1A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. 2Rationalizing phonon dispersion for lattice thermal conductivity of solids显示文摘Lattice thermal conductivity(κL) is one of the most fundamental properties of solids. The acoustic–elastic-wave assumption, proposed by Debye(Debye P. Ann Phys 1912; 344: 789–839), has led to linear phonon dispersion being the most common approximation for understanding phonon transport over the past century. Such an assumption does not take into account the effect of a periodic boundary condition on the phonon dispersion, originating from the nature of periodicity on atomic arrangements. Driven by modern demands on the thermal functionality of materials, with κLranging from ultra-low to ultra-high,any deviation from the Debye approximation in real materials becomes more and more significant. This work takes into account the periodic boundary condition, and therefore rationalizes the phonon dispersion to be more realistic. This significantly improves the precision for quickly predicting κLwithout any fitting parameters, as demonstrated in hundreds of materials, and offers a theoretical basis rationalizing κLto be lower than the minimum currently accepted based on the Debye dispersion. 3The Epitope Study on the SARS-CoV Nucleocapsid Protein显示文摘The nucleocapsid protein (N protein) has been found to be an antigenic protein in a number of coronaviruses. Whether the N protein in severe acute respiratory syndrome-associated coronavirus (SARS-CoV) is antigenic remains to be elucidated. Using Western blot and Enzyme-linked Immunosorbent Assay (ELISA),the recombinant N proteins and the synthesized peptides derived from the N protein were screened in sera from SARS patients. All patient sera in this study displayed strong positive immunoreactivities against the recombinant N proteins,whereas normal sera gave negative immunoresponses to these proteins, indicating that the N protein of SARS-CoV is an antigenic protein. Furthermore, the epitope sites in the N protein were determined by competition experiments, in which the recombinant proteins or the synthesized peptides competed against the SARS-CoV proteins to bind to the antibodies raised in SARS sera. One epitope site located at the C-terminus was confirmed as the most antigenic region in this protein. A detailed screening of peptide with ELISA demonstrated that the amino sequence from Codons 371 to 407 was the epitope site at the C-terminus of the N protein. 4Manipulation of Band Degeneracy and Lattice Strain for Extraordinary PbTe Thermoelectrics显示文摘Maximizing band degeneracy and minimizing phonon relaxation time are proven to be successful for advancing thermoelectrics.Alloying with monotellurides has been known to be an effective approach for converging the valence bands of PbTe for electronic improvements,while the lattice thermal conductivity of the materials remains available room for being further reduced.It is recently revealed that the broadening of phonon dispersion measures the strength of phonon scattering,and lattice dislocations are particularly effective sources for such broadening through lattice strain fluctuations.In this work,a fine control of MnTe and EuTe alloying enables a significant increase in density of electron states near the valence band edge of PbTe due to involvement of multiple transporting bands,while the creation of dense in-grain dislocations leads to an effective broadening in phonon dispersion for reduced phonon lifetime due to the large strain fluctuations of dislocations as confirmed by synchrotron X-ray diffraction.The synergy of both electronic and thermal improvements successfully leads the average thermoelectric figure of merit to be higher than that ever reported for p-type PbTe at working temperatures.Yixuan Wu Pengfei Nan Zhiwei Chen Zezhu Zeng Siqi Lin Xinyue Zhang Hongliang Dong Zhiqiang Chen Hongkai Gu Wen Li Yue Chen Binghui Ge Yanzhong Pei 2020Research2020,,1:6
