| 1 | Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART. | Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu | 2017 | Journal of Genetics and Genomics2017,44,10: | 5 |
| 2 | Clinical and radiological features of medullary infarction caused by spontaneous vertebral artery dissection显示文摘Background and purpose Medullary infarction(MI)caused by spontaneous vertebral artery dissection(sVAD)is an important type of stroke.It is important to distinguish sVAD from other causes of stroke since the treatment strategies and prognosis were different between them.In this study,we aimed to explore the clinical and radiological features of MI in patients with acute MI caused by sVAD.Methods Patients with acute MI caused by sVAD and non-sVAD in a single tertiary hospital were enrolled from 2010 to 2020.Epidemiologic,clinical and image features were collected and analysed.MI lesions were categorised into three levels rostrocaudally and four arterial groups:anteromedial,anterolateral,lateral and posterior.Results A total of 128 patients with MI were enrolled with 47 cases of sVAD and 81 cases of non-sVAD.Patients with sVAD were younger than those with non-sVAD(med 44 years old vs 58 years old).The sVAD group was less likely to have hypertension(44.68%vs 67.90%;p=0.010)and diabetes(19.15%vs 45.69%;p=0.003),but more likely to have non-sudden onset(27.66%vs 9.87%,p=0.009),minor neck injury(19.15%vs 1.23%;p=0.001)and headache(46.81%vs 7.41%;p=0.000).Vertically,sVAD became more common in caudal medulla than in rostral medulla.Horizontally,the sVAD group was more likely to have lateral MI(91.48%vs 2.96%,p=0.000).In multivariable logistic regression analysis,age,non-sudden onset and headache were independently associated with sVAD with ORs of 0.935(95%CI 0.892 to 0.981,p=0.006),3.507(95%CI 1.060 to 11.599,p=0.040)and 5.426(95%CI 1.673 to 17.599,p=0.005).Conclusion sVAD was not uncommon in patients with MI,especially in patients with lateral MI.Young patients with headache and non-sudden onset should remind clinician the possibility of sVAD. | Chun Yu Zhu Zhu Siying Li Yi Xu Wei Yan Xiaocui Kang Yao Li Qiang Dong Weijun Tang Xiang Han | 2022 | Stroke & Vascular Neurology2022,7,3: | 3 |
| 3 | Inhibition of p53 and/or AKT as a new therapeutic approach specifically targeting ALT cancers显示文摘While the majority of all human cancers court teract telomere shortening by expressing telomerase,-15%of all cancers maintain telomere length by a telomerase?independent mechanism known as alternative lengthening of telomeres(ALT).Here,we show that high load of intrinsic DNA damage is present in ALT cancer cells,leading to apoptosis stress by activating p53-independent,but JNK/c-IVIyc-dependent apoptotic pathway.Notably,ALT cells expressing wild-type p53 show much lower apoptosis than p53-deficient ALT cells.Mechanistically,we find that intrinsic DNA damage in ALT cells induces low level of p53 that is insufficient to initiate the transcription of apoptosis-related genes,but is sufficient to stimulate the expression of key components of mTORC2(mTOR and Rictor),which in turn leads to phosphorylation of AKT.Activated AKT(p-AKT)thereby stimulates downstream anti-apoptotic events.Therefore,p53 and AKT are the key factors that suppress sponta?neous apoptosis in ALT cells.Indeed,inhibition of p53 or AKT selectively induces rapid death of ALT cells in vitro,and p53 inhibitor severely suppresses the growth of ALT-cell xenograft tumors in mice.These findings reveal a previously unrecognized function of p53 in antiapoptosis and identify that the inhibition of p53 or AKT has a potential as therapeutics for specifically targeting ALT cancers. | Yuanlong Ge Shu Wu Zepeng Zhang Xiaocui Li Feng Li Siyu Yan Haiying Liu Junjiu Huang Yong Zhao | 2019 | Protein & Cell2019,10,11: | 2 |

