1Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART.Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu 2017Journal of Genetics and Genomics2017,44,10:5

3Prognostic Nomogram for Patients with Hepatitis E Virus-related Acute Liver Failure:A Multicenter Study in China显示文摘Background and Aims:Timely and effective assessment scoring systems for predicting the mortality of patients with hepatitis E virus-related acute liver failure(HEV-ALF)are urgently needed.The present study aimed to establish an effective nomogram for predicting the mortality of HEV-ALF patients.Methods:The nomogram was based on a cross-sectional set of 404 HEV-ALF patients who were identified and enrolled from a cohort of 650 patients with liver failure.To compare the performance with that of the model for end-stage liver disease(MELD)scoring and CLIF-Consortiumacute-on-chronic liver failure score(CLIF-C-ACLFs)models,we assessed the predictive accuracy of the nomogram using the concordance index(C-index),and its discriminative ability using time-dependent receiver operating characteristics(td-ROC)analysis,respectively.Results:Multivariate logistic regression analysis of the development set carried out to predict mortality revealed that γ-glutamyl transpeptidase,albumin,total bilirubin,urea nitrogen,creatinine,international normalized ratio,and neutrophil-to-lymphocyte ratio were independent factors,all of which were incorporated into the new nomogram to predict the mortality of HEV-ALF patients.The area under the curve of this nomogram for mortality prediction was 0.671(95%confidence interval:0.602-0.740),which was higher than that of the MELD and CLIF-C-ACLFs models.Moreover,the td-ROC and decision curves analysis showed that both discriminative ability and threshold probabilities of the nomogram were superior to those of the MELD and CLIF-C-ACLFs models.A similar trend was observed in the validation set.Conclusions:The novel nomogram is an accurate and efficient mortality prediction method for HEV-ALF patients.Jian Wu Cuifen Shi Xinyu Sheng Yanping Xu Jinrong Zhang Xinguo Zhao Jiong Yu Xinhui Shi Gongqi Li Hongcui Cao Lanjuan Li 2021Journal of Clinical and Translational Hepatology2021,9,6:2
4Targeting TRMT5 suppresses hepatocellular carcinoma progression via inhibiting the HIF-1αpathways显示文摘Accumulating evidence has confirmed the links between transfer RNA(tRNA)modifications and tumor progression.The present study is the first to explore the role of tRNA methyltransferase 5(TRMT5),which catalyzes the m1G37 modification of mitochondrial tRNAs in hepatocellular carcinoma(HCC)progression.Here,based on bioinformatics and clinical analyses,we identified that TRMT5 expression was upregulated in HCC,which correlated with poor prognosis.Silencing TRMT5 attenuated HCC proliferation and metastasis both in vivo and in vitro,which may be partially explained by declined extracellular acidification rate(ECAR)and oxygen consumption rate(OCR).Mechanistically,we discovered that knockdown of TRMT5 inactivated the hypoxia-inducible factor-1(HIF-1)signaling pathway by preventing HIF-1αstability through the enhancement of cellular oxygen content.Moreover,our data indicated that inhibition of TRMT5 sensitized HCC to doxorubicin by adjusting HIF-1α.In conclusion,our study revealed that targeting TRMT5 could inhibit HCC progression and increase the susceptibility of tumor cells to chemotherapy drugs.Thus,TRMT5 might be a carcinogenesis candidate gene that could serve as a potential target for HCC therapy.Qiong ZHAO Luwen ZHANG Qiufen HE Hui CHANG Zhiqiang WANG Hongcui CAO Ying ZHOU Ruolang PAN Ye CHEN 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,1:1