5A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 6Solute manipulation enabled band and defect engineering for thermoelectric enhancements of SnTe显示文摘With years of development, SnTe as a homologue of PbTe has shown great potentialfor thermoelectric applications in p-type conduction, and the most successfulstrategy is typified by alloying for maximizing the valence band degeneracy.Among the known alloy agents, MnTe has been found to be one of the most effectiveenabling a band convergence for an enhancement in electronic performance ofSnTe, yet its solubility of only ~15 at% unfortunately prevents a full optimizationin the valence band structure. This work reveals that additional PbTe alloying notonly promotes the MnTe solubility to locate the optimal valence band structure butalso increases the overall substitutional defects in the material for a substantialreduction in lattice thermal conductivity. In addition, PbTe alloying simultaneouslyoptimizes the carrier concentration due to the cation size effect. 7Human umbilical cord mesenchymal stem cells for psoriasis:a phase 1/2a,single-arm study显示文摘Psoriasis is a common,chronic immune-mediated systemic disease that had no effective and durable treatment.Mesenchymal stem cells(MSCs)have immunomodulatory properties.Therefore,we performed a phase 1/2a,single-arm clinical trial to evaluate the safety and efficacy of human umbilical cord-derived MSCs(UMSCs)in the treatment of psoriasis and to preliminarily explore the possible mechanisms.Seventeen patients with psoriasis were enrolled and received UMsC infusions.Adverse events,laboratory parameters,PASl,and PGA were analyzed.We did not observe obvious side effects during the treatment and 6-month follow-up.A total of 47.1%(8/17)of the psoriasis patients had at least 40%improvement in the PASl score,and 17.6%(3/17)had no sign of disease or minimal disease based on the PGA score.And the efficiency was 25%(2/8)for males and 66.7%(6/9)for females.After UMSC transplantation(UMSCT),the frequencies of Tregs and CD4^(+)memory T cells were significantly increased,and the frequencies of T helper(Th)17 and CD4^(+)naive T cells were significantly decreased in peripheral blood(PB)of psoriasis patients.And all responders showed significant increases in Tregs and CD4^(+)memory T cells,and significant decreases in Th17 cells and serum IL-17 level after UMsCT.And baseline level of Tregs in responders were significantly lower than those in nonresponders.In conclusion,allogeneic UMSCT is safe and partially effective in psoriasis patients,and level of Tregs may be used as a potent biomarker to predict the clinical efficacy of UMSCT.Trial registration Clinical Trials NCT03765957.Lamei Cheng Siqi Wang Cong Peng Xiao Zou Chao Yang Hua Mei Chuang Li Xian Su Na Xiao Qi Ouyang Mi Zhang Qiaolin Wang Yan Luo Minxue Shen Qun Qin Honglin Wang Wu Zhu Guangxiu Lu Ge Lin Yehong Kuang Xiang Chen 2022Signal Transduction and Targeted Therapy2022,7,9:2
8Ambient air pollution and low temperature associated with case fatality of COVID-19: A nationwide retrospective cohort study in China显示文摘The evidence for the effects of environmental factors on COVID-19 case fatality remains controversial,and it is crucial to understand the role of preventable environmental factors in driving COVID-19 fatality.We thus conducted a nationwide cohort study to estimate the effects of environmental factors(temperature,particulate matter[PM2.5,PM10],sulfur dioxide[SO2],nitrogen dioxide[NO2],and ozone[O3])on COVID-19 case fatality.A total of 71,808 confirmed COVID-19 cases were identified and followed up for their vital status through April 25,2020.Exposures to ambient air pollution and temperature were estimated by linking the city-and county-level monitoring data to the residential community of each participant.For each participant,two windows were defined:the period from symptom onset to diagnosis(exposure window I)and the period from diagnosis date to date of death/recovery or end of the study period(exposure window II).Cox proportional hazards models were used to estimate the associations between these environmental factors and COVID-19 case fatality.COVID-19 case fatality increased in association with environmental factors for the two exposure windows.For example,each 10 mg/m^(3) increase in PM2.5,PM10,O3,and NO2 in window I was associated with a hazard ratio of 1.11(95%CI 1.09,1.13),1.10(95%CI 1.08,1.13),1.09(95 CI 1.03,1.14),and 1.27(95%CI 1.19,1.35)for COVID-19 fatality,respectively.A significant effect was also observed for low temperature,with a hazard ratio of 1.03(95%CI 1.01,1.04)for COVID-19 case fatality per 1C decrease.Subgroup