1Selection and development of representative simple sequence repeat primers and multiplex SSR sets for high throughput automated genotyping in maize显示文摘In the current study, 1900 maize simple sequence repeat (SSR) primers published in MaizeGDB were screened utilizing reference literature, 15 representative Chinese maize inbred lines and 15 Chinese maize hybrids from national regional testing. In total, 500 highly polymorphic primers were identified and used to construct a genetic map. 100 evenly distributed primers, 10 primers per chromosome, were further selected as a set of universal SSR core primers, recommended as preferred primers for general studies. These core primers were then redesigned and used to construct a high throughput multiplex PCR system based on a five-color fluorescence capillary detection system. 2Multi-objective evolutionary design of selective triple modular redundancy systems against SEUs显示文摘To improve the reliability of spaceborne electronic systems,a fault-tolerant strategy of selective triple modular redundancy(STMR)based on multi-objective optimization and evolvable hardware(EHW)against single-event upsets(SEUs)for circuits implemented on field programmable gate arrays(FPGAs)based on static random access memory(SRAM)is presented in this paper.Various topologies of circuit with the same functionality are evolved using EHW firstly.Then the SEU-sensitive gates of each circuit are identified using signal probabilities of all the lines in it,and each circuit is hardened against SEUs by selectively applying triple modular redundancy(TMR)to these SEU-sensitive gates.Afterward,each circuit hardened has been evaluated by SEU Simulation,and the multi-objective optimization technology is introduced to optimize the area overhead and the number of functional errors of all the circuits.The proposed fault-tolerant strategy is tested on four circuits from microelectronics center of North Carolina(MCNC)benchmark suite.The experimental results show that it can generate innovative trade-off solutions to compromise between hardware resource consumption and system reliability.The maximum savings in the area overhead of the STMR circuit over the full TMR design is 58%with the same SEU immunity.Yao Rui Chen Qinqin Li Zengwu Sun Yanmei 2015Chinese Journal of Aeronautics2015,28,3:6
3Water-soluble supramolecular polymers constructed by macrocycle-based host-guest interactions显示文摘Supramolecular polymers, which combined of the supramolecular chemistry and polymer science, are constructed from monomers together by reversible noncovalent interactions. Supramolecular polymers in aqueous solution are especially important due to their superior biocompatibility and environmental adaptation, which determined they have wide applications in various areas, such as drug delivery, selfhealing, shape memory. Macrocyclic compounds, such as cyclodextrins, calixarenes, cucurbiturils and pillar[n]arenes, are the most used building blocks in the preparation of supramolecular polymers.Macrocycle-based supramolecular polymers, which introduce the host-guest interaction in the system,endow these polymers with interesting and smart physical-chemical properties. 4Generation of tooth-like structures from integration-free human urine induced pluripotent stem cells显示文摘Background:Tooth is vital not only for a good smile,but also good health.Yet,we lose tooth regularly due to accidents or diseases.An ideal solution to this problem is to regenerate tooth with patients’own cells.Here we describe the generation of tooth-like structures from integration-free human urine induced pluripotent stem cells(ifhU-iPSCs).Results:We first differentiated ifhU-iPSCs to epithelial sheets,which were then recombined with E14.5 mouse dental mesenchymes.Tooth-like structures were recovered from these recombinants in 3 weeks with success rate up to 30%for 8 different iPSC lines,comparable to H1 hESC.We further detected that ifhU-iPSC derived epithelial sheets differentiated into enamel-secreting ameloblasts in the tooth-like structures,possessing physical properties such as elastic modulus and hardness found in the regular human tooth.Conclusion:Our results demonstrate that ifhU-iPSCs can be used to regenerate patient specific dental tissues or even tooth for further drug screening or regenerative therapies.Jinglei Cai Yanmei Zhang Pengfei Liu Shubin Chen Xuan Wu Yuhua Sun Ang Li Ke Huang Rongping Luo Lihui Wang Ying Liu Ting Zhou Shicheng Wei Guangjin Pan Duanqing Pei 2013Cell Regeneration2013,2,1:4
