1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. 3Bone marrow mesenchymal stem cells with Nogo-66 receptor gene silencing for repair of spinal cord injury显示文摘We hypothesized that RNA interference to silence Nogo-66 receptor gene expression in bone marrow mesenchymal stem cells before transplantation might further improve neurological function in rats with spinal cord transection injury. After 2 weeks, the number of neurons and BrdU-positive cells in the Nogo-66 receptor gene silencing group was higher than in the bone marrow mesenchymal stem cell group, and significantly greater compared with the model group. After 4 weeks, behavioral performance was significantly enhanced in the model group. After 8 weeks, the number of horseradish peroxidase-labeled nerve fibers was higher in the Nogo-66 receptor gene silencing group than in the bone marrow mesenchymal stem cell group, and significantly higher than in the model group. 5Induced electric fields and plasmonic interactions between a metallic nanotube and a thin metallic film显示文摘We have numerically simulated the induced electric fields and the plasmonic interactions of a metallic nanotube near a thin metallic film. Our study shows that the energies and intensities of the plasmon resonances depend strongly on the aspect ratio (the ratio of the inner to outer radius) of the nanotube as well as the separation between the center of the nanotube and the upper surface of the metallic film and the thickness of the film. The enhancement of the induced electric field of this system reaches as high as 10 4 orders of magnitude and its field distribution is characterized by waveguide-mode resonance. 6Towards smart optical focusing:deep learningempowered dynamic wavefront shaping through nonstationary scattering media显示文摘Optical focusing through scattering media is of great significance yet challenging in lots of scenarios,including biomedical imaging,optical communication,cybersecurity,three-dimensional displays,etc.Wavefront shaping is a promising approach to solve this problem,but most implementations thus far have only dealt with static media,which,however,deviates from realistic applications.Herein,we put forward a deep learning-empowered adaptive framework,which is specifically implemented by a proposed Timely-Focusing-Optical-Transformation-Net(TFOTNet),and it effectively tackles the grand challenge of real-time light focusing and refocusing through time-variant media without complicated computation.The introduction of recursive fine-tuning allows timely focusing recovery,and the adaptive adjustment of hyperparameters of TFOTNet on the basis of medium changing speed efficiently handles the spatiotemporal non-stationarity of the medium.Simulation and experimental results demonstrate that the adaptive recursive algorithm with the proposed network significantly improves light focusing and tracking performance over traditional methods,permitting rapid recovery of an optical focus from degradation.It is believed that the proposed deep learning-empowered framework delivers a promising platform towards smart optical focusing implementations requiring dynamic wavefront control.YUNQI Luo SUxIA YAN HUANHAO LI PUXIANG LAI YUANJIN ZHENG 2021Photonics Research2021,9,8:3
7Effects of the local environment on plasmonic coupling of metallic nanotube arrays显示文摘We have theoretically investigated the effect of the dielectric core and the dielectric embedding medium separately on the transmission spectra and plasmonic properties of coupled metallic nanotube arrays.It is found that the plasmonic properties of the nanotube arrays are strongly influenced by the presence of the dielectric which induces additional screening charges.We show that instead of one single photonic bandgap for the hollow nanotube arrays placed in air,an additional photonic bandgap arises from the presence of dielectric media in the transmission spectra.Based on the localized nature of the electric field distributions,we also clearly show the presence of the local plasmonic resonant modes that originate from multipolar plasmon polaritons in the cross section of these nanotube arrays,and that a large amount of opposing surface charges are built up in the gap between adjacent nanotubes.LI HongJian FU ShaoLi XIE SuXia XU HaiQing ZHOU Xin WU JinJun 2011Science China(Physics,Mechanics & Astronomy)2011,54,1:2
8Repurposing econazole as a pharmacological autophagy inhibitor to treat pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma(PDAC)is characterized by the highest mortality among carcinomas.The pathogenesis of PDAC requires elevated autophagy,inhibition of which using hydroxychloroquine has shown promise.However,current realization is impeded by its suboptimal use and unpredictable toxicity.Attempts to identify novel autophagy-modulating agents from already approved drugs offer a rapid and accessible approach.Here,using a patient-derived organoid model,we performed a comparative analysis of therapeutic responses among various antimalarial/fungal/parasitic/viral agents,through which econazole(ECON),an antifungal compound,emerged as the top candidate.Further testing in cell-line and xenograft models of PDAC validated this activity,which occurred as a direct consequence of dysfunctional autophagy.More specifically,ECON boosted autophagy initiation but blocked lysosome biogenesis.RNA sequencing analysis revealed that this autophagic induction was largely attributed to the altered expression of activation transcription factor 3(ATF3).Increased nuclear import of ATF3 and its transcriptional repression of inhibitor of differentiation-1(ID-1)led to inactivation of the AKT/mammalian target of rapamycin(m TOR)pathway,thus giving rise to autophagosome accumulation in PDAC cells.The magnitude of the increase in autophagosomes was sufficient to elicit ER stress-mediated apoptosis.Furthermore,ECON,as an autophagy inhibitor,exhibited synergistic effects with trametinib on PDAC.This study provides direct preclinical and experimental evidence for the therapeutic efficacy of ECON in PDAC treatment and reveals a mechanism whereby ECON inhibits PDAC growth.Ningna Weng Siyuan Qin Jiayang Liu Xing Huang Jingwen Jiang Li Zhou Zhe Zhang Na Xie Kui Wang Ping Jin Maochao Luo Liyuan Peng Edouard C.Nice Ajay Goel Suxia Han Canhua Huang Qing Zhu 2022Acta Pharmaceutica Sinica B2022,12,7:2

