| 2 | Induced earthquakes in the development of unconventional energy resources显示文摘It has long been known that human activities such as waste fluid disposal and reservoir impoundment may cause earthquakes. Recently, anthropogenic activities to tackle the increasing energy demand and to address climate change issues are also reported to induce earthquakes. These activities have a common attribute in that fluids are injected and extracted underground and induce spatiotemporal changes of pore pressure and stress, which may cause slip on faults. Induced earthquakes not only pose significant impacts on seismic hazard assessment and preparation, but also raise the question to the society as how to balance the economic needs of resources development and the public's concerns about potential environmental impacts. Here we review the observations of fluid-injection/extraction induced earthquakes, ground deformation associated with these activities, and their physical mechanisms. [Methods]With Rhodiola crenulata as raw material,ethanol as extractant,ultrasonic extraction as the extraction method,the single factor method was first used for preliminary investigation of effect of ethanol volume fraction,solid-liquid ratio,extraction temperature and extraction time on the flavonoids extraction rate,and then the Box-Behnken response surface method was used to optimize the extraction process of total flavonoids in R. crenulata. [Results] The optimal extraction conditions: ethanol concentration of 72%;solid-liquid ratio of 1∶ 43; extraction temperature of 66℃; extraction time of 50 min. Under these conditions,the extraction rate of total flavonoids from R. crenulata was 2. 591%. | 6 | Protective effect of Xuebijing injection on myocardial injury in patients with sepsis: a randomized clinical trial显示文摘OBJECTIVE: To investigate the protective effect and possible mechanism of Xuebijing Injection on myocardial injury in patients with sepsis, and to evaluate its prognostic implications.METHODS: Patients with septic myocardial injury were recruited, and were randomly divided into two groups: treatment group and control group. All patients in two groups received conventional cluster treatment, the patients in treatment group additional received Xuebijing injection dissolved in0.9% sodium chloride injection, and the patients in control group received the same amount of 0.9%sodium chloride injection. At the beginning of treatment and 3, 7 and 10-day after treatment, lab-oratory indicators of cardiac troponin Ⅰ(cTnI),N-terminal pro B-type natriuretic peptide(NT-pro BNP) and procalcitonin(PCT) were respectively tested in venous blood. The patient's length of stay in Intensive Care Unit(ICU) and the mortality in 28 days were recorded.RESULTS: At 3, 7 and 10-day after treatment, the improvements of c Tn I, NT-pro BNP and PCT in treatment group were better than those in control group, and the differences were statistically significant(P < 0.05). The mortality of treatment group in28 days was not significantly different from that of control group(P > 0.05). | 7 | Effects of feeding untreated, pasteurized and acidified waste milk and bunk tank milk on the performance, serum metabolic profiles, immunity, and intestinal development in Holstein calves显示文摘Background: The present experiment was performed to assess the effects of different sources of milk on the growth performance, serum metabolism, immunity, and intestinal development of calves. Eighty-four Holstein male neonatal calves were assigned to one of the following four treatment groups: those that received bunk tank milk(BTM), untreated waste milk(UWM), pasteurized waste milk(PWM), and acidified waste milk(AWM) for 21 d.Results: Calves in the BTM and AWM groups consumed more starter(P < 0.05) than those in the UWM group.Average daily gain in the UWM group was the highest(P < 0.05). Calves exhibited the highest(P < 0.05) serum total protein, albumin, total cholesterol, high density lipoprotein, triglycerides, growth hormone, immunoglobulin(Ig) A and Ig M concentrations in the UWM group, highest malondialdehyde and tumor necrosis factor-α in the PWM group(P < 0.05), and highest glutathione peroxidase and Ig G in the BTM group(P < 0.05). The jejunum and ileum of the calves in all treatments presented a slight inflammatory response. The jejunal inflammation scores were higher(P < 0.05) in the UWM and AWM groups than the BTM group; the ileal inflammation scores increased more(P < 0.05) in the AWM group than the BTM group. Jejunal immunohistochemical scores(IHS) were higher(P < 0.05) in the PWM and AWM groups than the BTM group. Compared to the other three groups, calves feeding on BTM had lower(P < 0.05) ileal IHS. Jejunal interleukin(IL)-1β, IL-8, and IL-10 mRNA expression in the UWM group was the highest(P < 0.05). Calves fed AWM increased(P < 0.05) mRNA expression of IL-8 and toll like receptor 4(TLR-4) in the jejunum and IL-8, IL-1β, IL-6, IL-8, and IL-10 in the mesenteric lymph nodes.Conclusions: Overall, bunk tank milk is the best choice for calf raising compared to waste milk. | 8 | An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. | 9 | Recombinant rabies virus expressing IL-15 enhances immunogenicity through promoting the activation of dendritic cells in mice显示文摘Rabies remains a public health threat that kills approximately 59,000 people worldwide each year,most of which are from the developing countries of Africa and Asia where dog rabies are endemic.Therefore, developing an affordable and efficacious vaccine is crucial for rabies control in these countries. Interleukin(IL)-15, an immunoregulatory cytokine, is a pluripotent molecule with therapeutic potential, which targets many cell types and links the innate and adaptive immune system. In this study, IL-15 gene was cloned and inserted into the genome of a recombinant rabies virus(RABV) strain LBNSE(designated as LBNSE-IL15), and the effect of over-expression of IL-15 on the immunogenicity of RABV was investigated. It was found that mice vaccinated with LBNSEIL15 could induce significantly higher level of virus-neutralizing antibody(VNA) than those immunized with LBNSE, resulting in the higher protection after challenge. Further investigation was performed to find out the