analysis indicated that these effects were stronger in the elderly,as well as in those with mild symptoms and living in Wuhan or Hubei.Overall,the sensitivity analyses also yielded consistent estimates.Short-term exposure to ambient air pollution and low temperature during the illness would play a nonnegligible part in causing case fatality due to COVID-19.Reduced exposures to high concentrations of PM2.5,PM10,O3,SO2,and NO2 and low temperature would help improve the prognosis and reduce public health burden.Fei Tian Xiaobo Liu Qingchen Chao Zhengmin(Min)Qian Siqi Zhang Li Qi Yanlin Niu Lauren DArnold Shiyu Zhang Huan Li Hualiang Lin Qiyong Liu 2021The Innovation2021,2,3:2

11Mapping of wheat stripe rust resistance gene Yr041133 by BSR-Seq analysis显示文摘Puccinia striiformis Westend. f. sp. tritici(Pst) pathotype CYR34 is widely virulent and prevalent in China.Here, we report identification of a strpie rust resistance(Yr) gene, designated Yr041133, in winter wheat line 041133. This line produced a hypersensitive reaction to CYR34 and conferred resistance to 13 other pathotypes. Resistance to CYR34 in line 041133 was controlled by a single dominant gene. Bulked segregant RNA sequencing(BSR-Seq) was performed on a pair of RNA bulks generated by pooling resistant and susceptible recombinant inbred lines. Yr041133 was mapped to a 1.7 c M genetic interval on the chromosome arm 7 BL that corresponded to a 0.8 Mb physical interval(608.9–609.7 Mb) in the Chinese Spring reference genome. 13The self-assembly and structural regulation of a hydrogen-bonded dimeric building block formed by two N-H…O hydrogen bonds on HOPG显示文摘The self-assembled behavior of an unsymmetric molecule(BCDTDA)with one imidazole group as center and benzoic acid group as functional group is studied,and the regulatory behaviors of coronene(COR)and three bipyridine derivatives(named BP,PEBP-C4 and PEBP-C8)on BCDTDA self-assembly structures are also investigated.Based on highly oriented pyrolytic graphite(HOPG)substrate,scanning tunneling microscopy(STM)is used to observe the variation of assembled behaviors at the solid-liquid interface.Because of the concentration effect,BCDTDA molecules can assemble into grids and Kagomés structures in the form of N–H…O hydrogen bonded dimers.BCDTDA molecules still maintain dimeric structures in the regulation of COR and BP molecules to BCDTDA self-assembly.However,PEBP-C4 and PEBP-C8 destroy the structure of the dimers,and form a variety of co-assembled structures with BCDTDA.Different guest molecules coordinate the host molecules differently,which makes the experiment more meaningful.Combined with density functional theory(DFT)calculation,the discovery of molecular interactions provides a promising strategy for the construction of functional nanostructures and devices.Peng Lei Lin Ma Siqi Zhang Jianqiao Li Linlin Gan Ke Deng Wubiao Duan Wei Li Qingdao Zeng 2023Chinese Chemical Letters2023,34,8:1
14A Strategy for Searching Antigenic Regions in the SARS-CoV Spike Protein显示文摘In the face of the worldwide threat of severe acute respiratory syndrome (SARS)to human life, some of the most urgent challenges are to develop fast and accurate analytical methods for early diagnosis of this disease as well as to create a safe anti-viral vaccine for prevention. To these ends, we investigated the antigenicity of the spike protein (S protein), a major structural protein in the SARS-coronavirus (SARS-CoV). Based upon the theoretical analysis for hydrophobicity of the S protein, 18 peptides were synthesized. Using Enzyme-Linked Immunosorbent Assay (ELISA), these peptides were screened in the sera from SARS patients. According to these results, two fragments of the S gene were amplified by PCR and cloned into pET-32a. Both S fragments were expressed in the BL-21 strain and further purified with an affinity chromatography. These recombinant S fragments were confirmed to have positive cross-reactions with SARS sera, either by Western blot or by ELISA. Our results demonstrated that the potential epitope regions were located at Codons 469-882 in the S protein, and one epitope site was located at Codons 599-620. 