| 5 | QTL mapping of adult plant resistance to stripe rust and leaf rust in a Fuyu 3/Zhengzhou 5389 wheat population显示文摘Stripe or yellow rust(YR)and leaf rust(LR)cause large losses in wheat production worldwide.Resistant cultivars curtail the levels of losses.The present study aimed to identify quantitative trait loci(QTL)for YR and LR resistance in 147 F2:6 recombinant inbred lines(RIL)derived from the cross Fuyu 3/Zhengzhou 5389.The RIL population and parents were genotyped with the Wheat55 K single nucleotide polymorphism(SNP)array and simple sequence repeat(SSR)markers.All materials were also phenotyped for YR severity at Mianyang in Sichuan province and Baoding in Hebei province in the 2015/2016,2016/2017,and 2017/2018 cropping seasons,and LR severity at Zhoukou in Henan province and at Baoding in 2017/2018.Eleven QTL for YR resistance and five for LR resistance were detected using inclusive composite interval mapping(Ici Mapping).Four of these QTL on chromosomes 1 BL,2 BS,3 AL,and 5 AL conferred resistance to both YR and LR.The QTL on 1 BL was Lr46/Yr29,and that on 7 BL might be Lr68.The QTL on chromosome 2 BS was detected at a similar position to previously detected loci.QYr.hebau-3 AL/QLr.hebau-3 AL,QYr.hebau-5 AL/QLr.hebau-5 AL,QYr.hebau-7 DL,QYr.hebau-4 BS,QYr.hebau-6 DL,and QYr.hebau-2 AS are likely to be new.An SSR marker for QYr.hebau-7 DL was developed and validated in a diverse wheat panel from China,suggesting effectiveness in different genetic backgrounds.These QTL with closely linked SNP and SSR markers could be useful for marker-assisted selection in wheat breeding programs targeting durable resistance to both diseases. | Takele Weldu Gebrewahid Peipei Zhang Yue Zhou Xiaocui Yan Xianchun Xia Zhonghu He Daqun Liu Zaifeng Li | 2020 | The Crop Journal2020,8,4: | 2 |

| 7 | Geological and Geochemical Characteristics of the Zhiluo Formation in the Bayinqinggeli Uranium Deposit, Northern Ordos Basin: Significance for Uranium Mineralization显示文摘The Bayinqinggeli deposit in the northern Ordos Basin, northwestern of China, is a recently discovered sandstone-type uranium deposit. The uranium(U) orebodies are generally hosted in the lower member of the Jurassic Zhiluo Formation(Fm.), and are primarily tabular or irregular in shape. In the study area, 23 sandstone samples were collected from the Zhiluo Fm. and analyzed for major, trace, and rare earth elements(REEs). The geochemical characteristics of these sandstones are used to evaluate the factors controlling U mineralization. The source rocks of the Zhiluo Fm. sandstones are mainly volcanic and felsic magmatic rocks formed in continental arc and active continentalmarginal arc environments, and they provided the material required for the mineralization. The index of compositional variability ranges from 1.02 to 3.29(average1.38), indicating that the Zhiluo Fm. sandstones are immature and composed of first-cycle sediments. The corrected chemical index of alteration averages 56, suggesting that the source rocks underwent weak chemical weathering. The ore host rocks are loose, providing favorable conditions for epigenetic oxidation and U precipitation and enrichment. | 9 | Short-term Effects of Nitrogen Deposition on Soil Respiration of Cinnamomum camphora Plantation显示文摘The nitrogen deposition experiments on Cinnamomum camphora plantation of Hunan provincial botanical garden were simulated from June the 7th to October the 7th,2010,the nitrogen deposition levels was respectively as control(CK, 0 g·m-2 a-1),low nitrogen(LN,5 g·m-2 a-1),medium nitrogen(MN,15 g·m-2 a-1) and high nitrogen(HN, 30 g·m -2 a-1).The soil respiration speed and soil surface temperature and the surface water content of the soil were determined by LI-8100 measuring instrument.The results show that the average values of soil respiration speed were CK (4.09±0.66μmol·m-2 s-1),LN(2.39±029μmol·m-2 s-1), MN(2.18±0.19μmol·m-2s-1),HN (2.28±0.25μmol·m-2 s-1),and the treatment of the CK was obviously higher than the other three treatments(P<0.01).With different nitrogen concentrations deposition treatments,the Q10 (temperature sensitivity coefficients) for soil respiration of CK,LN,MN and HN treatments were 1.84,1.71,1.83 and 1.56.It was positive correlation between the respiration speed and the surface(≤10 cm) temperature of tested soil.It was negative correlation between the respiration speed and the surface(≤5 cm) water content of tested soil(P>0.05).Volumetric water content of 5-cm soil was between 0.266 6-0.294 4(m3·m-3), and the monthly content did not vary too much during the research period(1.8%-9.4%).The findings suggest that the initial stage of nitrogen deposition obviously influenced the soil respiration of C.camphora plantation. | Zhang Xuyuan Yan Wende Ma Xiuhong Zheng Wei Wang Guangjun Liang Xiaocui | 2012 | Chinese Forestry Science and Technology2012,11,3: | 0 |
