| 1 | Effect of Yiqihuoxue prescription on myocardial energy metabolism after myocardial infarction via cross talk of liver kinase B1-depen-dent Notch1 and adenosine 5'-monophosphate-activated protein kinase显示文摘OBJECTIVE: To investigate the effect of Yiqihuoxue prescription(YQHX) from Traditional Chinese Medicine(TCM) on myocardial glucose and lipid metabolism after myocardial infarction via the cross talk between the liver kinase B1(LKB1)-dependent Notch1 and adenosine 5'-monophosphate(AMP)-activated protein kinase(AMPK). YQHX was prepared with substances with properties that benefit, to activate blood circulation based on the TCM theory.METHODS: Animal models of myocardial infarction were established by ligating Sprague Dawley rats' left anterior descending coronary arteries. The animals were randomly divided into a myocardial infarction(MI) group, a YQHX group, a perindopril group, a γ-secretase inhibitor, Notch signal inhibitor(DAPT) group, a DAPT+YQHX group and a sham group. The related drugs were administered on the second day after operation, and changes in the relevant indexes were examined on weeks 1 and 4.Changes in cardiac structure and function were examined by echocardiography. The glucose and free fatty acids(FFA) were examined by ELISA. The expression of Notch, LKB1 and AMPK m RNA was examined by a real-time fluorescence quantitative method. The expression of glucose transporter 4(GLUT4), and the expression of total acetyl-Co A carboxylase(ACC) and its phosphorylation were examined by western blotting.RESULTS: Compared with the sham group, the expression of Notch, LKB1 and AMPK m RNA in the MI group was lower. Compared with the MI group, the expression of these m RNAs in the YQHX and perindopril groups was higher, and their expression in the DAPT group was lower. At all time points, the protein expression of GLUT4 and p ACC decreased in the MI group. On week 1, the expression of p ACC protein was higher. In the DAPT group, the expression of p ACC protein decreased. Compared with the YQHX group, the expression of p ACC protein in the DAPT + YQHX group was lower. On week 4,compared with the MI group, the expression of GLUT4 protein in the YQHX group and the perindo-pril group was higher. The expression of GLUT4 protein in the DAPT group decreased. Compared with the YQHX group, the expression of GLUT4 protein in the DAPT+YQHX group was lower. There was no significant difference in the expression of ACC protein between the groups.CONCLUSION: YQHX promoted cross talk between the LKB1-dependent Notch1 and AMPK in myocardial tissue after myocardial infarction. | 3 | Periostin mediates epithelial-mesenchymal transition through the MAPK/ERK pathway in hepatoblastoma显示文摘Objective: The aim of the present study was to analyze the prognostic factors in patients with hepatoblastoma(HB) in our single center and to evaluate periostin(POSTN) expression in HB and its association with clinicopathological variables. In addition, the underlying mechanism of how POSTN promotes HB progression was discussed.Methods: POSTN expression was investigated in HB tumors by immunohistochemistry(IHC), immunofluorescence(IF) and Western blot(WB). The association among POSTN expression, clinicopathological features and overall survival(OS) was also evaluated. The migration and adhesion ability of HB cells were measured using chemotaxis and cell-matrix adhesion assays,respectively. Epithelial-mesenchymal transition(EMT)-associated markers and activation of the ERK pathway were detected by WB.Results: HB patients had poor prognosis which displayed lymph node metastasis, vascular invasion, POSTN and vimentin expression. POSTN expression was also associated with lymph node metastasis. Furthermore, overexpressed POSTN promoted migration and the adhesive ability of HB cells in vitro. In addition, we demonstrated that POSTN activated the MAPK/ERK pathway, upregulated the expression of Snail and decreased the expression of OVOL2. | 6 | In Situ Monitoring the Potassium‑Ion Storage Enhancement in Iron Selenide with Ether‑Based Electrolyte显示文摘As one of the promising anode materials,iron selenide has received much attention for potassium-ion batteries(KIBs).Nevertheless,volume expansion and sluggish kinetics of iron selenide result in the poor reversibility and stability during potassiation–depotassiation process.In this work,we develop iron selenide composite matching ether-based electrolyte for KIBs,which presents a reversible specific capacity of 356 mAh g^(−1) at 200 mA g^(−1) after 75 cycles.According to the measurement of mechanical properties,it is found that iron selenide composite also exhibits robust and elastic solid electrolyte interphase layer in ether-based electrolyte,contributing to the improvement in reversibility and stability for KIBs.To further investigate the electrochemical enhancement mechanism of ether-based electrolyte in KIBs,we also utilize in situ visualization technique to monitor the potassiation–depotassiation process.For comparison,iron selenide composite matching carbonate-based electrolyte presents vast morphology change during potassiation–depotassiation process.When changing to ether-based electrolyte,a few minor morphology changes can be observed.This phenomenon indicates an occurrence of homogeneous electrochemical reaction in ether-based electrolyte,which results in a stable performance for potassium-ion(K-ion)storage.We believe that our work will provide a new perspective to visually monitor the potassium-ion storage process and guide the improvement in electrode material performance. | Xiaodan Li Jinliang Li Wenchen Zhuo Zhibin Li Liang Ma Zhong Ji Likun Pan Wenjie Mai | 2021 | Nano-Micro Letters2021,13,11: | 2 |

| 16 | Injectable muscle-adhesive antioxidant conductive photothermal bioactive nanomatrix for efficiently promoting full-thickness skeletal muscle regeneration显示文摘The completed skeletal muscle regeneration resulted from severe injury and muscle-related disease is still a challenge.Here,we developed an injectable muscle-adhesive antioxidant conductive bioactive photothermo-responsive nanomatrix for regulating the myogenic differentiation and promoting the skeletal muscle regeneration in vivo.The multifunctional nanomatrix was composed of polypyrrole@polydopamine(PPy@PDA,342±5.6 nm)nanoparticles-crosslinked Pluronic F-127(F127)-polycitrate matrix(FPCP).The FPCP nanomatrix demonstrated inherent multifunctional properties including excellent photothermo-responsive and shear-thinning behavior,muscle-adhesive feature,injectable ability,electronic conductivity(0.48±0.03 S/m)and antioxidant activity and photothermal function.The FPCP nanomatrix displayed better photothermal performance with near-infrared irradiation,which could provide the photo-controlled release of protein(91%±2.6%of BSA was released after irradiated 3 times).Additionally,FPCP nanomatrix could significantly enhance the cell proliferation and myogenic differentiation of mouse myoblast cells(C2C12)by promoting the expressions of myogenic genes(MyoD and MyoG)and myosin heavy chain(MHC)protein with negligible cytotoxicity.Based on the multifunctional properties,FPCP nanomatrix efficiently promoted the full-thickness skeletal muscle repair and regeneration in vivo,through stimulating the angiogenesis and myotube formation.This study firstly indicated the vital role of multifunctional PPy@PDA nanoparticles in regulating myogenic differentiation and skeletal muscle regeneration.This work also suggests that rational design of bioactive matrix with multifunctional feature would greatly enhance the development of regenerative medicine. | Li Zhou Juan Ge Min Wang Mi Chen Wei Cheng Wenchen Ji Bo Lei | 2021 | Bioactive Materials2021,6,6: | 0 |