possible role of IL-15 plays in the process of antibody induction, and it was found that LBNSE-IL15 could enhance the maturation of dendritic cells(DCs) in immunized mice. Furthermore, the mice immunized with LBNSE-IL15 could promote the T_(FH) cells differentiation and the generation of germinal center B cells and plasma cells. | 11 | Arterial stiffness and stroke:de-stiffening strategy,a therapeutic target for stroke显示文摘Stroke is the second leading cause of mortality and morbidity worldwide.Early intervention is of great importance in reducing disease burden.Since the conventional risk factors cannot fully account for the pathogenesis of stroke,it is extremely important to detect useful biomarkers of the vascular disorder for appropriate intervention.Arterial stiffness,a newly recognised reliable feature of arterial structure and function,is demonstrated to be associated with stroke onset and serve as an independent predictor of stroke incidence and poststroke functional outcomes.In this review article,different measurements of arterial stiffness,especially pressure wave velocity,were discussed.We explained the association between arterial stiffness and stroke occurrence by discussing the secondary haemodynamic changes.We reviewed clinical data that support the prediction role of arterial stiffness on stroke.Despite the lack of long-term randomised double-blind controlled therapeutic trials,it is high potential to reduce stroke prevalence through a significant reduction of arterial stiffness(which is called de-stiffening therapy).Pharmacological interventions or lifestyle modification that can influence blood pressure,arterial function or structure in either the short or long term are promising de-stiffening therapies.Here,we summarised different de-stiffening strategies including antihypertension drugs,antihyperlipidaemic agents,chemicals that target arterial remodelling and exercise training.Large and well-designed clinical trials on de-stiffening strategy are needed to testify the prevention effect for stroke.Novel techniques such as modern microscopic imaging and reliable animal models would facilitate the mechanistic analyses in pathophysiology,pharmacology and therapeutics. | Yajing Chen Fanxia Shen Jianrong Liu Guo-Yuan Yang | 2017 | Stroke & Vascular Neurology2017,2,2: | 3 |

| 15 | Tet2 Regulates Osteoclast Differentiation by Interacting with Runx1 and Maintaining Genomic 5-Hydroxymethylcytosine(5hmC)显示文摘As a dioxygenase, Ten-Eleven Translocation 2(TET2) catalyzes subsequent steps of 5-methylcytosine(5 mC) oxidation. TET2 plays a critical role in the self-renewal, proliferation,and differentiation of hematopoietic stem cells, but its impact on mature hematopoietic cells is not well-characterized. Here we show that Tet2 plays an essential role in osteoclastogenesis. Deletion of Tet2 impairs the differentiation of osteoclast precursor cells(macrophages) and their maturation into bone-resorbing osteoclasts in vitro. Furthermore, Tet2^(-/-) mice exhibit mild osteopetrosis, accompanied by decreased number of osteoclasts in vivo. Tet2 loss in macrophages results in the altered expression of a set of genes implicated in osteoclast differentiation, such as Cebpa, Mafb, and Nfkbiz. Tet2 deletion also leads to a genome-wide alteration in the level of 5-hydroxymethylcytosine(5 hmC) and altered expression of a specific subset of macrophage genes associated with osteoclast differentiation. Furthermore, Tet2 interacts with Runx1 and negatively modulates its transcriptional activity. Our studies demonstrate a novel molecular mechanism controlling osteoclast differentiation and function by Tet2, that is, through interactions with Runx1 and the maintenance of genomic 5 hmC. | 17 | Ascorbate peroxidase 1 confers resistance to southern corn leaf blight in maize显示文摘Southern corn leaf blight (SCLB), caused by Bipolarismaydis, is one of the most devastatingdiseases affecting maize production. However,only one SLCB resistance gene, conferring partialresistance, is currently known, underscoring theimportance of isolating new SCLB resistancerelatedgenes. Here, we performed a comparativeproteomic analysis and identified 258 proteinsshowing differential abundance during the maizeresponse to B. maydis. These proteins included anascorbate peroxidase (Zea mays ascorbate peroxidase1 (ZmAPX1)) encoded by a gene locatedwithin the mapping interval of a previously identifiedquantitative trait locus associated with SCLBresistance. ZmAPX1 overexpression resulted inlower H_(2)O_(2) accumulation and enhanced resistanceagainst B. maydis. Jasmonic acid (JA)contents and transcript levels for JA biosynthesisand responsive genes increased in ZmAPX1-overexpressing plants infected with B. maydis,whereas Zmapx1 mutants showed the oppositeeffects. We further determined that low levels of H_(2)O_(2) are accompanied by an accumulation of JAthat enhances SCLB resistance. These resultsdemonstrate that ZmAPX1 positively regulatesSCLB resistance by decreasing H_(2)O_(2) accumulationand activating the JA-mediated defensesignaling pathway. This study identified ZmAPX1as a potentially useful gene for increasing SCLBresistance. | 19 | Generation of isogenic single and multiplex gene knockout mice by base editing-induced STOP显示文摘Although CRISPR/Cas9 has been widely used to generate knockout mice, two major limitations remain:the founders usually carry a mixture of genotypes, and mosaicism harboring multiple genotypes.Therefore, it takes a long time to get homozygous mutants. Recently developed base editing(BE) system,which introduces C-to-T conversion without double strand DNA cleavage, has been used to introduce artificial stop codons(i-STOP) to prematurely terminate translation, providing a cleaner strategy for genome engineering. Using this strategy, we generated CD160 KO and VISTA/CD160 double KO mice by microinjection of a single sg RNA targeting CD160 and a mixture of sg RNAs targeting VISTA and CD160,respectively. The BE system induced STOP efficiently in mouse embryos and consequently in founder mice without detectable off-target. Most interestingly, the majority of the mutants harbor same genetic modifications, indicating we generated isogenic single and multiplex gene mutant mice by BE-induced STOP.
