative medicine.To date,available studies on pesticide-induced ingredient variations of herbal medicine are limited only on a few compounds and remain most others unexamined.In this study,a plant metabolomics-based strategy was performed to systematically explore the effects of two frequently used insecticides on the comprehensive constituents of Lonicerae Japonicae Flos(LJF),the flower buds of Lonicera japonica Thunb..Field trials were designed on a cultivating plot of L.japonica with controls and treatments of imidacloprid(IMI)and compound flonicamid and acetamiprid(CFA).Unbiased metabolite profiling was conducted by ultra-high performance liquid chromatography/quadrupole-Orbitrap mass spectrometer.After data pretreatment by automatic extraction and screening,a data matrix of metabolite features was submitted for statistical analyses.Consequently,29 metabolic markers,including chlorogenic acids,iridoids and organic acid-glucosides were obtained and characterized.The relative quantitative assay was subsequently performed to monitor their variations across flowering developments.This is the first study that systematically explored the insecticide-induced metabolite variations of LJF while taking into account the inherent variability of flowering development.The results were beneficial for holistic quality assessment of LJF and significant for guiding scientific use of pesticides in the large-scale cultivation.PAN Hui-Qin ZHOU Heng MIAO Shui GUO De-An ZHANG Xiao-Li HU Qing MAO Xiu-Hong JI Shen 2021Chinese Journal of Natural Medicines2021,19,1:1

15Calculating the Number of Pregnant Women Receiving Standardized Services for PMTCT of HIV in Liangshan Prefecture,Based on the Equivalent Method显示文摘Objective To calculate the number of pregnant women who receive standardized prevention of mother-to-child transmission(PMTCT)services for HIV annually.Methods HIV-positive pregnant women in six counties of Liangshan Prefecture in 2017 were selected as study subjects.The entire process,from when the subjects first received the PMTCT of HIV services to the end,was divided into four stages,which were further divided into 25 phases.The equivalent coefficient was used to indicate the weight of workload in each phase.Seven experts were invited to score the equivalent coefficient;the number of pregnant women who received standardized services to prevent the transmission of HIV was calculated.Results A total of 663 HIV-positive pregnant women were registered in six Liangshan Prefecture counties in 2017.This figure was converted into 7,780 person-months devoted to HIV-positive pregnant women,with 260 person-months(3.34%)spent on the first antenatal care,1,510 person-months(19.41%)during pregnancy,378 person-months(4.86%)on delivery,and 5,632 person-months(72.39%)on post-partum period.The equivalent coefficient calculation showed that 314 HIV-positive pregnant women received standardized PMTCT services.Conclusion The number of pregnant women receiving standardized services for the PMTCT of HIV can be calculated accurately using the equivalent method to identify the gap between the level of PMTCT of HIV intervention services needed and the actual workload.QU Shui Ling WANG Ai Ling PAN Xiao Ping WANG Xiao Yan LUO Hui Ming ZHANG Tong 2020Biomedical and Environmental Sciences2020,33,10:1