6Ginsenoside Rb1 inhibits oxidative stress-induced ovarian granulosa cell injury through Akt-FoxO1 interaction显示文摘Ginsenoside Rb1 shows a strong antioxidant effect and has potential activation effects on Akt.The aim of the present study was to investigate the protective effect of Rb1 on age-related ovarian granulosa cell injury.Ovarian granulosa cells(GCs)were obtained from 50 young women(≤30 years)and 50 aged women(≥38 years)at an IVF center.Young and aged ICR mice were administered with or without Rb1(10 mg kg^(-1),i.p.)for 2 weeks.The protective effects of Rb1 were investigated and the role of Rb1 on the modulation of Akt-FoxO1 interaction was determined with immunofluorescence,Western blotting,immunoprecipitation,si RNA silencing and pharmacological inhibitor.Rb1 effectively decreased LDH and MDA,and reversed the apoptotic-related protein levels in h GL cells from old patients.Similar results were found in mice.In addition,the mitochondrial membrane potential was restored and the overaccumulation of ROS was reversed by Rb1.Rb1 preserved peroxide-impaired Akt activation,to some extent,by increasing phosphorylation at Ser473.Rb1 also facilitated p-Akt binding to FoxO1 and promoted the phosphorylation of FoxO1.Si RNA silencing of Akt,Akt inhibitor LY294002,and FoxO1 inhibitor AS1842856 attenuated the effects of Rb1.Ginsenoside Rb1 inhibits age-related GCs oxidative damage by activating Akt phosphorylation at Ser473 and by further interaction with FoxO1.Ping Zhou Feng Deng Zi Yang Canhui Cao Hongcui Zhao Fenting Liu Ke Zhong Lin Fu Tianliu Peng Di Sun Hui Liu Rong Li Yang Yu 2022Science China(Life Sciences)2022,65,11:1
7Powdery mildew disease resistance and markerassisted screening at the Pm60 locus in wild diploid wheat Triticum urartu显示文摘Triticum urartu,a diploid wild wheat and progenitor of the A genome of bread wheat,is an important resource for resistance to powdery mildew fungus caused by Blumeria graminis f.sp.tritici(Bgt).In the present study we systematically characterized the interaction between the Bgt fungus and T.urartu at the microscopic level.We also tested 227 T.urartu accessions for reaction to Bgt isolate E09 and discovered previously uncategorized powdery mildew resistance in this collection.Pm60 is a CC-NB-LRR type powdery mildew resistance gene that has at least three functional alleles,Pm60,Pm60a,and Pm60b.A marker-assisted screen targeting the Pm60 locus identified a non-functional allele of Pm60a,designated as Pm60a′.A sequence comparison of Pm60a′and Pm60a revealed that they differed by 58 SNPs and one 3-nucleotide deletion.Based on the sequence variations two molecular markers were developed to differentiate the functional Pm60a allele from the non-functional Pm60a′.Our screen revealed the presence of a previously uncharacterized powdery mildew resistance in T.urartu and provides new insights into the Pm60 locus.We believe that the two molecular markers developed here and new T.urartu resistant accessions will facilitate further identification of novel powdery mildew resistance genes and benefit breeding for powdery mildew resistance.Fukai Zhao Yinghui Li Baoju Yang Hongbo Yuan Cong Jin Lixun Zhou Hongcui Pei Lifang Zhao Yiwen Li Yilin Zhou Jiankun Xie Qian-Hua Shen 2020The Crop Journal2020,8,2:0
8Photothermal‑Triggered Structural Change of Nanofiber Scaffold Integrating with Graded Mineralization to Promote Tendon–Bone Healing显示文摘Scaffolds functionalized with graded changes in both fiber alignment and mineral content are more appealing for tendon-bone healing.This study reports the healing of rotator cuff injury using a heterogeneous nanofiber scaffold,which is associated with a structural gradating from aligned to random and an increasing gradient of mineral content in the same orientation.The photothermal-triggered structural change of a nanofiber scaffold followed by graded mineralization is key to constructing such scaffolds.This type of scaffold was found to be biocompatible and provide beneficial contact guidance in the manipulation of tendon-derived stem cell morphologies in vitro.Specifically,tenogenic and osteogenic differentiation of tendon-derived stem cells were simultaneously achieved using the fabricated scaffold.In vivo investigation also showed the improved healing of rabbit rotator cuff injuries based on immunohistochemical analysis and biomechanical investigation that indicates the promising potential of a dual-gradient nanofiber scaffold in clinical tendon-bone healing.Chenghao Yu Tianrui Wang Hongcui Diao Na Liu Yi Zhang Hongyuan Jiang Peng Zhao Zhengyi Shan Zewen Sun Tong Wu Xiumei Mo Tengbo Yu 2022Advanced Fiber Materials2022,4,4:0
