1The Role of Exportin-5 in MicroRNA Biogenesis and Cancer显示文摘Micro RNAs(mi RNAs) are conserved small non-coding RNAs that play an important role in the regulation of gene expression and participate in a variety of biological processes. The biogenesis of mi RNAs is tightly controlled at multiple steps, such as transcription of mi RNA genes,processing by Drosha and Dicer, and transportation of precursor mi RNAs(pre-mi RNAs) from the nucleus to the cytoplasm by exportin-5(XPO5). Given the critical role of nuclear export of premi RNAs in mi RNA biogenesis, any alterations of XPO5, resulting from either genetic mutation,epigenetic change, abnormal expression level or posttranslational modification, could affect mi RNA expression and thus have profound effects on tumorigenesis. Importantly, XPO5 phosphorylation by ERK kinase and its cis/trans isomerization by the prolyl isomerase Pin1 impair XPO50 s nucleo-to-cytoplasmic transport ability of pre-mi RNAs, leading to downregulation of mature mi RNAs in hepatocellular carcinoma. 2Periostin mediates epithelial-mesenchymal transition through the MAPK/ERK pathway in hepatoblastoma显示文摘Objective: The aim of the present study was to analyze the prognostic factors in patients with hepatoblastoma(HB) in our single center and to evaluate periostin(POSTN) expression in HB and its association with clinicopathological variables. In addition, the underlying mechanism of how POSTN promotes HB progression was discussed.Methods: POSTN expression was investigated in HB tumors by immunohistochemistry(IHC), immunofluorescence(IF) and Western blot(WB). The association among POSTN expression, clinicopathological features and overall survival(OS) was also evaluated. The migration and adhesion ability of HB cells were measured using chemotaxis and cell-matrix adhesion assays,respectively. Epithelial-mesenchymal transition(EMT)-associated markers and activation of the ERK pathway were detected by WB.Results: HB patients had poor prognosis which displayed lymph node metastasis, vascular invasion, POSTN and vimentin expression. POSTN expression was also associated with lymph node metastasis. Furthermore, overexpressed POSTN promoted migration and the adhesive ability of HB cells in vitro. In addition, we demonstrated that POSTN activated the MAPK/ERK pathway, upregulated the expression of Snail and decreased the expression of OVOL2. 9QoS-Aware and Resource-Efficient Dynamic Slicing Mechanism for Internet of Things显示文摘With the popularization of terminal devices and services in Internet of things(IoT),it will be a challenge to design a network resource allocation method meeting various QoS requirements and effectively using substrate resources.In this paper,a dynamic network slicing mechanism including virtual network(VN)mapping and VN reconfiguration is proposed to provide network slices for services.Firstly,a service priority model is defined to create queue for resource allocation.Then a slice including Virtual Network Function(VNF)placement and routing with optimal cost is generated by VN mapping.Next,considering temporal variations of service resource requirements,the size of network slice is adjusted dynamically to guarantee resource utilization in VN reconfiguration.Additionally,load balancing factors are designed to make traffic balanced.Simulation results show that dynamic slicing mechanism not only saves 22%and 31%cost than static slicing mechanism with extending shortest path(SS_ESP)and dynamic slicing mechanism with embedding single path(DS_ESP),but also maintains high service acceptance rate.Wenchen He Shaoyong Guo Yun Liang Rui Ma Xuesong Qiu Lei Shi 2019Computers, Materials & Continua2019,,9:0