11Strong optical transmission through the ellipsoid metal-film nanohole arrays显示文摘The transmission characteristics of a metallic film with subwavelength ellipsoid nanohole arrays are investigated by using the three-dimensional finite-difference time-domain (3D-FDTD) method. The extraordinary transmission is attributed to the collaboration of localized waveguide resonance and surface plasmon resonance. The influences of the lattice constant and the hole shape on the transmission are studied. 12The expression of Fas, FasL and their biological behavior in human cervical carcinoma显示文摘Objective: To investigate the relationship between the expression of Fas and Fas ligand (FasL) and its biological behavior in human cervix carcinoma. Methods: Immunohistochemisty technique was used to detect the expression of Fas and FasL in 47 cases of cervical carcinoma, 16 cases of cervical interaepithelial neoplasia, 10 cases of chronic cervicitis and 10 cases of normal cervix. TUNEL technique was used to observe the apoptic cells in 47 cases of cervical carcinoma. Retrospective study was carried out to find the relationship between the expression of Fas and FasL and cell apoptosis, clinical stage, pathological classification, lymph node metastasis, prognosis and age. Results: The expression of Fas and FasL was significantly different in different cervix (P < 0.01), and also related to the degree of differentiation, lymph node metastasis and prognosis (P < 0.05). But had no relation with clinical stage or age (P > 0.05); Cervix carcinoma cells apoptosis in different pathological classification appeared negative relation(Rs=-0.35, P < 0.05). Cervix carcinoma cell apoptosis was significantly higher in Fas-positive and FasL-positive than that in Fas-negative and FasL-negative (P < 0.05). By retrospective investigation, Fas-negative and FasL-positive were related to poor prognoses of the patients with cervical carcinoma (P < 0.05). Conclusion: The development of apoptosis in cervix carcinoma has a promoting regulation function in Fas and FasL expression. Gene treatment can alter apoptosis abnormality, thus induce apoptosis in cancerous cell expressing Fas and FasL. 17Study on the Bonding Performance of the Moso Bamboo Dowel Welded to a Poplar Substrate Joint by High-Speed Rotation显示文摘The wood friction welding technique with its high bonding strength,low cost,high efficiency,and without any adhesive has been increasing concern in China.Moso bamboo(Phyllostachys pubescens)and poplar(Populus sp.)are widely planted and used in the furniture industry,interior decoration,and wood structure construction in China.The aim of this work was to investigate the bonding performance of moso bamboo dowel rotation welded joints with different dowel/receiving hole diameter ratios.The results indicated that the ratio of dowel/receiving hole diameter was an important parameter that influenced the welding performance.The bonding strength of the bamboo-to-poplar welded joints at the optimal ratio of 10/7 was as high as 7.50 MPa,which was higher than that of the beech(Fagus sylvatica,L.),schima(Schima superba)dowels and PVAc glued joints.The temperature measurement results showed a peak temperature of bamboo dowel welding as high as 350–360°C.Some differences in the temperature curves between each dowel/hole diameter ratio group were observed at the three different hole depths,such as the friction time,peak temperatures,and stabilization time at the maximum temperature,which could explain the differences in welding strengths between different ratios.The SEM results showed the temperature-induced softening,melting and flowing of cell-interconnected polymer material in the wood and bamboo structure.In addition,the bamboo fibers(mainly vascular bundles)were wrapped to form a dense continuous bonding layer,similar to the reinforced concrete,thus producing a good bonding effect.The Fourier transform-infrared spectroscopy(FT-IR)analyses showed that the high temperature resulted in the increase of the lignin relative content due to the degradation reaction of cellulose in the welding zone,which improved the bonding properties.Suxia Li Haiyang Zhang Biqing Shu Liangsong Cheng Zehui Ju Xiaoning Lu 2021Journal of Renewable Materials2021,9,7:1
18Study on the Tangential Tensile Mechanical Properties of Moso Bamboo显示文摘In this work,we used tensile tests to analyze the tangential failure forms of raw bamboo and determine a relationship between tangential tensile strength,elastic modulus,position,density,and moisture content.We found that the tangential mechanical properties of the culm wall were mainly dependent on the mechanical properties of the basic structure of the thin wall.Formulas for calculating the tangential tensile strength of moso bamboo and adjusting the moisture content were also determined.The tangential tensile strength and the tangential tensile modulus of elasticity(TTMOE)followed:outer>middle>inner,and diaphragm>bamboo node>culm wall.Below the fiber saturation point,the tangential tensile strength and TTMOE values of the bamboo gradually decreased with increasing moisture content.When the moisture content was 15%,the tangential tensile strengths of the inner,middle,outer,culm wall,bamboo node,and diaphragm samples of the five-year-old moso bamboo were 3.17,3.29,3.31,3.24,3.67,and 8.85 MPa,respectively.Furthermore,their TTMOE values were 215.09,227.98,238.45,224.04,267.21,and 559.27 MPa,respectively.Hence,this study provides a theoretical basis for future research on bamboo cracking.Biqing Shu Lu Hong Suxia Li Yupeng Tao Jianxin Cui Naiqiang Fu Junbao Yu Chen Li Xiaoning Lu 2022Journal of Renewable Materials2022,10,8:1