5Transformation:how do nematode sperm become activated and crawl?显示文摘Nematode sperm undergo a drastic physiological change during spermiogenesis(sperm activation).Unlike mammalian flagellated sperm,nematode sperm are amoeboid cells and their motility is driven by the dynamics of a cytoskeleton composed of major sperm protein(MSP)rather than actin found in other crawling cells.This review focuses on sperm from Caenorhabditis elegans and Ascaris suum to address the roles of external and internal factors that trigger sperm activation and power sperm motility.Nematode sperm can be activated in vitro by several factors,including Pronase and ionophores,and in vivo through the TRY-5 and SPE-8 pathways.Moreover,protease and protease inhibitors are crucial regulators of sperm maturation.MSP-based sperm motility involves a coupled process of protrusion and retraction,both of which have been reconstituted in vitro.Sperm motility is mediated by phosphorylation signals,as illustrated by identification of several key components(MPOP,MFPs and MPAK)in Ascaris and the characterization of GSP-3/4 in C.elegans.Xuan Ma Yanmei Zhao Wei Sun Katsuya Shimabukuro Long Miao 2012Protein & Cell2012,3,10:4
6Interaction of epicatechin with bovine serum albumin using fluorescence quenching combined with chemometrics显示文摘The interaction between BSA and epicatechin was studied using fluorescence quenching titrations combined with trilinear decomposition method and excitation-emission matrix(EEM)fluorescence.The resolved spectra were highly similar with the actual ones which indicated that the resolved results were reliable.The relevant parameters of the binding process were obtained by quantifying each substance in the complicated mixtures in situ.The quenching was static quenching,epicatechin had a weak interaction with BSA and the binding site was one.The total concentration and the free concentration of quenchers had different effect on the system.The results demonstrated that the method exploited in this article is a useful tool to investigate complicated interactions,avoiding complicated pretreatment and simplify experimental procedure.ZHAI Min WU HaiLong ZHANG ShuRong ZHANG XiHua SUN YanMei YU RuQin 2014Science China Chemistry2014,57,5:2
7Determination of minimum overburden depth for underwater shield tunnel in sands:Comparison between circular and rectangular tunnels显示文摘With the development of global urbanization,the utilization of underground space is more critical and attractive for civil purposes.Various shapes of shield tunnels have been gradually proposed to cope with different geological conditions and service purposes of underground structures.Generally,reducing the burial depth of shield tunnel is conducive to construction and cost saving.However,extremely small overburden depth cannot provide sufficient uplift resistance to maintain the stability and serviceability of the tunnel.To this end,this paper firstly reviewed the status of deriving the minimum sand over-burden depth of circular shield tunnel using mechanical equilibrium(ME)method.It revealed that the estimated depth is rather conservative.Then,the uplift resistance mechanism of both circular and rectangular tunnels was deduced theoretically and verified with the model tests.The theoretical uplift resistance is consistent with the experimental values,indicating the feasibility of the proposed equations.Furthermore,the determination of the minimum soil overburden depth of rectangular shield tunnel under various working conditions was presented through integrated ME method,which can provide more reasonable estimations of suggested tunnel burial depth for practical construction.Additionally,optimizations were made for calculating the uplift resistance,and the soil thickness providing uplift resistance is suggested to be adjusted according to the testing results.The results can provide reference for the design and construction of various shapes of shield tunnels in urban underground space exploitation.Weixin Sun Fucheng Han Hanlong Liu Wengang Zhang Yanmei Zhang Weijia Su Songlin Liu 2023Journal of Rock Mechanics and Geotechnical Engineering2023,15,7:2
8Experimental analysis on the interaction between underground structures and sand layer under groundwater level change显示文摘Groundwater level change stands a momentous role in affecting the geotechnical construction stability and safety of underground structures.Global warming and active underground construction cause conspicuous changes in the groundwater level,which further leaves an impact on the underground structures’serviceability.To reveal the interaction between underground structure and soil under groundwater level change in the sand layer,model tests of circular transportation tunnel and rectangular utility tunnel were carried out.With the self-designed experimental equipment and innovative experimental methods,the changes in tunnel stress,bending moment,buoyancy,and vertical displacement of the rise and drawdown of the groundwater level in the sand layer were studied.The results revealed the developments of concentrated structural forces during the rising and falling process of the groundwater water level,indi-cating critical locations that should be strengthened.Meanwhile,both tunnels showed the same movement trend:settling first,floating afterwards and settling at last.And it is concluded that no reduction is required when calculating buoyancy in sands using measured pore pressure.Conclusions can provide a notable reference for future related research and engineering designs.Hanlong Liu Weixin Sun Yanmei Zhang Wengang Zhang Fucheng Han Weijia Su 2023Underground Space2023,,3:1