163D Printing of Nacre-Inspired Structures with Exceptional Mechanical and Flame-Retardant Properties显示文摘Flame-retardant and thermal management structures have attracted great attention duc to the requirement of high-temperature exposure in industrial,aerospace,and thermal power fields,but the development of protoctive fire-retardant structures with complex shapes to fit arbitrary surfaces is still challenging.Herein,we reported a rotation-blade casting-assisted 3D printing process to fabricate nacre-inspired structures with exceptional mechanical and flame-retardant properties,and the relatedi fundamental mechanisms are studied.3-(Trimethoxysilyl)propyl methacrylate(TMSPMA)modified boron nitride nanoplatelets(BNs)were aligned by rotation-blade casting during the 3D printing process to build the'hrick and mortar'architecture.The 3D printed structures are more lightweight,while having higher fracture toughness than the natural nacre,which is attributed to the crack deflection,aligned BN(a-BNs)bridging,and pull-outs reinforced structures by the covalent bonding between TMSPMA grafted a-BNs and polymer matrix.Thermal conductivity is enhanced by 25.5 times compared with pure polymer and 5.8 times of anisotropy due to the interconnection of a-BNs.3D printed heat-exchange structures with vertically aligned BNs in complex shapes were demonstrated for efficient thermal control of high-power light-emitting diocles.3D printed helmet and armor with a-BNs show exceptional mechanical and fire-retardant properties,demonstrating integrated mechanical and thermal protection.Yang Yang Ziyu Wang Qingqing He Xiangjia Li Gengxi Lu Laiming Jjiang Yushun Zeng Brandon Bethers Jie Jin Shuang Lin Siqi Xiao Yizhen Zhu Xianke Wu Wenwu Xu Qiming Wang Yong Chen 2022Research2022,,2:1

18Tetrahedral DNA nanostructures synergize with MnO_(2) to enhance antitumor immunity via promoting STING activation and M1 polarization显示文摘Stimulator of interferon genes(STING) is a cytosolic DNA sensor which is regarded as a potential target for antitumor immunotherapy. However, clinical trials of STING agonists display limited anti-tumor effects and dose-dependent side-effects like inflammatory damage and cell toxicity. Here,we showed that tetrahedral DNA nanostructures(TDNs) actively enter macrophages to promote STING activation and M1 polarization in a size-dependent manner, and synergized with Mn^(2+) to enhance the expressions of IFN-β and iNOS, as well as the co-stimulatory molecules for antigen presentation. Moreover, to reduce the cytotoxicity of Mn^(2+),we constructed a TDN-MnO_(2) complex and found that it displayed a much higher efficacy than TDN plus Mn^(2+) to initiate macrophage activation and anti-tumor response both in vitro and in vivo. 19Domain knowledge discovery from abstracts of scientific literature on Nickel-based single crystal superalloys显示文摘Despite the huge accumulation of scientific literature,it is inefficient and laborious to manually search it for useful information to investigate structure-activity relationships.Here,we propose an efficient text-mining framework for the discovery of credible and valuable domain knowledge from abstracts of scientific literature focusing on Nickel-based single crystal superalloys.Firstly,the credibility of abstracts is quantified in terms of source timeliness,publication authority and author’s academic standing.Next,eight entity types and domain dictionaries describing Nickel-based single crystal superalloys are predefined to realize the named entity recognition from the abstracts,achieving an accuracy of 85.10%.Thirdly,by formulating 12 naming rules for the alloy brands derived from the recognized entities,we extract the target entities and refine them as domain knowledge through the credibility analysis.Following this,we also map out the academic cooperative“Author-Literature-Institute”network,characterize the generations of Nickel-based single crystal superalloys,as well as obtain the fractions of the most important chemical elements in superalloys.The extracted rich and diverse knowledge of Nickel-based single crystal superalloys provides important insights toward understanding the structure-activity relationships for Nickel-based single crystal superalloys and is expected to accelerate the design and discovery of novel superalloys.LIU Yue DING Lin YANG ZhengWei GE XianYuan LIU DaHui LIU Wei YU Tao AVDEEV Maxim SHI SiQi 2023Science China(Technological Sciences)2023,66,6:1
